目的:观察经白藜芦醇培养的胚鼠腹侧中脑组织移植至帕金森病大鼠模型脑纹状体神经元的凋亡情况,同时探讨白藜芦醇浓度的影响。方法:实验于2004-03/2006-01在南京中医药大学解剖实验室完成。①用神经毒剂6-羟基多巴胺单侧损毁SD大鼠左侧中脑被盖腹侧区和黑质致密部建立帕金森病动物模型。②实验分为白藜芦醇培养组和对照组。白藜芦醇培养组又分为3个亚组:20,40,60mg/L组,每组各9只,分别将胚鼠腹侧中脑组织预置于20,40,60mg/L白藜芦醇培养液中;对照组9只,将腹侧中脑组织预置于DMEM营养液中。③预置12h后移植至大鼠左侧纹状体尾壳核头部。④于移植后5,10,15d将动物处死,采用原位末端标记法结合流式细胞术及免疫组织化学方法对移植神经元凋亡和存活情况进行定量分析。结果:①脑纹状体组织观察酪氨酸羟化酶免疫组织化学结果:白藜芦醇培养组和对照组纹状体移植针道和针道周围都可见大量酪氨酸羟化酶阳性细胞;细胞计数显示白藜芦醇培养组的酪氨酸羟化酶阳性细胞数量较对照组多(P<0.01);白藜芦醇培养组中,40mg/L白藜芦醇组中的存活的酪氨酸羟化酶阳性细胞数量较20mg/L组和60mg/L组多([5d:(4804.77±427.37)比(3069.89±148.95),(3038.20±118.95)个/200倍视野;10d:(4608.97±274.64)比(2850.83±48.06),(2875.92±210.54)个/200倍视野;15d:(4217.42±325.93)比(2623.39±350.68),(2568.39±49.67)个/200倍视野;P<0.01]。②脑纹状体组织观察原位末端标记法结果:白藜芦醇培养组较对照组移植神经元凋亡细胞数减少;白藜芦醇培养组中,40mg/L白藜芦醇组中的凋亡细胞数量少于其他两组(P<0.01)。③脑纹状体组织流式细胞仪检测结果:白藜芦醇培养组较对照组移植神经元凋亡率降低。结论:白藜芦醇具有促移植神经元存活作用,作用效果与培养液中白藜芦醇含量有关。

Irradiation Testing of Coated Particle Fuel at HAnARO

TRISO （Tri-structural iso-tropic）-coated particle fuel is being developed to support the development of a VHTR （very high temperature reactor） in Korea. From August 2013, the first irradiation testing of coated particle fuel was begun to demonstrate and qualify TRISO fuel for use in the VHTR in HAnARO （high-flux advanced neutron application reactor） at KAERI （Korea Atomic Energy Research Institute）. This experiment is currently undergoing under an atmosphere of a mixed inert gas without on-line temperature monitoring and control combined with on-line fission product monitoring of the sweep gas. The irradiation device contains two test rods, one contains nine fuel compacts and the other five compacts and eight graphite specimens. Each compact has 263 coated particles. After a peak bum-up of about 4% and a peak fast neutron fluence of about 1.7 × 1021 n/cm2, PIE （post irradiation examination） will be carried out at KAERI＇s irradiated material examination facility. This paper describes the characteristics of coated particle fuels, and the design of the test rod and irradiation device for the coated particle fuels, and discusses the technical results of irradiation testing at HAnARO.

Background:This study aimed to determine the effect of different carbohydrate(cho)doses on exercise capacity in patients with McArdle disease—the paradigm of“exercise intolerance”,characterized by complete muscle glycogen unavailability—and to determine whether higher exogenous glucose levels affect metabolic responses at the McArdle muscle cell(in vitro)***:Patients with McArdle disease(n=8)and healthy controls(n=9)underwent a 12-min submaximal cycling constant-load bout followed by a maximal ramp test 15 min after ingesting a non-caloric *** a randomized,double-blinded,cross-over design,patients repeated the tests after consuming either 75 g or 150 g of cho(glucose:fructose=2:1).Cardiorespiratory,biochemical,perceptual,and electromyographic(EMG)variables were ***,glucose uptake and lactate appearance were studied in vitro in wild-type and McArdle mouse myotubes cultured with increasing glucose concentrations(0.35,1.00,4.50,and 10.00 g/L).Results:Compared with controls,patients showed the“classical”second-wind phenomenon(after prior disproportionate tachycardia,myalgia,and excess electromyographic activity during submaximal exercise,all pnd an impaired endurance exercise capacity(-51%ventilatory threshold and55%peak power output,both pcho dose(pcho intake increased blood glucose and lactate levels,decreased fat oxidation rates,and attenuated the second wind in the ***,only the higher dose increased ventilatory threshold(+27%,p=0.010)and peak power output(+18%,p=0.007).In vitro analyses revealed no differences in lactate levels across glucose concentrations in wild-type myotubes,whereas a doseresponse effect was observed in McArdle ***:cho intake exerts beneficial effects on exercise capacity in McArdle disease,a condition associated with total muscle glycogen *** of these benefits are dose dependent.

