Objective:To explore the possible effects and mechanism of Zhizhu Decoction(ZZD)on the pathophysiology of slow transit constipation(STC).Methods A total of 54 C57BL/6 mice was randomly divided into the following 6 groups by a random number table,including control,STC model(model),positive control,and low-,medium-and high-doses ZZD treatment groups(5,10,20 g/kg,namely L,M-,and H-ZZD,respectively),9 mice in each group.Following 2-week treatment,intestinal transport rate(ITR)and fecal water content were determined,and blood and colon tissue samples were collected.Hematoxylin-eosin and periodic acid-Schiff staining were performed to evaluate the morphology of colon tissues and calculate the number of goblet cells.To determine intestinal permeability,serum levels of lipopolysaccharide(LPS),low-density lipoprotein(LDL)and mannose were measured using enzyme-linked immunosorbent assay(ELISA).Western blot analysis was carried out to detect the expression levels of intestinal tight junction proteins zona-occludens-1(ZO-1),claudin-1,occludin and recombinant mucin 2(MUC2).The mRNA expression levels of inflammatory cytokines including tumor necrosis factor(TNF)-α,interleukin(IL)-1β,IL-6,IL-4,IL-10 and IL-22 were determined using reverse transcription-quantitative reverse transcription reaction.Colon indexes of oxidative stress were measured by ELISA,and protein expression levels of colon silent information regulator 1/forkhead box O transcription factor 1(SIRT1/FoxO1)antioxidant signaling pathway were detected by Western blot.Results Compared with the model group,ITR and fecal moisture were significantly enhanced in STC mice in the M-ZZD and H-ZZD groups(P<0.01).Additionally,ZZD treatment notably increased the thickness of mucosal and muscular tissue,elevated the number of goblet cells in the colon of STC mice,reduced the secretion levels of LPS,LDL and mannose,and upregulated ZO-1,claudin-1,occludin and MUC2 expressions in the colon in a dose-dependent manner,compared with the model group(P<0.05 or P<0.01).In addition,ZZD si

Electrolyte effect on electrochemical behaviors of manganese fluoride material for aqueous asymmetric and symmetric supercapacitors

Exploring wide voltage window materials is not only an available measure to enhance the energy density of hybrid supercapacitor(HSCs),but also avoids the dynamic mismatch caused by different energy storage mechanisms of two electrodes in assembled symmetrical HSC.However,there are few reports about the wide potential window materials except Bi_(2)O_(3)and VO_(2).Therefore,the MnF_(2)synthesized by solvothermal reaction was served as the electrode for HSC.The MnF_(2)exhibited electrochemical activity in alkaline solution in three-electrode system,especially with a wide potential window from-0.8 to+0.5 V in 2 mol·L^(-1)NaOH.Furthermore,the assembled MnF_(2)//MnF_(2)symmetrical HSC had a potential window of 1.5 V,and it exhibited outstanding long-cycle capability.Meanwhile,when MnF_(2)was taken as the negative and positive respectively,the potential windows of asymmetric devices CoMoO_(4)//MnF_(2)and MnF_(2)//Activated Carbon(AC)could reach 1.3 and 1.45 V,respectively,showing excellent cycle stability.This work shows that MnF_(2)material has great research value in HSC,and provides a new research direction for developing high-performance devices.

Biotoxicity dynamic change and key toxic organics identification of coal chemical wastewater along a novel full-scale treatment process

It is particularly important to comprehensively assess the biotoxicity variation of industrial wastewater along the treatment process for ensuring the water environment security.However,intensive studies on the biotoxicity reduction of industrial wastewater are still limited.In this study,the toxic organics removal and biotoxicity reduction of coal chemical wastewater(CCW)along a novel full-scale treatment process based on the pretreatment process-anaerobic process-biological enhanced(BE)process-anoxic/oxic(A/O)process-advanced treatment process was evaluated.This process performed great removal efficiency of COD,total phenol,NH_(4)^(+)-N and total nitrogen.And the biotoxicity variation along the treatment units was analyzed from the perspective of acute biotoxicity,genotixicity and oxidative damage.The results indicated that the effluent of pretreatment process presented relatively high acute biotoxicity to Tetrahymena thermophila.But the acute biotoxicity was significantly reduced in BE-A/O process.And the genotoxicity and oxidative damage to Tetrahymena thermophila were significantly decreased after advanced treatment.The polar organics in CCW were identified as the main biotoxicity contributors.Phenols were positively correlated with acute biotoxicity,while the nitrogenous heterocyclic compounds and polycyclic aromatic hydrocarbons were positively correlated with genotoxicity.Although the biotoxicity was effectively reduced in the novel full-scale treatment process,the effluent still performed potential biotoxicity,which need to be further explored in order to reduce environmental risk.

