Effects of plant growth regulators, carbon sources and pH values on callus induction in Aquilaria malaccensis leaf explants and characteristics of the resultant calli

The endangered tropical tree, Aquilaria malaccensis, produces agarwood for use in fragrance and medicines. Efforts are currently un-derway to produce valuable agarwood compoundsn tissue culture. The purpose of this study was to develop an optimal growth medium, specif-ically, the best hormone combination for callus suspension culture. Using nursery-grown A. malaccensis, sterilized leaf explants were first incu-bated on basic Murashige and Skoog （MS） gel medium containing 15g/L sucrose and at pH 5.7. Different auxin types including 1-naphthaleneacetic acid （NAA）, 2,4-dichlorophenoxyacetic acid （2,4-D）, and indole-3-butyric acid （IBA）, were tested at various concentrations （0.55, 1.1 and 1.65 μM） using the basic medium. Leaf explants were incubated for 30 days in the dark. Callus induced by 1.1 μM NAA had the highest biomass dry weight （DW） of 17.3 mg;however the callus was of a compact type. This auxin concentration was then combined with either 6-benzylaminopurine （BAP） or kinetin at 0.55, 1.1, 2.2 or 3.3 μM to induce growth of friable callus. The 1.1μM NAA＋2.2μM BAP com-bination produced friable callus with the highest biomass （93.3mg DW）. When testing the different carbon sources and pHs, sucrose at 15g/L and pH at 5.7 yielded highest biomasses at 87.7mg and 83mg DW, respec-tively. Microscopic observations revealed the arrangement of the friable cells as loosely packed with relatively large cells, while for the compact callus, the cells were small and densely packed. We concluded that MS medium containing 15 g/L sucrose, 1.1 μM NAA ＋ 2.2 μM BAP hor-mone combination, and a pH of 5.7 was highly effective for inducing friable callus from leaf explants of A. malaccensis for the purpose of establishing cell suspension culture.

Forecasting the Municipal Solid Waste Using GSO-XGBoost Model

Waste production rises in tandem with population growth and increased *** indecorous disposal of waste paves the way for huge disaster named as climate *** National Environment Agency(NEA)of Singapore oversees the sustainable management of waste across the *** three main contributors to the solid waste of Singapore are paper and cardboard(P&C),plastic,and food ***,they have a negligible rate of *** this study,Machine Learning techniques were utilized to forecast the amount of garbage also known as waste *** waste audit would aid the authorities to plan their waste *** applied models were k-nearest neighbors,Support Vector Regressor,ExtraTrees,CatBoost,and *** XGBoost model with its default parameters performed better with a lower Mean Absolute Percentage Error(MAPE)of 8.3093(P&C waste),8.3217(plastic waste),and 6.9495(food waste).However,Grid Search Optimization(GSO)was used to enhance the parameters of the XGBoost model,increasing its ***,the optimized XGBoost algorithm performs the best for P&C,plastics,and food waste with MAPE of 4.9349,6.7967,and 5.9626,*** proposed GSO-XGBoost model yields better results than the other employed models in predicting municipal solid waste.

Home-based Detection and Prediction of Diabetic Foot Ulcers at Early Stage Using Sensor Technology and Supervised Learning

For years,foot ulcers linked with diabetes mellitus and neuropathy have significantly impacted diabetic patients’ health-related quality of life(HRQoL). Diabetes foot ulcers impact15% of all diabetic patients at some point in their lives. The facilities and resources used for DFU detection and treatment are only available at hospitals and clinics,which results in the unavailability of feasible and timely detection at an early stage. This necessitates the development of an at-home DFU detection system that enables timely predictions and seamless communication with users,thereby preventing amputations due to neglect and severity. This paper proposes a feasible system consisting of three major modules:an IoT device that works to sense foot nodes to send vibrations onto a foot sole,a machine learning model based on supervised learning which predicts the level of severity of the DFU using four different classification techniques including XGBoost,K-SVM,Random Forest,and Decision tree,and a mobile application that acts as an interface between the sensors and the patient. Based on the severity levels,necessary steps for prevention,treatment,and medications are recommended via the application.

