Ligustroflavone ameliorates CCl_(4)-induced liver fibrosis through down-regulating the TGF-β/Smad signaling pathway

Liver fibrosis is a pathological process characterized by excess deposition of extracellular matrix(ECM)that are mainly derived from activated hepatic stellate *** studies suggested that ligustroflavone(LF)was an ingredient of Ligustrum lucidum *** activities of anti-inflammation and *** this study,we investigated whether LF had any effect on liver *** our study,we established a mouse model of carbon tetrachloride(CCl4)-induced liver fibrosis and used TGF-β1-stimulated human hepatic stellate cell line(LX-2)to explore the effect of LF and associated underlying *** was used in vivo with low dose(L-LF,5 mg·kg^(-1),i.p.,3 times each week)and high dose(H-LF,20 mg·kg^(-1),i.p.,3 times each week)and in vitro(25μmol·L^(-1)).Histopathological and biochemical assays investigations showed that LF delayed the formation of liver fibrosis;decreased AST,ALT activities and increased Alb activity in serum;decreased Mda level,Hyp content and increased GSH-Px concentration,SOD activity in liver ***,immunohistochemical,immunofluorescent and Western blot results showed that LF reduced the expressions of hepatic stellate cells specific marker proteins,including collagen I andα-SMA in vivo and in *** addition,LF markedly suppressed TGF-β1-upregulated protein expressions of TβR I,TβR II,P-Smad2,P-Smad3 and Smad4 in LX-2 *** together,these findings demonstrated LF could decrease histopathological lesions,ameliorate oxidative injury,attenuate CCl4-induced liver fibrosis,which may be associated with down-regulating the TGF-β/Smad signaling pathway.

An algorithm for decomposing a polynomial system into normal ascending sets

We present an algorithm to decompose a polynomial system into a finite set of normal ascending sets such that the set of the zeros of the polynomial system is the union of the sets of the regular zeros of the normal ascending sets. If the polynomial system is zero dimensional, the set of the zeros of the polynomials is the union of the sets of the zeros of the normal ascending sets.

Principle of equivalency of bed structures and bed load motion

Step-pool systems and cobble clusters are structures composed of boulders and cobbles on mountain streambeds rearranged by flood flow to reach high resistance and high bed *** bed structures and bed load motion can protect the riverbed from *** each step-pool,the flow energy is transformed into turbulence,and finally into *** load motion also consumes flow energy and plays a role to protect the bed from *** collision of bed load particles with the bed results in a force,known as dispersive force,which balances the lift force and controls the erosion of bed *** investigations and field experiments were conducted in the Xiaojiang River basin on the Yunnan-Guizhou plateau of China,where there were incised streams and stable streams with bed load motion or with step-pool *** study reveals that for a given stream power, strong bed structures are associated with low or zero bed load transportation;and weak or no bed structures are associated with intensive bed load *** showed that for incised streams, the final bed profiles were the same if there was bed load motion or there were bed *** key stones that made up the bed structures,for instance the large boulders in steps,were removed,the flow immediately scoured the sediment *** bed load transportation sharply increased by 100 times and the median diameter of bed load increased by 2-20 *** structures and bed load motion are mutually replaceable for their effects on flow energy consumption and streambed incision *** is the principle of equivalency of bed load motion and bed *** is due to the principle that there was no bed load motion in the Yalutsangpo Grand Canyon,where a very strong step-pool system had developed,although the bed gradient and shear stress of flow were extremely high.A possible application of this principle for incision control of the downstream reaches of the Three Gorges dam is also discussed in t

Two-stage urethroplasty is a better choice for proximal hypospadias with severe chordee after urethral plate transection： a single-center experience

It is still debatable whether single- or two-stage urethroplasty is a more suitable technique for treating hypospadias with severe chordee after urethral plate transection. This retrospective study evaluated these two techniques. A total of 66 patients of proximal hypospadias with severe chordee were divided into two groups according to the techniques they underwent： 32 and 34 patients underwent single-stage （Duckett） or two-stage urethroplasty, respectively. Median ages at presentation were 7.5 years and 11.0 years in single-stage and two-stage repair groups, respectively. Median follow-ups were 28.5 months （20-60 months） and 35 months （18-60 months） in the single-stage and two-stage groups, respectively. The meatus of the neourethra was located at the top of the glans in all patients. No recurrence of chordee was found during follow-up, and all patients or parents were satisfied with the penile length and appearance. Complications were encountered in eight patients in both groups, with no statistically significant differences between the two techniques. The late complication rate of stricture was higher after the single-stage procedure （18.75% vs 0%）. The complication rate after single-stage repairs was significantly lower in the prepubescent subgroup （10.52%） than in the postpubescent cohort （46.15%）. These results indicate that the urethral plate transection effectively corrects severe chordee associated with proximal hypospadias during the intermediate follow-up period. Considering the higher rate of stricture after single-stage urethroplasty, two-stage urethroplasty is recommended for proximal hypospadias with severe chordee after urethral plate transection.

Drug discovery is a crucial part of human healthcare and has dramatically benefited human lifespan and life quality in recent centuries, however, it is usually time-and effort-consuming. Structural biology has been demonstrated as a powerful tool to accelerate drug development. Among different techniques, cryo-electron microscopy(cryo-EM) is emerging as the mainstream of structure determination of biomacromolecules in the past decade and has received increasing attention from the pharmaceutical industry. Although cryo-EM still has limitations in resolution, speed and throughput, a growing number of innovative drugs are being developed with the help of cryo-EM. Here, we aim to provide an overview of how cryo-EM techniques are applied to facilitate drug discovery. The development and typical workflow of cryo-EM technique will be briefly introduced, followed by its specific applications in structure-based drug design, fragment-based drug discovery, proteolysis targeting chimeras, antibody drug development and drug repurposing. Besides cryo-EM, drug discovery innovation usually involves other state-of-the-art techniques such as artificial intelligence(AI), which is increasingly active in diverse areas. The combination of cryo-EM and AI provides an opportunity to minimize limitations of cryo-EM such as automation, throughput and interpretation of mediumresolution maps, and tends to be the new direction of future development of cryo-EM. The rapid development of cryo-EM will make it as an indispensable part of modern drug discovery.
