Involvement of Sphingosine 1-Phosphate (S1P) Receptor Type 1 and Type 4 in Migratory Response of Mouse T Cells toward S1P

Sphingosine 1-phosphate （S1P）, a pleiotropic lysophospholipid, regulates signal transduction pathway via Gprotein-coupled receptors termed S1P1-5 in several types of the cells including lymphocytes. Higher levels of S1P4 mRNA as well as S1P1 mRNA are expressed in lymphoid tissues such as the spleen, thymus, lymph nodes, and Payer＇s patches. In contrast to S1P1 that plays an essential role in lymphocyte egress, little is known about the role of S1P4 in immune system. In this study, we found that S1P at 10 to 100 nM significantly induced the cell migration and the significant levels of S1P1 and S1P4 mRNA were expressed in mouse CD4 T cells, D10.G4.1 mouse Th2 cells, and ***-2 mouse thymoma cells. In D10.G4.1 and ***-2 cells, S1P-induced migration was almost completely inhibited by pretreatment with pertussis toxin, Clostoridium difficile toxin B, and （S）-enantiomer of FTY720-phosphate, a potent agonist at S1P1 and S1P4. The members of the Rho family small GTPase, Cdc42 and Rac were activated by S1P stimulation in these cells. The transfection with dominant negative or constitutively active forms of Cdc42 and Rac revealed that the activation of both Cdc42 and Rac is essential for S1P-induced migration of these cells. The immunoprecipitation assays using CHO cells co-expressing both S1P4 and S1P1 receptors indicated that S1P4 and S1P1 are associated on the cell surface. These results suggest that the association of S1P4 and S1PI plays an important role in migratory response of mouse T cells toward S1P.

Recurrent renal cell carcinoma leading to a misdiagnosis of polycystic liver disease: A case report

BACKGROUND Polycystic liver disease(PCLD) with a large cystic volume deteriorates the quality of life of patients through substantial effects on the adjacent organs,recurrent cyst infections, cyst rupture, and hemorrhage. Surgical or radiological intervention is usually needed to alleviate these symptoms. We report a rare case of the cystic metastasis of renal cell carcinoma(RCC), which was misdiagnosed as PCLD, as a result of the clinical and radiological similarity between these *** SUMMARY A 74-year-old female who had undergone nephrectomy for papillary-type RCC(PRCC) was suffering from abdominal pain and the recurrent intracystic hemorrhage of multiple cysts in the liver. Imaging studies and aspiration cytology of the cysts showed no evidence of malignancy. With a diagnosis of autosomal dominant polycystic liver disease, the patient received hepatectomy for the purpose of mass reduction and infectious cyst removal. Surgery was performed without complications, and the patient was discharged on postoperative day 14. Postoperatively, the pathology revealed a diagnosis of recurrent PRCC with cystic *** This case demonstrates the importance of excluding the cystic metastasis of a cancer when liver cysts are observed.

Confirmation of Novel Quantitative Trait Loci for Seed Dormancy at Different Ripening Stages in Rice

Seed dormancy contributes resistance to pre-harvest sprouting. Effects on respective quantitative trait loci （QTLs） for dormancy should be assessed by using fresh seeds before germinability altered through storage. We investigated QTLs related to seed dormancy using backcross inbred lines derived from a cross between Nipponbare and Kasalath. Four putative QTLs for seed dormancy were detected immediately after harvest using composite interval mapping. These putative QTLs were mapped near C1488 on chromosome 3 （qSD-3.1）, R2171 on chromosome 6 （qSD-6.1）, R1245 on chromosome 7 （qSD-7.1） and C488 on chromosome 10 （qSD-IO.1）. Kasalath alleles promoted dormancy for qSD-3.1, qSD-6.1 and qSD-7.1, and the respective proportions of phenotypic variation explained by each QTL were 12.9%, 9.3% and 8.1%. We evaluated the seed dormancy harvested at different ripening stages during seed development using chromosome segment substitution lines （CSSLs） to confirm gene effects. The germination rates of CSSL27 and CSSL28 substituted with the region including qSD-6.1 were significantly lower than those of Nipponbare and other CSSLs at the late ripening stage. Therefore, qSD-6.1 is considered the most effective novel QTL for pre-harvest sprouting resistance among the QTLs detected in this study.

Evaluation of the Validity of SARS-CoV-2 Infection Control Measures through Antibody Testing for Employees of a University and Hospital

Introduction: Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), is spread through person-to-person transmission and has become a global pandemic. At Saitama Medical University Hospital, many medical staff members have been involved in treating patients with COVID-19. The Care Task Force was established in collaboration with physicians, medical staff, and clerical staff in the various hospital departments to strengthen infection control measures based on standard precautions. Methods: To determine the outcome of infection control measures, we administered anti-SARS-CoV-2 antibody tests and questionnaires to all 2461 employees including nonhospital workers, as a local standard, between June 29 and July 10, 2020. Results: Among the hospital workers, 698 (33.99%) had contact with patients with COVID-19 and 325 healthcare workers worked in specialized wards for the COVID-19, intensive care unit, and high-fever outpatient clinics. Positive for the anti-SARS-CoV-2 antibody were only 4 (0.16%) employees. Among them, the past histories of two employees were unknown, while the other two had a history of COVID-19 before the test and were not involved in the medical care of COVID-19 patients at our hospital. Conclusion: It is the first study assessing the seropositive rate in Saitama-prefecture, a bed-town of Tokyo. Compared with the local standard, we found that health care workers are not at risk for viral droplet transmission, especially with SARS-CoV-2 and even with the current pandemic, with infection control measures based on standard precautions. Based on our findings and with no clusters formed in our university and hospital, we continued current infection control measures.