Pulsed laser interference patterning of transition-metal carbides for stable alkaline water electrolysis kinetics

We investigated the role of metal atomization and solvent decomposition into reductive species and carbon clusters in the phase formation of transition-metal carbides(TMCs;namely,Co_(3)C,Fe_(3)C,TiC,and MoC)by pulsed laser ablation of Co,Fe,Ti,and Mo metals in *** interaction between carbon s-p-orbitals and metal d-orbitals causes a redistribution of valence structure through charge transfer,leading to the formation of surface defects as observed by X-ray photoelectron *** defects influence the evolved TMCs,making them effective for hydrogen and oxygen evolution reactions(HER and OER)in an alkaline ***_(3)C with more oxygen affinity promoted CoO(OH)intermediates,and the electrochemical surface oxidation to Co_(3)O_(4)was captured via in situ/operando electrochemical Raman probes,increasing the number of active sites for OER *** with more d-vacancies exhibits strong hydrogen binding,promoting HER kinetics,whereas Fe_(3)C and TiC with more defect states to trap charge carriers may hinder both OER and HER *** results show that the assembled membrane-less electrolyzer with Co_(3)C∥Co_(3)C and MoC∥MoC electrodes requires~2.01 and 1.99 V,respectively,to deliver a 10 mA cm−2 with excellent electrochemical and structural *** addition,the ascertained pulsed laser synthesis mechanism and unit-cell packing relations will open up sustainable pathways for obtaining highly stable electrocatalysts for electrolyzers.

Accurate quantitation standards of glutathione via traceable sulfur measurement by inductively coupled plasma optical emission spectrometry and ion chromatography

The quantitative analysis of glutathione （GSH） is important in different fields like medicine, biology, and biotechnology. Accurate quantitative measurements of this analyte have been hampered by the lack of well characterized reference standards. The proposed procedure is intended to provide an accurate and definitive method for the quantitation of GSH for reference measurements. Measurement of the stoichiometrically existing sulfur content in purified GSH offers an approach for its quantitation and calibration through an appropriate characterized reference material （CRM） for sulfur would provide a methodology for the certification of GSH quantity, that is traceable to SI （International system of units）. The inductively coupled plasma optical emission spectrometry （ICP-OES） approach negates the need for any sample digestion. The sulfur content of the purified GSH is quantitatively converted into sulfate ions by microwave-assisted UV digestion in the presence of hydrogen peroxide prior to ion chromatography （IC） measurements. The measurement of sulfur by ICP-OES and IC （as sulfate） using the ＂high performance＂ methodology could be useful for characterizing primary calibration standards and certified reference materials with low uncertainties. The relative expanded uncertainties （% U） expressed at 95% confidence interval for ICP-OES analyses varied from 0.1% to 0.3%, while in the case of IC, they were between 0.2% and 1.2%. The described methods are more suitable for characterizing primary calibration standards and certifying reference materials of GSH, than for routine measurements.

Effects of derived meals from juncea(Brassica juncea),yellow and black seeded canola(Brassica napus)and multicarbohydrase enzymes supplementation on apparent metabolizable energy in broiler chickens

