A biosynthetic gene cluster for the bioactive fungal sesterterpenoids variecolin(1)and variecolactone(2)was identified in Aspergillus aculeatus ATCC *** production of 1 and 2 was achieved in Aspergillus oryzae by expressing the sesterterpene synthase VrcA and the cytochrome P450 ***,the replacement of VrcB with homologous P450s from other fungal terpenoid pathways yielded three new variecolin analogues(5-7).Analysis of the compounds’anticancer activity in vitro and in vivo revealed that although 5 and 1 had comparable activities,5 was associated with significantly reduced toxic side effects in cancer-bearing mice,indicating its potentially broader therapeutic *** study describes the first tests of variecolin and its analogues in animals and demonstrates the utility of synthetic biology for creating molecules with improved biological activities.

Retinitis pigmentosa(RP)is a group of heredofamilial retinal diseases which is characterized by night blindness and progressive visual field *** study aims to study the treatment effect of Lycium barbarum(LB)on retinal functions and structure of RP *** study is a double-masked randomized controlled *** subjects received scheduled eye examination including visual acuity(VA),Humphrey field analysis(HFA),ganzfeld flash electroretinogram(ffERG)and optical coherence tomography(OCT).The suitable subjects were randomly allocated to either LB-treatment or placebo groups with the supply of LB or placebo for 12 *** were total 41 RP subjects(22 in LB group and 19 in placebo group)completed the 12 months *** compliance rates for LB and placebo groups were 89.8%±12.5%and 85.3%±7.7%*** compared with placebo group,there were no deteriorations of both high and low contrast VA in LB group(P<0.01).In addition,certain improvements of scotopic rod response and photopic cone response of ffERG were obtained in LB group(P<0.05).In the OCT measurement,an obvious thinning of macular thickness was observed in placebo group but not found in LB group(P<0.05).However,there were no changes found in the sensitivity of central visual field between two *** results confirm that the 12-month LB treatment for RP patients had neuroprotective effect on retina and is believed to delay or minimize the deterioration of visual function in RP.

An organic-inorganic hybrid cathode interfacial layer(CIL) was developed by doping ZnO with the naphthalene-diimide based derivative NDI-PFNBr. It was found the resulting organic-inorganic hybrid CIL showed apparently improved conductivity and could act as an effective cathode interlayer to modify indium tin oxide(ITO) transparent electrodes. As a result, by employing the blend of PTB7-Th:PC71BM as the photoactive layer, the inverted polymer solar cells(PSCs) exhibited a remarkable enhancement of power conversion efficiency(PCE) from 8.52% for the control device to 10.04% for the device fabricated with the hybrid CIL. Moreover, all device parameters were simultaneously improved by using this hybrid CIL. The improved open-circuit voltage(VOC) was attributed to the reduced work function of the ITO cathode, whereas the enhancements in fill factor(FF) and short-circuit current density(JSC) were assigned to the increased conductivity and more effective charge extraction and collection at interface. Encouragingly, when the thickness of the hybrid CIL was increased to 80 nm, the resulting device could still keep a PCE of 8.81%, exhibiting less thickness dependence. Considering these advantages, 16 and 93 cm2large-area PSCs modules were successfully fabricated from the hybrid CIL by using doctor-blade coating techniques and yielded a remarkable PCE of8.05% and 4.49%, respectively. These results indicated that the hybrid CIL could be a promising candidate to serve as the cathode interlayer for high-performance large-area inverted PSCs.

Life skill-based training is essential for intellectual disabled individuals to regain autonomy and social *** reality(VR)could deliver a nascent application that is more engaging and *** objective of this study was to evaluate the training effects of the VR-based multiple life skill training program on life skill perfor-mance,self-efficacy,memory,cognitive and behavioral functions via a multicenter randomized controlled trial.A total of 145 intellectual disabled participants were recruited for a randomized controlled trial with three in-tervention arms:VR(n=42);traditional(n=53);and control(n=50).The life skill tasks for the interventions included 1)grocery shopping,2)cooking,and 3)kitchen *** measures were performance scores for the three tasks,self-efficacy scale,digit span score,and Frontal Assessment Battery score,graded by blinded ***-after effects of each group were evaluated using separated Wilcoxon Signed-rank *** sig-nificantly improved cooking,cleaning performance,and memory span,while traditional training significantly improved shopping,cooking,and cleaning *** generalized estimating equation evaluated effects between groups adjusted for age,gender and intelligence quotient(IQ).VR had significantly larger improvement effects in cooking and cleaning than the control group and memory span compared to traditional training and controls *** appeared to be a significant confounder on the training *** work may consider developing artificial intelligence to customize programs depending on IQ level.

