Selective deletion of the soluble Colony-Stimulating Factor 1 isoform in vivo prevents estrogen-deficiency bone loss in mice

Neutralizing CSF1 in vivo completely prevents ovariectomy （OVX）-induced bone loss in mice. There are two isoforms of CSF1, soluble （sCSF1）, and membrane-bound （mCSF1）, but their individual biological functions are unclear. It had been previously reported that mCSF1 knockout （K/O） and wild type （Wt） female mice experience the same degree of bone loss following OVX. In Wt mice the expression of sCSF1 was elevated fourfold in skeletal tissue following OVX while expression of mCSF1 was unchanged. To examine the role of sCSF1 in OVX-induced bone loss, mice were engineered in which sCSF1 was not expressed but expression of mCSF1 was unaffected （sCSF1 K/O）. Isoform-specific reverse transcription PCR confirmed the absence of transcripts for sCSF1 in bone tissue isolated from these animals and no circulating CSF1 was detected by ElISA. Surprisingly, there were no significant differences in bone mineral density （BMD） between sCSF1 K/O mice and Wt controls as assessed by dual-energy X-ray absorptiometry and micro-CT. However, one month after OVX, femoral, spinal and total BMD had declined by 11.2%, 8.9%, and 8.7% respectively in OVX-Wt animals as compared to Sham-OVX. In contrast OVX sCSF1 K/O mice showed changes of ＋0.1%, - 2.4%, and ＋2.3% at the same 3 sites compared to Sham-OVX sCSF1 K/O mice. These data indicate important non-redundant functions for the two isoforms of CSF1 and suggest that sCSF1, but not mCSF1, plays a key role in estrogen-deficiency bone loss.

Agrobacterium tumefaciens mediated plant transformation is a versatile tool for plant genetic engineering following its discovery nearly half a century ago.Numerous modifications were made in its application to increase efficiency,especially in the recalcitrant major cereals plants.Recent breakthroughs in transformation efficiency continue its role as a mainstream technique in CRISPR/Cas-based genome editing and gene stacking.These modifications led to higher transformation frequency and lower but more stable transgene copies with the capability to revolutionize modern agriculture.In this review,we provide a brief overview of the history of Agrobacterium-mediated plant transformation and focus on the most recent progress to improve the system in both the Agrobacterium and the host recipient.A promising future for transformation in biotechnology and agriculture is predicted.

Background Optimising the immunogenicity of COVID-19 vaccines to improve their protection against disease is necessary.Fractional dosing by intradermal(ID)administration has been shown to be equally immunogenic as intramuscular(IM)administration for several vaccines,but the immunogenicity of ID inactivated whole severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)at the full dose is unknown.This study(NCT04800133)investigated the superiority of antibody and T-cell responses of full-dose CoronaVac by ID over IM administration in adolescents.Methods Participants aged 11–17 years received two doses of IM or ID vaccine,followed by the 3rd dose 13–42 days later.Humoral and cellular immunogenicity outcomes were measured post-dose 2(IM-CC versus ID-CC)and post-dose 3(IMCCC versus ID-CCC).Doses 2 and 3 were administered to 173 and 104 adolescents,respectively.Results Spike protein(S)immunoglobulin G(IgG),S-receptor-binding domain(RBD)IgG,S IgG Fcγreceptor IIIa(FcγRIIIa)-binding,SNM[sum of individual(S),nucleocapsid protein(N),and membrane protein(M)peptide pool]-specific interleukin-2(Il-2)+CD4+,SNM-specific Il-2+CD8+,S-specific Il-2+CD8+,N-specific Il-2+CD4+,N-specific Il-2+CD8+and M-specific Il-2+CD4+responses fulfilled the superior and non-inferior criteria for ID-CC compared to IM-CC,whereas IgG avidity was inferior.For ID-CCC,S-RBD IgG,surrogate virus neutralisation test,90%plaque reduction neutralisation titre(PRNT90),PRNT50,S IgG avidity,S IgG FcγRIIIa-binding,M-specific Il-2+CD4+,interferon-γ+CD8+and Il-2+CD8+responses were superior and non-inferior to IM-CCC.The estimated vaccine efficacies were 49%,52%,66%and 79%for IM-CC,ID-CC,IM-CCC and ID-CCC,respectively.The ID groups reported more local,mild adverse reactions.Conclusion This is the first study to demonstrate superior antibody and M-specific T-cell responses by ID inactivated SARSCoV-2 vaccination and serves as the basis for future research to improve the immunogenicity of inactivated vaccines.

