Role of calmodulin in neuronal Kv7/KCNQ potassium channels and epilepsy

Neuronal Kv7/KCNQ channels are critical regulators of neuronal excitability since they potently suppress repetitive firing of action potentials. These voltage-dependent potassium channels are composed mostly of Kv7.2 / KCNQ2 and KvT.3 / KCNQ3 subunits that show overlapping distribution throughout the brain and in the peripheral nervous system. They are also called ＇m-channels＇ since their inhibition by muscarinic agonists leads to a profound increase in action potential firing. Consistent with their ability to suppress seizures and attenuate chronic inflammatory and neuropathic pain, mutations in the KCNQ2 and KCNQ3 genes are associated with benign familial neonatal convulsions, a dominantly-inherited epilepsy in infancy. Recently, de novo mutations in the KCNQ2 gene have been linked to early onset epileptic encephalopathy. Notably, some of these mutations are clustered in a region of the intracellular cytoplasmic tail of Kv7.2 that interacts with a ubiquitous calcium sensor, calmodulin. In this review, we highlight the recent advances in understanding the role of calmodulin in modulating physiological function of neuronal Kv7 channels including their biophysical properties, assembly, and trafficking. We also summarize recent studies that have investigated functional impact of epilepsy-associated mutations localized to the calmodulin binding domains of Kv7.2.

Recently,ultra-precision polishing processes using smart materials such as magnetorheological(mR)fluids have been used for optics parts such as aspheric *** mR fluid applied in mR polishing is a phase controllable suspension comprising a mixture of CI particles and non-magnetic fluids such as mineral oil and *** fluids are a kind of smart material that can be controlled according to their flow properties such as viscosity and stiffness. Water-based mR fluids are easily oxidized if the fluids are exposed and this oxidation can bring irregular polishing results. Also,this phenomenon is one of several factors that reduce the life time of mR *** a previous work,CI particles were coated with a polymer or a composite for the improvement of dispersion stability and prevention of corrosion due to *** this study,the rheological properties such as viscosity and sedimentation of coated CI particles with silica were investigated with a rotational rheometer;mR polishing characteristics of BK7 glass were studied in order to investigate the surface *** wheel speed and magnetic field intensity were chosen as variables.

Decellularization to produce bioscaffolds composed of the extracellular matrix(ECm)uses enzymatic,chemical and physical methods to remove antigens and cellular com.onents from *** decellularization methods depend on the characteristics of tissues,and in particular,tissues with dense,complex structure and abundant lipid content are difficult to completely *** study enables future research on the development of methods and treatments for fabricating bioscaffolds via decellularization of complex and rigid skin tissues,which are not com.only considered for decellularization to date as their structural and functional characteristics could not be preserved after severe *** this study,decellularization of human dermal tissue was done by a combination of both chemical(0.05%trypsin-EDTA,2%SDS and 1%Triton X-100)and physical methods(electroporation and sonication).After decellularization,the content of DNA remaining in the tissue was quantitatively confirmed,and the structural change of the tissue and the retention and distribution of ECm com.onents were evaluated through histological and histochemical analysis,*** of the chemical pretreatment that increase the efficiency of physical stimulation as well as decellularization,and conditions for electroporation and sonication without the use of detergents,unlike the methods performed in previous studies,were established to enable the complete decellularization of the skin *** combinatorial decellularization treatment formed micropores in the lipid bilayers of the skin tissues while removing all cell and cellular residues without affecting the ECm ***,this procedure can be widely used to fabricate bioscaffolds by decellularizing biological tissues with dense and complex structures.

Dear Editor,Somatic cells can be reprogrammed into pluripotent stem cells,called induced pluripotent stem cells(iPSCs),by defined transcription factors(Takahashi et al.,2006).The reprogramming of somatic cells may be a continuous stochastic process in which nearly all somatic donor cells have the ability to give rise to iPSCs with continuous passaging and the expression of defined factors(Hanna et al.,2009).However,the reprogramming process can be divided into discrete stages,including the generation of dedifferentiated pre-pluripotent cells followed by authentic pluripotent cells(Silva et al.,2009).

Sojucktang (SJT) has long been used for the treatment of endometrial diseases in Korea. However, the mechanisms responsible for the SJT-induced apoptosis in endometrial cancer cells remain unclear. In the present study, SJT was demonstrated to show cytotoxic effect and induce apoptotic cell death via mitochondrial regulation in KLE endometrial cancer cells. Linderae Radix, Glycyrrhizae Radix, Zedoariae Rhizoma, Trogopterorum Faeces and Agelicae Gigantis Radix were found to be the potent constituent herbs of SJT to significantly decrease the viability of KLE cells by a tetra zolium salt (XTT) assay. Apoptotic bodies were observed in SJT-treated KLE cells by 4′-6-diamidino-2-phenylindole (DAPI) and TdT-mediated-dUTP nick-end labeling (TUNEL) assay. SJT also increased sub-G1 DNA contents of the cell cycle undergoing apoptosis in a dose-dependent manner. Furthermore, it was observed that SJT activated caspase-3 and cleaved poly (ADP-ribose) polymerase (PARP), and decreased mitochondrial membrane potential in a dose-dependent manner. Taken together, this study shows that SJT exerts anti-tumor activity against KLE endometrial cancer cells via mitochondrial dependent apoptosis induction.

