Targeting multiple immune mechanisms may overcome therapy resistance and further improve cancer immunotherapy for ***,we describe the application of virus-like vesicles(VLV)for delivery of three immunomodulators alone and in combination,as a promising approach for cancer *** vectors were designed to deliver single chain interleukin(IL)-12,shorthairpin RNA(shRNA)targeting programmed death ligand 1(pD-L1),and a dominant-negative form of IL-17 receptor A(dn-IL17RA)as a single payload or as a combination *** delivery of the VLV vector expressing IL-12 alone,as well as the trivalent vector(designated CARG-2020)eradicated large established ***,only CARG-2020 prevented tumor recurrence and provided long-term survival benefit to the tumor-bearing mice,indicating a benefit of the combined *** abscopal effects of CARG-2020 on the non-injected contralateral tumors,as well as protection from the tumor cell re-challenge,suggest immune-mediated mechanism of protection and establishment of immunological ***,CARG-2020 potently activates Th1 immune mechanisms and inhibits expression of genes related to T cell exhaustion and cancer-promoting *** ability of CARG-2020 to prevent tumor recurrence and to provide survival benefit makes it a promising candidate for its development for human cancer immunotherapy.

BACkGROUND Celiac disease(CeD)is an autoimmune disorder triggered by the immune response to gluten in genetically predisposed *** research has unveiled a heightened risk of developing specific malignant neoplasms(MN)and various malignancies,including gastrointestinal,lymphomas,skin,and others,in individuals with *** To investigate the prevalence of MN in hospitalized CeD patients in the United *** Using data from the National Inpatient Sample spanning two decades,from january 2000 to December 2019,we identified 529842 CeD patients,of which 78128(14.75%)had *** score matching,based on age,sex,race,and calendar year,was employed to compare CeD patients with the general non-CeD population at a 1:1 *** positive associations were observed for several malignancies,including small intestine,lymphoma,nonmelanoma skin,liver,melanoma skin,pancreas myelodysplastic syndrome,biliary,stomach,and other neuroendocrine tumors(excluding small and large intestine malignant carcinoid),leukemia,uterus,and ***,CeD patients exhibited a reduced risk of respiratory and secondary ***,certain malignancies showed null associations with CeD,including head and neck,nervous system,esophagus,colorectal,anus,breast,malignant carcinoids,bone and connective tissues,myeloma,cervix,and ovary *** Our study is unique in highlighting the detailed results of positive,negative,or null associations between different hematologic and solid malignancies and ***,it offers insights into evolving trends in CeD hospital outcomes,shedding light on advancements in its management over the past two *** findings contribute valuable information to the understanding of CeD’s impact on health and healthcare utilization.

Oncologic outcomes with and without amniotic membranes in robotic-assisted radical prostatectomy:A propensity score matched analysis

Objective:placement of human placenta derived grafts during robotic-assisted radical prostatectomy(RARp)hastens the return of continence and *** long-term impact on the oncologic outcomes remains to be *** objective was to determine the oncologic outcomes of patients with dehydrated human amnion chorion membrane(dHACM)at RARp compared to a matched ***:In a referral centre,from August 2013 to October 2019,599 patients used dHACM in bilateral nerve-sparing *** excluded patients with less than 12 months follow-up,simple prostatectomy,and unilateral *** with dHACM(amnio group)were 529,and were propensity score matched 1:1 to 2465 patients without dHACM(non-amnio group)and a minimum follow-up of 36 *** the time of RARp,dHACM was placed around the neurovascular bundle in the amnio *** and categorical variables in matched groups was tested by two-sample kolmogorov-Smirnov test and Fisher's exact test *** measured were biochemical recurrence(BCR),adjuvant and salvage therapy ***:propensity score matching resulted in two groups of 444 *** incidence functions for BCR did not show a difference between the groups(p=0.3).patients in the non-amnio group required salvage therapy more frequently than the amnio group,particularly after partial nerve-sparing RARp(6.3%vs.2.3%,p=0.001).Limitations are the absence of prospective ***:The data suggest that using dHACM does not have a negative impact on BCR in *** of cancer specific and overall survival will require follow-up study to increase our understanding of these grafts’impact on prostate cancer biology.

