Monitoring adenoviral based gene delivery in rat glioma by molecular imaging

AIM:To determine whether endothelial progenitor cells(EPCs)can be used as delivery vehicle for adenoviral vectors and imaging probes for gene therapy in ***:To use cord blood derived EPCs as delivery vehicle for adenoviral vectors and imaging probes for glioma gene therapy,a rat model of human glioma was made by implanting U251 cells *** were transfected with an adenovirus(AD5/carrying hnIS gene)and labeled with iron oxide and inoculated them directly into the tumor 14 d following implantation of U251 *** resonance imaging(MRI)was used to in vivo track the migration of EPCs in the *** expression of gene products was determined by in vivo Tc-99m single photon emission computed tomography(SPECT).The findings were validated with immunohistochemistry(IHC).RESULTS:EPCs were successfully transfected with the adenoviral vectors carrying hnIS which was proved by significantly(Pn transfected *** of EPCs following transfection and iron labeling was not *** vivo imaging showed the presence of iron positive cells and the expression of transgene(hnIS)product on MRI and SPECT,respectively,all over the tumors following administration of transfected and iron labeled EPCs in the *** confirmed the distribution of EPC around the tumor away from the injection site and also showed transgene expression in the *** results indicated the EPCs’ability to deliver adenoviral vectors into the glioma upon intratumor ***:EPCs can be used as vehicle to deliver adenoviral vector to glioma and also act as imaging probe at the same time.

Social determinants of health(SDOH)affect quality of *** investigated SDOH impacts on self-perceived resilience among people with adult congenital heart disease(ACHD).Secondary analysis of data from two com-plementary studies:a survey study conducted May 2021–June 2022 and a qualitative study conducted June 2020–August *** was assessed through CD-RISC10 score(range 0–40,higher scores reﬂect greater self-perceived resilience)and interview *** and SDOH(education,employment,living situa-tion,monetary stability,ﬁnancial dependency,area deprivation index)data were collected by healthcare record review and *** used linear regression with robust standard errors to analyze survey data and performed a thematic analysis of interview *** participants(n=127)mean age was 42±14 years;51%were female,87%*** was moderate(75%)or complex(25%);41%functional class C or ***(mean 30±7)varied by monetary stability:compared to people with difﬁculty paying bills,resilience was 15.0 points higher(95%CI:6.9–23.1,png having enough money and 14.2 points higher(95%CI:5.9–22.4,p=0.001)for those reporting just enough *** participants’(n=25)mean age was 32 years(range 22–44);52%were female,72%*** was moderate(56%)or complex(44%);76%functional class C or *** discussed factors affecting resilience aligned with each of the major SDOH,prominently,economic stability and healthcare access and *** stability may be important for supporting self-perceived resi-lience in *** knowledge can inform the development of resilience interventions for this population.

Pest preference and subsequent susceptibility of a host individual is likely related to previous growth patterns in that *** ash borer(Agrilus planipennis Fairmaire)is a pestiferous beetle introduced to north America from *** all species of ash are susceptible to attack,some individual trees appear to survive *** selected ash trees in southeastern Michigan,collected cores and categorized trees as high tolerance to emerald ash borer attack(high overall health,low crown dieback),low tolerance(low overall health,high crown dieback)and intermediate tolerance(in-between the other categories).We artificially wounded trees and measured wound closure after 3 *** width indices were not correlated between high and low tolerance *** slopes comparing growth and years were significantly different between the three tolerance categories,with high tolerance trees having the steepest *** closure was greatest in high tolerance *** tolerance trees demonstrating more rapid(steeper regression slope),consistent(lower variance),and effective(greater wound closure)*** vigorously growing trees likely had more capacity to repair damage caused by emerald ash borer,leading to healthier trees in our *** previous host growth patterns to health may have implications related to identifying individual trees potentially tolerant to attack.

