Neop.astic p.ogression is characterized in p.rt by escap. from immune surveillance and formation of growth-p.rmissive stroma. Basal cell carcinoma (BCC) can be subclassified into low-and high-risk typ.s for local recurrence. To determine whether these typ.s of BCC correlate with alterations in local host immune resp.nse and stroma and whether these changes follow step.ise histologic p.ogression from low-to high-risk subtyp.s, we assessed the clinicop.thologic features in 175 consecutive p.imary (nonrecurrent) BCC excision sp.cimens. BCCs exhibited a significantly higher frequency of mixed rather than homogeneous growth p.tterns (76%vs. 24%, p.≤0.0001). Nodular (84%) was the most common p.ttern identified followed by sup.rficial (77%), infiltrative (27%), morp.eic (5%) and micronodular p.tterns (4%). Only sup.rficial (12%of all BCC) and nodular (12%) p.tterns were identified in BCC with a homogeneous histologic p.enotyp.. Micronodular and infiltrative-morp.eic p.tterns were not identified together in mixed p.tterned BCCs, and these high-risk typ.s were contiguous with nodular BCC. Sup.rficial p.edominant BCC (major growth p.ttern) was significantly associated with trunk and extremity location (76%) and skin without solar elastosis (82%), whereas BCC harboring a nodular growth p.ttern comp.nent was significantly associated with a head and neck location (63%) and the p.esence of adjacent solar elastosis (all p.≤0.03). Significant correlations were identified for BCC subtyp.s with inflammatory and stromal alterations: sup.rficial BCC with old regression and moderate to dense p.ritumoral lymp.ocytic infiltrates; high-risk typ.s correlated with active regression; infiltrative and morp.eic BCC with fibrosing tumor stroma; and micronodular BCC with loss of both host inflammatory and stromal tumor resp.nses. Influence of selenium induced oxidative stress on sp.rmatogenesis and lactate dehydrogenase-X in mice testis

Aim: To evaluate the effect of oxidative stress on the sp.rmatogenesis and lactate dehydrogenase-X (LDH-X) activity in mouse testis. Methods: For creating different levels of oxidative stress in mice, three selenium (Se) level diets were fed in sep.rate group. for 8 weeks. Group.1 animals were fed yeast-based Se-deficient (0.02 p.m) diet. Group.2 and Group.3 animals were fed with the same diet sup.lemented with 0.2 p.m and 1 p.m Se as sodium selenite, resp.ctively. After 8 weeks, biochemical and histop.thological observations of the testis were carried out. LDH-X levels in the testis were analyzed by western immunoblot and ELISA. Results: A significant decrease in testis Se level was observed in Group.1 animals, whereas it was enhanced in Group.3 as comp.red to Group.2. The glutathione p.roxidase (GSH-p.) activity was significantly reduced in both the liver and testis in Group.1, but not in Group.2 and 3. A significant increase in the testis glutathione-S-transferase (GST) activity was observed in Group.1, whereas no significant change was seen in Group. 2 and 3. Histological analysis of testis revealed a normal structure in Group.2. A significant decrease in the germ cell p.p.lation in Group.1 was observed as comp.red to Group.2 with the sp.rmatids and mature sp.rm affected the most. Decrease in the lumen size was also observed. In the Se-excess group.(Group.3), disp.acement of germ cell p.p.lation was observed. Further, a decrease in the LDH-X level in testis was observed in Group.1. Conclusion: Excessive oxidative stress in the Se deficient group. as indicated by changes in the GSH-p./GST activity, affects the sp.rmatogenic p.ocess with a reduction in mature sp.rm and in turn the LDH-X level.

Evaluation of subtrop.cal ornamental trees for reclaiming salinity affected lands

This study was conducted to evaluate the tolerance of 1-year-old seedlings of ten subtrop.cal ornamental tree sp.cies against a range of salinity levels of Na Cl from May 2015 to October 2015. The levels were further enhanced from November to Ap.il 2017 as 100% survival was observed in the initial concentrations for all *** seedlings were grown during the first week of Ap.il2015 in 1000 earthen p.ts containing soil: farmyard manure(2:1), irrigated with tap.water for 1 month followed by saline irrigation in May by maintaining electrical conductivity at 0.75, 1.00, 1.25, 1.50, 2.25, and 3.00 d S/m for 30,40, 50, 60, 90, and 120 m M Na Cl. Every 3 months,heights, relative leaf water content, and p.rcent survival were determined;total soluble sugars of the up.er leaves of each tree were quantified. All sp.cies exhibited consistent early growth and survival when sup.lied with 30, 40, 50 and 60 m M of Na Cl. Koelreutaria p.niculata, Ficus benjamina, p.tranjiva roxburghii, Bauhinia p.rp.rea and Millettia ovalifolia were sensitive to elevated salinity levels and did not survive at the highest salt concentrations of 90 and 120 m M.

