Establishment of induced pluripotent stem cells from aged mice using bone marrow-derived myeloid cells

If induced pluripotent stem(iPS)cells are to be used to treat damaged tissues or repair organs in elderly patients,it will be necessary to establish iPS cells from their *** determine the feasibility of using this technology with elderly patients,we asked if it was indeed possible to establish iPS cells from the bone marrow(BM)of aged *** cells from aged C57BL/6 mice carrying the green fluorescence protein(GFP)gene were cultured with granulocyte macrophage-colony stimulating factor(GM-CSF)for 4 *** factors(Oct3/4,Sox2,Klf4 and c-Myc)were introduced into the BM-derived myeloid(BM-M)*** efficiency of generating iPS cells from aged BM cultured in GM-CSF was ***,we succeeded in obtaining BM-M-iPS cells from aged C57BL/6 mice,which carried *** BM-M-iPS cells expressed SSEA-1 and Pou5f1 and were positive for alkaline phosphatase *** iPS cells did make teratoma with three germ layers following injection into syngeneic C57BL/6 mice,and can be differentiated to three germ layers in *** co-culturing with OP9,the BM-M-iPS cells can be differentiated to the myeloid *** differentiated BM-M-iPS cells proliferated well in the presence of GM-CSF,and lost expression of Nanog and Pou5f1,at least in part,due to methylation of their *** the contrary,Tnf and Il1b gene expression was upregulated and their promoters were hypomethylated.

The proportion of elderly people rises in the developed countries. The increased susceptibility of the elderly to infectious diseases is caused by immune dysfunction, especially T cell functional decline. Age-related hematopoietic stem cells deviate from lymphoid lineage to myeloid lineage. Thymus shrinks early in life, which is followed by the decline of na?ve T cells. T-cell receptor repertoire diversity declines by aging, which is caused by cytomegalovirus-driven T cell clonal expansion. Functional decline of B cell induces antibody affinity declines by aging. Many effector functions including phagocytosis of myeloid cells are down regulated by aging. The studies of aging of myeloid cells have some controversial results. Although M1 macrophages have been shown to be replaced by antiinflammatory(M2) macrophages by advanced age, many human studies showed that pro-inflammatory cytokines are elevated in older human. To solve this discrepancy here we divide age-related pathological changes into two categories. One is an aging of immune cell itself. Second is involvement of immune cells to age-related pathological changes. Cellular senescence and damaged cells in aged tissue recruit pro-inflammatory M1 macrophages, which produce pro-inflammatory cytokines and proceed to agerelated diseases. Underlying biochemical and metabolic studies will open nutritional treatment.

MXenes,a new family of two-dimensional transition metal carbides or nitrides,have attracted tremendous attention for various applications due to their unique properties such as good electrical conductivity,hydrophilicity,and ion *** this work,Ti_(3)C_(2) MXene,or MX,is converted to MX-TiO_(2) composites using a simple and rapid microwave hydrothermal treatment in HCl/NaCl mixture solution that induces formation of fine TiO_(2) particles on the MX parent structure and imparts photocatalytic activity to the resulting MX-TiO_(2) *** composites were used for enrofloxacin(ENR),a frequently found contaminating antibiotic,removal from *** relative amount of the MX and TiO_(2) can be controlled by controlling the hydrothermal temperature resulting in composites with tunable adsorption/photocatalytic *** addition was found to play important role as composites synthesized without NaCl could not adsorb enrofloxacin *** NaCl into the hydrothermal treatment causes sodium ions to be simultaneously intercalated into the composite structure,improving ENR adsorption greatly from 1 to 6 mg ENR/g *** also slows down the MX to TiO2 conversion leading to a smaller and more uniform distribution of TiO_(2) particles on the ***-TiO_(2)/NaCl composites,which have sodium intercalated in their structures,showed both higher ENR adsorption and photocatalytic activity than composites without NaCl despite the latter having higher TiO2 *** ENR on the composites can be efficiently degraded by free radicals generated from the photoexcited TiO2 particles,leading to high photocatalytic degradation *** demonstrates the synergetic effect between adsorption and photocatalytic degradation of the synthesized compounds.

Detailed analysis of fan-shaped jets in three dimensional numerical simulation

We performed three dimensional resistive magnetohydrodynamic simulations to study the magnetic reconnection using an initially shearing magnetic field configuration（force free field with a current sheet in the middle of the computational box）.It is shown that there are two types of reconnection jets：the ordinary reconnection jets and fan-shaped jets,which are formed along the guide magnetic *** fan-shaped jets are significantly different from the ordinary reconnection jets which are ejected by magnetic tension *** are two driving forces for accelerating the fan-shaped *** is the Lorentz force which initially dominates the motion of fluid elements,and then the gas pressure gradient force accelerates the fluid elements in the later *** dependence on magnetic reconnection angle and resistivity value has also been *** formation and evolution of these jets provide a new understanding of dynamic magnetohydrodynamic jets.

