Objective: Osteoarthritis is a degenerative disease that affects synovial joints. Micro-injuries of articular structures initiate inflammatory processes, leading to persistent pain. Due to various risk factors,osteoarthritis is often diagnosed in multimorbid patients. This makes pain management one of the key challenges, with a consistent need for new therapeutic strategies. Hence, complementary and integrative methods such as hirudotherapy have become increasingly important, even though their mechanisms of action are not entirely ***: We retrospectively analyzed the longitudinal effect of a single leech application on osteoarthritic joints in a heterogenic cohort of 24 cases with various chronic pain syndromes. We assessed articular pain intensity ratings and movability of the treated joint after one-time leeching for up to 12 *** further investigated the effect of hirudotherapy on the systemic pain status and multimodal treatment strategies of the ***: There was a significant reduction in pain intensity ratings at the joint of leech application for up to 12 months after treatment. The improvements in pain intensities were independent of the form of osteoarthritis treated. In addition, we saw a considerable enhancement in local movability of the treated joint. Hirudotherapy did not seem to influence the systemic pain status as well as the previously established individualized multimodal treatment model of the ***: Leeching as an adjuvant therapy has a great potential especially in terms of safety and longterm outcome.

Assisted reproductive technologies invoIving the use of spermatozoa and eggs for in vitro fertilization(IVF)have come as the solution for many infertile couples to become ***,in some cases,the use of ejaculated spermatozoa delivers poor IVF *** studies have suggested the use of testicular spermatozoa in severe male in fertility cases,but no guideli nes regarding their utilization are currently *** the present study,we found the mRNA protamine 1/protamine 2(P1/P2)ratio to be a valuable biomarker of poor sperm function that could be used as a diagnostic key for the identification of cases that would benefit from the use of testicular spermatozoa.A total of 23 couples undergoing egg donation cycles with at least one previous cycle failure were *** couples underwent two consecutive intracytoplasmic sperm injection(ICSI)cycles with either ejaculated or testicular spermatozoa(TESA).The sperm mRNA P1/P2 ratio,fertilization rate,blastocyst rate,and pregnancy and live birth rate were *** showed improved ICSI and clinical outcomes in cycles with testicular spermatozoa in men with altered mRNA P1/P2 *** cycles presented significantly higher rates of fertilization(mean±standard deviation:76.1%±15.1%vs 65.5%±18.8%),blastocyst formation(55.0%±20.3%vs 30.8%±23.8%),and good morphological quality blastocyst(28.9%±22.9%vs 13.5%±17.9%)and also improvements on pregnancy(60.9%vs 0%)and healthy birth rates(56.5%vs 0%)than EJACULATE *** results described here suggest that in patients with previous IVF/ICSI failures and aberrant mRNA protamine ratios,the use of testicular spermatozoa may be a good alternative to improve clinical outcomes.

In this paper,we introduce an extension of a splitting method for singularly perturbed equations,the socalled RS-IMEX splitting[Kaiser et al.,Journal of Scientific Computing,70(3),1390–1407],to deal with the fully compressible Euler *** straightforward application of the splitting yields sub-equations that are,due to the occurrence of complex eigenvalues,not hyperbolic.A modification,slightly changing the convective flux,is introduced that overcomes this *** is shown that the splitting gives rise to a discretization that respects the low-Mach number limit of the Euler equations;numerical results using finite volume and discontinuous Galerkin schemes show the potential of the discretization.

DICER-like （DCL） proteins produce small RNAs that silence genes involved in development and defenses against viruses and pathogens. Which DCLs participate in plant-herbivore interactions remains unstudied. We identified and stably silenced four distinct DCL genes by RNAi in Nicotiana attenuata （Torrey ex. Watson）, a model for the study of plant-herbivore interactions. Silencing DCL1 expression was lethal. Manduca sexta larvae performed significantly better on ir-dcl3 and ir-dcl4 plants, but not on ir-dcl2 plants compared to wild type plants. Phytohormones, defense metabolites and microarray analyses revealed that when DCL3 and DCL4 were silenced separately, herbivore resistance traits were regulated in distinctly different ways. Crossing of the lines revealed complex interactions in the patterns of regulation. Single ir-dcl4 and double ir-dcl2 ir-dcl3 plants were impaired in JA accumulation, while JA- Ile was increased in ir-dcl3 plants. Ir-dcl3 and ir-dcl4 plants were impaired in nicotine accumulation; silencing DCL2 in combination with either DCL3 or DCL4 restored nicotine levels to those of WT. Trypsin proteinase inhibitor activity and transcripts were only silenced in ir-dcl3 plants. We conclude that DCL2/3/4 interact in a complex manner to regulate anti-herbivore defenses and that these interactions significantly complicate the already challenging task of understanding smRNA function in the regulation of biotic interactions.

