Global Analysis of Cytosine Methylation and Proteome Under Cold Treatment in Brassica napus

Cytosine methylation/demethylation plays pivotal roles in regulating gene expression at a genome-wide level. However, limited reports are available to reveal correlating changes of cytosine methylation and proteomic expression in Brassica napus so far. Therefore, in the present study, global cytosine methylation and proteome were analysed in B. napus after cold treatment by methylation-sensitive amplified polymorphism （MSAP） and two-dimensional protein electrophoresis technology （2-DE）. The results showed that the lowered genome-wide DNA methylation status was revealed after cold treatment, and about 0.88% of discrepancy in DNA methylation was detected between the non-flowering and flowering plants after cold treatment. Moreover, the 52 significantly up-regulated proteins emerged in comparison with the 36 down-regulated proteins, as well as the 14 proteins exclusively detected in the flowering plants. Intriguingly the 8 specifically expressed proteins in the non-flowering plants disappeared in the flowering plants with cold treatment. Therefore, these present data proved that the correlating changes of cytosine methylation and proteomic expression were evidenced under cold treatment in B. napus.

Background Fenvalerate （FEN） has been demonstrated to be a reproductive toxicant in humans and rodents. However,little is known about whether short-term exposure to low-dose FEN produces reproductive *** We administered FEN （0.009 375, 0.1875, 3.750, or 45.00 mg·kg-1d-1 by gavage for 30 days） to male ICR mice and compared reproductive toxicity parameters between groups receiving different concentrations of *** toxicity was evaluated by computer-assisted semen quality analysis （CASA）, chlortetracycline （CTC） assay,and *** The sperm morphology and testis histology of FEN-exposed mice （all doses） were similar to that in controlling mice. The long-term success of gene therapy for cancer relies heavily on the development of effective targeting systems. We investigate the possibility of targeted gene therapy using promoter of carcinoembryonic antigen (CEA) gene. By using luciferase reporter gene, we found that CEA promoter exhibit 16 times high activity in CEA-producing lung cancer cells, A549 than in nonproducing cells, Hela. We also constructed a recombinant expression plasmid pCEATK, in which CEA promoter drives the effector gene, thymidine kinase gene of Herpes Simplex Virus (HSVTK). A549 cells transfected with pCEATK became 865 times more sensitive to ganciclovir (GCV) than the control cells. However, Hela cells transfected with this plasmid remained resistant to GCV. These data indicate the potential for targeted gene therapy using the CEA promoter against CEA-producing tumor cells, such as lung cancer cells.

In this work, novel phosphors Na5Gd(WO4)4: Tb^3+/Yb^3+ and Na5Gd(WO4)4: Tb^3+/Er^3+/Yb^3+ phosphors were synthesized by the solid state reaction method. The photoluminescence properties were investigated. The introduction of Er^3+ to NGW: Tb^3+/Yb^3+ was used to modify the chromaticity coordinates, then providing a good color tunable property. The change in the chromaticity coordinates induces the shift of emission color from yellow-green to blue-green for NGW: Tb^3+/Er^3+/Yb^3+ as the temperature increases. All the results indicate that the NGW: Er^3+/Tb^3+/Yb^3+ phosphors may be used as optical temperature sensing materials.

