通过检测188份尤氏泰勒虫阳性样品和198份已知阴性样品,对基于重组表达TuIP抗原的间接ELISA方法检测性能进行了评价。利用two-graphic receiver-operating characteristic(TG-ROC)方法分别在95%和90%准确性的前提下,推断出了该方法的检测阈值为32.7%的阳性百分比率,检测敏感性和特异性均为98.4%。通过重复检测同样的阳性和阴性对照样品证实了该方法的稳定性和可靠性。对实验室吕氏泰勒虫感染的血清进行动态监测,发现该方法能在早期感染到两个月的感染期内检测到抗体,说明这种方法也可以用于吕氏泰勒虫感染血清学检测。该方法对来自已知的羊泰勒虫在随州和贵阳流行区域的样品阳性率分别为17.8%和11.6%,并且从一些相应的阳性样品中扩增到了18S rRNA基因片段,进一步证实了该方法检测的可靠性。以上数据表明FuIP ELASA可用于由尤氏泰勒虫和(或)吕氏泰勒虫引起的羊的泰勒虫病的血清学流行病学调查。

Pistachio is a nut crop domesticated in the Fertile Crescent and a dioecious species with ZW sex *** sequenced the genomes of Pistacia vera cultivar(cv.)Siirt,the female parent,and *** ***,the male *** chromosome-level reference genomes of pistachio were generated,and Z and W chromosomes were *** ZW chromosomes originated from an autosome following the first inversion,which occurred approximately 8.18 *** inversion events in the W chromosome led to the formation of a 12.7-Mb(22.8%of the W chromosome)non-recombining *** W-specific sequences contain several genes of interest that may have played a pivotal role in sex determination and contributed to the initiation and evolution of a ZW sex chromosome system in *** W-specific genes,including defA,defA-like,DYT1,two PTEN1,and two tandem duplications of six VPS13A paralogs,are strong candidates for sex determination or *** history analysis of resequenced genomes suggest that cultivated pistachio underwent severe domestication bottlenecks approximately 7640 years ago,dating the domestication event close to the archeological record of pistachio domestication in *** identified 390,211,and 290 potential selective sweeps in 3 cultivar subgroups that underlie agronomic traits such as nut development and quality,grafting success,flowering time shift,and drought *** findings have improved our understanding of the genomic basis of sex determination/differentiation and horticulturally important traits and will accelerate the improvement of pistachio cultivars and rootstocks.

A consensus meeting of national experts from all major national hepatobiliary centres in the country was held on May 26,2023,at the Pakistan Kidney and Liver Institute&Research Centre(PKLI&RC)after initial consultations with the *** Pakistan Society for the Study of Liver Diseases(PSSLD)and PKLI&RC jointly organised this *** effort was based on a comprehensive literature review to establish national practice guidelines for hilar cholangiocarcinoma(hCCA).The consensus was that hCCA is a complex disease and requires a multidisciplinary team approach to best manage these *** coordinated effort can minimise delays and give patients a chance for curative treatment and effective *** diagnostic and staging workup includes high-quality computed tomography,magnetic resonance imaging,and magnetic resonance *** cytology or biopsy utilizing endoscopic retrograde cholangiopancreatography is a mainstay for ***,histopathologic confirmation is not always required before *** ultrasound with fine needle aspiration of regional lymph nodes and positron emission tomography scan are valuable adjuncts for *** only curative treatment is the surgical resection of the biliary tree based on the Bismuth-Corlette *** patients with unresectable hCCA can be considered for liver *** chemotherapy should be offered to patients with a high risk of *** use of preoperative biliary drainage and the need for portal vein embolisation should be based on local multidisciplinary *** with acute cholangitis can be drained with endoscopic or percutaneous biliary *** chemotherapy with cisplatin and gemcitabine has shown improved survival in patients with irresectable and recurrent hCCA.

