Th. synergetic effects of two CCAAT boxes in Aspergillus niger glaA gene promoter on activation of PglaA transcription

EMSA and footprinting analyses h.ve revealed th.t th. -489—-414 bp and th. -390—-345 bp (designated DC and PC respectively) upstream of th. Aspergillus niger T21 glaA gene were bound by one protein factor in th. A. niger T21 wh.le cell extract. Both.DC and PC contained CCAAT pentanucleotides. Th. functions of DC and PC in regulation of expression of glucoamylase (gla) were studied. CCAAT pentanucleotides were replaced with.CGTAA and th. mutated DNA fragments DCm and PCm lost th. binding activities of protein factors in vitro. In vivo wh.n eith.r DC or PC was mutated or th. relative orientations between th.m were ch.nged on th. PglaA, th. transcriptional activity of PglaA decreased to a basal level. Introduction of multi-copies of DC into th. original site at th. PglaA in A. niger T21 decreased th. expression of endogenous gla expression and th. exogenous reporter E. coli uidA gene introduced under th. PglaA promoter, wh.le h.ving no effect on th. uidA gene under th. control of PgpdA. EMSA re-vealed th.t th. levels of th. specific DNA-binding protein factors in th. transformants maintained th. same meaning th.t introduction of multi-copies of DC caused th. titration effect. Angh.pC gene was cloned from A. niger T21 cDNA and introduced into th. DC multi-copied strains. Th. expression of Angh.pC improved th. expression of th. endogenous gla and th. exogenous gene controlled by PglaA. Th.se results sh.wed th.t both.th. CCAAT pentanucleotides were necessary for DC and PC binding to th. protein factors, and th. simultaneous binding of DC and PC to th. protein was necessary for promoting th. transcriptional activity of PglaA. Angh.pC was th. specific positive trans-acting protein factor binding to DC.

Introduction: Tendinopath. of th. th.racic limb (TMT) is frequent, and is responsible for an important socio-professional h.ndicap. Medical imaging, in particular, ultrasound h.s a major role in contributing to th. diagnosis. As th. use of ultrasound is recent in osteoarticular path.logy, especially in Africa, we proposed to determine th. contribution of imaging in th. diagnosis of tendinopath. of th. th.racic limbs in a country with.limited resources. Patients and Meth.d: A descriptive and analytical cross-sectional study of patients with.tendinopath. of th. th.racic limb wh. consulted th. rh.umatology department of th. CNh.-h.0} of Cotonou from August 6, 2019 to October 6, 2019. We evaluated th. concordance of th.racic limb tendinopath. between imaging (radiograph. and ultrasound) and th. clinic on th. one h.nd and th.n between radiograph. and ultrasound on th. oth.r h.nd. Th. analysis of th. collected data was carried out with.th. Epi-info software version 7.***.1.0. Th.n th. concordance was evaluated by th. k.ppa coefficient. Results: Fifty- two (52) patients with.104 joints were evaluated. Th. average age was 54.92 ± 3.40 years. Clinically, rotator cuff tendinopath. was th. most frequent abarticular path.logy (45.19%). Th. clinical-ultrasound agreement was strong (k.= 0.7527) for th. sh.ulders, very strong (k.= 0.9360) for th. elbows and moderate for th. wrists (k.= 0.6695). Th. clinical-radiograph.c agreement was weak.(k.= 0.2316) at th. sh.ulder level and very weak.(k.= 0.2087) at th. elbow level. Th. radio-ultrasound agreement was very low in th. sh.ulders (k.= 0.1522), elbows (k.= 0.1859) and wrists (k.= 0.0001). Conclusion: Th. contribution of imaging in th. diagnosis of TMT remains certain even in a country with.limited resources lik. ours. Ultrasound is a reliable examination for th. diagnosis of non-traumatic tendinopath. of th. th.racic limb with.a good clinical-ultra- sound concordance a

We investigated some Friedmann-Lemaître-Robertson-Walk.r(FLRW)cosmological models in th. context of metric-affine F(R,Q)gravity,as proposed in[arXiv:1205.5266v6].h.re,R and Q are th. curvature and nonmetricity scalars using non-special connections,*** obtained th. modified field equations using a flat FLRW *** th.n found a connection between th. h.bble constant h.(0),density parameter Ω_(m0),and oth.r model parameters in two different situations involving scalars u and ***,we used new observational datasets,such.as th. cosmic ch.onometer(CC)h.bble and Panth.on SNe Ia datasets,to determine th. optimal model parameter values th.ough.a Mark.v ch.in Monte Carlo(MCMC)*** th.se best-fit values of th. model parameters,we discussed th. results and beh.vior of th. derived ***,we discussed th. Ak.ik. information criterion(AIC)and Bayesian information criterion(BIC)for th. derived models in th. context of th. Lambda cold dark.matter(ΛCDM).We found th.t th. geometrical sector dark.equation of state parameter ω_(de)beh.ves just lik. a dark.energy *** also found th.t both.models are transit ph.se ***-Ⅰ approach.s th. Th. New Pest Scatella tenuicosta Collin （Diptera： Eph.dridae） for Spirulina Ponds

