Cerebral Palsy and Stroke—Early and Late Brain Lesion Present Differences in Systemic Biomarkers and Gene Expression Related to Muscle Contractures

Background: CNS lesions that are acquired early in life e.g. cerebral palsy (CP) disturb muscle development and growth, while CNS injuries acquired later in life e.g. stroke, affect fully matured muscles and cause paresis and atrophy. These differences may result in different contracture phenotypes. Aim: The purpose of this study was to compare systemic biomarkers and gene expression levels in muscle of individuals with CNS lesions acquired early and later in life. Methods: Blood samples and muscle biopsies were analyzed using Enzyme-linked immunosorbent assay and Real-time PCR from n = 24 control participants, n = 14 individuals with cerebral palsy, and n = 12 stroke survivors. Results: Systemic markers: Myostatin was significantly decreased in both the cerebral palsy (p = 0.0051), and the stroke group (p = 0.036). Creatine Kinase-MB and C-Reactive Protein were significantly elevated in stroke patients only (p 0.034 respectively). Gene expressions: The expression of myostatin (MSTN) was significantly lower in both the ST and the CP group when compared to Ctrl (p = 0.02). In addition, collagen type 4A1 (COL4A1) was significantly lower in the CP group compared to the other groups (p = 0.015). Finally, the troponin 1 slow skeletal muscle type was significantly increased in the ST group when compared to both CP and Ctrl (p = 0.03). Conclusion: The downregulation of myostatin in individuals with both early and late CNS injury is likely a compensatory reaction to muscle weakness, reduced muscle mass and/or muscle atrophy. Changes in gene expression may reflect a specific alteration depending on when in life the CNS lesions were acquired.

Sortilins in the blood-brain barrier:impact on barrier integrity

Sortilin was discovered in 1997 by petersen and his *** single transmembrane receptor was isolated as a 95 kDa membrane glycoprotein by receptor-associated protein affinity chromatography and therefore initially named *** cDNA library screening revealed sequence homology between the luminal segment of gp95/sortilin and the yeast vacuolar sorting protein 10 protein(Vps10p)domain(petersen et al.,1997).The Vps10p domain was first identified in the Saccharomyces cerevisiae protein Vps10p that directs lysosomal enzymes from the Golgi to the vacuole(Marcusson et al.,1994).

Cytokine responses in infants infected with respiratory syncytial virus

Introduction: Variability in severity of Respiratory Syncytial Virus (RSV) infection is reportedly due to differences in inflammatory response. Objective: To characterize the cytokine response in RSV+ infants aged 0 - 36 months and to relate their responses to disease severity. Methods: Nasopharyngeal aspirations (NPAs) were analyzed for RSV and IL-1β, IL-2, IL-4, IL-5, IL-6, IL-10, IL-12, IL-1RA, IL-4R, IFN-γ, sTNFR1, sTNFR2, and TNF-α. Clinical data were collected from the medical records. Results: We included 331 infants of whom 214 were RSV+. In comparison to RSV- infants, they had significantly higher levels of TNF-α, IL-6, IL-1β, and IFN-γ (p α, IL-6, and IL-1β. sTNFR1/2 were significantly increased in RSV+ infants. Hospitalized patients had significantly higher levels of TNF-α, sTNFR2, and IL-10 (p < 0.05) than non-hospitalized patients. The cytokine response could not be related to disease severity. We found no evidence of a skewed Th1/Th2 immune profile. Conclusion: In acute RSV disease, infected infants’ NPAs contain a significant amount of pro-inflammatory cytokines. Whether this response is beneficial or deleterious remains unanswered. Interpersonal variations in cytokine responses might be linked to an inherited tendency to variations in disease severity.

DSDP 524区中生代最晚期和第三纪最早期的沉积物中,保存着白垩纪末期环境变化和生物演化的详细纪录。我们对该孔进行了加密取样,样品间距大致相当于100年或1000年的时间间隔,同时对样品进行了碳酸盐含量、微量元素成分及碳氧同位素分析。我们所得到的数据可以解释为白垩纪末期生物大批死亡的结果,当时大量远洋地区的生物扫地以尽。养料锐减导致部分从河流带来的溶解CO_2转移到大气圈,致使在长达40,000年的时间内,空气中的CO_2剧增。由此产生的温室效应就是氧同位素所揭示的温度升高的原因。第三纪最早期沉积物中,包括来自“白垩纪生物群”中的化石,有的已经过再沉积作用,有的则是这场灾难的最后幸存者。我们的资料表明,大量远洋生物的灭绝是在第三纪最早期的不利环境中发生的。而这种异常的环境则是生物大批死亡的结果,且持续有几千年之久。恐龙就是因为温度灾难性升高产生的热异常而灭绝的。大洋中生物大批灭绝的原因,与巨大的天外星体的陨落有关,界线附近的铱异常即其标志,我们的资料与许靖华不久前发表的彗星冲击说是不抵触的。

The identification of jets originating from quarks and gluons,often referred to as quark/gluon tagging,plays an important role in various analyses performed at the Large Hadron Collider,as Standard Model measurements and searches for new particles decaying to quarks often rely on suppressing a large gluon-induced *** paper describes the measurement of the efficiencies of quark/gluon taggers developed within the ATLAS Collaboration,using √s=13 TeV proton–proton collision data with an integrated luminosity of 140 fb^(-1) collected by the ATLAS *** taggers with high performances in rejecting jets from gluon over jets from quarks are studied:one tagger is based on requirements on the number of inner-detector tracks associated with the jet,and the other combines several jet substructure observables using a boosted decision tree.A method is established to determine the quark/gluon fraction in data,by using quark/gluon-enriched subsamples defined by the jet *** in tagging efficiency between data and simulation are provided for jets with transverse momentum between 500 GeV and 2 TeV and for multiple tagger working points.
