Conserved arginine residue in the membrane-spanning domain of HIV-1 gp41 is required for efficient membrane fusion

Despite the high mutation rate of HIV-1,the amino acid sequences of the membrane-spanning domain(MSD)of HIV-1 gp41 are well *** residues are rarely found in single membrane-spanning domains,yet an arginine residue,R696(the numbering is based on that of HXB2),is highly conserved in HIV-1 *** examine the role of R696,it was mutated to K,A,I,L,D,E,n,and *** of these substitutions did not affect the expression,processing or surface distribution of the envelope protein(Env).However,a syncytia formation assay showed that the substitution of R696 with amino acid residues other than K,a naturally observed mutation in the gp41 MSD,decreased fusion *** with hydrophobic amino acid residues(A,I,and L)resulted in a modest decrease,while substitution with D or E,potentially negatively-charged residues,almost abolished the syncytia *** the fusion-defective mutants showed slower kinetics with the cell-based dual split protein(DSP)assay that scores the degree of membrane fusion based on pore formation between fusing ***,the D and E substitutions did show some fusion activity in the DSP assays,suggesting that proteins containing D or E substitutions retained some fusion pore-forming ***,nascent pores failed to develop,due probably to impaired activity in the pore enlargement *** data show the importance of this conserved arginine residue for efficient membrane fusion.

Solid-State Interfacial Reaction between Eutectic Au-Ge Solder and Cu/ni(P)/Au Metalized Ceramic Substrate and Its Suppression

Eutectic Au-12 Ge solder was employed to bond the SiC power devices to a Si3n4/Cu/ni（P）/Au multilayered *** high-temperature reliability of the bond was investigated in detail at 200,250,300 and 330 ℃,*** and ni5Ge3intermetallic compounds（IMCs） were identified at the Au-12Ge/ni（P） interface by micro X-ray diffraction（μXRD） and scanning electron microscopy（SEM）equipped with energy dispersive X-ray analysis（EDX）.The growth of the ni-Ge IMCs was dominated by ni5Ge3layer,which formed at the ni5Ge3/niGe interface by outward diffusion of ni from the ni（P） *** activation energy of the total ni-Ge IMCs growth was 66 kJ/*** shear strength of the bond was tested at both 25 ℃ and the aging temperatures,*** shear strength decreased slightly after aging at 200 and 250 ℃.The shear strength tested at 250 ℃ was 46 MPa after aging at 250 ℃ for 3000 *** shear strength aged at 300 and 330 ℃ rapidly decreased with aging time due to the rapid growth of the *** slow down the interfacial reaction between the high temperature solder and the ni（P）layer,an approximately 200 nm-thick Ta/Tan/Ta new diffusion barrier（DB） was deposited on the *** by transmission electron microscopy（TEM） equipped with EDX,reveals that the Ta/Tan/Ta DB was bonded well to the ni（P） layer and the *** temperature storage test at 330 ℃ for1500 h reveals that the bond of the SiC devices maintained its high shear strength of approximately56 MPa without *** new DB effectively suppressed the interfacial reaction between the Au-12 Ge solder and the ni（P） layer of the substrate.

Suppressive Effect of Brassica juncea on the Root-Lesion nematode （ Pratylenchus penetrans）

Incorporation of sulfur-rich crucifer tissues into soil is known to suppress a variety of soil-bome plant pathogens and pests. The potentials of using Brassica juncea as green manure to kill the root lesion nematode Pratylenchus penetrans m soil and to decrease damages to subsequent crops were assessed in field and pot experiments. In the first trial, green manures containing B. juncea were grown and incorporated during spring to summer 2009. Japanese radish was then cultivated in each plot. In the second trial in spring 2010, green manure was grown and incorporated during summer to autumn 2009, and greater burdock was cultivated in pots containing soil sampled from each plot. neither trial showed clear effects on nematode populations in the soil. However, in the first trial, Japanese radish grown following a B. juncea breeding line with a high content of sinigrin had a lower root lesion index and a higher number of marketable taproots than grown in the fallow soil. In the second trail, greater burdock cultivated in pots following incorporation ofB. juncea had a lower root lesion index with the incorporation of white mustard, which is widely used as a landscape plant. These findings suggest that B. juncea used as green manure can potentially decrease damage to subsequent crops caused by the root-lesion nematode, although it had no positive effect on decreasing populations of the root-lesion nematode in the soil.

