Ouabain-Induced Auditory Nerve Degeneration in Congenic Ly5.1 Mice

The Ly5.1 mouse,also termed ***-Ptprca Pepcb/BoyJ,is a congenic strain widely used as a recipient in animal studies of bone marrow *** previous study documented that a majority of type I spiral ganglion neurons (SGNs) in the apical turns of Ly5.1 mice are unmyelinated and aggregate into neuronal clusters,similar to the spiral ganglion in the human ***,a well known Na-K ATPase inhibitor,has been shown to induce neuronal degeneration in a variety of neural tissues including the adult gerbil and CBA/CaJ mouse spiral ***,functional and pathological changes of the auditory nerves in young-adult Ly5.1 mice were examined at 3,7 and 14 days after ouabain *** to observations in CBA/CaJ mice,ouabain application selectively removed type I SGNs,resulting in an immense decline of the auditory nerve *** of glial cells was seen in the injured auditory nerves at 7 days after ouabain *** data indicate that the 'human-like' features of unmyelinated type I SGNs have no protective impact on the fate of SGNs after ouabain *** incorporating bromodeoxyuridine (BrdU) and expressing Sox2 were also counted in the auditory nerves of control and ouabain-treated *** number of Sox2+ glial cells significantly increased at 3 and 7 days ***,the highest density of BrdU+ cells appeared in the apical turn of the injured auditory nerve shortly after ouabain exposure,suggesting that the pattern of SGN loss at the apical turn in Ly5.1 mouse may have some impact on the reaction of non-neuronal cells in response to acute ototoxic drug exposure in the auditory nerve.

Self-synchronization of reinjected droplets for highefficiency droplet pairing and merging

Droplet merging serves as a powerful tool to add reagents to moving droplets for biological and chemical ***,unsynchronized droplet pairing impedes high-effciency ***,we develop a microfluidic design for the self-synchronization of reinjected droplets.A periodic increase in the hydrodynamic resistance caused by droplet blocking a T-junction enables automatic pairing of *** inducing spacing,the paired droplets merge downstream under an electric *** blockage-based design can achieve a 100%synchronization efficiency even when the mismatch rate of droplet frequencies reaches 10%.Over 98%of the droplets can still be synchronized at nonuniform droplet sizes and fluctuating reinjection flow ***,the droplet pairing ratio can be adjusted flexibly for on-demand sample *** this system,we merge two groups of droplets encapsulating enzyme/substrate,demonstrating its capacity to conduct multi-step *** also combine droplet sorting and merging to coencapsulate single cells and single beads,providing a basis for high-efficiency single-cell *** expect that this system can be integrated with other droplet manipulation systems for a broad range of chemical and biological applications.

A Versatile Flow-Profile Engineering Method in the Stokes Flow Regime for Complex-Shaped Flows

Flow profiles are frequently engineered in microfluidic channels for enhanced mixing,reaction control,and material ***,flow profiles are engineered by inducing inertial secondary flow to redistribute the streams,which can hardly be reproduced in microfluidic environments with negligible inertial *** employed symmetric channel structures also limit the variety of achievable flow ***,each of the flow profiles specifically corresponds to a strictly defined flow condition and cannot be generalized to other flow *** address these issues,we present a systematic method to engineer the flow profile using inertialess secondary *** flow is manipulated in the Stokes regime by deploying a cascaded series of microsteps with various morphologies inside a microchannel to shape the flow *** tuning the shapes of the microsteps,arbitrary outflow profiles can be customized.A numerical profile-transformation program is developed for rapid prediction of the output profiles of arbitrary sequences of predefined *** proposed method allows the engineering of stable flow profiles,including asymmetric ones,over a wide range of flow conditions for complex microfluidic environmental prediction and design.

Oil-mediated high-throughput generation and sorting of water-in-water droplets

Aqueous two-phase system(ATPS)droplets have demonstrated superior compatibility over conventional water-in-oil droplets for various biological ***,the ultralow interfacial tension hampers efficient and stable droplet generation,limiting further development and more extensive use of such ***,we present a simple strategy to employ oil as a transient medium for ATPS droplet *** methods based on passive flow focusing and active pico-injection are demonstrated to generate water-water-oil double emulsions,achieving a high generation frequency of~2.4 *** evaporation of the oil to break the double emulsions,the aqueous core can be released to form uniform-sized water-in-water ***,this technique can be used to fabricate aqueous microgels,and the introduction of the oil medium enables integration of droplet sorting to produce single-cell-laden hydrogels with a harvest rate of over 90%.We believe that the demonstrated high-throughput generation and sorting of ATPS droplets represent an important tool to advance droplet-based tissue engineering and single-cell analyses.