Aim: The harmful effects of pesticides have been largely documented in recent times. But effective therapeutic solutions to pesticide related male infertility are yet to be established. This study investigated the curative effects of lannea acida on imidacloprid (IMI)-induced hypofertility in male Wistar rats. Methods: Rats of 150 – 200 g were administered IMI (22.5 mg/kg) for two weeks and partitioned into control (distilled water, vitamin E, clomiphene citrate) or test (aqueous (340 mg/kg), methanol (170 mg/kg) extract) groups for eight weeks treatment. Animals were sacrificed at the end of the treatment and samples were collected for sperm, antioxidant and hormonal analysis. Fertility tests were performed from treatment day 47 for fertility indices estimation. Results were expressed as mean ± SEM and one way ANOVA was applied using STATISTICA Software. Results: Exposition to IMI resulted in a significant decrease in sperm count, motility, viability and normality, testosterone and lH, coupled to an increase in oxidative stress markers. Moreover, IMI impaired male fertility evidenced by a significant drop in fertility index and litter size. Similar to clomiphene citrate and vitamin E, plant extracts significantly improved the sperm parameters, sexual hormones and decreased the oxidative stress markers. More importantly, the fertility index and litter size were restored, especially with the aqueous extract. Conclusion: Present results indicate that l. acida possesses curative potentials against IMI-induced hypofertility through its androgenic and antioxidant properties. However, the effects the extract on spermatozoa DNA structure and the fertility of offsprings from exposed parents are yet to be studied to conclude on total recovery from IMI toxicity.

Maximizing recovery time between knock-out races improves sprint cross-country skiing performance

Background:In a sprint cross-country(XC)ski competition,the difference in recovery times separating the first and the second semi-final(SF)heats from the final(F)may affect performance.The aim of the current study was to compare the effects of longer vs.shorter recovery periods prescribed between the 3 knock-out races of a simulated sprint XC ski competition involving a prologue(P),quarter-final(QF),SF,and F.Methods:Eleven well-trained XC ski athletes completed 2 simulated sprint XC ski competitions on a treadmill involving 4×883-m roller-ski bouts at a 4°incline using the gear 3 ski-skating sub-technique.The first 3 bouts were completed at a fixed speed(PFIX,QFFIX,and SFFIX)corresponding to~96%of each individual’s previously determined maximal effort.The final bout was performed as a self-paced sprint time trial(FSTT).Test conditions differed by the time durations prescribed between the QFFIX,SFFIX,and FSTT,which simulated real-world XC ski competition conditions using maximum(MAX-REC)or minimum(MIN-REC)recovery periods.Results:The FSTT was completed 5.4±5.5 s faster(p=0.009)during MAX-REC(179.2±18.1 s)compared to MIN-REC(184.6±20.0 s),and this was linked to a significantly higher power output(p=0.010)and total metabolic rate(p=0.009).The pre FSTT blood lactate(Bla)concentration was significantly lower during MAX-REC compared to MIN-REC(2.5±0.8 mmol/l vs.3.6±1.6 mmol/l,respectively;p=0.027),and the pre-to-post FSTT increase in Bla was greater(8.8±2.1 mmol/l vs.7.1±2.3 mmol/l,respectively;p=0.024).No other differences for MAX-REC vs.MIN-REC reached significance(p>0.05).Conclusion:Performance in a group of well-trained XC skiers is negatively affected when recovery times between sprint heats are minimized which,in competition conditions,would occur when selecting the last QF heat.This result is combined with a higher pre-race Bla concentration and a reduced rise in Bla concentration under shorter recovery conditions.These findings may help inform decision making when XC skiers are faced with selecting a QF heat wi

A Primal-Dual SGD Algorithm for Distributed Nonconvex Optimization

The distributed nonconvex optimization problem of minimizing a global cost function formed by a sum of n local cost functions by using local information exchange is considered.This problem is an important component of many machine learning techniques with data parallelism,such as deep learning and federated learning.We propose a distributed primal-dual stochastic gradient descent(SGD)algorithm,suitable for arbitrarily connected communication networks and any smooth(possibly nonconvex)cost functions.We show that the proposed algorithm achieves the linear speedup convergence rate O(1/(√nT))for general nonconvex cost functions and the linear speedup convergence rate O(1/(nT)) when the global cost function satisfies the Polyak-lojasiewicz(P-l)condition,where T is the total number of iterations.We also show that the output of the proposed algorithm with constant parameters linearly converges to a neighborhood of a global optimum.We demonstrate through numerical experiments the efficiency of our algorithm in comparison with the baseline centralized SGD and recently proposed distributed SGD algorithms.

Suitability of Hermetia illucens larvae meal and fat in broiler diets:effects on animal performance,apparent ileal digestibility,gut histology,and microbial metabolites

