Miniature fluorescence microscopes are a standard tool in systems ***,widefield miniature microscopes capture only 2D information,and modifications that enable 3D capabilities increase the size and weight and have poor resolution outside a narrow depth ***,we achieve the 3D capability by replacing the tube lens of a conventional 2D Miniscope with an optimized multifocal phase mask at the objective’s aperture *** the phase mask at the aperture stop significantly reduces the size of the device,and varying the focal lengths enables a uniform resolution across a wide depth *** phase mask encodes the 3D fluorescence intensity into a single 2D measurement,and the 3D volume is recovered by solving a sparsity-constrained inverse *** provide methods for designing and fabricating the phase mask and an efficient forward model that accounts for the fieldvarying aberrations in miniature *** demonstrate a prototype that is 17mm tall and weighs 2.5 grams,achieving 2.76μm lateral,and 15μm axial resolution across most of the 900×700×390μm^(3) volume at 40 volumes per *** performance is validated experimentally on resolution targets,dynamic biological samples,and mouse brain *** with existing miniature single-shot volume-capture implementations,our system is smaller and lighter and achieves a more than 2×better lateral and axial resolution throughout a 10×larger usable depth *** microscope design provides single-shot 3D imaging for applications where a compact platform matters,such as volumetric neural imaging in freely moving animals and 3D motion studies of dynamic samples in incubators and lab-on-a-chip devices.

A deep learning-enabled portable imaging flow cytometer for cost-effective, highthroughput, and label-free analysis of natural water samples

We report a deep learning-enabled field-portable and cost-effective imaging flow cytometer that automatically captures phase-contrast color images of the contents of a continuously flowing water sample at a throughput of 100 mL/*** device is based on partially coherent lens-free holographic microscopy and acquires the diffraction patterns of flowing micro-objects inside a microfluidic *** holographic diffraction patterns are reconstructed in real time using a deep learning-based phase-recovery and image-reconstruction method to produce a color image of each micro-object without the use of external *** blur is eliminated by simultaneously illuminating the sample with red,green,and blue light-emitting diodes that are *** by a laptop computer,this portable device measures 15.5 cm×15 cm×12.5 cm,weighs 1 kg,and compared to standard imaging flow cytometers,it provides extreme reductions of cost,size and weight while also providing a high volumetric throughput over a large object size *** demonstrated the capabilities of this device by measuring ocean samples at the Los Angeles coastline and obtaining images of its micro-and nanoplankton ***,we measured the concentration of a potentially toxic alga(Pseudo-nitzschia)in six public beaches in Los Angeles and achieved good agreement with measurements conducted by the California Department of Public *** cost-effectiveness,compactness,and simplicity of this computational platform might lead to the creation of a network of imaging flow cytometers for largescale and continuous monitoring of the ocean microbiome,including its plankton composition.

Eight-week ledipasvir/sofosbuvir in non-cirrhotic, treatment-na?ve hepatitis C genotype-1 patients with hepatitis C virus-RNA < 6 million: Single center, real world effectiveness and safety

AIM To evaluate sustained viral response(SVR) of 8-wk ledipasvir/sofosbuvir therapy among non-cirrhotic, genotype-1 hepatitis C virus(HCV) patients with RNA < 6 million IU/m *** We performed a retrospective cohort study to examine SVR rates, predictors of treatment failure and safety analysis of 8-wk ledipasvir/sofosbuvir(LDV/SOF) therapy among non-cirrhotic, genotype 1 HCV patients with viral load < 6 million IU/m L. Primary outcome was an achievement of SVR at 12 wk after treatment. Secondary outcomes were identifying predictors of treatment failure and adverse events during *** Total 736 patients: 55% males, 51% Caucasians and 65% were genotype 1a. Non-cirrhotic state of 53% was determined by clinical judgment(imaging, AST, platelet count) and 47% had documented liver fibrosis testing(biopsy, vibration-controlled transient elastography, serum biomarkers). Overall SVR12 was 96%. No difference in SVR12 was seen between patients whose non-cirrhotic state was determined by clinical judgment and patients who had fibrosis testing. Age groups, gender, ethnicity and genotype 1 subtype did not predict SVR. Non-cirrhotic state determined by clinical judgment based on simple, non-invasive tests were not associated with lower SVR [OR = 1.02, 95%CI: 0.48-2.17, P = 0.962]. The AUROC for hepatitis C RNA viral load was 0.734(P < 0.001, 95%CI: 0.66-0.82). HCV RNA 2.2 million IU/m L was identified as the cutoff value with sensitivity 73% and specificity 64%. HCV RNA < 2.2 million IU/m L was associated with significantly higher SVR 98% with OR = 0.22(95%CI: 0.1-0.49, P < 0.001) compared to SVR 92% in HCV RNA ≥ 2.2 million IU/m L. No death or morbidities were *** Our outcomes validate safety and effectiveness of 8-wk LDV/SOF therapy in non-cirrhotic, untreated HCV genotype 1 patients with HCV RNA < 6 million IU/m L.