GROWTH AND MORPHOLOGY OF POLYMER-ACTIN COMPLEXES

F-actins are semi-flexible polyelectrolytes and can be assembled into large polymer-actin complex with polymorphism through electrostatic interaction with polycations. This study investigates the structural phase behavior and the growth of polymer-actin complexes in terms of its longitudinal and lateral sizes. Our results show that formation of polymer-actin complexes is cooperative, and morphology and growth of polymer-actin complexes depend on polycation species and concentrations of polycation and salt in a constant actin concentration. We found that the longitudinal growth and lateral growth of polymer-actin complexes are dominated by different factors. This induces the structural polymorphism of polymer-actin complexes. Major factors to influence the polymorphism of polymer-actin complexes in polyelectrolytc system have been discussed. Our results indicate that the semi-flexible polyelectrolyte nature of F-actins is important for controlling the morphology and growth ofactin architectures in cell.

In Vivo Investigation of Zr-Based Bulk Metallic Glasses Sub-Periosteally Implanted on the Bone Surface

Bulk metallic glasses (BMG) show higher strength and lower Young’s modulus than SUS 316L stainless steel and Ti-6Al-4V alloys. This study aimed to investigate the reaction of Zr-based BMG sub-periosteally implanted on the surface of the rat femur, thereby evaluate the possibility of the BMG as biomaterials for osteosynthetic devices. Zr65Al7.5Ni10Cu17.5 BMG ribbons with 10 mm length, 2 mm width and 0.5 mm thickness were implanted sub-periosteally on the femur surface in three male Wistar rats for 6 weeks. Systemic effects were evaluated by measuring Cu and Ni levels in the blood, and local effects were evaluated by the histological observation of the surrounding soft tissues in contact with the BMG. The reaction of the surface of the BMG was examined with scanning electron microscopy. No increase of Cu and Ni levels in the blood was recognized. In the scanning electron microscopy observation, spherical deposits which were considered as sodium chloride crystals were observed. Neither breakage nor pitting corrosion was noted. BMG will be a promising metallic biomaterial for osteosynthetic device that must be removed.

Aspirin-induced small bowel injuries and the preventive effect of rebamipide

AIM:To evaluate the influence of taking low-dose aspirin for 4 wk on small intestinal complications and to examine the preventive effect of ***:This study was conducted as a single-center,randomized,double-blind,cross-over,placebo-controlled *** healthy male subjects were *** subject underwent video capsule endos-copy after 1 and 4 wk of taking aspirin and omepra-zole,along with either rebamipide or placebo *** primary endpoint was to evaluate small bowel damage in healthy subjects before and after taking low-dose aspirin for 4 ***:The number of subjects with mucosal breaks(defined as multiple erosions and/or ulcers)were 1 at 1 wk and 1 at 4 wk on the jejunum,and 6 at 1 wk(P = 0.0061)and 7 at 4 wk on the ileum(P =0.0019).Rebamipide significantly prevented mucosal breaks on the ileum compared with the placebo group(P = 0.0173 at 1 wk and P = 0.0266 at 4 wk).CONCLUSION:Longer-term,low-dose aspirin adminis-tration induced damage in the small *** prevented this damage,and may be a candidate drug for treating aspirin-induced small bowel complications.

Common Management Process Model of New TQM Based on the Situation Analysis

In the previous study, we suggested the concept of new TQM based on the consideration of basic concept of Quality Control. Also, in the previous study, we suggested the target domains and entities of product and process based on the TQM Matrix and view point of Three Dimensional Unification Value Models for managing quality of organization systems. Furthermore, in the previous study, we suggest the Common Management Process of organizations. Based on the above suggestion, in this paper, we would like to propose the Common Management Process Model of Total Quality Management based on the consideration of situation analysis and more precise definition of TQM Matrix and Three Dimensional Unification Value Model of “Product and Process”. Improvement of quality and efficiency of organization management can be expected by the integration of conventional different management such as quality assurance, quality improvement, risk management, investment individually from the view point of common management process.

Long-term effects of lamivudine treatment in Japanese chronic hepatitis B patients

AIM:To analyze the association between the emergence of tyrosine-methionine-asparatate-asparatate(YMDD) mutants(reverse transcription;rtM204I/V) and deterioration of liver function during long-term lamivudine treatment of Japanese patients with chronic hepatitis B virus(HBV) ***:The data of 61 consecutive Japanese patients with chronic hepatitis B who underwent continuous lamivudine treatment for more than 24 mo and had a virological response were *** of YMDD mutants was done by real-time polymerase chain reaction with LightCycler probe hybridization assay for up to 90 mo(mean,50.8 mo;range,24-90 mo).RESULTS:A mixed mutant-type(YMDD + tyrosine-isoleucine-asparatate-asparatate:YIDD or tyrosine-valineasparatate-asparatate:YVDD) or a mutant-type(YIDD or YVDD) were found in 57.4% of 61 patients at 1 year,78.7% of 61 patients at 2 years,79.6% of 49 patients at 3 years,70.5% of 34 patients at 4 years,68.4% of 19 patients at 5 years,57.1% of 14 patients at 6 years,and 33.3% of 6 patients at 7 *** the 61 patients,56(92%) had mixed mutant-or a *** 5(8%) had no mutants at each observation *** breakthrough was found in 26(46.4%) of 56 patients with YMDD mutants,20 of whom had a hepatitis flare-up:the remaining 30(53.6%) had neither a virological breakthrough nor a *** 20 patients who developed a hepatitis flare-up had a biochemical and virological response after adefovir was added to the lamivudine ***:Our results suggest that it is possible to continue lamivudine treatment,even after the emergence of YMDD mutants,up to the time that the patients develop a hepatitis flare-up.
