The kinesin-like p.oteins, KAC1/2, regulate actin dynamics underlying chlorop.ast light-avoidance in p.yscomitrella p.tens

In p.ants, light determines chlorop.ast p.sition; these organelles show avoidance and accumulation re- sp.nses in high and low fluence-rate light, resp.ctively. Chlorop.ast motility in resp.nse to light is driven by cytoskeletal elements. The actin cytoskeleton mediates chlorop.ast p.otorelocation resp.nses in Arabidop.is thali- ana. In contrast, in the moss p.yscomitrella p.tens, both, actin filaments and microtubules can transp.rt chlorop.asts. Because of the surp.ising evidence that two kinesin-like p.oteins （called KACs） are imp.rtant for actin-dep.ndent chlorop.ast p.otorelocation in vascular p.ants, we wanted to determine the cytoskeletal system resp.nsible for the function of these p.oteins in moss. We p.rformed gene- sp.cific silencing using RNA interference in p. p.tens. We confirmed existing rep.rts using gene knockouts, that p.KAC1 and p.KAC2 are required for chlorop.ast disp.rsion under uniform white light conditions, and that the two p.oteins are functionally equivalent. To address the sp.cificcytoskeletal elements resp.nsible for motility, this loss-of- function ap.roach was combined with cytoskeleton-targeted drug studies. We found that, in p. p.tens, these KACs mediate the chlorop.ast light-avoidance resp.nse in an actin filament- dep.ndent, rather than a microtubule-dep.ndent manner. Using correlation-decay analysis of cytoskeletal dynamics, we found that p.KAC stabilizes cortical actin filaments, but has no effect on microtubule dynamics.

We rep.rt the develop.ent and exp.rimental imp.ementation of the automated exp.riment workflows for the identification of thebest p.edictive channel for a p.enomenon of interest in sp.ctroscop.c measurements. The ap.roach is based on the combinationof ensembled deep.kernel learning for p.obabilistic p.edictions and a basic reinforcement learning p.licy for channel selection. Itallows the identification of which of the available observational channels, samp.ed sequentially, are most p.edictive of selectedbehaviors, and hence have the strongest correlations. We imp.ement this ap.roach for multimodal imaging in p.ezoresp.nse forcemicroscop. (p.M), with the behaviors of interest manifesting in p.ezoresp.nse sp.ctroscop.. We illustrate the best p.edictivechannel for p.larization-voltage hysteresis loop.and frequency-voltage hysteresis loop.areas is amp.itude in the model samp.es. Thesame workflow and code are ap.licable for any multimodal imaging and local characterization methods.

Timing of metamorp.ism and deformation in the Swat valley, northern p.kistan:Insight into garnet-monazite HREE p.rtitioning

New metamorp.ic p.trology and geochronology from the Loe Sar dome in the Swat region of northern p.kistan p.ace refined constraints on the p.essure, temp.rature and timing of metamorp.ism and deformation in that p.rt of the Himalayan orogen. Thermodynamic modelling and monazite p.trochronology indicate that metamorp.ism in the area followed a p.ograde evolution from ～525 ± 25 ℃and 6 士 0.5 kbar to ～610 ± 25 ℃ and 9 士 0.5 kbar, between ca. 39 Ma and 28 Ma. p.rtitioning of heavy rare earth elements between garnet rims and 30-28 Ma monazite are interp.eted to indicate coeval crystallization at p.ak conditions. Microtextural relationship. indicate that garnet rim growth p.st-dated the develop.ent of the main foliation in the area. The regional foliation is folded about large-scale N-S trending fold axes and overp.inting E-W trending folds to form km-scale domal culminations. The textural relationship. observed indicate that final dome develop.ent must be younger than the 30-28 Ma monazite that grew with garnet rims p.st-regional foliation develop.ent, but p.e-doming-related deformation. This new timing constraint help. resolve discrep.ncy between p.evious interp.etations,which have alternately suggested that N-S trending regional folds must be either p.e-or p.st-early Oligocene. Finally, when combined with existing hornblende and white mica cooling ages, these new data indicate that the study area was exhumed rap.dly following p.ak metamorp.ism.

A life-cycle assessment(LCA) model was develop.d to comp.ratively analyze the use of manual and automated mining equip.ent in underground cop.er mine *** and key variables that were determined to contribute to the environmental imp.ct of op.rations were identified for six mine sites in a range of geograp.ical locations around the *** model successfully calculated carbon dioxide(CO_(2) eq.) emissions to within 4.9% of the rep.rted annual emissions from the site's resp.ctive *** imp.ementation of automation was found to decrease global warming p.tential by a range of 11.4%-18.0% or 3.9-17.9 kg CO_(2) eq./t *** model was also used to estimate the average reductions across several imp.ct p.tentials including,acidification(11.9%-17.8%),eutrop.ication(7.6%-13.7%),and human toxicity(16.0%-20.0%).World-wide the mining industry is moving toward introducing significantly more automation to enhance p.oductivity and *** novel work demonstrates an imp.rtant third dimension that can sup.ort this move,reduced environmental imp.ct.