BACkGROUND prediabetes is a well-established risk factor for major adverse cardiac and cerebrovascular events(MACCE).However,the relationship between prediabetes and MACCE in atrial fibrillation(AF)patients has not been extensively ***,this study aimed to establish a link between prediabetes and MACCE in AF *** To investigate a link between prediabetes and MACCE in AF *** We used the National Inpatient Sample(2019)and relevant ICD-10 CM codes to identify hospitalizations with AF and categorized them into groups with and without prediabetes,excluding *** primary outcome was MACCE(all-cause inpatient mortality,cardiac arrest including ventricular fibrillation,and stroke)in AF-related *** Of the 2965875 AF-related hospitalizations for MACCE,47505(1.6%)were among patients with *** prediabetes cohort was relatively younger(median 75 vs 78 years),and often consisted of males(56.3%vs 51.4%),blacks(9.8%vs 7.9%),Hispanics(7.3%vs 4.3%),and Asians(4.7%vs 1.6%)than the non-prediabetic cohort(pprediabetes group had significantly higher rates of hypertension,hyperlipidemia,smoking,obesity,drug abuse,prior myocardial infarction,peripheral vascular disease,and hyperthyroidism(all pprediabetes cohort was often discharged routinely(51.1%vs 41.1%),but more frequently required home health care(23.6%vs 21.0%)and had higher *** adjusting for baseline characteristics or comorbidities,the prediabetes cohort with AF admissions showed a higher rate and significantly higher odds of MACCE compared to the non-prediabetic cohort[18.6%vs 14.7%,odds ratio(OR)1.34,95%confidence interval 1.26-1.42,pp analyses,males had a stronger association(aOR 1.43)compared to females(aOR 1.22),whereas on the race-wise comparison,Hispanics(aOR 1.43)and Asians(aOR 1.36)had a stronger association with MACCE with prediabetes vs whites(aOR 1.33)and blacks(aOR 1.21).CONCLUSION This population-

Non-alcoholic steatohepatitis(NASH),an advanced form of non-alcoholic fatty liver disease(NAFLD),has emerged as the leading cause of liver failure and related ***,no medication is specifically approved to treat NAFLD or *** we report that oral administration of honey vesiclelike nanoparticles(H-VLNs)to naturally aged mice protects the liver from NASH development.H-VLNs are dominantly taken up by kupffer cells in the liver and suppress hepatic chronic inflammation and further development of fibrosis and nodule formation in aged *** their reported antiinflammasome function,H-VLNs are found to inhibit the transcriptional activities of C-jUN and nuclear factor-kappa B(NF-κB).MicroRNAs miR5119 and miR5108 and phenolic compound luteolin in H-VLNs are identified in suppressing both the C-jUN and NF-kB ***,oral intake of H-VLNs represents a promising new user-friendly modality to prevent the development of NASH.

The role of cysteine-rich secretory proteins in male fertility

The cysteine-rich secretory proteins （CRISps） are a subgroup of the CRISp, antigen 5 and pr-1 （CAp） protein superfamily, and are found only in vertebrates. They show a strong expression bias to the mammalian male reproductive tract and the venom of poisonous reptiles. Within the male reproductive tract CRISps have been implicated in many aspects of male germ cell biology spanning haploid germ cell development, epididymal maturation, capacitation, motility and the actual processes of fertilization. At a structural level, CRISps are composed of two domains, a CAp domain, which has been implicated in cell-cell adhesion, and a CRISp domain, which has been shown to regulate several classes of ion channels across multiple species. Herein, we will review the current literature on the role of CRISps in male fertility, and by inference to related non-mammalian protein, infer potential biochemical functions.