Bedding structural planes significantly influence the mechanical properties and stability of engineering rock *** study conducts uniaxial compression tests on layered sandstone with various bedding angles(0°,15°,30°,45°,60°,75°and 90°)to explore the impact of bedding angle on the deformational mechanical response,failure mode,and damage evolution processes of *** develops a damage model based on the Logistic equation derived from the modulus’s degradation considering the combined effect of the sandstone bedding dip angle and *** model is employed to study the damage accumulation state and its evolution within the layered rock *** research also introduces a piecewise constitutive model that considers the initial compaction characteristics to simulate the whole deformation process of layered sandstone under uniaxial *** results revealed that as the bedding angle increases from 0°to 90°,the uniaxial compressive strength and elastic modulus of layered sandstone significantly decrease,slightly increase,and then decline *** corresponding failure modes transition from splitting tensile failure to slipping shear failure and back to splitting tensile *** indicated by the modulus’s degradation,the damage characteristics can be categorized into four stages:initial no damage,damage initiation,damage acceleration,and damage deceleration *** theoretical damage model based on the Logistic equation effectively simulates and predicts the entire damage evolution ***,the theoretical constitutive model curves closely align with the actual stress−strain curves of layered sandstone under uniaxial *** introduced constitutive model is concise,with fewer parameters,a straightforward parameter determination process,and a clear physical *** study offers valuable insights into the theory of layered rock mechanics and holds implications for ensuring the safety of rock engineering.

Equal channel angle pressing is recognized for its ability to refine alloy grains and alter grain orientation, thereby achieve better mechanical performance of the magnesium alloy. This study investigates the microstructures,dynamic recrystallization mechanism, texture development, and mechanical performance of GW94K(Mg–8.7Gd–4.18Y–0.42Zr wt. %) Mg alloy following ECAP-4 passes at 400°C and 3 mm/min. Results show that when high-temperature deformation is undertaken, twin formation is suppressed while dislocation slip is facilitated, increasing dislocation density during deformation. Following ECAP deformation, the sample displayed higher fracture elongation, TYS, and UTS than the as-solutioned GW94K alloy. In particular, the GW94K alloy performed well mechanically after ECAP-4 passes, with an ultimate TYS of 231 MPa, an UTS of 290 MPa, and an elongation of 14.8 %. DDRX and shear bands induce CDRX, both of which are important in plastic deformation. as well as in modifying microstructure and grain orientation during ECAP deformation.

Coniopteris simplex is a common component in many Jurassic ***,due to morphological variations in sterile fronds and incomplete preservation of reproductive organs,its classification has been controversial for a long ***,we present new material collected from the Middle Jurassic Yan’an Formation in the Ordos Basin,Inner *** new material reveals important morphological characters,including hemi-dimorphic fronds,two forms of sterile pinnae,short-stalked sori and sporangia on fertile fronds,annulus of sporangium composed of about 30 cells,in situ rounded triangular spores with protruded suture,and the lips of trilete laesura encircled by one or two rows of small *** on these observations,we emended the diagnosis of this common *** spatio-temporal distribution of Coniopteris simplex shows that the iconic species was restricted to the paleolatitudes ranging from 19.5°N to 48.9°N in the Northern Hemisphere during the Early—Late Jurassic,mainly distributed in the tropical to paratropical zones,and the paleolongitude ranging from 9.6°E to 129.3°E,which may be limited by the opening of the Viking Corridor and the splitting of the Central Atlantic Ocean.

Biological structural materials not only exhibit remarkable mechanical properties but also often embody dynamic characteristics such as environmental responsiveness,autonomy,and self-healing,which are difficult to achieve in conventional engineering *** merging materials science,synthetic biology,and other disciplines,engineered living materials(ELMs)provide a promising solution to combine living organisms with abiotic components,thus facilitating the construction of functional“living”*** natural materials,ELMs possess vitality and hold immense application potential in areas such as medicine,electronics,and construction,captivating increasing research attention *** an emerging branch of ELMs,structural ELMs aim to mimic living biological structural materials by achieving desired mechanical performance while maintaining important“living”*** we summarize the recent progress and provide our perspectives for this emerging research *** first summarize the superiority of structural ELMs by reviewing biological structural materials and biomimetic material design ***,we provide a systematic discussion on the definition and classifications of structural ELMs,their mechanical performance,and physiological ***,we summarize some critical challenges faced by structural ELMs and highlight directions of future *** hope this review article can provide a timely summary of the state of the art and relevant perspectives for future development of structural ELMs.
