Unveiling phosphofructokinase as the key target: an innovative strategy combining pull-down assays with optical imaging for snail-killing mechanism exploration

The Pomacea canaliculata, also known as apple snail, is an invasive species in China, posing a serious threat to agricultural production and public health. To prevent its rampant infestation, we developed a highly efficient snail-killing lead compound, 1-(4-chlorophenyl)-3-(pyridin-3-yl)urea (PBQ). However, the killing mechanism of PBQ remains unclear. To elucidate this, two novel compounds, B-PBQ and J-PBQ, were developed through structural modification of PBQ and were used, through both protein pull-down assay and confocal laser scanning microscopy imaging, to identify and confirm that phosphofructokinase(PFK) in the apple snail is the primary target protein of PBQ. Further studies revealed that PBQ successfully inhibits the activity,content, and expression of PFK in apple snail, disrupting the glycolysis pathway and leading to abnormal energy *** disruptions impair the snail’s tolerance to low oxygen, resulting in its efficient eradication. These findings offer a promising direction for developing next-generation PFK-targeted snail-killing pesticides. Furthermore, the strategy of combining pull-down assays with optical imaging used in this work provides a novel approach for the exploration of mechanisms of other pesticide candidates.

Skeletal muscle plays a significant role in both local and systemic energy metabolism. The current investigation aims to explore the role of the Bambi gene in skeletal muscle, focusing on its implications for muscle hypertrophy and systemic metabolism. We hypothesize that skeletal muscle-specific deletion of Bambi induces muscle hypertrophy, improves metabolic performance, and activates thermogenic adipocytes via the reprogramming of progenitor of i WAT, offering potential therapeutic strategies for metabolic syndromes. Leveraging the Chromatin immunoprecipitation(ChIP)-seq and bioinformatics analysis, Bambi gene is shown to be a direct target of HIF2α, which is further confirmed by ChIP-qPCR and promoter luciferase assay. Skeletal muscle-specific Bambi deletion led to significant muscle hypertrophy and improved metabolic parameters, even under high-fat diet conditions. This deletion induced metabolic reprogramming of stromal vascular fractions(SVFs) into thermogenic adipocytes, contributing to systemic metabolic improvements, potentially through the secretory ***, mice with skeletal muscle-specific Bambi deletion demonstrate resistance to high-fat diet-induced metabolic disorders, highlighting a potential therapeutic pathway for metabolic syndrome management. Thus, skeletal muscle-specific deletion of Bambi triggers muscle growth, enhances metabolic performance, and activates thermogenic adipocytes. These findings suggest Bambi as a novel therapeutic target for metabolic syndromes, providing new insights into the interaction between muscle hypertrophy and systemic metabolic improvement. The study underscores the potential of manipulating muscle physiology to regulate whole-body metabolism, offering a novel perspective on treating metabolic disorders.

Ni-catalyzed cross-electrophile alkyl-alkyl coupling reactions

Introducing saturated carbon-carbon bonds into organic molecules significantly increases the three-dimensional configuration,thus altering the physical and chemical properties as well as the functions of molecules. Transition-metal-catalyzed carboncarbon cross-couplings represent one of the most straightforward, efficient and modular alternatives to build saturation in a single step. However, the efficient construction of alkyl-alkyl bonds remains a formidable challenge due to the precise manipulation of multiple alkyl metallic intermediates as well as associated competitive side reactions and selectivity control. Over the past years, Ni-catalyzed cross-coupling under reductive conditions has gained more and more attention for alkyl-alkyl crosscoupling partially attributing to the low-valent state of metal centers under reducing conditions, which suppresses the competitive β-hydride elimination process, thus facilitating the formation of alkyl-alkyl bonds. In this review, the initiative and progress of Ni-catalyzed cross-electrophile alkyl-alkyl cross-coupling, including the efforts in racemic and enantioselective formation of alkyl-alkyl bonds, have been summarized and discussed. Moreover, the challenges and further opportunities in this area have also been discussed.