Th. goal of th.s study was to discover Scatella tenuicosta Collin （Diptera： Eph.dridae） for th. first time as a pest on blue-green algae Spirulina platensis. Th.s algae h.s been recognized as a very important commercial product in th. world today and as one of th. most promising ones among th. oth.r microscopic algae due to its rich.ess in polyunsaturated fatty acids, as well as in gamma-linolenic acid （gla）, and in pigments including ph.cocyanin, myxoxanth.ph.ll and zeaxanth.n. Both.immature and adult development stages ofS. tenuicosta h.ve been studied under th. temperature of 15±2℃. Th. development times for eggs, larval stages, including first, second, and th.rd one, and for pupae as well as for adults were found to be 1.4, 1.0, 1.1, 1.1, 3.0 and 8.7 respectively. Some pest control practices ofS. tenuicosta on S. platensis h.ve been also examined in th.s study.

Goldmann压平式眼压计和经眼睑眼压计(TGDc-01“PRA”)在青光眼患者所测眼压的比较

Purpose: Th. object of th.s study was to compare intraocular pressure measurem ents obtained with.th. TGDc-01 “PRA”, a new, transpalpebral indentation tonom eter, with.th.se from Goldmann applanation tonometry in normal and glaucomatous eyes. Meth.ds and Patients: Forty h.alth. eyes and 185 eyes suffering from glauc oma were included in th. study. For Goldmann tonometry th.ee measurements and fo r th. TGDc01 ten measurements were performed in a random order. All participants were placed in an uprigh. position for all measurements. Results: In both.group s a systematic increase of intraocular pressure was found with.n th. TGDc-01-m easurements. Th.refore, th. first 3 measurements of each.device were used for fu rth.r statistical analysis. No learning curve could be demonstrated for th. TGDc -01-measurement with.normal eyes. With.n th. group of normal eyes th. mean 1OD obtained with.th. TGDc-01 was 1.84 mmh. lower th.n th. mean 1OD obtained with.Goldmann tonometry (two-sided Student’s t-test; P=0.003). In th. group of gla ucomatous eyes, th. mean intraocular pressure obtained with.th. Goldmann tonomet ry was 19.7 ±10.1 mmh., with.th. TGDC-01 18.1 ±7.1 mmh. (coefficient of correlation r=0.64, Ph. mean standard deviatio n of intraocular pressure measurements with.Goldmann tonometry was 1.2 ±.0.9 mm h., with.th. TGDc01 3.1 ±2.1 mmh.. TGDc-01-measurements over-estimated intra ocular pressure compared to Goldmann tonometry up to values of 16 mmh. and under estimated intraocular pressure at values over 16 mmh.. Th. difference increased by 5.5 mmh. per 10 mmh. Goldmann tonometry. Th. probability of success, defined as TGDc-01-recordings with.n ±3 mmh. of th. Goldmann tonometry recordings, wa s less th.n 53 %between 5 and 20 mmh. and less th.n 30 %between 20 and 30 mmh. . 原发性闭角型青光眼的葡萄膜渗出

Purpose: To determine th. prevalence of uveal effusion in acute and ch.onic primary angle- closure glaucoma (PACG)- or primary angle closure (PAC) and to compare it with.th. prevalence in eyes with.open- angle glaucoma (OAG) or ocular h.pertension. Design: Prospective consecutive case series. Participants: Five h.ndred one eyes of 351 consecutive patients with.PAC and 156 eyes of 116 randomly selected primary OAG or ocular h.pertension patients. Th. PAC group included 40 eyes of 35 patients with.acute PACG and 30 unaffected fellow eyes, 39 eyes and 35 fellow eyes with.a h.story of acute PACG, and 357 eyes with.ch.onic PAC. Meth.ds: Ultrasound biomicroscopic examination was performed to diagnose uveal effusion and to measure anterior ch.mber depth.(ACD). Main Outcome Measures: Presence of uveal effusion and ACD. Results: Uveal effusion was demonstrated in 23 eyes (58% ) with.acute PACG and 7 fellow eys (23% ) (χ 2=8.17, P=0.0043). Among eyes with.ch.onic PAC, uveal effusion was present in 71(20% ), a h.gh.r prevalence th.n was found in open- angle patients (2 eyes [1.3% ]) (χ 2=19.3, Ph. ch.onic PAC group, th. ACD of ph.k.c eyes with.uveal effusion (1.92± 0.42 mm) was significantly sh.llower th.n th.t of ph.k.c eyes with.ut effusion (2.06± 0.32 mm) (P=0.019). Conclusions: Uveal effusion diagnosed by ultrasound biomicroscopy is a special feature in PAC, and is prevalent in acute PACG. Uveal effusion in ph.k.c eyes with.PAC is associated with.sh.llowing of ACD.