Validity of electrocardiographic criteria for increased left ventricular mass in young patients in the general population

AIMTo investigate validity of electrocardiographic(ECG)criteria for left ventricular hypertrophy(LVH)in young *** Retrospectively,echocardiograms showing LVH and concomitant electrocardiograms were collected in patients 18 to 39 years old.A control group of patients without LVH was *** echocardiogram as the gold standard,electrocardiograms were analyzed using common voltage *** Study included 100 subjects(52%male,mean age=28±6.8 years,96%Hispanic or African-American)with 50%LVH *** and specificity for SokolowLyon criteria were 24%(95%CI:13.5%-38.4%)and 88%(95%CI:74.9%-95%).For Cornell criteria,sensitivity was 32%(95%CI:19.9%-46.8%)and specificity 98%(95%CI:87.9%-99.8%).For R in a VL criteria,sensitivity was 12%(95%CI:4.9%-25%)and specificity 100%(95%CI:91.1%-100%).COnCLUSIOn In young adults common ECG voltage criteria have low sensitivities and high specificities similar to other age *** sensitivities preclude these ECG criteria from serving as effective screening tests.

Effects of protein deprivation and re-feeding on P2X_2 receptors in enteric neurons

AIM:To investigate the effects of malnutrition and refeeding on the P2X2 receptor,nitric oxide synthase(nOS),calretinin,calbindin and choline acetyltransferase(ChAT) in neurons of the rat ***:We analyzed the co-localization,numbers and sizes of P2X2-expressing neurons in relation to nOS-immunoreactive(IR),calbindin-IR,ChAT-IR,and calretinin-IR neurons of the myenteric and submucosal *** experimental groups consisted of:(1) rats maintained on normal feed throughout pregnancy until 42 d post-parturition(n);(2) rats deprived of protein throughout pregnancy and 42 d post-parturition(D);and(3) rats undernourished for 21 d post-parturition and then given a protein diet from days 22 to 42(DR).The myenteric and submucosal plexuses were evaluated by double labeling by immunohistochemical methods for P2X2 receptor,nOS,ChAT,calbindin and ***:We found similar P2X2 receptor immunoreactivity in the cytoplasm and surface membranes of myenteric and submucosal neurons from the n,D and DR *** labeling of the myenteric plexus demonstrated that approximately 100% of nOS-IR,calbindin-IR,calretinin-IR and ChAT-IR neurons in all groups also expressed the P2X2 *** the submucosal plexus,the calretinin-IR,ChAT-IR and calbindin-IR neurons were nearly all immunoreactive for the P2X2 *** the myenteric plexus,there was a 19% increase in numbers per cm2 for P2X2 receptor-IR neurons,64% for nOS-IR,84% for calretinin-IR and 26% for ChAT-IR neurons in the D *** spatial density of calbindin-IR neurons,however,did not differ among the three *** submucosal neuronal density increased for calbindin-IR,calretinin-IR and ChAT-IR *** average size of neurons in the myenteric plexus neurons in the D group was less than that in the controls and,in the re-fed rats;there was a 34% reduction in size only for the calretinin-IR ***:This work demonstrates that expression of the P2X2 receptor is present in inhibitory,intrins

目的评价经颈静脉肝内门腔分流术（TIPS）专用覆膜支架（Viatorr支架）在TIPS中的应用价值。方法回顾性分析37例在美国俄勒冈州健康生命科技大学Dotter介入放射学研究所采用Viatorr支架行’nPs治疗的患者资料，随访时间为（15．2±9．3）个月（3～42个月）。TIPS指征包括门静脉高压相关性的急慢性消化道出血，经药物及内镜治疗无效者17例；顽固性肝源性胸、腹水18例，***综合征2例。采用配对t检验比较手术前后门腔静脉压力差（PSG）的变化，以Kaplan-Meier曲线分析支架开通率。结果37例共置入41枚Viatorr支架，其中3枚直径为8mm，38枚为10mm，支架带膜长度为4～8cm，无相关手术并发症。PSG由术前的（22．4±8．4）mmHg（1mmHg=0．133kPa）降为（8．1±3．2）mmHg，差异有统计学意义（t=12．754，P〈0．01）。17例出血患者术后出血均停止，1例于术后17个月复发。18例严重顽固性腹水及肝性胸水患者中，4例术后腹水不消退，其余14例随访期间有2例腹水复发。2例（5．4％）发生分流道阻塞，Kaplan-Meier曲线分析结果显示1年的开通率为97．0％。术后1个月内无病死患者，2例分别于术后3个月及15个月死于多器官功能衰竭，晚期病死率为5．4％，死亡前1周内复查支架均通畅。肝移植患者5例（13．5％）。结论Viatorr支架能明显提高TIPS术后开通率，选择合适的支架，采用正确的释放技术能进一步提高疗效，但长期效果评价尚需验证。
