SARS-CoV2 Nsp3 protein triggers cell death and exacerbates amyloid β42-mediated neurodegeneration

Infection caused by the severe acute respiratory syndrome coronavirus 2(SARS-CoV2) virus, responsible for the coronavirus disease 2019(COVID-19) pandemic, induces symptoms including increased inflammatory response, severe acute respiratory syndrome(SARS), cognitive dysfunction like brain fog, and cardiovascular defects. Long-term effects of SARS-CoV2 COVID-19 syndrome referred to as post-COVID-19 syndrome on age-related progressive neurodegenerative disorders such as Alzheimer’s disease remain understudied. Using the targeted misexpression of individual SARS-CoV2 proteins in the retinal neurons of the Drosophila melanogaster eye, we found that misexpression of nonstructural protein 3(Nsp3), a papain-like protease, ablates the eye and generates dark necrotic spots. Targeted misexpression of Nsp3 in the eye triggers reactive oxygen species production and leads to apoptosis as shown by cell death reporters, terminal deoxynucleotidyl transferase(TdT) dUTP Nick-end labeling(TUNEL) assay, and dihydroethidium staining. Furthermore, Nsp3 misexpression activates both apoptosis and autophagy mechanism(s) to regulate tissue homeostasis. Transient expression of SARS-CoV2 Nsp3 in murine neuroblastoma, Neuro-2a cells, significantly reduced the metabolic activity of these cells and triggers cell death. Misexpression of SARS-CoV2 Nsp3 in an Alzheimer’s disease transgenic fly eye model(glass multiple repeats [GMR]>amyloid β42) further enhances the neurodegenerative rough eye phenotype due to increased cell death. These findings suggest that SARS-CoV2 utilizes Nsp3 protein to potentiate cell death response in a neurodegenerative disease background that has high pre-existing levels of neuroinflammation and cell death.

This research paper presents a black hole solution with a rational nonlinear electrodynamics source within the Rastall gravity framework.The paper analyzes the thermodynamic properties of the solution in normal phase space and explores its critical behavior.The phase structure is examined using the extended first law of thermodynamics,with the cosmological constantΛserving as pressure P.The isotherms exhibit van der Waals behavior at small values of horizon r_(+).The paper also investigates the Gibbs free energy behavior and finds two critical points with two pressures where the re-entrant phase transition occurs and disappears.We also explore the prevalent microstructure of black holes in Ruppeiner geometry,uncovering significant deviations in the nature of particle interactions from conventional practice.Moreover,the thermodynamic geometry is analyzed using the Ruppeiner formalism,with the normalized Ricci scalar indicating possible point-phase transitions of the heat capacity,and the normalized extrinsic curvature having the same sign as the normalized Ricci scalar.The three-phase transitions of the heat capacity are those that we find for the normalized Ruppeiner curvatures.Thus,there is an absolute correspondence.

Malaria presents a significant global public health challenge,with severe malarial anaemia being a primary manifestation of the disease.The understanding of anaemia caused by malaria remains incomplete,making the treatment more complex.Iron is a crucial micronutrient essential for haemoglobin synthesis,oxygen delivery,and other vital metabolic functions in the body.It is indispensable for the growth of human beings,as well as bacteria,protozoa,and viruses in vitro and in vivo.Iron deficiency is among the most common nutritional deficiencies and can have detrimental effects during developmental stages of life.Malaria-induced iron deficiency occurs due to the hemolysis of erythrocytes and the suppression of erythropoiesis,leading to anaemia.Meeting iron requirements is particularly critical during pivotal life stages such as pregnancy,infancy,and childhood.Dietary intake alone may not suffice to meet adequate iron requirements,thus highlighting the vital role of iron supplementation.While iron supplementation can alleviate iron deficiency,it can exacerbate malaria infection by providing additional iron for the parasites.However,in the context of pregnancy and childhood,iron supplementation combined with malaria prevention and treatment has been shown to be beneficial in improving birth outcomes and ensuring proper growth and development,respectively.This review aims to identify the role and impact of iron supplementation in malaria infection during the life stages of pregnancy and childhood.

This review paper explores the efficacy of magnesium ferrite-based catalysts in photocatalytic degradation of organic contaminates(antibiotic and dyes).We report the influence of different doping strategies,synthesis methods,and composite materials on the degradation efficiency of these pollutants.Our analysis reveals the versatile and promising nature of magnesium ferrite-based catalysts,offering the valuable insights into their practical application for restoring the environment.Due to the smaller band gap and magnetic nature of magnesium ferrite,it holds the benefit of utilising the broader spectrum of light while also being recoverable.The in-depth analysis of magnesium ferrites'photocatalytic mechanism could lead to the development of cheap and reliable photocatalyst for the wastewater treatment.This concise review offers a thorough summary of the key advancements in this field,highlighting the pivotal role of the magnesium ferrite based photocatalysts in addressing the pressing global issue of organic pollutants in wastewater.

The coronavirus disease 2019(COVID-19)pandemic has had a tremendous adverse impact on the global health system,public sector,and social aspects.It is unarguably the worst pandemic of the century.However,COVID-19 management is a mystery in front of us,and an authentic treatment is urgently needed.Various repurposed drugs,like ivermectin,remdesivir,tocilizumab,baricitinib,etc.,have been used to treat COVID-19,but none are promising.Antibody therapy and their combinations are emerging modalities for treating moderate COVID-19,and they have shown the potential to reduce hospitalisations.One antibody monotherapy,bamlanivimab,and two cocktails,casirivimab/imdevimab and bamlanivimab/esterivimab,have received authorization for emergency use by the United States Food and Drug Administration for the treatment of mild COVID-19 in high risk individuals.The European Emergency has made similar recommendations for use of the drug in COVID-19 patients without oxygen therapy.This brief review will focus on monoclonal antibodies and their combination cocktail therapy in managing COVID-19 infection.