A bis-benzospiropyran（BBSP） and nitro-substituted indoline spirobenzopyrans（SBP） were prepared by following a simple procedure. The reaction mechanism of synthesizing bis-benzospiropyran is discussed. A comparative analysis of the spectral and photochromic properties between SBP and BBSP was carried out with the aid of a PMMA film. The results indicate that chemical structures and UV irradiation played an important role in the spectral and photochromic properties of SBP and BBSP, which would readily affect λmax of the colored forms and the reversibility. The IR spectra and ^1H NMR spectra of these compounds are used to interpret the photochromic phenomena under different UV irradiation and determine the configurations of SBP and BBSP.

We evaluated the efficacy and safety of as-needed tadalafil in a diverse clinical population （with varying patient demographics, disease severity, and comorbid medical conditions） of Asian men with erectile dysfunction （ED）. An integrated analysis of five double-blind, placebo-controlled trials （N = 1 046） was performed. Patients were randomly assigned to receive 10 mg tadalafil （N = 185）, 20 mg tadalafil （N = 510）, or placebo （N = 351）. Efficacy assessments included the International Index of Erectile Function （IIEF）, Sexual Encounter Profile （SEP） diary and Global Assessment Question （GAQ）. Patients receiving 10 mg or 20 mg tadalafil showed significant improvement from baseline-to-end point on the Erectile Function domain of the International Index of Erectile Function （IIEF-EF） domain score in all clinical sub-populations analyzed, compared with patients receiving placebo （P 〈 0.001）. The 10-mg and 20-mg tadalafil groups showed a mean success rate of 64.1% and 70.5% for sexual intercourse attempts （SEP3, successful intercourse）, respectively, compared with 33.4% in the placebo group （P 〈 0.001）, and 85.5% and 85.4% reported improved erections at end point GAQ, respectively, versus 43.5% in the placebo group （P 〈 0.001）. Tadalafil was well tolerated across all groups studied. Headache and back pain were the most frequently reported adverse events. Overall, tadalafil was effective and well tolerated across a diverse clinical spectrum of Asian men with ED.

The mean sea surface heights (sea surface topography) of the South China, East China, Yellow and Bohai Seas are derived from an ocean general circulation model and surface air pressure. The circulation model covers the global oceans, with fine grid (1/6°)? covering the East Asian marginal seas and coarse grid (3°) covering the rest part of the global oceans. The result shows that the China 1985 Na-tional Altitude Datum is 24.7 cm above the mean sea surface height of the world oceans. The mean sea surface in the coastal ocean adjacent to China is higher in the south than in the north. Intercomparison of the model results with the geodetic leveling measurements at 28 coastal tidal stations shows a standard deviation of 4.8 cm and a fitting coefficient of 95.3%. After correction through linear regression, the standard deviation is reduced to 4.5 cm. This indicates that the accuracy of model results is sufficient for practical application. Association between smoking,pancreatic insulin secretion and insulin resistance in Chinese subjects with or without glucose intolerance

Background There are studies suggesting smoking may increase the risk of type 2 diabetes. Effects of smoking on insulin secretion and insulin resistance （IR） are, however, controversial. Methods This is a cross-sectional study. Since there were very few smokers among Hong Kong Chinese women, only men （n=-1068） were analyzed in this report. Fasting and 2-hour plasma glucose and insulin were measured. Insulinogenic index as well as beta-cell function and IR based on homeostatic model assessment （HOMA） by computer model （HOMA Calculator v2.2） were calculated. Results Of the 1068 men, 147 had newly diagnosed diabetes, 131 newly diagnosed impaired glucose tolerance （IGT） and 790 were non-diabetic normal controls. Smokers had similar fasting and 2-hour insulin levels, insulinogenic index and HOMA derived beta-cell function as compared to non-smokers in the groups with diabetes, IGT or normal oral glucose tolerance test （OGTT）. IR was also similar between smokers, ex-smokers and non-smokers in those with normal OGTT. In men with IGT or diabetes, after adjustment for age and body mass index, smokers were more insulin resistant as compared to non-smokers （IR, IGT： 1.59±1.07 vs 1.03±0.54, P〈0.05; diabetes： 1.96±1.36 vs 1.06±0.45, P〈0.01）. With Logistic regression analysis, comparing smokers and non-smokers, IR was independently associated with smoking （odds ratio （95% C/）, IGT： 2.23 （1.05, 4.71）; diabetes： 3.92 （1.22, 12.58））. None of the other insulin parameters enter into the model among those with normal OGTT or comparing ex-smokers and non-smoker or smokers and ex-smokers. Conclusions In Chinese men, smoking did not show any direct association with insulin levels and pancreatic insulin secretion. Smoking men with IGT or diabetes appeared more insulin resistant than their non-smoking counterparts.