This study examined whether exposure to a firm＇s report of corporate social responsibility （CSR） activities affected attitudes toward the firm and increased behavioral intent to purchase products and services from it. Findings showed that increased knowledge of a company＇s CSR deeds fosters positive attitudes and favorable purchase intentions toward the company. Reports that compared the firm＇s CSR activities to the competitor＇s activities favorably affected consumers＇ attitudes and purchase intentions positively. On the other hand, self-promotional reports affected neither attitudes nor purchase intentions significantly.

The effects of extrusion temperature on the microstructure and tensile properties of extruded AZ61 and AZ91 alloys are investigated by subjecting them to hot extrusion at 300 and 400℃.Although the average grain size of the extruded AZ61 alloy slightly increases from 9.5 to 12.6μm with increasing extrusion temperature,its resultant microstructural variation is *** contrast,the average grain size of the extruded AZ91 alloy significantly increases from 5.7 to 22.5μm with increasing extrusion temperature,and the type of mg17Al12 precipitates formed in it changes from fine dynamic precipitates with a spherical shape to coarse static precipitates with a lamellar *** the extrusion temperature increases,the tensile yield strength of the extruded AZ61 alloy increases from 183 to 197 mPa while that of the extruded AZ91 alloy decreases from 232 to 224 *** tensile elongations of the extruded AZ61 and AZ91 alloys decrease with increasing extrusion temperature,but the degree of decrease is significant in the latter *** different extrusion temperature dependences of the tensile properties of the extruded AZ61 and AZ91 alloys are discussed in terms of their microstructural characteristics,strengthening mechanisms,and crack initiation sites.

High-risk human papillomaviruses(HPVs) are involved in the development of several human cancers, including oropharyngeal squamous cell carcinomas. However, many studies have demonstrated that HPV alone is not sufficient for the oncogenic transformation of normal human epithelial cells, indicating that additional cofactors are required for the oncogenic conversion of HPV-infected cells. Inasmuch as chronic inflammation is also closely associated with carcinogenesis, we investigated the effect of chronic exposure to tumor necrosis factor α(TNFα), the major proinflammatory cytokine, on oncogenesis in two immortalized oral keratinocyte cell lines, namely, HPV16-immortalized and human telomerase reverse transcriptase(h TERT)-immortalized cells. TNFαtreatment led to the acquisition of malignant growth properties in HPV16-immortalized cells, such as(1) calcium resistance,(2)anchorage independence, and(3) increased cell proliferation in vivo. moreover, TNFα increased the cancer stem cell-like population and stemness phenotype in HPV16-immortalized cells. However, such transforming effects were not observed in h TERTimmortalized cells, suggesting an HPV-specific role in TNFα-promoted oncogenesis. We also generated h TERT-immortalized cells that express HPV16 E6 and E7. Chronic TNFα exposure successfully induced the malignant growth and stemness phenotype in the E6-expressing cells but not in the control and E7-expressing cells. We further demonstrated that HPV16 E6 played a key role in TNFα-induced cancer stemness via suppression of the stemness-inhibiting micro RNAs mi R-203 and mi R-200 c. Overexpression of mi R-203 and mi R-200 c suppressed cancer stemness in TNFα-treated HPV16-immortalized cells. Overall, our study suggests that chronic inflammation promotes cancer stemness in HPV-infected cells, thereby promoting HPV-associated oral carcinogenesis.

Purpose: The purpose of this study was to investigate the effects of obesity and aerobic exercise training on oxidant-antioxidant balance,neurotrophic factor levels, and blood-brain barrier(BBB) ***: Ten non-obese healthy men(body mass index m2) and 10 obese men(body mass index ≥ 25 kg/m2) were included in the *** groups performed treadmill exercise for 40 min 3 times weekly for 8 weeks at 70% heart rate reserve. Blood samples were collected to examine oxidant-antioxidant balance(reactive oxygen species(ROS) and superoxide dismutase(SOD) activity levels), neurotrophic factors(brain-derived neurotrophic factor(BDNF), nerve growth factor, and glial cell line-derived neurotrophic factor levels), and BBB function(S100βand neuron-specific enolase(NSE) levels) before and after exercise ***: The obese group showed significantly greater changes than the non-obese group in serum ROS(-0.46 ± 0.31 mmol/L vs.-0.10 ±0.17 mmol/L,p=0.005),serum S100 p levels(-8.50 ± 5.92 ng/L vs.-0.78 ± 5.45 ng/L,p=0.007),and serum NSE levels(-0.89 ± 0.54 μg/L vs.-0.01 ± 0.74 μg/L,p= 0.007) after training. At baseline,the obese group showed significantly higher serum ROS and S100β levels and significantly lower serum SOD activity and BDNF levels than the non-obese group(p m ROS, S100β,and NSE levels and significantly higher serum SOD activity and BDNF levels after training compared with baseline(p m neurotrophic factor levels and can induce BBB dysfunction. On the other hand,aerobic exercise can improve an oxidant-antioxidant imbalance in obese subjects and limit BBB dysfunction.