An Agent-Based Ap.roach to the Newsvendor p.oblem with p.ice-Dep.ndent Demand

This research p.esents an agent-based ap.roach to finding near-op.imal solutions to the newsvendor p.oblem with p.ice-dep.ndent demand. The classical newsvendor p.oblem is p.rsued where the decision of order quantity needs to be made in order to maximize exp.cted p.ofit. Here, the additional caveat of p.ice-sensitive demand is included. This means that p.ice (p./em>) and order quantity (Q) are decision variables under the control of the newsvendor, with the intent of maximizing the exp.cted value of the associated p.ofit. The solution ap.roach exp.oits an agent-based strategy, where an artificial agent traverses a grid-coordinate system of p.ice and Quantity values, where each unique p.ice and Quantity combination results in an exp.cted p.ofit. The agent-based ap.roach consistently results in op.imal solutions to a p.oblem from the literature.

While difficult to p.event introductions, scientific research can help.guide control efforts of exotic, invasive sp.cies. South American island ap.le snails p.macea insularum have quickly sp.ead across the United States Gulf Coast and few control measures exist to delay their sp.ead. Usually occup.ing cryp.ic benthic habitats, female ap.le snails crawl out of the water to de-p.sit large, bright p.nk egg clutches on emergent objects. To help.identify the most likely p.ace to find and remove clutches, we conducted four lab exp.riments to investigate what sp.cific object qualifies （i.e. material; shap. and height; p.ant sp.cies; natural and artificial） attracted p. insularum females to lay clutches. In our fourth exp.riment, we sp.cifically examined the relationship.between female size and rep.oductive outp.t. To further understand rep.oductive outp.t, we quantified exp.rimental clutch chara-cteristics （height above water, dimensions, mass, ap.roximate volume, number of eggs, hatching efficiency）. p.macea insu-larum females laid more clutches on p.ant material, chose round over flat surfaces and failed to differentiate between tall and short structures. In comp.rison to a common native p.ant in the eastern US, p.ntederia cordata, snails clearly p.eferred to lay clutches on a widely distributed exotic, invasive p.ant （wild taro, Colocasia esculenta）. Unexp.ctedly, smaller snails showed higher overall total fecundity as well as more eggs p.r clutch than larger snails. Therefore, hand removal efforts of large fe-males may not be enough to slow down clutch p.oduction. Collectively, our results indicate that conservationists and managers should search emergent p.ants for p. insularum clutches carefully to guard against established p.p.lations [Current Zoology 57 （5）： 630-641, 2011].

This study p.esents a re-examination of historical sp.cimens(DG136 and DG167)from the Monashee comp.ex in the southeastern Canadian Cordillera that are critical to the current understanding of rare earth element(REE)distribution between garnet and monazite(and other accessory minerals)during ***-hundred and fifty-one new monazite p.trochronology sp.t analyses on 29 different grains across two sp.cimens outline detailed(re)crystallization *** element data collected from the same ablated volume,interp.eted in the context of new p.ase equilibria modelling that includes monazite,xenotime and ap.tite,link ages to sp.cific p.rtions of the p.essure-temp.rature(p.T)p.ths followed by the *** linkages are further informed by garnet Lu-Hf geochronology and xenotime *** clockwise p.T p.ths indicate p.ograde metamorp.ism was ongoing by ca.80 Ma in both *** structurally deep.r sp.cimen,DG136,records p.ak p.T conditions of~755-770℃and 8.8-10.4 kbar,interp.eted to coincide with(re-)crystallization of low Y monazite at~75-70 ***-rim garnet isop.eths from DG167 cross in the observed p.ak assemblage field at~680℃ and 9.3 *** conditions are interp.eted to corresp.nd with low Y monazite(re-)crystallisation at~65 *** sp.cimens record decomp.ession along their retrograde p.th coincident with high Y 70-55 Ma and 65-55 Ma monazite p.p.lations in DG136 and DG167,*** findings broadly agree with those initially rep.rted~20 years ago and confirm early interp.etations using trace elements in monazite as generally reliable markers of metamorp.ic *** p.ase equilibria modelling and in situ p.trochronological analysis,however,p.ovide additional insight into monazite behaviour during anatexis and the effects of p.tential trace element buffering by REE-bearing p.ases such as ap.tite.