The use of dyes such as tartrazine (E102) and sunset yellow (E102) in food, beverages and health products for technological and commercial purposes is common. The adverse effects caused by these dyes, such as allergies and hyperactivity disorder have been reported, especially in children. In the present study, a chromatographic method was developed and validated for simultaneous determination of tartrazine and sunset yellow. The chromatographic separation was performed on a Lichrocart® C18 column (125 × 4.6 mm;5 μm) with a security Guard-C18 column (4 × 2.0 mm, 5 μm;Phenomenex, Torrance, CA, USA) maintained at 30°C. The mobile phase consisted of a mixture of acetonitrile/ammonium acetate buffer pH 6.8 in gradient mode with a flow rate of 1 mL/min. The injection volume was 10 μL. The detection wavelength was set at 455 nm. The parameters of specificity, linearity, precision, repeatability, accuracy and sensitivity were examined for validation. The developed method is linear in the range of 1 μg/mL to 100 μg/mL with a R2> 0.998. The intra-day and inter-day precisions (RSD) were less than 0.6% and 3.1% respectively. The detection limit was 0.03 μg/mL and the quantification limit was 0.1 μg/mL. The retention time of tartrazine was 2.86 min, while sunset yellow was detected at 5.67 min. A simple, rapid, accurate and robust HPLC/UV-Visible method was developed and validated for simultaneous identification and quantification of tartrazine and sunset yellow. This developed method was successfully applied for the simultaneous determination of tartrazine and sunset yellow in soft drinks sold in Benin.

The use of well water by disadvantaged populations in most African cities often presents health risks. The purpose of this study is to determine the physico-chemical and microbiological quality of M’bahiakro well water. A series of physico-chemical, microbiological and piezometric analyses were carried out on the well water in the area. The results of these analyses were processed using Piper and Schoeller-Berkaloff diagrams and the software R 3.1.2 for Principal Component Analysis. Physico-chemical analysis results show that the water has an average temperature of 28°C. They are acidic, with a pH that generally varies from 4 to 6. M’bahiakro well waters are highly mineralized, with an average electrical conductivity of 369.1 μS/cm and 984.1 μS/cm and pass from sodium and potassium chlorinated facies in the dry season (February) to sodium chlorinated facies in the rainy season (October) with K+, na+ and nO3- ions dominating. These ions would be the result of soil rainfall and the intervention of anthropogenic activities in the pollution of shallow groundwater. Bacteriological results exploitation shows that the well waters studied host high densities of enteritis bacteria (E. coli, Fecal streptococcus, and Clostridium perfringens) during the rainy period (October). M’Bahiakro’s well water is of poor quality and unsuitable for human consumption without prior treatment. However, they deserve particular attention and would require further treatment in rainy periods.

AIM： To compare small sphincterotomy combined with endoscopic papillary large balloon dilation （SES ＋ ELBD） and endoscopic sphincterotomy （EST） for large bile duct stones. METHODS： We compared prospectively SES ＋ ELBD （group A, n = 27） with conventional EST （group B, n = 28） for the treatment of large bile duct stones （≥ 15 mm）. When the stone could not be removed with a normal basket, mechanical lithotripsy was performed. We compared the rates of complete stone removal with one session and application of mechanical lithotripsy. RESULTS： no significant differences were observed in the mean largest stone size （A： 20.8 mm, B： 21.3 mm）, bile duct diameter （A： 21.4 turn, B： 20.5 ram）, number of stones （A： 2.2, B： 2.3）, or procedure time （A： 18 min, B： 19 rain） between the two groups. The rates of complete stone removal with one session was 85% in group A and 86% in group B （P = 0.473）. Mechanical lithotripsy was required for stone removal in nine of 27 patients （33%） in group A and nine of 28 patients （32%, P = 0.527） in group ***： SES ＋ ELBD did not show significant benefits compared to conventional EST, especially for the removal of large （≥ 15 mm） bile duct stones.

Objective:To demonstrate the effect of dieckol from Eisenia bicyclis on osteoclastogenesis using RAW 264.7 ***:Murine macrophage RAW 264.7 cells were subjected to dieckol treatment,followed by treatment with receptor activator of nuclear factor kappa-B ligand(RAnKL)to induce ***-resistant acid phosphatase(TRAP)activity was examined using a TRAP activity *** blotting analysis was conducted to examine the level of osteoclast-related factors,including TRAP and calcitonin receptor(CTR),transcriptional factors,including c-Fos,c-Jun,and nuclear factor of activated T cells cytoplasmic 1(nFATc1),nuclear factor kappa-B(nF-κB),extracellular signal-regulated kinase(ERK),and c-Jun n-terminal kinase(JnK).Immunofluorescence staining was conducted to examine the expression of c-Fos,c-Jun,and ***:Among the four phlorotannin compounds present in Eisenia bicyclis,dieckol significantly hindered osteoclast differentiation and expression of RAnKL-induced TRAP and *** addition,dieckol downregulated the expression levels of c-Fos,c-Jun,nFATc1,ERK,and JnK,and suppressed nF-κB ***:Dieckol can suppress RAnKL-induced ***,it has therapeutic potential in treating osteoclastogenesis-associated diseases.