Overview of enhancing biological treatment of coal chemical wastewater:New strategies and future directions

Coal chemical wastewater(CCW)is a type of refractory industrial wastewater,and its treatment has become the main bottleneck restricting the sustainable development of novel coal chemical industry.Biological treatment is considered as an economical,effective and environmentally friendly technology for CCW treatment.However,conventional biological process is difficult to achieve the efficient removal of refractory organics because of CCW with the characteristics of composition complexity and high toxicity.Therefore,seeking the novel enhancement strategy appears to be a favorable solution for enhancing biological treatment efficiency of CCW.This review focuses on presenting a comprehensive picture about the exogenous enhancement strategies for CCW biological treatment.The performance and potential application of exogenous enhancement strategies,including co-metabolic substrate enhancement,biofilm filler enhancement,adsorption material enhancement and conductive mediator enhancement,were expounded.Meanwhile,the enhancing mechanisms of different strategies were comprehensively discussed from a biological perspective.Furthermore,the prospects of enhancement strategies based on the engineering performance,economic cost and environmental impact(3E)evaluation were introduced.And novel enhancement strategy based on“low carbon emissions”,“resource recycling”and“water environment security”in the context of carbon neutrality was proposed.Taken together,this review provides technical reference and new direction to facilitate the regulation and optimization of typical industrial wastewater biological treatment.

Coupling of RANS and BEM Solvers for Simulation of Propeller Behind the Hull in Full Appended Model

To design a propeller for ship power plant,the interaction between ship hull and propeller must be taken into account.The main concern is to apply the wake effect of ship stern on the propeller performance.In this paper,a coupled BEM(Boundary Element Method)/RANS(Renolds-Averaged Navier−Stokes)solver is used to simulate propeller behind the hull in the self-propulsion test.The motivation of this work is to develop a practical tool to design marine propulsion system without suffering long computational time.An unsteady boundary element method which is also known as panel method is chosen to estimate the propeller forces.Propeller wakes are treated using a time marching wake alignment method.Also,a RANS code coupled with VoF equation is developed to consider the ship motions and wake field effects in the problem.A coupling algorithm is developed to interchange ship wake field to the potential flow solver and propeller thrust to the RANS code.Based on the difference between hull resistance and the propeller thrust,a PI controller is developed to compute the propeller RPM in every time step.Verification of the solver is carried out using the towing tank test report of a 50 m oceanography research vessel.Wake factor and trust deduction coefficient are estimated numerically.Also,the wake rollup pattern of the propeller in open water is compared with the propeller in real wake field.

Mucosal immune responses to Ichthyophthirius multifiliis in the ocular mucosa of rainbow trout(Oncorhynchus mykiss,Walbaum),an ancient teleost fish

The eye,as a specialized visual organ,is directly exposed to the external environment,and,therefore,it faces constant chal-lenges from external pathogenic organisms and toxins.In the ocular mucosa(OM)of mammals,mucosal-associated lymphoid tissues(MALTs)constitute the primary line of defense.However,the immune defense role of the OM remains unknown in aquatic vertebrates.To gain insights into the immune processes within the OM of teleost fish,we developed an infection model of rainbow trout(Oncorhynchus mykiss)OM using a parasite,Ichthyophthirius multifiliis(Ich).Immunofluorescence,qPCR,and H&E staining revealed that Ich successfully infiltrates the OM of rainbow trout,leading to pathological struc-tural changes,as evidenced by A&B staining.Importantly,the qPCR results indicate an up-regulation of immune-related genes following Ich infection in the OM.Moreover,transcriptome analyses were conducted to detect immune responses and impairments in eye function within the OM of rainbow trout with Ich infection.The results of the transcriptome analysis that Ich infection can cause an extensive immune response in the OM,ultimately affecting ocular function.To the best of our knowledge,our findings represent for the first time that the teleost OM could act as an invasion site for parasites and trigger a strong mucosal immune response to parasitic infection.

LetΩbe a bounded smooth domain in RN(N≥3).Assuming that 00 are constants,we consider the existence results for positive solutions of a class of fractional elliptic system below,{(a+b[u]^(2)_(s))(-Δ)^(s)u=vp+h_(1)(x,u,v,▽u,▽v),x∈Ω,(-Δ)^(s)v=u^(q)+h_(2)(x,u,▽,▽u,▽v),x∈Q,u,v>0,x∈Ω,u=v=0,x∈RN\Ω.Under some assumptions of hi(x,u,v,▽u,▽v)(i=1,2),we get a priori bounds of the positive solutions to the problem(1.1)by the blow-up methods and rescaling argument.Based on these estimates and degree theory,we establish the existence of positive solutions to problem(1.1).

