Introduction Alpha-fetoprotein(AFP)is the most abundant serum protein found in the human fetus,produced by the yolk sac and the liver[1].The levels of maternal serum AFP reach a peak value at28-32 weeks of gestation and decrease rapidly after birth and usually drop to the normal at 8-12 months of *** normal serum AFP level for an adult isml[2].In contrast,livertumor cells usually synthesize and secrete an increased level of *** ultrasound and AFP are generally recommended as the screening modality for HCC for patients at risk for development of hepatocellular carcinoma(HCC)[1].AFP-L3,an isoform of AFP that binds Lens culinaris agglutinin,can be particularly useful in the early identification of aggressive HCC[2].These assays,however,can generate false-positive and false-negative AFP *** patients with advanced HCC,20%had normal AFP levels,whereas some patients with liver diseases had significant AFP elevation without liver cancer in long-term surveillance[3].Elevated AFP levels could be associated with active liver diseases with hepatocyte regeneration[4,5].In that setting,the AFP level will decline with improvement of the underlying liver *** case illustrated the dilemma and uncertainties as a result of persistent abnormal AFP values after extensive clinical investigations.

To expand the single-dose duration over which noninvasive clinical and preclinical cancer imaging can be conducted with high sensitivity,and well-defined spatial and temporal resolutions,a facile strategy to prepare ultrasmall nanoparticulate X-ray contrast media(nano-XRCM)as dual-modality imaging agents for positron emission tomography(PET)and computed tomography(CT)has been *** from controlled copolymerization of triiodobenzoyl ethyl acrylate and oligo(ethylene oxide)acrylate monomers,the amphiphilic statistical iodocopolymers(ICPs)could directly dissolve in water to afford thermodynamically stable solutions with high aqueous iodine concentrations(>140 mg iodine/ml water)and comparable viscosities to conventional small molecule *** formation of ultrasmall iodinated nanoparticles with hydrodynamic diameters of ca.10 nm in water was confirmed by dynamic and static light scattering *** a breast cancer mouse model,in vivo biodistribution studies revealed that the64Cu-chelator-functionalized iodinated nano-XRCM exhibited extended blood residency and higher tumor accumulation compared to typical small molecule imaging ***/CT imaging of tumor over 3 days showed good correlation between PET and CT signals,while CT imaging allowed continuous observation of tumor retention even after 10 days post-injection,enabling longitudinal monitoring of tumor retention for imaging or potentially therapeutic effect after a single administration of nano-XRCM.

Aim: The effectiveness of nutritional supplementation on drug-seeking behavior of amphetamine-addicted rats during withdrawal was investigated using a biased conditioned place preference (CPP) paradigm. Method: Twenty-four male Sprague-Dawley rats exhibiting baseline preference for the black chamber during a 20-minute pre-conditioning exploration of the CPP box completed the study. On alternate days of an 8-day schedule, twelve rats (Group AMP) were randomly selected, given either amphetamine sulfate (5 mg/ml, i.p.) and confined to the white chamber;or vehicle (1 ml saline, i.p.) and confined to the black chamber. A significant increased percentage time spent and number of entries made by Group AMP to the drug-paired, white chamber on the test day confirmed amphetamine addiction. Group AMP subsequently received increasing doses of amphetamine over 6 days. Following acute drug withdrawal, their CPP performance was compared with that of vehicle treated rats (Group SAL). Groups AMP and SAL were equally divided and randomly assigned to animals fed chow reconstituted with the nutritional supplement (AMP-S and SAL-S) over 8 weeks or standard rat chow (AMP-N and SAL-N). CPP performances for all rats were determined blindly from video recordings following this period. Results: Nutritionally supplemented, amphetamine withdrawn rats (AMP-S) exhibited significantly decreased percentage entries and time spent in the white chamber (p ?ve, control animals. Conclusion: Drug-seeking behavior by amphetamine-addicted animals was eliminated after treatment with a nutritionally supplemented diet.

