Xiyanping injection,a traditional chinese medicine injection made of andrographolide sulfonate,consisting of well-defined ingredients with antiviral,antibacterial,anti-inflammatory and antipyretic efficacy,has been widely used for treating infectious diseases of respiratory and digestive ***,its wide applications may easily lead to unreasonable clinical *** order to guide the precise clinical application and rational use of Xiyanping injection,experts in related fields conducted systematically literature review,evaluated and deliberated the application of Xiyanping injection in treating acute infectious diseases using evidence-based medicine method,and jointly drafted the consensus to summarize types of acute infectious diseases in children and adults that can be treated with Xiyanping injection,and recommend the intervention time,usage and dosage,course of treatment and combined medication of the ***,the consensus elucidates the safety,precautions and contraindications of the injection,so as to provide guidance for clinical use.

Effect of chinese Herbal Treatment on Th1- and Th2-Type Cytokines,Progesterone andβ-Human Chorionic Gonadotropin in Early Pregnant Women of Threatened Abortion

Objective： To study the clinical effects of Prescription Zhuyun-Ⅲ （助孕3号方, ZYⅢ） on early pregnant women diagnosed as threatened abortion, and its mechanism in immunity and endocrine by determining serum Th1- and Th2-type cytokines, progesterone, and β-human chorionic gonadotropin （β-HCG）. Methods： The treatment group comprised 30 early pregnant women diagnosed as threatened abortion of deficiency syndrome of Pi （脾）, Shen （肾）, or both. The control group consisted of 20 normal early pregnant women of similar gestational age. Patients in the treatment group were administered with ZYⅢ for 4 weeks. Peripheral blood samples were collected pre- and post-treatment from both the treatment and the control groups. Serum Thl-type cytokine [interleukin-2 （IL-2）] and Th2-type cytokine [interleukin-10 （IL-10）] were determined by flow cytometry, and serum progesterone and 13-HCG were determined by ELISA. Results：（1） The treatment was effective in 26 and ineffective in 4 patients of the treatment group. Therefore, the cure percentage was 86.67%. （2） In the treatment group before the treatment, IL-2 was significantly higher, IL-10 tended to be less, and the Th1/Th2 balance shifted toward Thl compared with those in the control group. （3） After the treatment, IL-2 was decreased, IL-10 was increased, and IL-2/IL-10 was decreased. Both progesterone and β-HCG were increased. Changes of progesterone were positively correlated with changes of IL-10, whereas changes of β-HCG were negatively correlated with changes of IL-2. Conclusions： Our study suggests that ZYⅢ has an evident function of protecting the fetus, and one of its mechanisms is inhibiting the secretion of Thl cytokines, promoting the secretion of Th2 cytokines, and recovering the pathological shift of the Th1/Th2 balance. The other possible mechanism is increasing serum progesterone and β-HCG concentrations. Moreover, there are some correlations between the above two effects.

