Defamiliarizing Reality for Mental and Physical Subjection:Rereading Harold Pinter's The Room and The Dumb Waiter as Political Comedies

While Pinter's earliest plays have been recognized in the modernist history of theatre as comedies of menace and his later plays as political comedies,this article argues that his earliest plays are equally very liable to be interpreted as political comedies.Regardless of their absurdist dramatization of people's helpless exposure to external,unidentifiable threats,a common post-WWII characteristic feature of human experience,I claim that The Room and The Dumb Waiter(both written 1957,staged 1960),two model examples of Pinter's earliest oeuvres,do not simply follow the aesthetic of absurdist theatre to express human futility.The audience's experience of viewing the theatrical performances of both plays in terms of discursive cyclicality or character normality is subverted into one of changeability,strangeness,and contradiction.To foreground the political implications of such revolutionary theatrical experience,Pinter's plays are examined in the light of his unique use of defamiliarization,relying not on Brecht's traditional techniques of singing,dancing,image-projecting,or captioning,but on a simple,dual technique of image destruction and creation.It consists of divesting characters of their normality and portraying them instead as individuals who identify only with unusual images of place,time,body,and consciousness.Using this special technique of defamiliarization,both plays are examined to reveal Pinter's central political theme of undermining reality for purposes of mental and physical subjections.

The continuous development of cyberattacks is threatening digital transformation endeavors worldwide and leadsto wide losses for various organizations. These dangers have proven that signature-based approaches are insufficientto prevent emerging and polymorphic attacks. Therefore, this paper is proposing a Robust Malicious ExecutableDetection (RMED) using Host-based Machine Learning Classifier to discover malicious Portable Executable (PE)files in hosts using Windows operating systems through collecting PE headers and applying machine learningmechanisms to detect unknown infected files. The authors have collected a novel reliable dataset containing 116,031benign files and 179,071 malware samples from diverse sources to ensure the efficiency of RMED approach.The most effective PE headers that can highly differentiate between benign and malware files were selected totrain the model on 15 PE features to speed up the classification process and achieve real-time detection formalicious executables. The evaluation results showed that RMED succeeded in shrinking the classification timeto 91 milliseconds for each file while reaching an accuracy of 98.42% with a false positive rate equal to 1.58. Inconclusion, this paper contributes to the field of cybersecurity by presenting a comprehensive framework thatleverages Artificial Intelligence (AI) methods to proactively detect and prevent cyber-attacks.

This work carried out a measurement study of the Ethereum Peer-to-Peer(P2P)network to gain a better understanding of the underlying nodes.Ethereum was applied because it pioneered distributed applications,smart contracts,and Web3.Moreover,its application layer language“Solidity”is widely used in smart contracts across different public and private blockchains.To this end,we wrote a new Ethereum client based on Geth to collect Ethereum node information.Moreover,various web scrapers have been written to collect nodes’historical data fromthe Internet Archive and the Wayback Machine project.The collected data has been compared with two other services that harvest the number of Ethereumnodes.Ourmethod has collectedmore than 30% more than the other services.The data trained a neural network model regarding time series to predict the number of online nodes in the future.Our findings show that there are less than 20% of the same nodes daily,indicating thatmost nodes in the network change frequently.It poses a question of the stability of the network.Furthermore,historical data shows that the top ten countries with Ethereum clients have not changed since 2016.The popular operating system of the underlying nodes has shifted from Windows to Linux over time,increasing node security.The results have also shown that the number of Middle East and North Africa(MENA)Ethereum nodes is neglected compared with nodes recorded from other regions.It opens the door for developing new mechanisms to encourage users from these regions to contribute to this technology.Finally,the model has been trained and demonstrated an accuracy of 92% in predicting the future number of nodes in the Ethereum network.

Gastric cancers are caused primarily due to the activation and amplification of the EGFR or HER2 kinases resulting in cell proliferation,adhesion,angiogenesis,and metastasis.Conventional therapies are ineffective due to the intra-tumoral heterogeneity and concomitant genetic mutations.Hence,dual inhibition strategies are recommended to increase potency and reduce cytotoxicity.In this study,we have conducted computational high-throughput screening of the ChemBridge library followed by in vitro assays and identified novel selective inhibitors that have a dual impediment of EGFR/HER2 kinase activities.Diversity-based High-throughput Virtual Screening(D-HTVS)was used to screen the whole ChemBridge small molecular library against EGFR and HER2.The atomistic molecular dynamic simulation was conducted to understand the dynamics and stability of the protein-ligand complexes.EGFR/HER2 kinase enzymes,KATOIII,and Snu-5 cells were used for in vitro validations.The atomistic Molecular Dynamics simulations followed by solvent-based Gibbs binding free energy calculation of top molecules,identified compound C3(5-(4-oxo-4H-3,1-benzoxazin-2-yl)-2-[3-(4-oxo-4H-3,1-benzoxazin-2-yl)phenyl]-1H-isoindole-1,3(2H)-dione)to have a good affinity for both EGFR and HER2.The predicted compound,C3,was promising with better binding energy,good binding pose,and optimum interactions with the EGFR and HER2 residues.C3 inhibited EGFR and HER2 kinases with IC50 values of 37.24 and 45.83 nM,respectively.The GI50 values of C3 to inhibit KATOIII and Snu-5 cells were 84.76 and 48.26 nM,respectively.Based on these findings,we conclude that the identified compound C3 showed a conceivable dual inhibitory activity on EGFR/HER2 kinase,and therefore can be considered as a plausible lead-like molecule for treating gastric cancers with minimal side effects,though testing in higher models with pharmacokinetic approach is required.