Autophagy is an intracellular degradation mechanism involved in antiviral defense,but the strategies employed by plant viruses to counteract autophagy-related defense remain unknown for the majority of the ***,we describe how the Chinese wheat mosaic virus(CWMV,genus Furovirus)interferes with autophagy and enhances its infection in nicotiana *** two-hybrid screening and in vivo/in vitro assays revealed that the 19 k Da coat protein(CP19 K)of CWMV interacts with cytosolic glyceraldehyde-3-phosphate dehydrogenases(GAPCs),negative regulators of autophagy,which bind autophagy-related protein 3(ATG3),a key factor in ***19 K also directly interacts with ATG3,possibly leading to the formation of a CP19 K–GAPC–ATG3 ***19 K–GAPC interaction appeared to intensify CP19 K–ATG3 ***,CP19 K expression upregulated GAPC gene transcripts and reduced autophagic ***,the silencing of GAPC genes in transgenic *** reduced CWMV accumulation,whereas CP19 K overexpression enhanced ***,our results suggest that CWMV CP19 K interferes with autophagy through the promotion and utilization of the GAPC role as a negative regulator of autophagy.

Silicon nitride(Si_(3)n_(4))based ceramics are one of the most attractive advanced engineering materials,which have been widely used under high-speed rotational operation or for mechanical contacts across a curved *** the present study,rotationally symmetric texturing of Si_(3)n_(4),with radial grain align-ment,was obtained by centripetal sinter-forging(CSF)of a partially sintered *** average values of the included angles between the c-axis of the local Si_(3)n_(4)grain and radial direction were approxi-mately 16.4°and 11.0°,on the section plane perpendicular to the pressing direction,and parallel to both the pressing and radial directions,*** compressive strain in the pressing direction forced the ceramic body to flow towards the central axis,resulting in compressive strain in the tangential di-rection and tensile strain in the radial direction.A fundamental physical model was created to simulate the grain rotation during the 3-dimentional strain reorientation,which revealed the rod-like grain would preferentially rotate toward the center of the sample under the CSF *** addition,due to the fric-tion between the sample surface and the pressing punch,the increased shear strain could enhance the Si_(3)n_(4)grain ***,ceramics with rod-like grains perpendicular to the curved side sur-face could be anticipated by applying the centripetal forming concept in a controlled manner.

Incorporating nitrogen(n)atom in graphene is considered a key technique for tuning its electrical ***,this is still a great challenge,and it is unclear how to build n-graphene with desired nitrogen *** is a lack of experimental evidence to explain the influence and mechanism of structural defects for nitrogen incorporation into graphene compared to the derived DFT ***,this gap is bridged through a systematic study of different nitrogen-containing gaseous plasma post-treatments on graphene nanowalls(CnWs)to produce n-CnWs with incorporated and substituted *** structural and morphological analyses describe a remarkable difference in the plasma–surface interaction,nitrogen concentration and nitrogen incorporation mechanism in CnWs by using different nitrogen-containing *** conductivity measurements revealed that the conductivity of the n-graphene is strongly influenced by the position and concentration of C–n bonding *** findings open up a new pathway for the synthesis of n-graphene using plasma post-treatment to control the concentration and configuration of incorporated nitrogen for application-specific properties.

Effect of an extract of Ganoderma lucidum in men with lower urinary tract symptoms： a double-blind, placebo-controlled randomized and dose-ranging study

Aim： To conduct a double-blind, placebo-controlled randomized and dose-ranging study to evaluate the safety and efficacy of the extract of Ganoderma lucidum （G. lucidum） in men with lower urinary tract symptoms （LUTS）. Methods： We enrolled male volunteers （〉 50 years） with an International Prostate Symptom Score （IPSS; questions 1-7）〉 5 and a prostate-specific antigen （PSA） value 〈 4 ng/mL. Volunteers were randomized into groups of placebo （n = 12）, G. lucidum of 0.6 mg （n = 12）, 6 mg （n = 12） or 60 mg （n = 14）, administered once daily. Efficacy was measured as a change from baseline in IPSS and the peak urine flow rate （Qmax）. Prostate volume and residual urine were estimated by ultrasonography, and blood tests, including PSA levels, were measured at baseline and at the end of the treatment. Results： The overall administration was well tolerated, with no major adverse effects. Statistical significances in the magnitude of changes between the experimental groups were observed at weeks 4 and 8. no changes were observed with respect to Qmax, residual urine, prostate volume or PSA levels. Conclusion： The extract of G. lucidum was well tolerated and an improvement in IPSS was observed. The recommended dose of the extract of G. lucidum is 6 mg in men with LUTS.