Advances in cell sources of hepatocytes for bioartificial liver

BACKGROUND: Orthotopic liver transplantation (OLT) is the most effective therapy for liver failure. However, OLT is severely limited by the shortage of liver donors. Bioartificial liver (BAL) shows great potential as an alternative therapy for liver failure In recent years, progress has been made in BAL regarding genetically engineered cell lines, immortalized human hepatocytes, methods for preserving the phenotype of primary human hepatocytes, and other functional hepatocytes derived from stem cells. DATA SOURCES: A systematic search of PubMed and ISI Web of Science was performed to identify relevant studies in English language literature using the key words such as liver failure bioartificial liver, hepatocyte, stem cells, differentiation, and immortalization. More than 200 articles related to the cell sources of hepatocyte in BAL were systematically reviewed. RESULTS: Methods for preserving the phenotype of primary human hepatocytes have been successfully developed. Many genetically engineered cell lines and immortalized human hepatocytes have also been established. Among these cell lines the incorporation of BAL with GS-HepG2 cells or alginate encapsulated HepG2 cells could prolong the survival time and improve pathophysiological parameters in an animal model of liver failure. The cBAL111 cells were evaluated using the AMC-BAL bioreactor, which could eliminate ammonia and lidocaine, and produce albumin. Importantly, BAL loading with HepLi-4 cells could significantly improve the blood biochemical parameters, and prolong the survival time in pigs with liver failure. Other functional hepatocytes differentiated from stem cells, such as human liver progenitor cells, have been successfully achieved. CONCLUSIONS: Aside from genetically modified liver cell lines and immortalized human hepatocytes, other functionalhepatocytes derived from stem cells show great potential as cell sources for BAL. BACKGROUND: Ideal model of poststroke depression (PSD) may be induced in rats guided by the theoretical evidence that 'primary endogenous mechanism' and 'reactivity mechanism' theories for PSD in human being. OBJECTIVE: To investigate the feasibility of comprehensive methods to induce PSD models in rats. DESIGN: A randomized controlled animal trial. SETTING: Department of Neurology, Affiliated hospital of Chengdu University of traditional chinese medicine. MATERIALS: Male SD rats of SPF degree, weighing 350-500 g, were provided by the experimental animal center of Chengdu University of traditional chinese medicine. The rats were raised for 1 week adaptively, then screened behaviorally by open-field test and passive avoidance test. Forty-five rats with close scores were randomly divided into normal control group (n =10), simple stroke group (n =10), stress group (n =10) and PSD group (n =15). METHODS: The experiments were carried out in the laboratory of Chengdu University of traditional chinese medicine from July 2002 to February 2003. ① Rat models of focal cerebral ischemia were induced by thread embolization, then treated with separate raising and unpredictable stress to induce PSD models. ②The neurologic deficit was evaluated by Longa 5-grade standard (the higher the score, the severer the neurologic deficit) and horizontal round rod test (normal rat could stay on it for at least 3 minutes). ③ The behavioral changes of PSD rats were evaluated by the saccharin water test, open-field text and passive avoidance test, including the changes of interest, spontaneous and exploratory activities, etc. ④ The levels of monoamine neurotransmitters, including norepinephrine (NE), serotonin (5-HT) and dopamine, in brain were determined using fluorospectrophotometry. 我国原发性肝癌(肝癌)疾病负担严重,预后不理想。通过简便的方法对肝癌高危人群进行早期筛查,是实现早期诊断和早期治疗,提高肝癌生存率的关键。目前在肝癌的早期筛查方面,甲胎蛋白等血液学检查仍是主要手段,但是其灵敏度及特异度有限,存在高漏诊风险。近年来随着科学技术的不断发展,对传统检测方法的改进以及新兴标志物如甲基化DNA、微小RNA等的出现,为进一步提高早期肝癌筛查的灵敏度和特异度带来了希望。此专家共识总结了传统及新型血液学检查手段的研究进展,并提出了在肝癌早期筛查中应用的专家指导意见,旨在为肝癌早期筛查中血液学检查的应用提供依据,改善我国肝癌防控水平。

