Analysis of the Effects of Different Synthesis and Processing Methods on Circular RNA Integrity,Protein Expression,and Removal of Immunogenic Impurities

Circular RNAs(circRNAs)are emerging as a promising alternative to messenger RNAs(mRNAs)in gene delivery applications due to their enhanced stability and *** circRNA-based therapeutic platforms requires efficient manufacturing of circRNA with broad ***,the permuted intron-exon(PIE)-based circRNA production commonly used to date involves complex RNA synthesis,circularization,precursor RNA digestion,and impurity removal steps that have limited practical *** co-transcriptional circularization could effectively streamline circRNA production,and both cellulose/phosphatase treatment and high-performance liquid chromatography(HPLC)have demonstrated their reliability in mRNA manufacturing,their potential effects on the quality,translation,and reactogenicity of circRNA remained to be fully ***,using circRNAs systematically manufactured through three independent workflows,we comprehensively examined the utilities of these RNA synthesis and processing methods in circRNA production by comparing the integrity,translation,and immunogenicity of their circRNA *** began by manufacturing a mNeonGreen(mNG)-encoding circRNA through these workflows and subsequently assessed circRNA integrity via E-gel EX *** expression was then monitored in HEK 293T,A549,and DC2.4 cells at 72 hours ***,we evaluated the immunogenicity of these circRNAs by measuring their interferon beta(IFN-β)induction in A549 cells at 4 hours *** HPLC purification over cellulose and phosphatase treatment resulted in 10-14%higher circRNA enrichment by reducing nicking associated with processing *** expression remained consistent across circRNAs from different workflows(P>0.05),demonstrating that co-transcriptional circularization produces circRNA with translation levels comparable to those obtained from the conventional PIE ***,both cellulose/phosphatase treatment

Hepatocellular carcinoma （HCC） metastasizes to the mandible is infrequently seen. Solitary bony metastasis to the mandible is rarer. The intractable bleeding caused by rupture of the metastatic HCC is challenging to clinicians. We present a case of a 74-year-old woman with HCC under control without progression for 3 years. Left facial swelling and episodes of bleeding developed recently and biopsy revealed a metastatic HCC. Computer tomography showed a large tumor in parapharyngeal space with evident mandibular ramus destruction. Bleeding occurred from the metastatic tumor but could not be controlled by electrocauterization, SurgicelTM, tissue glue, and bone wax and angiographic embolization. Palliative radiotherapy （2400 cGy in 6 fractions） was tried and the intractable bleeding was successfully stopped after the radiotherapy. Because of the hypervascular and osteolytic nature of the solitary mandibular metastatic lesion, the bleeding was troublesome. Radiotherapy provided successful control of intractable bleeding from the metastatic tumor.

Effect of UV-ozone treatment on the performance of isopropanol dehydration by pervaporation through the PDMS membranes was investigated.X-ray photoelectron spectrometer（XPS）and water contact angle were used to characterize the chemical compositions and hydrophilicity of the surface layer of the modified PDMS *** was found that the O/Si atomic ratio of PDMS surface layer increased with increasing UV-ozone treatment time,but C/Si atomic ratio decreased with treatment *** formation of silica like surface layer improved the selectivity of PDMS membrane.

Flexible optoelectronics that can be bent,wrapped,and stretched have attracted interest for wearable and mobile *** this work,we demonstrate a transparent 360°omnidirectional photodetector(PD)that can be stretched and wrapped around flexible or curved *** embedding interlaced ZnO and Ag nanowires(NWs)in thermoplastic polyurethane via inkjet printing,the device featured>75%transmittance in the visible region,showing high photoresponsivity and response time(10-30 A/W and 0.8 s,respectively).Moreover,the flexible PD performs well under deformation(only 9%decay in the photocurrent under 60%strain and 8%loss when the device is bent at a radius of 5 mm),which allows it to be readily applied on curved surfaces,such as skin or optical *** study opens the door for the development of flexible optoelectronics that could be implemented in fiber optics,wearable electronics,self-powered systems,bio-signal monitors,and epidermal electronics.

Lymphoepithelioma-like cholangiocarcinoma:A mimic of hepatocellular carcinoma on imaging features

Primary lymphoepithelioma-like carcinoma in the liver is extremely rare. A few cases of lymphoepithelioma-like cholangiocarcinoma have been reported, but few radiologic features were described. We reviewed 23 cases of lymphoepithelioma-like cholangiocarcinoma reported between 1996 and 2014 and describe a rare case of a 35-year-old woman in our hospital who was diagnosed with lymphoepithelioma-like cholangiocarcinoma of the liver and was a hepatitis B carrier. The tumor(1.6 cm) in our patient appeared to be hypoechoic in sonographic images and hypodense in computed tomography(CT) images. In addition, it was homogeneous hypointense in T1-weighted magnetic resonance(MR) images(MRI) and hyperintense in T2-weighted MRI. Dynamic gadolinium-enhanced MRI showed typical image pattern of hepatocellular carcinoma(HCC). The patient underwent a laparoscopic left hepatic lobectomy, and the resected tumor consisted of well-differentiated glandular cells with extensive lymphocytic infiltration that were immunoreactive to CK(AE1/AE3), CD3, and CD20. In addition, the tumor was positive for Epstein-Barr virus-encoded RNA in situ hybridization. Finally, lymphoepithelioma-like cholangiocarcinoma was diagnosed. In previous studies, the incidence is highest among middle-aged people. Most tumors appeared to be hypodense with either hypovascular or hypervascular patterns in CT images. This case report is the first study to address sonography, CT, and MRI observations and delineate pathologic correlations. We suggest that the imaging pattern of lymphoepithelioma-like cholangiocarcinoma, either the typical cholangiocarcinoma pattern or a mimic of HCC, should be considered in the differential lists for HCC.

目的评估“病人关怀与实践课程”结束后学生体验的效果。方法课程活动包括由专业辅导员带领辅导团体进行同理心体验,尔后让学生至附设医院陪伴病人及家属以及陪伴后分享相关经验三部分。参加学生为2005年入学的医学系一年级新生118人。结果学生回馈数据显示,经由课程设计之团体辅导及陪伴病人后,学生更能体验何谓同理心,并能体会到陪伴病人的正向情绪经验。结论学生参加“病人关怀与实践课程”,能使学生在正式接触医学专业之前,更了解如何同理病人、帮助别人,进而提升医学生的人文关怀。