BACKGROUND:Hepatitis C virus (HCV) infection is thought to be chronic and the factors leading to viral clearance or persistence are poorly understood.This study was undertaken to investigate the possibility of a significant relationship between the spontaneous clearance or the persistence of hepatitis C virus (HCV) infection and cytokine and apoptosis gene polymorphisms in Tunisian patients on hemodialysis.METHODS:Polymorphisms of the genes IL-1 (-889 IL-1α,-511 and +3954 IL-1β,IL-1Ra),IL-18 (-137 and-607),IL-12 (-1188) and Apo1/Fas (-670) were determined by PCR-RFLP,PCR-SSP and PCR-VNTR in 100 healthy blood donors and 100 patients infected with HCV and undergoing hemodialysis.The patients were classified into two groups:G1 consisted of 76 active chronic hepatitis patients (positive for HCV RNA) and G2 consisted of 24 hemodialysed patients who spontaneously eliminated the virus (negative for HCV RNA).RESULTS:The frequency of genotype association [-137GC/-607CA] IL-18 was higher in G2 (41.7%) than in G1 (15.8%) (P=0.008;OR=0.26;95% CI,0.10-0.73).We also found a higher frequency of the AA genotype of the Apo1/Fas gene in G2 (41.6%) than in G1 (17.5%) (P=0.026;OR=3.49;95% CI,1.13-10.69).Adjustment for known covariate factors (age,gender and genotype) confirmed these univariate findings and revealed that the genotype association GC-CA of the (-137 and-607) IL-18 gene and the AA genotype of the Apo1/Fas gene were associated with the clearance of HCV (P=0.041 and 0.017,respectively).CONCLUSION:The two genotypes GC-CA of the (-137 and-607) IL-18 polymorphism and the AA genotype of the Apo1/Fas gene influence the outcome of HCV infection in Tunisian patients on hemodialysis.

Bioelectronic interventions,specifically trigeminal nerve st imulat ion(TNS),have attracted considerable attention in conditions where cortical spreading depolarizations(CSDs)accompanied by compromised cerebral perfusion may exacerbate neurological damage.While pharmacological interventions have demonstrated initial potential in addressing CSDs,a standardized treatment approach has not yet been established.The objective of this perspective is to explore emerging bioelectronic methodologies for addressing CSDs,particularly emphasizing TNS,and to underscore TNS’s capacity to enhance neurovascular coupling and cerebral perfusion.

Reducing the effects of external disturbance on overhead crane systems is crucial,as they can impair the controller performance and cause excessive vibrations or oscillations of the payloads.One such external disturbance is the inclination of the supporting track of the crane trolley,which causes the system dynamics model to change.An open-loop control strategy is widely utilized to control the payload sway motion and generally does not require any alterations in the physical structure of a system or the installation of sensors and/or actuators.Input and command shaping are two common open-loop control techniques applied to control overhead cranes.In this paper,the effect of moving an overhead crane system along an inclined supporting track is investigated.In addition,the ability of different types of input-and command-shaping control schemes to suppress the residual vibrations due to trolley track inclination is demonstrated.Two types of input-shaping controllers,which are double-step,zero vibration,and one command waveform(WF)shaper based on a trigonometric function,are used and tested.A linear equation of motion of the overhead crane resting on an inclined surface is developed to simulate the overhead crane and payload motion.The effectiveness of the different types of open-loop controllers to suppress residual vibrations is verified by both simulation and experimental results.In addition,a new WF command shaper is proposed and designed to overcome track inclination while eliminating payload residual vibration.A comprehensive comparative analysis,both numerically and experimentally,is performed on the new proposed shaper to measure its effectiveness in handling inclination when compared to other types of open-loop controllers.The new shaper outperforms other controllers in eliminating payload residual vibration for a wider range of inclination angles.

Association of Fas/Apo1 gene promoter (-670 A/G) polymorphism in Tunisian patients with IBD

AIM: To detect a possible association between the polymorphism of the (-670 A/G) Fas/Apo1 gene promoter and susceptibility to Crohn's disease (CD) and ulcerative colitis (UC) in the Tunisian population. METHODS: The (-670 A/G) Fas polymorphism was analyzed in 105 patients with CD, 59 patients with UC, and 100 controls using the polymerase chain reaction restriction fragment length polymorphism method. RESULTS: Significantly lower frequencies of the Fas -670 A allele and A/A homozygous individuals were observed in CD and UC patients when compared with controls. Analysis of (-670 A/G) Fas polymorphism with respect to sex in CD and UC showed a significant difference in A/A genotypes between female patients and controls (P corrected = 0.004 in CD patients and P corrected = 0.02 in UC patients, respectively). Analysis also showed a statistically significant association between genotype AA of the (-670 A/G) polymorphism and the ileum localization of the lesions (P corrected = 0.048) and between genotype GG and the colon localization (Pcorrected = 0.009). The analysis ofinflammatory bowel disease patients according to clinical behavior revealed no difference. CONCLUSION: Fas-670 polymorphism was associated with the development of CD and UC in the Tunisian population.
