Defatted Hickory Meal Hydrolysate's Impact on Memory Impairment Induced by D-Galactose in Mice

Defatted hickory meal (DHM)， a by-product of hickory oil production， is a protein source rich in essential amino acids. In this study， the functional properties of DHM hydrolysate (DHMH) were assessed using in vitro and in vivo assays in context to its antioxidant and memory-enhancing effects. To induce memory impairment， D-galactose (D-gal) was administered to mice at a dose of 120 mg/kg body weight per day， and DHMH was orally administered at doses of 300， 600， and 1000 mg/kg body weight per day for 8 weeks. DHMH treatment led to improved memory performance in D-gal-induced memory-impaired mice， as observed in the Morris water maze test. Furthermore， DHMH mitigated the accumulation of amyloid beta 1-42 triggered by D-gal exposure. Notably， high-dose DHMH significantly reduced the elevation of pro-inflammatory markers， including tumor necrosis factor alpha， interleukin 1β， and interleukin 6. Additionally， DHMH prevented the decline in total superoxide dismutase activity， glutathione peroxidase activity， and glutathione levels， while reducing malondialdehyde content in D-gal-induced mice， indicative of its antioxidant properties. Moreover， DHMH treatment effectively prevented histological alterations in neurons within the hippocampal CA1 area induced by D-gal. Collectively， our findings suggest that DHMH may counteract memory dysfunctions resulting from oxidative stress injury in the brain， positioning it as a potential candidate for use as a functional food.

Numerous studies have shown that cell replacement therapy can replenish lost cells and rebuild neural circuitry in animal models of Parkinson’s disease.Transplantation of midbrain dopaminergic progenitor cells is a promising treatment for Parkinson’s disease.However,transplanted cells can be injured by mechanical damage during handling and by changes in the transplantation niche.Here,we developed a one-step biomanufacturing platform that uses small-aperture gelatin microcarriers to produce beads carrying midbrain dopaminergic progenitor cells.These beads allow midbrain dopaminergic progenitor cell differentiation and cryopreservation without digestion,effectively maintaining axonal integrity in vitro.Importantly,midbrain dopaminergic progenitor cell bead grafts showed increased survival and only mild immunoreactivity in vivo compared with suspended midbrain dopaminergic progenitor cell grafts.Overall,our findings show that these midbrain dopaminergic progenitor cell beads enhance the effectiveness of neuronal cell transplantation.

Soft electrochemical actuators that can generate mechanical motions in response to electrical stimuli can be used as artificial muscles in microscale robots.The electrode materials are the key component of the electrochemical actuator because they limit the ion diffusion and accumulation processes and thus affect the deformation in fast response-time and potential engineering applications.Low-dimensional materials,such as one-dimensional(1D)CNT and two-dimensional(2D)vertically stood WS_(2),are great electrode due to their good electrochemical stability and high surface-to-volume ratios,but show limited performances when being used on their own in the actuators because of low reconstruction or low conductivity.A combination of 1D CNT and 2D WS_(2)could form a heterojunction with both high electrochemical activity and great mechanical stability.Herein,by depositing highly disordered graphene nanosheets(HDGNs)on CNT films and then WS_(2)on the formed low-dimensional nanocarbon film,a hybrid WS_(2)/HDGN/CNT film was synthesized through a multistep CVD method.Vertically grown WS_(2)nanosheets were achieved by taking advantage of HDGNs which limits CNTs sliding.So,the volume expansion caused by the transition from WO_(3)to WS-2can force the vertical growth of WS_(2).This hybrid film shows excellent electrochemical properties,such as high specific capacitance and high rate performance.Soft actuators based on the WS_(2)/HDGN/CNT films show good actuating performance,including stability and reconfigurability.

BACKGROUND Cognitive reserve(CR)and the catechol-O-methyltransferase(COMT)Val/Met polymorphism are reportedly linked to negative symptoms in schizophrenia.However,the regulatory effect of the COMT genotype on the relationship between CR and negative symptoms is still unexamined.AIM To investigate whether the relationship between CR and negative symptoms could be regulated by the COMT Val/Met polymorphism.METHODS In a cross-sectional study,54 clinically stable patients with schizophrenia underwent assessments for the COMT genotype,CR,and negative symptoms.CR was estimated using scores in the information and similarities subtests of a short form of the Chinese version of the Wechsler Adult Intelligence Scale.RESULTS COMT Met-carriers exhibited fewer negative symptoms than Val homozygotes.In the total sample,significant negative correlations were found between negative symptoms and information,similarities.Associations between information,similarities and negative symptoms were observed in Val homozygotes only,with information and similarities showing interaction effects with the COMT genotype in relation to negative symptoms(information,β=-0.282,95%CI:-0.552 to-0.011,P=0.042;similarities,β=-0.250,95%CI:-0.495 to-0.004,P=0.046).CONCLUSION This study provides initial evidence that the association between negative symptoms and CR is under the regulation of the COMT genotype in schizophrenia.

As a complementary and alternative medicine(CAM),traditional Chinese medicine(TCM)represents an established medical system with a rich history and abundant clinical experience.TCM is an empirical medicine,the process of which is analogous to ChatGPT’s learning and development model.In TCM,inquiry is a relatively objective way of traditional syndrome differentiation.Although various artificial intelligence systems related to TCM consultation exist,their interactive abilities remain limited.The study standardized the primary complaint and instructed ChatGPT to simulate a TCM practitioner,conducting three comprehensive inquiry tests.The results yielded unexpected conclusions,revealing that ChatGPT could simulate a TCM practitioner’s inquiry with patients,confirming its potential in the field of TCM inquiry.However,current applications still pose certain limitations and risks.Hence,to integrate ChatGPT-like language models with traditional TCM AI to establish an associative mode that can facilitate TCM diagnosis and treatment with more convenience and standardization is crucial,yet at the same time,it should be treated very carefully.