＜正＞ In vivo endothelin type B （ETB） receptor activation induced an acute rise in mean arterial pressure （MAP） accompanied by increased superoxide （O2-） production in sympathetic ganglia. The goal of our present study was to determine if this elevated O2- anion concentration participates in the pathogenesis of ET-dependent hypertension by facilitating nicotinic neurotransmission through the ganglion. We used chlorisondamine （CHL） to block nicotinic input in autonomic ganglia. Sprague-Dawley rats were assigned to one of 3 treatments: 1) 2h infusion of the specific ETB receptor agonist sarafotoxin 6c （S6c）, 2) CHL followed by 2h S6c. 3) CHL followed by 2h saline. MAP increased significantly following S6c and CHL-S6c treatment. To measure CY levels,we removed ganglia following infusion and stained them with dihydroethidine （DHE）. The DHE fluorescence intensities were significantly greater in both S6c and CHL-S6c rats compared to CHL saline infused rats. Our results show that hypertension and elevated O2- production following ETB receptor activation persist after ganglionic blockade,suggesting that ET-dependent hypertension maybe impartial to preganglionic input.

Candidate members of the Pal 5, GD-1, Cetus Polar and Orphan tidal stellar halo streams from SDSS DR9, LAMOST DR3 and APOGEE catalogs

We present candidate members of the Pal 5, GD-1, Cetus Polar and Orphan tidal stellar streams found in LAMOST DR3, SDSS DR9 and APOGEE catalogs. In LAMOST DR3, we find 20, 4 and 24 high confidence candidates of tidal streams GD-1, Cetus Polar and Orphan respectively. We also list 59, 118 and 10 high confidence candidates of tidal streams Cetus Polar, Orphan and Pal 5, respectively from the SDSS DR9 spectroscopic catalog. Furthermore, we find seven high confidence candidates of the Pal 5 tidal stream in the APOGEE data. Compared with SDSS, the new candidates from LAMOST DR3 are brighter, so that together, more of the color-magnitude diagram, including the giant branch, can be explored. Analysis of the SDSS data shows that there are three metallicity peaks associated with the Orphan stream which also exhibit some spatial separation. The LAMOST data confirm multiple metallicities in this stream. The metallicity, given by the higher resolution APOGEE instrument, of the Pal 5 tidal stream is [Fe/H] ,- -1.2, higher than that given earlier by SDSS spectra. Many previously unidentified stream members are tabulated here for the first time, along with existing members, allowing future researchers to further constrain the orbits of these objects as they move within the Galaxy＇s dark matter potential.

中国目前正在建设两个打破世界纪录的特高压直流输电工程,一是±800kV云南—广东直流输电工程,二是±800kV向家坝—上海直流输电工程。文章探讨了这两项直流输电工程的换流变压器在型式试验期间遇到的一些技术问题。该试验是按照客户基于相关的IEC标准提出的项目设备规范要求实施的,因此研究结果与相关的IEC标准及该试验的实际经验有关。基于这些研究结果,KEMA提出了对将来工程中换流变压器的试验要求的建议以及针对个别具体试验的改进措施。有一点要强调的,KEMA所推荐的变压器试验顺序值得遵循,因为在这一方面IEC标准是不明确的。

We quantify and correct systematic errors in PPMXL proper motions using extragalactic sources from the first two LAMOST data releases and the Vbron-Cetty ＆ Vebron Catalog of Quasars. Although the majority of the sources are from the Veron catalog, LAMOST makes important contributions in regions that are not well-sampled by previous catalogs, particularly at low Galactic latitudes and in the south Galactic cap. We show that quasars in PPMXL have measurable and significant proper motions, which reflect the systematic zero-point offsets present in the catalog. We confirm the global proper motion shifts seen by Wu et al., and additionally find smaller-scale fluctuations of the QSO-derived corrections to an absolute frame. We average the proper motions of 158 106 extragalactic objects in bins of 3° ×3° and present a table of proper motion corrections.

We describe the current plans for a spectroscopic survey of millions of stars in the Milky Way galaxy using the Guo Shou Jing Telescope （GSJT, formerly calledthe Large sky Area Multi-Object fiber Spectroscopic Telescope -- LAMOST）. The survey will obtain spectra for 2.5 million stars brighter than r 〈 19 during dark/grey time, and 5 million stars brighter than r 〈 17 or J 〈 16 on nights that are moonlit or have low transparency. The survey will begin in the fall of 2012, and will run for at least four years. The telescope＇s design constrains the optimal declination range for observations to 10~ 〈 di 〈 50~, and site conditions lead to an emphasis on stars in the direction of the Galactic anticenter. The survey is divided into three parts with different target selection strategies： disk, anticenter, and spheroid. The resulting dataset will be used to study the merger history of the Milky Way, the substructure and evolution of the disks, the nature of the first generation of stars through identification of the lowest metallicity stars, and star formation through study of open clusters and OB associations. Detailed design of the LAMOST Experiment for Galactic Understanding and Exploration （LEGUE） survey will be completed in summer 2012, after a review of the results of the pilot survey.