成环恒温扩增方法(LAMP)是一个新型的核酸检测方法,该方法需要两对或三对引物与目标DNA模板结合,在恒温条件下反应,这一方法具有高效,敏感和特异等特点。本研究中,我们以引起我国羊泰勒虫病的病原尤氏泰勒虫(Theileria uilenbergi)和吕氏泰勒虫(***)同源基因(UTRu6,UTRlu8)的保守区为模板,设计了LAMP引物。这组引物能同时扩增这两种病原,反应条件为63摄氏度,反应时间为15分钟。反应产物经EcoRⅠ酶切消化和序列测定证明了反应的特异性,另外,通过测定10倍稀释的DNA样品,证明了该方法具有从0.1皮克DNA样品中扩增到目标基因的敏感性。最后,通过比较LAMP和反向线性应迹法(RLB)测定86份野外样品的结果,确定了LAMP的敏感性和特异性分别为66.0%和97.4%。由于该方法快速,简便,廉价,敏感且特异,所以它将具有对我国羊泰勒虫病的诊断和流行病学调查的潜在应用价值。

不锈钢和碳素钢力学性能不同,但不锈钢连接设计规范只是在碳素钢规范基础上做一些修正。对于净截面承载力,欧洲不锈钢连接规范EN1993-1-4沿用欧洲冷弯碳素钢规范EN1993-1-3及SCI/Inox设计手册中的规定。对不锈钢连接净截面的破坏性能进行研究,建立奥氏体及马氏体不锈钢数值模型,用试验结果进行验证;并利用此模型对影响螺栓连接净截面破坏的主要参数进行研究,包括边缘距离e2和螺栓构造。通过研究不同e2和螺栓布置时净截面的应力分布,可发现不锈钢具有足够韧性,保证断裂前内力重分布。因此,提出基于EN1993-1-3的净截面承载力修正公式,并通过静力分析验证其可靠性。

根除幽门螺杆菌对胃组织学严重程度的影响:一项随访研究

Helicobacter pylori genetic diversity and geographic distribution affect the severity of gastric histology; while eradication heals gastritis, the improvement of atrophy and intestinal metaplasia (IM) is still controversial. We investigated whether H. pylori infection and genotypes (cagA- vacA) influence the histological changes and whether eradication resolves these changes. Twenty- one patients (11 duodenal ulcer, 2 gastric ulcer, 8 gastritis) received treatment. Biopsies for CLO, PCR, histology, and culture were collected before and at 1 and 12 months after treatment, and serum samples at 0, 1, 2, 6, and 12 months. H. pylori eradication was achieved in 71% of the patients. Histological scores for H. pylori densities were significantly higher in the antrum and incisura angularis. Scores for mononuclear cell and neutrophil infiltration were significantly higher in regions with a high H. pylori density and improved progressively after eradication. Eight patients with atrophy including five with IM showed no significant changes 12 months after eradication. The cagA gene, detected in 13 (62% ), the vacA- s1a gene, in 20 (95% ), and the vacA- m1 gene, in 12 (57% ) of 21 patients were significantly associated with duodenal ulcer. A gradual decline in antibody titer reached an average of 67% 12 months after eradication. H. pylori infection and the associated genotypes (cagA of Western type) affect the severity of the gastric histology (mild forms of atrophy and IM) and the disease outcome. Eradication of H. pyloriresulted in healing of gastritis, but with no significant improvement in atrophy or IM.

Background and Study Aims: Routine coagulation screening prior to gastrointestinal endoscopy is performed in many centres in the UK, despite the lack of any evidence to support the practice. The aim of this study was to assess the benefits of routine pre-endoscopy coagulation screening in children and to assess how widespread this practice is in the UK. Patients and Methods: We performed a retrospective analysis of the case notes of 250 consecutive patients who had undergone routine coagulation screening prior to endoscopy and biopsy, in accordance with our unit’s protocol, looking for evidence of abnormal results or episodes of bleeding. We also performed a telephone survey of the protocols for coagulation screening at other paediatric units in the UK which are known to perform gastro-intestinal endoscopy on a routine basis. Results: According to our hospital’s laboratory reference ranges, 16.8%of the children who underwent endoscopy and biopsy had abnormal clotting. This was neither clinically significant nor associated with an increased bleeding risk in any patient. Of the 23 UK paediatric gastroenterology centres surveyed, including our own, five (21.7%) perform routine coagulation screening before endoscopy. Conclusions: This study suggests that, although it is a relatively common practice, routine coagulation screening is not indicated in children who are undergoing gastrointestinal endoscopy and biopsy, and that it does not predict those at risk of significant bleeding. We would therefore suggest that if pre-endoscopy screening is to be performed, it should be reserved for those who are potentially at high risk of bleeding.