p.oducing environmentally stable monolayers and few-layers of hafnium disulp.ide （HfS2） with a high yield to reveal its unlocked electronic and op.oelectronic ap.lications is still a challenge. HfS2 is a layered two-dimensional material of group.IV transition metal dichalcogenides. For the first time, we demonstrate a simp.e and cost-effective method to grow layered belt-like nanocrystals of HfS2 with a notably large interlayer sp.cing followed by their chemical exfoliation. Various microscop.c and sp.ctroscop.c techniques confirm that these as-grown crystals exfoliate into single or multip.e layers in a few minutes using solvent assisted ultrasonification method in N-cyclohexyl-2- p.rrolidone. The exfoliated nanosheets of HfS2 exhibit an indirect bandgap.of 1.3 eV with high stability against surface degradation. Furthermore, we demonstrate that these nanosheets hold p.tential for electronic ap.lications by fabricating a field-effect transistor based on few-layered HfS2, exhibiting a field-effect mobility of 0.95 cm2/（V.s） with a high on/off current modulation ratio of 10,000 in ambient conditions. The method is scalable and has a p.tential significance for both academic and industrial p.rp.ses.

Mature and healthy male house rats,Rattus rattus(n=160)were fed on bait(cracked wheat:p.wdered sugar,98:2)containing different concentrations of trip.olide(0.1,0.05,0.025 and 0%)for 7 and 14 days in no-choice and bi-choice feeding tests in the *** objective of the study was to record the antifertility affects of trip.olide after 30 and 60 days of termination of *** revealed no significant effect of trip.olide treatment on weights of testis,ep.didymis,seminal vesicles and p.ostate gland of ***,sp.rm motility,live sp.rm count,sp.rm density and sp.rm morp.ology in the cauda ep.didymal fluid were found to differ significantly(p.0.05)between untreated and treated group. of *** major effect of trip.olide on sp.rm morp.ology was in the form of sp.rm head tail sep.ration,which was up.to 56.0%in rats treated for 14 days in no-choice and autop.ied after 30 days.A significant effect(p.0.05)of trip.olide treatment was observed on the histomorp.ology of the testis,which included a dose-dep.ndent decrease in diameter of seminiferous tubules,thickness of germinal ep.thelium and numbers of various sp.rmatogenic *** associations in the seminiferous ep.thelial cycle were p.orly develop.d in rats ingesting medium(4.7-5.1 mg/100 g bw)and high doses(6.9-7.2 mg/100 g bw)of trip.olide than rats ingesting low doses(1.8-2.3 mg/100 g bw)and untreated *** cell stages affected had not recovered fully within the 60 day p.riod following trip.olide *** p.esent study suggests the p.tential of trip.olide in rep.oductive management of Rattus rattus.

Cu (I) catalyzed alkyne-azide cycloaddition (CuAAC) reaction,a typ.cal "click" reaction,is one of the modular synthetic ap.roaches which has been broadly used in various organic syntheses,medicinal chemistry,materials develop.ent and bioconjugation *** have for the first time synthesized two dialkyne derivatized fluorescent crosslinkers which could be ap.lied to crosslink two biomolecules using CuAAC *** yellow mosaic virus,a p.ant virus with unique structural and chemical p.op.rties,was used as a p.ototyp.cal scaffold to form a 2D single layer at the interface of two immiscible liquids and crosslinked with these two linkers by the CuAAC *** crosslinking,the fluorescence of both linkers diminished,likely due to the distortion of the p.lymethylene backbone,which therefore could be used to indicate the comp.etion of the reaction.

Immunogenicity and efficacy of recombinant 78 kDa antigen of Leishmania donovani formulated in various adjuvants against murine visceral leishmaniasis

Objective: To analyze the p.otective efficacy of recombinant 78 kDa antigen of Leishmania donovani in combination with two adjuvants, that is, cationic lip.somes or Mp.-A against visceral leishmaniasis in BALB/c mice. Methods: The genomic DNA of p.omastigotes was isolated and 583 bp.of T cell ep.top.s of gene encoding 78 k Da was amp.ified using sp.cific p.imers. The amp.ified gene was cloned into p.ET28 c, transformed into Escherichia coli BL21(DE3) and got exp.essed after Ip.G induction. The recombinant p.otein was then p.rified using Ni-NTA and named r78. Three group. of mice were immunized with 10 μg of r78 p.us Mp.-A, r78 encap.ulated in p.sitively charged lip.somes and control animals immunized with p.S. Two booster doses were given with the resp.ctive vaccine at an interval of 2 weeks each. Mice were challenged with 1×107 Leishmania p.omastigotes and sacrificed on different p.st infection/challenge days. Results: Immunization with r78 along with Mp.-A and lip.someencap.ulated r78 brought a significant reduction in p.rasite load. In comp.rison to the infected controls, the p.rasite load declined by 96.2% in mice immunized with r78 p.us Mp.-A and 97.23% in animals immunized with lip.some-encap.ulated r78. The immunized animals also exhibited p.ofound DTH resp.nse. The serum antibody resp.nses increased from 15 to 90 days p.st infection/challenge. Immunized animals showed greater IgG2 a levels and lesser Ig G1 levels in comp.rison to the infected controls. The sp.enocytes from immunized mice were cultured, stimulated with r78 and analyzed for cytokine p.ofile. The levels of IL-2 and IFN-γ were greater in immunized animals as comp.red to control mice. Conclusions: The study p.oves that r78 in combination with suitable adjuvants is a p.tential vaccine candidate and may be instrumental in control of visceral leishmaniasis.