Multi-mode controller IC for soft-switched flyback converter with high efficiency over the entire load range

This paper presents a multi-mode control scheme for a soft-switched flyback converter to achieve high efficiency and excellent load regulation over the entire load range. At heavy load, critical conduction mode with valley switching (CCMVS) is employed to realize soft switching so as to reduce turn-on loss of power switch as well as conducted electromagnetic interference (EMI). At light load, the converter operates in discontinuous conduction mode (DCM) with valley switching and adaptive off-time control (AOT) to limit the switching frequency range and maintain load regulation. At extremely light load or in standby mode, burst mode operation is adopted to provide low power consumption through reducing both switching frequency and static power dissipation of the controller. The multi-mode control is implemented by an oscillator whose pulse duration is adjusted by output feedback. An accurate valley switching control circuit guarantees the minimum turn-on voltage drop of power switch. The pro-totype of the controller IC was fabricated in a 1.5-μm BiCMOS process and applied to a 310 V/20 V, 90 W flyback DC/DC converter circuitry. Experimental results showed that all expected functions were realized successfully. The flyback converter achieved a high efficiency of over 80% from full load down to 2.5 W, with the maximum reaching 88.8%, while the total power consumption in standby mode was about 300 mW.

Association between the level of CD4^+T lymphocyte microRNA-155 and coronary artery disease in patients with unstable angina pectoris

Objective To study the association between the expression of microRNA-155(miRNA-155)in peripheral blood CD4^+T lymphocytes and the level of semrn interferon-7(IFN-7)concentration and the severity of coronary artery disease (CAD).Methods After coronary angiography,252patients with suspected unstable angina pectoris (UAP)were divided into the UAP group (128patients with CAD confirmed by angiography)and the control group (124patients without CAD confirmed by angiography).Fresh peripheral blood was extracted 16-24h before coronary angiography,CD4^+T lymphocytes was tested using immunomagnetic beads,the expression ofmiRNA-155was tested using quantitative PCR and the expression of IFN-7was tested using enzyme-linked immunosorbent assay (ELISA).According to the results of angiography,Gensini score of coronary artery lesions was ***,we also analysis the association between the level of miRNA-155in peripheral blood CD4^+T lymphocytes,the level of serum IFN-γand Gensini score of coronary *** The levels ofmiRNA-155(0.49±0.08vs.0.23±0.09)and IFN-7(227.58±26.01vs.141.23±17.89)in the UAP group were significantly higher than that of the control group,the difference was statistically *** level of miRNA-155and IFN-γwere positively correlated with Gensini score of CAD (r =0.534,r =0.713,respectively,all P <0.05).The level of miRNA-155was positively correlated with the level of IFN-γ,(r =0.686,P <0.05).Conclusions The level of miRNA-155in peripheral blood CD4^+T lymphocytes and the level of IFN-γ are closely correlated with the severity of CAD.

A novel rose flower-like SnO hierarchical structure synthesized by a hydrothermal method in an ethanol/water system

Rose flower-like SnO hierarchical 3D architectures consisting of well-ordered microsheets were synthesized by a template-free and surfactant-free hydrothermal method based on the reaction between SnCl2 and NaOH in ethanol/water. The ethanol amount is a crucial factor in controlling the size and morphology of the microsheets. Ethanol can accelerate the delamination of the SnO blocks to the microsheets, and the microsheets changed from a quasi square to an octodecahedral shape with the increase of the ethanol ratio. When the volume ratio of ethanol/water is 2：1, the microstructure of SnO presents a rose flower-like hierarchical architecture with a diameter of about 40 μm, which is regularly composed of numerous well-ordered thin microsheets with octodecahedral shape, and the microsheets were formed by countless nanoparticles. A possible growth mechanism of the architectures was proposed. Moreover, SnO2 nanospheres were synthesized by a similar experiment only without NaOH, which showed that NaOH was crucial to the formation of SnOx.