Effect of Frequency on Degradation in BOPP Films Under Repetitively Pulsed Voltage

Operating conditions of film capacitors are complex,and the problem of film insulation failure caused by repetitively pulsed voltage is becoming ever serious.Degradation of the film under repetitively pulsed voltage cannot be accurately evaluated by the average breakdown electric field.In this paper,the effects of pulsed electric field and pulse repetition frequency on the breakdown in biaxially oriented polypropylene(BOPP)films are investigated.Three phases of BOPP degradation are proposed based on the voltage amplitude,i.e.,maintenance(M),decline(D),and near-zero(N).Evolution of the BOPP film from degradation to breakdown at different frequencies is presented.Meanwhile,transition of discharge mode and elemental composition of the film are analyzed.Experimental results show continuous heat generation under repetitive microsecond pulses is the dominant factor for degradation of BOPP film.The number of applied pulses and the repetitive stressing time decrease exponentially with increase of frequency.This research can be contributed to the safe and reliable operation of capacitors.

Distribution of per-and polyfluoroalkyl substances in blood,serum,and urine of patients with liver cancer and associations with liver function biomarkers

Studies have shown that per-and polyfluoroalkyl substances(PFASs)may be hepatotoxic in animals or humans.However,data on clinical epidemiology are very limited.In this study,21PFASs were determined in patients with liver diseases,with the highest median concentrations detected in the serum sample(26.7 ng/mL),followed by blood(10.7 ng/mL)and urine(5.02 ng/mL).Higher total PFAS concentrations were found in hepatocellular carcinoma(HCC)patients compared to non-HCC patients,with significant discrepancies in serum and blood samples.Besides,significant correlations were also found among PFAS concentrations and age,gender,body mass index(BMI),and liver function biomarkers levels.For example,PFAS concentrations are significantly higher in males than in females;Several serum PFASs concentrations increase with age and BMI,while the serum perfluorohexane sulfonic acid(PFHxS)concentrations are negatively correlated with age.In addition,multiple regression models adjusted for age,gender and BMI found that increased serum perfluorobutane sulfonic acid(PFBS),perfluoroheptane sulfonic acid(PFHpS)and perfluorohexylphosphonic acid(PFHxPA)conentrations are correlated with elevated alkaline phosphatase(ALP),aspartate aminotransferase(AST),and alpha-fetoprotein(AFP)(p<0.05).Our results provide epidemiological support for the future study on the potential clinical hepatotoxicity of PFAS.

Inhibition of SLC26A4 regulated by electroacupuncture suppresses the progression of myocardial ischemia-reperfusion injury

Introduction:Myocardial ischemia-reperfusion(IR)injury has received widespread attention due to its damaging effects.Electroacupuncture(EA)pretreatment has preventive effects on myocardial IR injury.SLC26A4 is a Na+independent anion reverse transporter and has not been reported in myocardial IR injury.Objectives:Toﬁnd potential genes that may be regulated by EA and explore the role of this gene in myocardial IR injury.Methods:RNA sequencing and bioinformatics analysis were performed to obtain the differentially expressed genes in the myocardial tissue of IR rats with EA pretreatment.Myocardial infarction size was detected by TTC staining.Serum CK,creatinine kinase-myocardial band,Cardiac troponin I,and lactate dehydrogenase levels were determined by ELISA.The effect of SLC26A4 on cardiomyocyte apoptosis was explored by TUNEL staining and western blotting.The effects of SLC26A4 on inﬂammation were determined by HE staining,ELISA,and real-time PCR.The effect of SLC26A4 on the NF-κB pathway was determined by western blotting.Results:SLC26A4 was up-regulated in IR rats but downregulated in IR rats with EA pretreatment.Compared with IR rats,those with SLC26A4 knockdown exhibited improved cardiac function according to decreased myocardial infarction size,reduced serum LDH/CK/CK-MB/cTnI levels,and elevated left ventricular ejection fraction and fractional shortening.SLC26A4 silencing inhibited myocardial inﬂammation,cell apoptosis,phosphorylation,and nuclear translocation of NF-κB p65.Conclusion:SLC26A4 exhibited promoting effects on myocardial IR injury,while the SLC26A4 knockdown had an inhibitory effect on the NF-κB pathway.These results further unveil the role of SLC26A4 in IR injury.