Two experiments were conducted to determine the nitrogen-corrected apparent metabolizable energy(AMEn) of differently processed meals from Juncea(Brassica juncea), yellow and black seeded canola(Brassica napus), with or without supplementation of multi-carbohydrase enzymes(Enz) in diets for broiler chickens. The first experiment was a 3×2×2 factorial arrangement with the main factors being seed type(yellow [Yellow] or black [B1] canola seeds and Juncea seeds), processed at two temperatures(high temperature desolventized-toasted [HTDT] at 95℃ or low temperature desolventized-toasted[LTDT] at 57℃), with or without Enz. In Exp. 1, a total of 384 one-day-old male broiler chicks were randomly assigned to 64 battery cages, with 6 birds/cage. The second experiment was a 2×2×2 factorial arrangement with the main factors being seed type(Yellow or black [B2]), seed source(Scott,Saskatchewan or Truro, Nova Scotia) and Enz(with or without) supplementation. A total of 264 one-dayold male broiler chicks were randomly assigned to 44 battery cages, with 6 birds per cage. In Exp, 1 and 2,all birds were fed a common starter diet from 1 to 14 days of age. From d 15 to 21, the birds were fed one of the test treatments, a basal grower diet or the basal grower diet replaced with 30% test ingredient with celite(0.8%) added as an inert marker. Excreta was collected on d 20 and 21. In Exp. 1, there were no interactions(P > 0.05) among seed type, processing temperature and Enz. Processing temperature and dietary Enz did not affect(P > 0.05) AMEn of different canola meals. The AMEn of prepress solvent extracted canola and juncea meals(PSEM) from Yellow(11.2 MJ/kg) was higher(P 0.05) among seed color,location and Enz. Effects of dietary threonine:lysine ratioes and sanitary conditions on performance, plasma urea nitrogen, plasma-free threonine and lysine of weaned pigs

Two 21 d-experiments were conducted to determine the optimum standardized ileal digestible(SID)threonine:lysine ratio(Thr:Lys) for weaned piglets reared under clean(Exp. 1) or unclean(Exp. 2) sanitary conditions and fed antibiotic-free diets. In each experiment, 90 mixed-sex pigs(Duroc × [Yorkshire × Landrace]; initial BW 7.2 ± 0.3 kg) were randomly assigned to 5 dietary treatments each with 6 replicates(3 pigs per pen). The dietary treatments were 5 graded levels of SID Thr:Lys(55,59, 63, 67 and 71%). Diets were corn-wheat-soybean meal-based with a constant SID Lys of 1.18% that was set to be second limiting amino acid. In Exp. 1 and Exp. 2, plasma-free Thr increased(P = 0.05) with increasing dietary SID Thr:Lys. In Exp. 1, the SID Thr:Lys for gain-to-feed ratio(G:F) was optimized at 65%.In Exp. 2, the estimated optimal SID Thr:Lys for overall G:F was 66.5%. In conclusion, an average optimal SID Thr:Lys of 65 and 66.5% could be used to optimize feed efficiency for weaned pigs under clean and unclean sanitary conditions, respectively.

Electrokinetic-mechanism of water and furfural oxidation on pulsed laser-interlaced Cu_(2)O and CoO on nickel foam

The electrocatalytic oxidation of biomass-derived furfural(FF)feedstocks into 2-furoic acid(FA)holds immense industrial potential in optics,cosmetics,polymers,and ***,we fabricated Co O/Ni O/nickel foam(NF)and Cu_(2)O/Ni O/NF electrodes via in situ pulsed laser irradiation in liquids(PLIL)for the bifunctional electrocatalysis of oxygen evolution reaction(OER)and furfural oxidation reaction(FOR),*** oxidation of NF surface to NiO and deposition of CoO and/or Cu_(2)O on NF during PLIL offer distinct advantages for enhancing both the OER and ***/NiO/NF electrocatalyst provides a consistently low overpotential of~359 m V(OER)at 10 m A/cm^(2),achieving the maximum FA yield(~16.37 m M)with 61.5%selectivity,79.5%carbon balance,and a remarkable Faradaic efficiency of~90.1%during 2 h of FOR at 1.43 V(*** hydrogen electrode).Mechanistic pathway via in situ electrochemical-Raman spectroscopy on CoO/NiO/NF reveals the involvement of phase transition intermediates(NiOOH and CoOOH)as surface-active centers during electrochemical *** carbonyl carbon in FF is attacked by hydroxyl groups to form unstable hydrates that subsequently undergo further oxidation to yield FA *** method holds promise for large-scale applications,enabling simultaneous production of renewable building materials and fuel.
