Activin-Directed Differentiation of Hum.n Em.ryonic Stem.Cells Differentially m.dulates Alveolar Epithelial Wound Repair via Paracrine m.chanism

Differentiated em.ryonic stem.cells (ESC) can am.liorate lung inflam.ation and fibrosis in anim.l lung injury m.dels;therefore, ESC, or their products, could be candidates for regenerative therapy for incurable lung diseases, such as idiopathic pulm.nary fibrosis (IPF). In this study, we have investigated the paracrine effect of differentiated and undifferentiated hum.n ESC on alveolar epithelial cell (AEC) wound repair. hESC line, SHEF-2 cells were differentiated with Activin treatm.nt for 22 days in an em.ryoid body (EB) suspension culture. Conditioned m.dia (Cm. which contain cell secretory factors were collected at different tim. points of differentiation. Cm.were then tested onin vitro?wound repair m.del with hum.n type II AEC line, A549 cells (AEC). Our study dem.nstrated that Cm.originated from.undifferentiated hESC significantly inhibited AEC wound repair when com.ared to the control. Whereas, Cm.originated from.Activin-directed hESC differentiated cell population dem.nstrated a differential reparative effect on AEC wound repair m.del. Cm.obtained from.Day-11 of differentiation significantly enhanced AEC wound repair in com.arison to Cm.collected from.pre- and post-Day-11 of differentiation. Day-11 Cm.enhanced AEC wound repair through significant stim.lation of cell m.gration and cell proliferation. RT-PCR and im.unocytochem.stry confirm.d that Day-11 Cm.was originated form.a m.xed population of endoderm.l/m.soderm.l differentiated hESC. This report suggests a putative paracrine-m.diated epithelial injury healing m.chanism.by hESC secreted products, which is valuable in the developm.nt of novel stem.cell-based therapeutic strategies.

Adult neurogenesis,the process of creating new neurons,involves the coordinated division,m.gration,and differentiation of neural stem.*** process is restricted to neurogenic niches located in two distinct areas of the brain:the subgranular zone of the dentate gyrus of the hippocam.us and the subventricular zone of the lateral ventricle,where new neurons are generated and then m.grate to the olfactory *** has been thought to occur only during the em.ryonic and early postnatal stages and to decline with age due to a continuous depletion of neural stem.***,recent years have seen trem.ndous progress in our understanding of adult brain neurogenesis,bridging the knowledge gap between em.ryonic and adult ***,we discuss the current status of adult brain neurogenesis in light of what we know about neural stem.*** this notion,we talk about the im.ortance of intra cellular signaling m.lecules in m.bilizing endogenous neural stem.cell prolife *** on the current understanding,we can declare that these m.lecules play a role in targeting neurogenesis in the m.ture ***,to achieve this goal,we need to avoid the undesired proliferation of neural stem.cells by controlling the necessary checkpoints,which can lead to tum.rigenesis and prove to be a curse instead of a blessing or hope.