Electrospinning synthesis of novel lithium-rich 0.4li2MnO3.0.6liNi1/3CO1/3Mn1/3O2 nanotube and its electrochemical performance as cathode of lithium-ion battery

In this study, a lithium-rich 0.4li2MnO3.0.6liNi1/3Co1/3Mn1/3O2 nanotube layered cathode synthesized by novel electrospinning is reported, and the effects of temperature on the electrochemical performance and morphologies are investigated. The crystal structure is characterized by X-ray diffraction patterns, and refined by two sets of diffraction data （R-3m and C2/m）. Refined crystal structure is 0.4li2MnO3.0.6liNi1/3COl/3Mn13O2 compos- ite. The inductively coupled plasma optical emission spec- trometer and thermogravimetric and differential scanning calorimetry analysis measurement supply reference to opti- mize the calcination temperature and heat-treatment time. The morphology is characterized by scanning and high- resolution transmission electron microscope techniques, and the micro-nanostructured hollow tubes of li-rich 0.4li2MnO3-0.6liNil/3COl/3Mn1/3O2 composite with outer diameter of 200-400 nm and the wall thickness of 50-80 nm are synthesized successfully. The electrochemical evaluation shows that 0.4li2MnO3.0.6liNi1/3Cov3Mn1/3O2 sintered at 800 ℃ for 8 h delivers the highest capacity of the first dis- charge capacity of 267.7 mAh/g between 2.5 V and 4.8 V at 0.1C and remains 183.3 mAh/g after 50 cycles. The electro- spinning method with heat-treatment to get micro-nanos- tructured lithium-rich cathode shows promising application inlithium-ion batteries with stable electrochemical performance and higher C-rate performance for its shorter li ions transfer channels and stable designed structure.

A low redox potential and long life organic anode material for sodium-ion batteries

Sodium-ion batteries （SIBs） with organic electrodes are an emerging research direction due to the sustainability of organic materials based on elements like C,H,O,and sodium ***,organic electrode materials for SIBs are mainly used as cathodes because of their relatively high redox potentials（>1 V）.Organic electrodes with low redox potential that can be used as anode are ***,a novel organic anode material （tetrasodium 1,4,5,8-naphthalenetetracarboxylate,Na4TDC） has been developed with low redox potential （lic *** three-sodium storage mechanism was demonstrated with various in-situ/ex-situ spectroscopy and theoretical calculations,showing a high capacity of 208 mAh/g and an average decay rate of merely 0.022%per ***,the Na4TDC-hard carbon composite can further acquire improved capacity and cycling stability for 1200 cycles even with a high mass loading of up to 20 mg *** pairing with a thick Na3V2（PO4）3cathode (20.6 mg cm-2),the as-fabricated full cell exhibited high operating voltage （2.8 V）,excellent rate performance and cycling stability with a high capacity retention of 88.7% after 200 cycles,well highlighting the Na4TDC anode material for SIBs.

Fracture mechanism of a cylindrical shell cut by circumferential detonation collision

The failure mechanism of a cylindrical shell cut into fragments by circumferential detonation collision was experimentally and numerically investigated.A self-designed detonation wave regulator was used to control the detonation and cut the *** was found that the self-designed regulator controlled the fragment *** macrostructure and micro-characteristics of fragments revealed that shear fracture was a prior mechanism,the shell fractured not only at the position of detonation collision,but the crack also penetrated the shell at the first contact position of the Chapmen-Jouguet(C-J)*** effects of groove number and outer layer thickness on the fracture behavior were tested by *** the thickness of the outer layer was 5e18 mm,it has little effect on fragmentation of the shell,and shells all fractured at similar *** increase of the groove number reduced the fracture possibility of the first contact position of the C-J *** the groove number reached 7 with a 10 mm outer layer(1/4 model),the fracture only occurred at the position of detonation collision and the fragment width rebounded.