Multimodal medical image fusion has attained immense popularity in recent years due to its robust technology for clinical diagnosis.It fuses multiple images into a single image to improve the quality of images by retaining significant information and aiding diagnostic practitioners in diagnosing and treating many diseases.However,recent image fusion techniques have encountered several challenges,including fusion artifacts,algorithm complexity,and high computing costs.To solve these problems,this study presents a novel medical image fusion strategy by combining the benefits of pixel significance with edge-preserving processing to achieve the best fusion performance.First,the method employs a cross-bilateral filter(CBF)that utilizes one image to determine the kernel and the other for filtering,and vice versa,by considering both geometric closeness and the gray-level similarities of neighboring pixels of the images without smoothing edges.The outputs of CBF are then subtracted from the original images to obtain detailed images.It further proposes to use edge-preserving processing that combines linear lowpass filtering with a non-linear technique that enables the selection of relevant regions in detailed images while maintaining structural properties.These regions are selected using morphologically processed linear filter residuals to identify the significant regions with high-amplitude edges and adequate size.The outputs of low-pass filtering are fused with meaningfully restored regions to reconstruct the original shape of the edges.In addition,weight computations are performed using these reconstructed images,and these weights are then fused with the original input images to produce a final fusion result by estimating the strength of horizontal and vertical details.Numerous standard quality evaluation metrics with complementary properties are used for comparison with existing,well-known algorithms objectively to validate the fusion results.Experimental results from the proposed research article exhibit superior per

The discovery of efficient,selective,and stable electrocatalysts can be a key point to produce the largescale chemical fuels via electrochemical CO_(2) reduction(ECR).In this study,an earth-abundant and nontoxic ZnO-based electrocatalyst was developed for use in gas-diffusion electrodes(GDE),and the effect of nitrogen(N)doping on the ECR activity of ZnO electrocatalysts was investigated.Initially,a ZnO nanosheet was prepared via the hydrothermal method,and nitridation was performed at different times to control the N-doping content.With an increase in the N-doping content,the morphological properties of the nanosheet changed significantly,namely,the 2D nanosheets transformed into irregularly shaped nanoparticles.Furthermore,the ECR performance of Zn O electrocatalysts with different N-doping content was assessed in 1.0 M KHCO_(3) electrolyte using a gas-diffusion electrode-based ECR cell.While the ECR activity increased after a small amount of N doping,it decreased for higher N doping content.Among them,the N:ZnO-1 h electrocatalysts showed the best CO selectivity,with a faradaic efficiency(FE_(CO))of 92.7%at-0.73 V vs.reversible hydrogen electrode(RHE),which was greater than that of an undoped Zn O electrocatalyst(FE_(CO)of 63.4%at-0.78 V_(RHE)).Also,the N:ZnO-1 h electrocatalyst exhibited outstanding durability for 16 h,with a partial current density of-92.1 mA cm^(-2).This improvement of N:ZnO-1 h electrocatalyst can be explained by density functional theory calculations,demonstrating that this improvement of N:ZnO-1 h electrocatalyst comes from(ⅰ)the optimized active sites lowering the free energy barrier for the rate-determining step(RDS),and(ⅱ)the modification of electronic structure enhancing the electron transfer rate by N doping.

Mammogram Classification with HanmanNets Using Hanman Transform Classifier

Breast cancer is a deadly disease and radiologists recommend mammography to detect it at the early stages. This paper presents two types of HanmanNets using the information set concept for the derivation of deep information set features from ResNet by modifying its kernel functions to yield Type-1 HanmanNets and then AlexNet, GoogLeNet and VGG-16 by changing their feature maps to yield Type-2 HanmanNets. The two types of HanmanNets exploit the final feature maps of these architectures in the generation of deep information set features from mammograms for their classification using the Hanman Transform Classifier. In this work, the characteristics of the abnormality present in the mammograms are captured using the above network architectures that help derive the features of HanmanNets based on information set concept and their performance is compared via the classification accuracies. The highest accuracy of 100% is achieved for the multi-class classifications on the mini-MIAS database thus surpassing the results in the literature. Validation of the results is done by the expert radiologists to show their clinical relevance.

Perspectives of the Role of Chemotherapy in the Management of Osteosarcoma

Background: Multimodality management of osteosarcoma has significantly improved the 5-year-survival rate for localized disease over the past 40 years: from 5% - 10% (in historical controls) to 65% - 75% and 20% - 30% in metastatic disease. These results were achieved with doxorubicin, cisplatin, high-dose methotrexate and ifosfamide (or cyclophosphamide). In the absence of new and effective agents the results have remained stationary for at least the past 30 years. No standard second line therapy exists for patients who relapse. In these circumstances surgery when feasible, constitutes the main therapeutic option. Questions/Purposes: To understand the present approach to therapy and determine the possibilities for improvement a review of the chemotherapeutic agents currently deployed in the treatment of Osteosarcoma was undertaken. Methods: The review focused on the results achieved with the evolution of therapy following the discovery of effective chemotherapeutic agents. Results: There was an improvement in survival during the first decade following the introduction of effective chemotherapy and limb salvage replaced amputation in the majority of patients. Attempts to rescue pulmonary metastases patients with surgical intervention were also enhanced but produced only minor improvement in survival. An international collaborative study, EURAMOS has been launched to investigate the utility of neoadjuvant chemotherapeutic agents in improving survival based upon their efficacy in the treatment of the primary tumor. Conclusions: New agents and or new strategies are urgently required to improve the outcome in Osteosarcoma.
