Prolonged hypervolemic hemodilution decreases functional capillary density of ileal mucosa in pigs revealed by sidestream dark-field imaging

Objective:Hemodilution changes the physical properties of blood by reducing its hematocrit and blood *** tested whether prolonged hypervolemic hemodilution (HHD) impairs functional capillary density (FCD) of ileal mucosa in healthy mechanically-ventilated pigs and if there is any correlation between changes in FCD of ileal and sublingual mucosas during ***:Sixteen domestic female pigs were anesthetized,mechanically-ventilated,and randomly assigned to the HHD (20 ml/(kg·h) Hartmann's solution for 3 h) or fluid restrictive (5 ml/(kg·h) Hartmann's solution for 3 h) *** of sublingual and ileal mucosas via ileostomy were visualized using sidestream dark-field (SDF) imaging at baseline conditions (t=0 h) and at selected time intervals of fluid therapy (t=1,2,and 3 h).Results:A significant decrease of ileal FCD (285 (278-292) cm/cm 2) in the HHD group was observed after the third hour of HHD when compared to the baseline (360 (350-370) cm/cm 2) (Pleal mucosa FCD was significantly higher after the third hour of fluid therapy as compared to the HHD group (Plation between microhemodynamic parameters obtained from sublingual and ileal mucosas was found throughout the ***:Prolonged HHD established by crystalloid solution significantly decreased ileal villus FCD when compared to restrictive fluid *** inappropriate degree of HHD can be harmful during uncomplicated abdominal surgery.

Nisin Use to Control l. innocua in Experimentally Contaminated Gombasek Sausage

Meat is a rich nutrient matrix that provides a suitable environment for proliferation of meat spoilage microbiota and common food-born pathogens, therefore adequate preservation has to be applied in order to preserve its safety and quality. lantibiotic nisin is not commonly used in the meat industry, in spite of its beneficial effect in e. g. dairy industry. Therefore, here, the effect of nisin was searched in traditional fermented sausage Gombasek experimentally contaminated with listeria innocua li1 strain. The antimicrobial effect of nisin was demonstrated by the li1 strain decrease at day 2 (after contamination) with the difference 1.79 logarithmic (log) cycles comparing with the control sausage (Cli). At week 1, the difference 4.14 log cycles was noted in the experimental sausages comparing with Cli sausages with prolonging inhibitory effect up to week 2; although the difference was diminished (1.82 log cycles); but higher than at day 2. The pH values as well as lactic acid values were not influenced by the nisin treatment (pH value in the reference sausage was 5.0, in Cli 5.7 and in the experimental sausage E, 5.1; amount of lactic acid in E was 0.099 mmol l-1 and in Cli 0.107 mmol l-1).

Molecular recognition of live methicillin-resistant staphylococcus aureus cells using DNA aptamers

AIM: To generate DNA-aptamers binding to Methicillinresistant Staphylococcus aureus(MRSA).METHODS: The Cell-Systematic Evolution of ligands by Exponential Enrichment(SElEX) technology was used to run the selection against MRSA bacteria and develop target-specific aptamers. MRSA bacteria were targeted while Enterococcus faecalis bacteria were used for counter selection during that process. Binding assays to determine the right aptamer candidates as well as binding assays on clinical samples were performed through flow cytometry and analyzed using the FlowJ o software. The characterization of the aptamers was done by determination of their Kd values and determined by analysis of flow data at different aptamer concentration using Sigma Plot. Finally, the recognitionof the complex Gold-nanoparticle-aptamer to the bacteria cells was observed using transmission electron microscopy(TEM).RESUlTS: During the cell-SElEX selection process, 17 rounds were necessary to generate enrichment of the pool. While the selection was run using fixed cells, it was shown that the binding of the pools with live cells was giving similar results. After sequencing and analysis of the two last pools, four sequences were identified to be aptamer candidates. The characterization of those aptamers showed that based on their Kd values, DTMRSA4 presented the best binding with a Kd value of 94.61 ± 18.82 nmol/l. A total of ten clinical samples of MRSA, S. aureus and Enterococcus faecalis were obtained to test those aptamers and determine their binding on a panel of samples. DTMRSA1 and DTMRSA3 showed the best results regarding their specificity to MRSA, DTMRSA1 being the most specific of all. Finally, those aptamers were coupled with gold-nanoparticle and their binding to MRSA cells was visualized through TEM showing that adduction of nanoparticles on the aptamers did not change their binding ***: A total of four aptamers that bind to MRSA were obtained with Kd values ranking from 94