Positive postprandial blood glucose responses have been related to low glycemic index (GI) and low glycemic load (GL) diets. The objective was to determine the effect of protein profile on glycemic response of low glycinin soymilk (high in β-conglycinin) (LGS) and conventional soymilk (S) in overweight and obese men. Twenty-four subjects, 23 - 45 years old, average BMI of 29 (26 - 38) with triglycerides ml of LGS, S with identical macronutrient content, with bovine milk (M) as reference. Fasting blood samples were followed by samples at 15, 30, 45, 60, 90, and 120 min after the milk consumption. One-day diet record, anthropometrics, and a demographic interview were completed. LGS and S presented a GI of (average ± standard deviation) 41.0 ± 29.9 and 40.4 ± 37.2, respectively. M showed a GI of 29.2 ± 25.3, however, treatment effect was not significant (P = 0.338). Similarly, GL for LGS (4.52 ± 3.29), S (4.44 ± 4.09) and M (2.33 ± 1.01) were not significantly different (P = 0.107). Postprandial glucose concentration curves for LGS, S and M presented the same tendency throughout 120 min (P = 0.331). Fasting blood glucose correlated with GI (r = -0.553;P = 0.032) and GL (r = -0.567;P = 0.028). LGS, S and M are low GI products and postprandial glycemic responses were not different. The high content of the protein β-conglycinin in LGS did not have an effect in postprandial blood glucose response in over-weight and obese men.

BACKGROUND Hepatitis B virus(HBV)nucleos(t)ide analog(NA)therapy reduces liver disease but requires prolonged therapy to achieve hepatitis B surface antigen(HBsAg)*** is limited North American real-world data using non-invasive tools for fibrosis assessment and few have compared 1st generation NA or lamivudine(LAM)to tenofovir disoproxil fumarate(TDF).AIM To assess impact of NA on virological response and fibrosis regression using liver stiffness measurement(LSM)(i.e.,FibroScan®).METHODS Retrospective,observational cohort study from the Canadian HBV *** collected included demographics,NA,HBV DNA,alanine aminotransferase(ALT),and *** were HBV monoinfected patients,treatment naïve,and received 1 NA with minimum 1 year *** In 465(median 49 years,37%female,35%hepatitis B e antigen+at baseline,84%Asian,6%White,and 9%Black).Percentage of 64(n=299)received TDF and 166 were LAM-treated with similar median duration of 3.9 and 3.7 years,*** mean baseline LSM was 11.2 kPa(TDF)vs 8.3 kPa(LAM)(P=0.003).At 5-year follow-up,the mean LSM was 7.0 kPa in TDF vs 6.7 kPa in LAM(P=0.83).There was a significant difference in fibrosis regression between groups(i.e.,mean-4.2 kPa change in TDF and-1.6 kPa in LAM,Pml)(n=170/190,89%)vs LAM-treated(n=35/58,60%)(P<0.05).None cleared *** In this real-world North American study,approximately 5 years of NA achieves liver fibrosis regression rarely leads to HBsAg loss.

目的研究血小板α粒子蛋白包括血小板反应素１（ｔｈｒｏｍｂｏｓｐｏｎｄｉｎ１，ＴＳＰ１），血小板因子４（ｐｌａｔｅｌｅｔｆａｃｔｏｒ４，ＰＦ４），β血小板球蛋白（βＴｈｒｏｍｂｏｇｌｏｂｕｌｉｎ，βＴＧ）和血小板源性生长因子（ｐｌａｔｅｌｅｔｄｅｒｉｖｅｄｇｒｏｗｔｈｆａｃｔｏｒ，ＰＤＧＦ）对巨核细胞生成的影响和可能的作用机制。方法用血浆凝块巨核细胞集落培养，观察对生长的影响，并进一步使用免疫组化和Ｗｅｓｔｅｒｎｂｌｏｔ证实受体的存在和对ｃｆｏｓ表达的影响。结果（１）ＴＳＰ１和ＰＦ４显著地抑制巨核细胞集落（ＣＦＵＭＫ）的形成，有意义的起始抑制浓度为２５μｇ／ｍｌ（Ｐ＜００５）。比较相同浓度的作用ＴＳＰ１和ＰＦ４有相似的抑制效应，但βＴＧ则无显著的影响。而ＰＤＧＦ显著地刺激巨核细胞的生成，最大的刺激剂量为５０ｎｇ／ｍｌ；（２）血小板生长因子（ＴＰＯ）（２０ｎｇ／ｍｌ）显著地增加ＣＦＵＭＫ的形成，但当加上ＴＳＰ１（５μｇ／ｍｌ）时。ＴＰＯ对巨核细胞的生长作用能被ＴＳＰ１减弱（７１±５５／２×１０５细胞～４１±８０／２×１０５细胞，Ｐ＜００１）；（３）证实骨髓的巨核细胞表达ＴＳＰ受体（Ｃ?