Objectives:Sciadopitysin(SP)is aﬂavonoid in Ginkgo biloba that exhibits various pharmacological activities.This study aimed to investigate its antitumor effects and the underlying molecular mechanism of SP in hepatocellular carcinoma(HCC)cells.Methods:Network pharmacology was used for target prediction analysis.Cell Counting Kit-8(CCK-8)assay was used to test the cell viability.Flow cytometry was used to test the cell cycle distribution,apoptosis status,and reactive oxygen species(ROS)levels.Transwell and wound-healing assay was used to test the migration effect of SP on HepG2 cells.Western Blot assay was used to test the expression levels of proteins.Results:Network pharmacology analysis results showed that the mitogen-activated protein kinase(MAPK)and other signaling pathways are involved in the SP anti-HCC biological process.CCK-8 assay results demonstrated that SP showed an obvious killing effect on three types of HCC cells and low cytotoxic effect on normal cells.Western Blot andﬂow cytometry results showed that SP regulated MAPK/signal transducer and activator of transcription 3(STAT3)/nuclear factor kappa-B(NF-κB)signaling pathway to induce mitochondrion-dependent apoptosis in HepG2 cells.Additionally,SP can arrest the G0/G1 phase cell cycle via the protein kinase B(AKT)/p21/p27/cyclin-dependent kinase(CDK)/Cyclin signaling pathway.Wound healing and transwell assays showed that SP inhibited cell motility and invasion through the AKT/glycogen synthase kinase3β(GSK-3β)/vimentin/β-catenin signaling pathway.Conclusion:Theseﬁndings demonstrated that SP induced mitochondrion-dependent apoptosis,arrested cell cycle in the G0/G1 phase,and inhibited cell migration by regulating the ROS-mediated signaling pathway in HepG2 cells.Thus,SP could serve as a therapeutic agent for the treatment of human HCC.

Background:Radix Aconiti Lateralis Preparata(Fu-zi)is a traditional Chinese medicinal herb,which has been widely used in the clinic and has potent anti-inflammatory activities.we aimed to explore the mechanisms of extract containing alkaloids from different Fu-zi Processed Products(FPP)in treating inflammation,especially rheumatoid arthritis(RA).Methods:Firstly,using network pharmacology technology,the ingredients,and targets of Fu-zi were obtained by searching and screening,the targets involving RA were acquired,the intersection targets were constructed a"component-target-pathway"network.A comprehensive investigation was conducted on the anti-rheumatoid arthritis mechanisms of 5 FPPs in lipopolysaccharide(LPS)induced RAW264.7 cells,which serve as a model for RA.The production of NO and inflammatory cytokines were measured by ELISA kit.Quantitative Real-time PCR(qRT-PCR)was utilized to measure the mRNA levels.COX-2/PGE2 signaling pathway-associated proteins were determined by western blot.Results:According to a network pharmacological study,16 chemical components and 43 common targets were found in Fu-zi and 6 key targets including PTGS2 were closely related to the mechanism of Fu-zi in treating RA.The in vitro study revealed that the levels of NO,TNF-α,and IL-1βwere substantially decreased by the 5 FPPs.The 5 FPPs significantly suppressed the expression of proteins COX-2,iNOS,and NF-κB,with particularly notable effects observed for PFZ and XFZ.Conclusion:Altogether,these results demonstrated that the 5 PPS containing alkaloids have a good anti-RA-related inflammatory effect,and the mechanism may be related to COX-2/PGE2 signaling pathway,particularly,Fu-zi prepared utilizing a traditional Chinese technique.

Owing to the broad energy coverage of Insight-HXMT in the hard X-ray band,we detected the highest energy of pulsation exceeding 200 keV around the 2017–2018 outburst peak of the first Galactic pulsating ultraluminous X-ray source (PULX) Swift J0243.6+6124,which is the highest energy detected from PULXs to date.We also obtained the highest energy of pulsation of every exposure during the outburst in 2017–2018,and found the highest energy is roughly positively correlated with luminosity.Using our newly developed method,we identified the critical luminosity being 4×10~(38)erg s~(-1) when the main peaks of the low and high energy pulse profiles became aligned,which separates the fan-beam dominated and pencil-beam dominated accretion regimes.Above the critical luminosity,the phase of the main peak shifted gradually from 0.5 to 0.8 until the outburst peak in al energy bands is reached,which is in agreement with the phase shift found previously at low energies.Our result is consistent with what is derived from spectral analysis.

分析180 kW大功率直流充电桩在不同工况下的气动噪声特性,进行整桩的降噪优化设计.采用计算流体力学(CFD)与计算气动声学(CAA)分步耦合仿真的方法,分析整桩的噪声频谱和声场分布规律;开展整桩试验测试,验证仿真的精确性;对比、分析不同降噪方案的降噪性能,并进行方案的试验验证.结果显示,整桩噪声主频为830 Hz,整桩噪声次频为650 Hz,分别与电源模块风扇以及系统风扇的叶片通过频率重合;在电源模块进风口布置吸音棉能够有效消除整桩噪声第三、第四峰值频率幅值;在系统风扇出风口布置吸音棉能够使整桩A计权声压级总值降低约1.2 dB;相同布置位置,在保证散热的同时增加吸音棉厚度的降噪效果不显著,整桩A计权声压级总值降低不足1.0 dB.
