Acorn squash(Cucurbita pepo)is an iconic fall vegetable in the United States,known for its unique fruit shape and also prized for its culinary *** is known about the metabolism that underlies the development of fruit quality attributes such as color,sweetness,texture and nutritional qualities in acorn squash,or any other winter squash grown *** provide insight into winter squash fruit and seed development and add to the genomic resources in the Cucurbita genus,RNA sequencing was used to generate an acorn squash fruit and seed transcriptome from the cultivar Sweet REBA at critical points throughout fruit development.141838600 high-quality paired-end Illumina reads were assembled into 55949 unigenes.85%of unigenes with predicted open reading frames had homology with previously identified genes and over 62%could be functionally *** with the watermelon and cucumber genomes provided confirmation that the unigenes are full-length and comprehensive,covering an average of 90%of the coding sequence of their homologs and 72%of the cucumber and watermelon *** candidate genes associated with carotenoid and carbohydrate metabolism were identified toward a resource for winter squash fruit quality trait *** transcriptome represents a major advance in *** genomics,providing significant new sequence information and revealing the repertoire of genes expressed throughout winter squash fruit and seed *** studies on the genetic basis of fruit quality and future breeding efforts will be enhanced by tools and insights developed from this resource.

目的　了解跨膜蛋白 β1整合素对信号分子G蛋白细胞内构型的影响 ,为进一步研究 β1整合素在信号传导中的作用打下基础 .方法　采用荧光双标记及重叠合显微镜分析技术 ,观察野生型胚胎干细胞系 (D3细胞 ) ,β1整合素基因敲除胚胎干细胞 (G2 0 1细胞 ) ,β1整合素基因敲除后再植入胚胎干细胞 (G2 0 1N)及小鼠胚胎心肌细胞中多种G蛋白 (Gαs,Gαi1-3 ,Gαo,Gαi/o/t/z)细胞内构型特点和差异 .结果　Gαs,Gαo,Gαi/o/t/z在上述几种细胞系中均呈弥漫性分布 ,未能发现明显差异 ,与此不同的是Gαi1-3 在D3细胞源性心肌细胞内呈网状分布 ,在非心肌细胞内呈线条状分布 ,而在G2 0 1细胞中Gi 上述特异性构型被打乱 ,呈近似于弥漫样分布 ,值得注意的是在β1整合素基因重新恢复的G2 0 1N细胞Gi构型又呈网样或线条样分布 ,与野生型胚胎干细胞中Gi 的构型相一致 .结论　β1整合素能够影响信号分子Gi蛋白的细胞内定位 ,推测β1整合素可通过改变Gi 的分布而影响Gi

The composition of the gut microbiota is linked to multiple diseases,including Parkinson’s disease(PD).Abundance of bacteria producing short-chain fatty acids(SCFAs)and fecal SCFA concentrations are reduced in *** exert various beneficial functions in *** the interventional,monocentric,open-label clinical trial “Effects of Resistant Starch on Bowel Habits,Short Chain Fatty Acids and Gut Microbiota in Parkinson’s Disease”(RESISTA-PD;ID:NCT02784145),we aimed at altering fecal SCFAs by an 8-week prebiotic intervention with resistant starch(RS).We enrolled 87 subjects in three study-arms:32 PD patients received RS(PD+RS),30 control subjects received RS,and 25 PD patients received solely dietary *** performed paired-end 100 bp length metagenomic sequencing of fecal samples using the BGISEQ platform at an average of 9.9 *** was *** the PD+RS group,fecal butyrate concentrations increased significantly,and fecal calprotectin concentrations dropped significantly after 8 weeks of RS ***,we observed a reduction in non-motor symptom load in the PD+RS *** reference-based analysis of metagenomes highlighted stable alpha-diversity and beta-diversity across the three groups,including bacteria producing ***-free analysis suggested punctual,yet pronounced differences in the metagenomic signature in the PD+RS ***-PD highlights that a prebiotic treatment with RS is safe and well-tolerated in *** stable alpha-diversity and beta-diversity alongside altered fecal butyrate and calprotectin concentrations call for long-term studies,also investigating whether RS is able to modify the clinical course of PD.

AIM: To investigate whether NSAIDs/ASA lesions in the colon can histologically be diagnosed on the basis of ischemic necrosis similar to biopsy-based diagnosis of NSAIDs/ASA-induced erosions and ulcers of the ***: In the period between 1997 and 2002, we investigated biopsy materials obtained from 611 patients (415 women, 196 men, average age 60.5 years) with endoscopic focal erosions, ulcerations, strictures or diaphragms in the colon. In the biopsies obtained from these lesions, we always established the suspected diagnosis of NSAID-induced lesions whenever necroses of the ischemic type were found. Together with the histological report, we endosed a questionnaire to investigate the use of *** data provided by the questionnaire were then correlated with the endoscopic findings, the location, number and nature of the lesions, and the histological ***: At the time of their colonoscopy, 86.1% of the patients had indeed been taking NSAID/ASA medication for years (43.9%) or months (29.5%). The most common indication for the use of these drugs was pain (64.3%),and the most common indication for colonoscopy was bleeding (55.5%). Endoscopic inspection revealed multiple erosions and/or ulcers in 60.6%, strictures in 15.8%, and diaphragms in 3.0% of the patients. The lesions were located mainly in the right colon including the transverse colon (79.9%). A separate analysis of age and sex distribution,endoscopic and histological findings for NSAIDs alone,ASA alone, combined NSAID/ASA, and for patients denying the use of such drugs, revealed no significant differences among the ***: This uncontrolled retrospective study based on the histological finding of an ischemic necrosis shows that the histologically suspected diagnosis of NSAID-induced lesions in the colon is often correct. The true diagnostic validity of this finding and the differentiation from ischemic colitis should, however, be investigated in a prospective controlled

Purpose: The study investigated the rate of complications associated with sonographically guided lymph node biopsies and assessed potential risk factors. Methods: A total of 536 sonographically guided puncture procedures (283 males, 52.8%;253 females, 47.2%;average age 57.0 ± 16.0 years;range 14 - 87 years) were performed in 469 patients for the work-up of unclear lymphadenopathy. Events, complications and potential risk factors, were prospectively documented. Results: The 469 patients underwent a total of 536 puncture procedures (PP) including 663 punctures and 1485 passes. Lymph node localizations were intraabdominal (55.2%, n = 296), cervical (22.4%, n = 120), inguinal (12.9%, n = 69), axillary (7.8%, n = 42) and other (1.7%, n = 9). No complications were documented during the entire study period. There was no increased risk of complications documented for the potential risk factors number of punctures, the number of passes, the localization, diameter of the lymph node (s), puncture technique, needle gauge, as well as patients’ sex, age and coagulation parameters, and the experience of the examiner. Conclusions: Our findings confirm the safety of percutaneous sonographically guided lymph node biopsies in different regions of the body in patients with adequate coagulation parameters undergoing pre-interventional color Doppler ultrasound examination.