Danggui-Buxue-Tang(DBT) is a famous traditional chinese *** have reported that total glucosides of Danggui-Buxue-Tang(DBTG) can attenuate bleomycin-induced lung fibrosis through reduction of pro-inflammatory cytokines or the decrease of I collagen *** present study we further focuses on investigating mechanisms underlying the anti-fibrosis effects of DBTG on pulmonary fibrosis(PF) *** were randomly divided into six groups:(1) saline control group;(2) Bleomycin group in which rats were intratracheally instillated with bleomycin(5mg/kg);(3-5) Bleomycin and DBTG group,in which DBTG were given to rats daily(16,32 or 64 mg/kg/day,i.g.) one day after bleomycin instillation for 4 weeks until the end of the treatment;(6) Bleomycin and positive control group. Bleomycin stimulation resulted in a significant increase in the pathological scores for alveolitis and lung fibrosis and an evident increase in the contents of extracellular matrix(ECM) components in serum including hyaluronic acid(HA),laminin(LN), procollagen typeⅢ(PCⅢ) and collagenⅣ(CⅣ).Moreover,bleomycin-induced pulmonary injury and lung fibrosis was indicated by increased lung MMP-1,MMP-9 and TIMP-1 mRNA *** the other hand,DBTG treatment attenuated bleomycin-induced alveolitis and lung fibrosis,significant decreased the concentrations of HA,LN,PCⅢand CⅣin ***,administration of DBTG resulted in a significant reduction in metalloproteinase-1(MMP-1),MMP-9 and the tissue inhibitor of metalloproteinase-1(TIMP-1) mRNA levels in different stages of pulmonary fibrosis,and the balance between MMPs and ***,the findings of the present study provide evidence that DBTG inhibits ECM deposition and modulates MMPs and TIMPs expression in the lung tissue,which may be associated with the attenuation of experimental pulmonary fibrosis.

红丹爽身粉致儿童铅中毒的调查

目的对一兄妹铅中毒进行调查,查明其中毒的原因。方法在铅中毒患儿肘部静脉处抽取静脉血2mL,分别置于用0.2mol·L-1EDTA抗凝的试管内,用塞曼石墨炉原子吸收分光光谱仪测定血铅水平。铅中毒诊断标准依据卫生部2006年颁布的《儿童高铅血症和铅中毒分级和处理原则(试行)》。用自行设计的调查问卷对患儿进行铅暴露调查。用Innov-α4000便携式X线荧光分析仪(XRF)对其家庭中物品进行铅水平检测。抽取相关的爽身粉,每个样品加入浓硝酸,并经微波消化,用火焰原子吸收光谱仪(FAAS)检测爽身粉中的铅水平。结果2例患儿入院血铅水平分别为303g.L-1和385g.L-1,诊断为中度铅中毒。给予依地酸二钠钙静脉滴注驱铅,5d后血铅分别降至118g.L-1和88g.L-1。应用XRF对其家庭用品进行铅水平检测,食品、水、儿童玩具和家具未超标,但一种供2例患儿使用的散装爽身粉铅水平>100000mg·kg-1,经FAAS测得铅水平为214000mg·kg-1,超过国家化妆品卫生标准(40mg·kg-1)5000多倍,超过食品卫生标准(5mg·kg-1)40000多倍。经询问这种爽身粉来自江西省赣州农村,由中药红丹与滑石粉混合而成,抽查赣州市药店出售的红丹粉16份,铅水平平均为817375mg·kg-1。抽查红丹爽身粉13份,铅水平全部超标,平均为206784mg·kg-1,普通盒装爽身粉6种,铅水平均为5mg·kg-1。结论2例患儿铅中毒是由于使用红丹爽身粉引起,红丹爽身粉在我国南方农村地区普遍使用,应引起重视。

Five novel bisindole alkaloids,cyclovinblastine A～C(1～3), 4-desacetoxyvinblastine N-oxide(4),and 4’-deoxyvinblastine AVoxide(5),together with nine known alkaloids leurosine,cycloleurosine,vinblastine,leurosidine,vinblastine AVoxide, isoleurosine,4’-deoxyleurosidine,4’-deoxyleurosidine AVoxide and 4-desacetoxyvinblastine were isolated from the leaves of Catharanthus *** structures of 1～5 were established by analysis of their NMR and HR-ESI-MS spectroscopic *** alkaloids were evaluated for their cytotoxic activities against the human hepatocellular carcinoma(HepG2) cell line, human colorectal carcinoma(Lovo) cell line,and human breast carcinoma(MCF-7) cell line by the MTT method in vitro,*** results indicated that cytotoxic activities of alkaloids isoleurosine,4’-deoxyleurosidine and 4-desacetoxyvinblastine were much more potent than those of the positive control *** addition,the structure-activity relationships(SAR) were conducted on the basis of the cytotoxicity of these isolated alkaloids.
