Single-nucleotide editing for zebra3 and wsl5 phenotypes in rice using CRISPR/Cas9-mediated adenine base editors

The CRISPR/Cas9-mediated base editing technology can efficiently generate point mutations in the genome without introducing a double-strand break(DSB)or supplying a DNA donor template for homology-directed repair(HDR).In this study,adenine base editors(ABEs)were used for rapid generation of precise point mutations in two distinct genes,OsWSL5,and OsZEBRA3(Z3),in both rice protoplasts and regenerated *** precisely engineered point mutations were stably inherited to subsequent *** single nucleotide alterations resulted in single amino acid changes and associated wsl5 and z3 phenotypes as evidenced by white stripe leaf and light green/dark green leaf pattern,*** selfing and genetic segregation,transgene-free,base edited wsl5 and z3 mutants were obtained in a short period of *** noticed a novel mutation(V540A)in Z3 locus could also mimic the phenotype of Z3 mutation(S542P).Furthermore,we observed unexpected non-A/G or T/C mutations in the ABE editing window in a few of the edited *** ABE vectors and the method from this study could be used to simultaneously generate point mutations in multiple target genes in a single transformation and serve as a useful base editing tool for crop improvement as well as basic studies in plant biology.

Geodetic Analysis inside the South Korean Peninsula and Impact of the 2011 Tohoku–Oki(TO) Earthquake

The most powerful Tohoku–Oki(TO) earthquake that occurred in Japan on 11 March 2011 affected Japan as well as South Korea. In the current study, we investigated contemporary geodetic deformation inside South Korea before and after the TO earthquake using Global Navigation Satellite System(GNSS) measurements from 01 January 2008 to 31 December 2017. Measured velocities of GNSS sites are modeled by Auto-regressive moving average(ARMA) method to analyze the long-term GNSS time-series variation and to investigate the secular tectonic crustal deformation. We found that the maximum co-seismic displacements during the TO earthquake reached up to 36.82 ± 0.21 mm in the east and 5.90 ± 0.08 mm in the north directions. The geometric model of the co-seismic thrust surface was characterized by a rectangular plane with a dip of 12.0° and strike 200°. The thrust is situated at 25 km hypocenter depth, with an area roughly ~470 km long and ~120 km wide. The seismicity pattern after the earthquake indicated that the compressional strain started to be replaced by the extensional strain during the post TO earthquake period from 2011 to 2014. Further, the strain became predominantly extensional during the period 2015 to 2017, revealing an effective rotational change that occurred inside the Korean Peninsula.

In this paper, first a circular diaphragm is modeled using the classical plate theory. An analytical solution based on differential transformation method （DTM） and Runge-Kutta method is employed for solving the governing differential equation for the first time. Then the influences of various parameters on central deflection of the diaphragm, stress distribution and capacitance of pressure sensor with a time-dependent pressure are examined. Several case studies are compared with simulations to confirm the proposed method. The analytical results compared with ABAQUS simulation show excellent agreement with the simulation results. This method is very promising for time saving in calculating micro-device characteristics.

Neutron stars:a relativistic study

We study the inner structure of a neutron star from a theoretical point of view and the outcome results are compared with observed data. We propose a stiff equation of state relating pressure with matter density. From our study we calculate mass（M）,compactness（u） and surface redshift（Zs） for two binary millisecond pulsars,namely PSR J1614–2230 and PSR J1903＋327,and four X-ray binaries,namely Cen X-3,SMC X-1,Vela X-1 and Her X-1,and compare them with recent observational ***,we examine the stability for such a type of theoretical structure.

Non-Newtonian effect on natural convection flow over cylinder of elliptic cross section

The non-Newtonian effect in the boundary layer flow over a horizontal elliptical cylinder is investigated numerically. A modified power-law viscosity model is used to correlate the non-Newtonian characteristics of the fluid flow. For natural convectionflows, the surface of the cylinder is maintained by the uniform surface temperature(UST)or the uniform heat flux(UHF) condition. The governing equations corresponding to theflow are first transformed into a dimensionless non-similar form using suitable transformations. The resulting equations are solved numerically by an efficient finite difference scheme. The numerical results are presented for the skin friction coefficient and the local Nusselt number with the eccentric angle for different values of the power-law index n. The local skin friction coefficient and the local Nusselt number are found to be higher and lower, respectively, for the shear thickening fluids(n > 1) than the other fluids(n≤1).The effects of different elliptical configurations on the average Nusselt number are also presented and discussed for both conditions of the surface temperature.

Evaluation of fungal potentiality for bioconversion of domestic wastewater sludge

This study was undertaken to screen the filamentous fungi isolated from its relevant habitats(wastewater, sewage sludge and sludge cake) for the bioconversion of domestic wastewater sludge. A total of 35 fungal strains were tested against wastewater sludge (total suspended solids, TSS 1%—5% w/w) to evaluate its potentiality for enhancing the biodegradability and dewaterability using liquid state bioconversion(LSB) process. The strains were divided into five groups i.e. Penicillium, Aspergillus, Trichoderma, Basidiomycete and Miscellaneous, respectively. The strains WWZP1003, SCahmA103, SCahmT105 and PC-9 among their respective groups of Penicillium, Aspergillus, Trichoderma and Basidiomycete played potential roles in terms of separation(formation of pellets/flocs/filaments), biodegradation(removal of COD) and filtration(filterability) of treated domestic wastewater sludge. The Miscellaneous group was not considered due to its unsatisfactory results as compared to the other groups. The pH value was also influenced by the microbial treatment during fermentation process. The filterability of treated sludge was improved by fungal treatment, and lowest filtration time was recorded for the strain WWZP1003 and SCahmA103 of Penicillium and Aspergillus groups respectively compared with other strains.