Essential oils are secondary metabolites whose biological properties have been studied with emphasis on antimicrobial activity. Hyptis carpinifolia Benth (Rosmaninho) is used in folk medicine in the fight against colds and rheumatism. The objective of this study was to evaluate the antimicrobial activity of the essential oil from H. carpinifolia. The essential oil was extracted by hydrodistillation using a modified Clevenger apparatus. The biological activity was determined using the Agar Cavity Diffusion technique to evaluate the effect of concentrations of 500, 250, 125, 62.5, 31.25, 15.62, 7.81 and 3.9 μg·ml-1 on the bacteria Escherichia coli, Listeria monocytogenes, Salmonella Choleraesuis, Staphylococcus aureus and Pseudomonas aeruginosa. The fungicidal potential was evaluated by the method of dilution in agar, and the percentage inhibition of mycelial growth of Alternaria alternata, Botrytis cinerea and Fusarium oxysporum was evaluated. The dilutions tested were 1000, 750, 500, 250 and 100 μL·L-1. The minimum inhibitory concentrations (MIC) of the essential oil for L. monocytogenes, S. aureus and E. coli were 31.25;15.62 and 62.5 μg·ml-1, respectively. The MIC was 500 μg·ml-1 for Salmonella Choleraesuis and P. aeruginosa. The percentage inhibitions of mycelial growth of B. cinerea and A. alternate were 67% and 33%, respectively, at the dilution of 750 μL·L-1. The essential oil inhibited about 48% of the mycelial growth of F. oxysporum, at the dilution of 1000 μL·L-1. The essential oil of H. carpinifolia presented biological activity against all the microorganisms evaluated.

目的建立早产猪呼吸窘迫综合征 (respiratory distress syndrome, RDS)模型并探讨猪肺表面活性物质(pulmonary surfactant, PS)制剂的治疗效果. 方法选择相当于足月孕期85%(94～97 d)的早产猪作RDS模型,随机分成4组(C组:对照组;C50组:PS 50 mg/kg;C100组:PS 100 mg/kg; N组:自主呼吸组).除N组外,其余早产猪气管切开插管,压力控制模式通气,吸入高浓度氧和标准潮气量通气.分别在治疗0～6 h测血气与呼吸力学参数,6 h后处死动物留取肺组织和肺泡灌洗液(bronchoalveolar lavage, BALF)进行形态学与生化分析. 结果 RDS动物BALF中总磷脂约10 mg/kg;电子显微镜下板层小体仅零星可见;光学显微镜下可见肺泡基本处于萎陷状态.C100组PaO2/FiO2由(92±37)上升到(235±137)mm Hg,肺顺应性由(0.48±0.08)上升到(0.76±0.29)ml/(cm H2O·kg),肺泡扩张均匀,少见损伤;C50组肺氧合及呼吸力学改善不明显. 结论早产猪RDS模型用于研究新生儿呼吸系统疾病具有一定价值,100 mg/kg的PS对肺氧合、呼吸力学及形态学的改善具有明显作用.