Background:The hypothesis was tested that insect meal(IM)as protein source influences intermediary metabolism of growing *** test this,30 male,5-week-old crossbred pigs were randomly assigned to 3 groups of 10 pigs each with similar body weights(BW)and fed isonitrogenous diets either without(CON)or with 5%IM(IM5)or 10%IM(IM10)from Tenebrio molitor *** 4 weeks and key metabolic tissues(liver,muscle,plasma)were analyzed using ***:Most performance parameters did not differ across the groups,whereas ileal digestibilities of most amino acids were 6.7 to 15.6%-units lower in IM10 than in CON(P<0.05).Transcriptomics of liver and skeletal muscle revealed a total of 166 and 198,respectively,transcripts differentially expressed between IM10 and CON(P<0.05).Plasma metabolomics revealed higher concentrations of alanine,citrulline,glutamate,proline,serine,tyrosine and valine and a lower concentration of asparagine in IM10 than in CON(P<0.05).Only one out of fourteen quantifiable amino acid metabolites,namely methionine sulfoxide(MetS),in plasma was elevated by 45%and 71%in IM5 and IM10,respectively,compared to CON(P<0.05).Plasma concentrations of both,major carnitine/acylcarnitine species and bile acids were not different across *** of liver and plasma demonstrated no differences in the concentrations of triacylglycerols,cholesterol and the main phospholipids,lysophospholipids and sphingolipids between *** percentages of all individual phosphatidylcholine(PC)and phosphatidylethanolamine(PE)species in the liver showed no differences between groups,except those with 6 double bonds(PC 38:6,PC 40:6,PE 38:6,PE 40:6),which were markedly lower in IM10 than in CON(P<0.05).In line with this,the percentage of C22:6n-3 in hepatic total lipids was lower in IM10 than in the other groups(P<0.05).Conclusions:Comprehensive analyzes of the transcriptome,lipidome and metabolome of key metabolic tissues indicate that partial or complete replacement o

At the end of October 2018,a storm of unprecedented strength severely damaged the forests of the eastern sector of the Italian *** affected forest area covers 42,500 *** president of one of the damaged regions asked for help from the University of *** eight months of discussion,the authors of this article wrote a consensus *** sometimes asper debate brought to light some crucial aspects:1)even experienced specialists may have various opinions based on scientific knowledge that lead to conflicting proposals for *** some of them there is evidence that to restore a destroyed natural environment it is more judicious to do nothing;2)the soil corresponds to a living structure and every ecosystem’s management should be based on it;3)faced with a catastrophe,people and politicians find themselves unarmed,also because they rarely have the scientific background to understand natural *** politicians are the only persons who make the key decisions that drive the economy in play and therefore determine the near future of our *** article is an attempt to respond directly to the governor of a region who formally and prudently asked a university department called"Land,Environment,Agriculture and Forestry"for help before taking decisions;4)the authors also propose an artistic interpretation of facts(uncontrolled storm)and conclusions(listen to the soil).Briefly,the authors identify the soil as an indispensable source for the renewal of the destroyed forest,give indications on how to prepare a map of the soils of the damaged region,and suggest to anchor on this soil map a series of silvicultural and soil management actions that will promote the soil conservation and the faster recovery of the natural dynamic stability and resilience.

Protein-losing enteropathy(PLE) is characterized by loss of serum proteins into the gastrointestinal tract. It may lead to hypoproteinemia and clinically present as protein deficiency edema, ascites, pleural or pericardial effusion and/or malnutrition. In most cases the site of protein loss is the small intestine. Here we present an unusual case of severe PLE in a 55-year old female with a one-year history of recurrent diarrhea, crampy abdominal pain, and peripheral edema. Endoscopy and MRI showed a diffuse inflammatory thickening of the sigmoid colon and the rectum. Surgical resection of the involved colon was performed and the symptoms were significantly resolved. The final histologic evaluation confirmed a diagnosis of a pseudomembranous colitis with cap polyposis-like features. Such a cause of PLE has never been described before.

Strawberry(Fragaria×ananassa)fruits are an excellent source of L-ascorbic acid(AsA),a powerful antioxidant for plants and *** the genetic components underlying AsA accumulation is crucial for enhancing strawberry nutritional ***,we unravel the genetic architecture of AsA accumulation using an F1 population derived from parental lines‘Candonga’and‘Senga Sengana’,adapted to distinct Southern and Northern European *** account for environmental effects,the F1 and parental lines were grown and phenotyped in five locations across Europe(France,Germany,Italy,Poland and Spain).Fruit AsA content displayed normal distribution typical of quantitative traits and ranged five-fold,with significant differences among genotypes and *** content in each country and the average in all of them was used in combination with 6,974 markers for quantitative trait locus(QTL)*** stable QTLs for AsA content were detected in linkage group(LG)3A,LG 5A,LG 5B,LG 6B and LG *** genes were identified within stable QTL intervals and expression analysis in lines with contrasting AsA content suggested that GDP-L-Galactose Phosphorylase FaGGP(3A),and the chloroplast-located AsA transporter gene FaPHT4;4(7C)might be the underlying genetic factors for QTLs on LG 3A and 7C,*** show that recessive alleles of FaGGP(3A)inherited from both parental lines increase fruit AsA ***,expression of FaGGP(3A)was two-fold higher in lines with high *** here identified represent a useful resource for efficient selection of new strawberry cultivars with increased AsA content.