Unsteady RANS simulation of wind flow around a building shape obstacle

This work aims to find the origin and connection of the surface,near-wake,and far-wake structures in the flow encompassing a high-rise building for a high Reynolds *** origin and interconnection of the stream-wise tip vortices,with the other components of the wake,is analysed in this study for the current *** Unsteady Reynolds Averaged Navier-Stokes equations(URANS)together with the realizable k-εturbulence model have been used in this investigation to study the turbulent wake flow following a ground-surface-attached square shape building.A moderately big obstacle aspect ratio of 4,a Reynolds number of 12,000,and a thin evolving boundary layer thickness have been used in the flow *** designed flow addresses the reversed-flows at the outlet during computation to improve the accuracy of the realizable k-e *** Reynolds stress components are retrieved using the Boussinesq *** wake’s principal compositions,including span-wise-side eddies and area of high stream-wise vorticity in the uppermost portion of the wake,are illustrated by both three-dimensional(3D)representations and planner projections of the mean flow distributions.A braided vortex formation,composed of asymmetric hairpin vortexes,is witnessed in the far-wake *** association of the near-wake vortex structures with the far-wake and near-wall flow,which is associated with the flow strengths,is also *** this investigation,few areas of large stream-wise vorticity magnitude,like tip vortexes,are correlated to the 3D curving of the fluid motion,and tip vortices did not continuously reach to the free end part of the *** 3D fluid motion interpretation,which combined several measurements of the flow distribution encompassing the cylinder,shows that the time-averaged near-wake structures are formed of two segments of distinct source and section of ***,addressing reversed-flow during computation shows notable improvement in the results.

Naturally Occurring Self-Reactive CD4^+CD25^+ Regulatory T Cells: Universal Immune Code

Naturally occurring thymus-arisen CD4^＋CD25^＋ regulatory T （Treg） cells are considered to play a central role in self-tolerance. Precise signals that promote the development of Treg cells remain elusive, but considerable evidence suggests that costimulatory molecules, cytokines, the nature of the TCR and the niche or the context in which the T cell encounters antigen in the thymus play important roles. Analysis of TCR from Treg cells has demonstrated that a large proportion of this population has a higher avidity to self-antigen in comparison with TCR from CD4^＋CD25^＋ cells and that peripheral antigen is required for their development, maintenance, or expansion. Treg cells have been shown to undergo expansion in the periphery, likely regulated by the presence of self-antigen. Many studies have shown that the involvement of Treg cells in the tolerance induction is antigen-specific, even with MHC-mismatched, in transplantation/graft versus host disease （GVHD）, autoimmunity, cancer, and pregnancy. Theses studies concluded a vital role for self-reactive Treg cells in maintenance of the body integrity. Based on those studies, we hypothesize that self-reactive Treg cells are shared among all healthy individuals and recognize same self-antigens and their TCR encodes for few dominant antigens of each organ which defines the healthy self. These dominant self antigens can be regarded as ＂universal immune code＂. Cellular ＆ Molecular Immunology.

Effects of In-situ Generated Coinage Nanometals on Crystallization and Microstructure of Fluorophlogopite Mica Containing Glass-Ceramics

The effects of in-situ generated coinage nanometals（Cu,Ag and Au） on crystallization behavior,microstructure,thermal and mechanical properties of SiO2-MgO-Al2O3-B2O3-K2O-MgF2（BMAPS） glassceramics were systematically *** addition of coinage nanometai,the glass transition temperature（Tg） is increased by 20-30 ℃,crystallization temperature（Tc） by 30-50 ℃ and dilatometric softening temperature（Td） by 10-25 ℃.It was found that the density of Cu-containing glass was 2.59 g cm-3and for other glasses it was in the range of 2.56-2.57 g *** the non-isothermal differential scanning calorimetry study,the activation energy of crystallization for BMAPS base glass was calculated as 344 kJ/mol,and changed to 406,334 and 274 kJ/mol on addition of Cu,Ag and Au-nanometals,*** evolved in the opaque BMAPS glass-ceramics derived by controlled heat treatment,were identified as fluorophlogopite mica(KMg3(AlSi3O10)F2) by X-ray diffraction（XRD） technique and confirmed by Fourier transformed infrared *** of copper,silver and gold nanometals were also identified by XRD *** is found from field emission scanning electron microscopy that the interlocked grain like microstructure developed in BMAPS glass-ceramics（being heat-treated at 1050 ℃ for 4 h） changed to denser house-of-cards like microstructure（containing smaller sized mica crystals） on addition of coinage *** of BMAPS base glass-ceramic was 2.60 g cm-3and marginally changed to 2.61-2.62 g cm-3on addition of Cu,Ag and *** change in microstructure resulted in the decrease of Vickers micro hardness value from 5.37 to 4.12,4.20 and 4.58 GPa on addition of Cu,Ag and Au,*** nanometai doped mica glass-ceramics containing interlocked microstructure with higher thermal expansion coefficient,hence,is suitable for high temperature sealing application（like solid oxide fuel cell）.