Effects of gamma-ray irradiation on electronic and non-electronic equipment of Large Helical Device

In a deuterium operation on the Large Helical Device, the measurement and control equipment placed in the toms hall must survive under an environment of radiation, To study the effects of gamma-ray irradiation on the equipment, an irradiation experiment is performed at the Cobalt-60 irradiation facility of Nagoya University. Transient and permanent effects on a personal computer, media converters, programmable logic controllers, isolation amplifiers, a web camera, optical flow meters, and water sealing gaskets are experimentally surveyed. Transient noise appears on the web camera. Offset of the signal increases with an increase of the integrated dose on the programmable logic controller. The DeviceNet module on the programmable logic controller is broken at the integrated dose of 72 Gy, which is the expected range of the integrated dose of the toms hall. The other equipment can survive under the gamma-ray field in the toms hall.

Molecular targeting to treat gastric cancer

Trastuzumab that targets human epidermal growth factor receptor 2 (HER2) protein is the only approved molecular targeting agent for treating gastric cancer in Japan and the outcomes have been favorable. However, trastuzumab is effective for only 10% to 20% of the population with gastric cancer that expresses HER2 protein. Molecular targeting therapy with bevacizumab against vascular endothelial growth factors (VEGF) and with cetuximab and panitumumab against the epidermal growth factors pathway that have been approved for treating colorectal cancer are not considered effective for treating gastric cancer according to several clinical trials. However, ramucirumab that targets VEGF receptor-2 prolonged overall survival in a large phase III clinical trial and it might be an effective molecular targeting therapy for gastric cancer. The significance of molecular targeting therapy for gastric cancer remains controversial. A large-scale randomized clinical trial of novel molecular targeting agents with which to treat gastric cancer is needed.

Observation of MHD Instabilities Driven by Energetic Electrons in the Large Helical Device

Coherent magnetic fluctuations in an acoustic range of frequency have been regularly observed in low-density（n_e〈0.2×10^（19）m^（-3））plasmas with strong second harmonic electron cyclotron resonance heating（ECRH）on the Large Helical *** X-ray measurements indicated that energetic electrons are generated in these ECRH *** magnetic fluctuations are suppressed in higher density discharges where energetic electrons are not *** ECRH power modulation experiment indicated that the observed magnetohydrodynamic（MHD）mode has an acoustic nature rather than an Alfvenic nature.

Study of first orbit losses of 1 MeV tritons using the Lorentz orbit code in the LHD

Shot-integrated measurement of the triton burnup ratio has been performed in the Large Helical Device. It was reported that the triton burnup ratio, defined as total DT neutron yield divided by total DD neutron yield, increases significantly in inward shifted configurations. To understand the magnetic configuration dependence of the triton burnup ratio, the first orbit loss fraction of 1 MeV tritons is evaluated by means of the Lorentz orbit code for various magnetic configurations. The first orbit loss of 1 MeV tritons is seen at t of less than 10-5 s and loss points of the triton are concentrated on the side of the helical coil case where the magnetic field is relatively weak. The significant decrease of the first orbit loss fraction by 15% is obtained with the inward shift of the magnetic axis position from 3.90 to 3.55 m. It is found that the decrease of first orbit loss is due to the reduction of the first orbit loss of transition and helically trapped tritons.

Reinforcement Effect and Permeability Assessment of Gravel-Tire Chips Mixture (GTCM) for Use in Marine Landfill

The number of marine landfills in Japan has increased over the past decade due to the lack of suitable land. For marine landfills, protection of the alluvium clay layer and improvement of the drainage performance in waste inflow are important aspects. In this paper, an economical construction method for these problems is proposed using gravel-tire chips mixture (GTCM) as the horizontal reinforcement and drainage medium beneath the waste. The content and particle size of tire chips mixed with gravel are essential factors that affect the bearing capacity and permeability of the reinforcement layer. Therefore, a series of permeability tests are conducted using newly developed large-scale triaxial compression and permeability test apparatus to investigate the effect of tire chips particle size, the mass proportion of tire chips (MPTC), and triaxial stress on the permeability of GTCM. In addition, the effectiveness of this technique is evaluated by numerical simulations. The experimental results confirm that the shear strength of GTCM is influenced by tire chips content. Furthermore, permeability coefficient of GTCM is on the order of 0.02 cm/s to 0.08 cm/s, which is higher than the tolerable level of permeability of drainage layer in landfills. GTCM sample shows excellent permeability even on higher compression. Moreover, the Non-Darcy flow properties of GTCM (non-linear) are introduced in this study, and an approximate power function relationship between the permeability coefficient and the non-Darcy flow coefficient is developed. The numerical results confirm that GTCM performs better than the sand, a traditional reinforcement material.