Background:The possibility of partially replacing soybean meal(SBM)with Hermetia illucens(HI)defatted larvae meal in broiler nutrition has frequently been suggested.For sustainability reasons,however,the larvae fat produced during defatting should also be used and could be particularly beneficial regarding gut health due to its fatty acid composition.To evaluate the suitability of HI larvae as protein and fat source,a 2×3 factorial arrangement with two types of protein,i.e.SBM(S)or SBM and 15%of its crude protein replaced by HI larvae meal(l),and three levels of fat sources,namely 0(0 l),50%(50 l)or 100%HI larvae fat(100 l)at the expense of soybean oil was applied.Results:In the starter phase,an interaction showed higher body weight(BW),average daily gain(ADG)and improved feed conversion ratio(FCR)if 50%or 100%HI larvae fat was fed with HI larvae meal.Moreover,BW,ADG and FCR improved when feeding HI larvae meal as protein source.Additionally,we observed an increased average daily feed intake in the grower,finisher,and overall phase in the l groups and an improved FCR in 0 l compared to 50 l groups during the overall period.Regarding apparent ileal digestibility,HI larvae meal feeding increased dry matter,organic matter,and fat digestibility.Feeding HI larvae meal as protein source decreased the concentrations of agmatine,spermidine,spermine and ammonia in the caecal digesta,whereas fat source affected agmatine with higher concentrations in 50 l compared to 0 l in the colonic digesta.In contrast,caecal ethanolamine concentrations increased in HI larvae meal groups compared to SBM.Caecal butyric acid concentrations decreased with HI larvae meal feeding.An interaction was found for the jejunal villus area,being higher in l+100 l compared to S+100 l.Furthermore,l groups had greater villus width.Conclusions:A partial replacement of SBM with HI larvae meal and soybean oil with HI larvae fat in broiler diets without impairing animal performance or gut health seems possible.Feeding HI larvae meal affected broiler perform

The antimicrobial interventions of cilantro(Coriandrum sativum)in mitigating cross-contamination of foodborne pathogens during the retail soaking process

The efficacy of commercially available antimicrobials for fresh produce—electrolyzed water(EW;around 60 mg/l of free chlorine),a combination of lactic acid and phosphoric acid-based(lPA)and citric acid-based(CA)—was compared with tap water(TW)in preventing cross-contamination during the soaking step of crisping at a retail setting.A bunch of cilantro((103.7±14.9)g/bunch)was inoculated with a three-strain cocktail of nalidixic acid-resistant Salmonella enterica(S.enterica)and Escherichia coli(E.coli)O157:H7,S.enterica and listeria monocytogenes(l.monocytogenes),or E.coli O157:H7 and l.monocytogenes(around 5.0 log CFU/g).One inoculated and seven non-inoculated cilantro bunches were soaked in 76 l of TW,EW,lPA,and CA for 5 min.Two additional soakings,each with eight bunches of non-inoculated cilantro,were performed in the same soaking water.To determine the cross-contamination of inoculated foodborne pathogens via soaking water,the cilantro samples and soaking water following each soaking step were subjected to microbiological analyses using selective media supplemented with nalidixic acid(100μg/ml).During the first soaking,significantly greater reductions in Salmonella((2.9±0.5)log CFU/g),E.coli O157:H7((3.0±0.1)log CFU/g),and l.monocytogenes((2.7±0.3)log CFU/g)on cilantro were achieved with EW compared to soaking with TW,lPA,and CA(Plated cilantro to non-inoculated cilantro was completely mitigated by EW during three subsequent soaking events.With the exception of TW soaking water,no inoculated foodborne pathogens were detected in the 100 ml soaking water of EW,CA,and lPA collected.Including an appropriate concentration of chemical antimicrobial in water during the soaking step of crisping aids in mitigating cross-contamination of foodborne pathogen(s)in cilantro bunches.

Predefined-time distributed multiobjective optimization for network resource allocation

We consider the multiobjective optimization problem for the resource allocation of the multiagent network, where each agent contains multiple conflicting local objective functions. The goal is to find compromise solutions minimizing all local objective functions subject to resource constraints as much as possible, i.e., the Pareto optimums. To this end, we first reformulate the multiobjective optimization problem into one single-objective distributed optimization problem by using the weighted lppreference index,where the weighting factors of all local objective functions are obtained from the optimization procedure so that the optimizer of the latter is the desired Pareto optimum of the former. Next, we propose novel predefined-time algorithms to solve the reformulated problem by time-based generators. We show that the reformulated problem is solved within a predefined time if the local objective functions are strongly convex and smooth. Moreover, the settling time can be arbitrarily preset since it does not depend on the initial values and designed parameters. Finally, numerical simulations are presented to illustrate the effectiveness of the proposed algorithms.

Computational engineering analysis of external geometrical modifications on MQ-1 unmanned combat aerial vehicle

This paper focuses on the effects of external geometrical modifications on the aerodynamic characteristics of the MQ-1 predator Unmanned Combat Aerial Vehicle(UCAV)using computational fluid dynamics.The investigations are performed for 16 flight conditions at an altitude of7.6 km and at a constant speed of 56.32 m/s.Two models are analysed,namely the baseline model and the model with external geometrical modifications installed on it.Both the models are investigated for various angles of attack from-4°to 16°,angles of bank from 0°to 6°and angles of yaw from 0°to 4°.Due to the unavailability of any experimental(wind tunnel or flight test)data for this UCAV in the literature,a thorough verification of calculations process is presented to demonstrate confidence level in the numerical simulations.The analysis quantifies the loss of lift and increase in drag for the modified version of the MQ-1 predator UCAV along with the identification of stall conditions.local improvement(in drag)of up to 96%has been obtained by relocating external modifications,whereas global drag force reduction of roughly 0.5%is observed.The effects of external geometrical modifications on the control surfaces indicate the blanking phenomenon and reduction in forces on the control surfaces that can reduce the aerodynamic performance of the UCAV.
