Feeding behavior and hormoligosis associated with imidacloprid resistance in Asian citrus psyllid,Diaphorina citri

Imidacloprid is a neonicotinoid insecticide used for managing the Asian citrus psyllid,Diaphorina citri Kuwayama,which serves as vector of phytopathogens causing citrus ***,development of resistance to neonicotinoids among populations of *** has coincided with occasional control failures in the *** objectives of this research were to(1)survey current levels of imidacloprid resistance in Florida citrus;(2)compare feeding behavior between imidacloprid-resistant and susceptible *** using electrical penetration graph recordings,and(3)investigate the possible amplification of insecticide hormoligosis associated with *** surveys confirmed that the susceptibility of *** populations to imidacloprid has decreased in commercial Florida citrus groves compared with a laboratory-susceptible *** 12 generations of selection,resistance to imidacloprid increased by 438 fold compared with the susceptible ***-susceptible *** feeding on citrus exhibited significantly more bouts associated with intercellular pathway(C),phloem penetration(D),phloem salivation(E1),and nonprobing(Np)activities than imidacloprid-resistant ***,there were no differences observed in the frequency or duration of phloem ingestion or xylem feeding between susceptible and resistant *** was no statistical difference in fecundity between resistant and susceptible ***,the fecundity of imidacloprid-susceptible female *** treated with a sublethal concentration of imidacloprid(LC_(25))increased significantly compared with controls,while such hormoligosis was less pronounced among imidacloprid-resistant *** results suggest that imidacloprid-resistant psyllids may cease feeding sooner than susceptible counterparts following sublethal exposure to this insecticide,indicative of a behavioral resistance mechanism.

Sanguinarine-Mediated Sensitization of Cervical Cancer SiHa Cells to TRAIL

Introduction: Cervical cancer is primarily caused by the human papilloma virus (HPV), which transforms normal cervical cells into cancerous cells that are highly resistant to radiation and chemotherapy. Induction of apoptosis in transformed cells is a key strategy in successfully treating HPV-induced cervical cancer. TRAIL (tumor necrosis factor related apoptosis-inducing ligand) has been shown to selectively induce apoptosis in cancer cells by binding to death receptors and activating extrinsic pathways for apoptosis. However, certain cervical cancers—such as the cultured cell line SiHa—are remarkably resistant to TRAIL. In this study, SiHa cells were sensitized to TRAIL by using sanguinarine—derived from the plant Sanguinaria Canadensis—which is known to induce oxidative stress and lead to the upregulation of receptors for TRAIL. Methods: Cultured SiHa cells were exposed to sub-lethal doses of sanguinarine in combination with TRAIL. Cell viability changes as well as the production of reactive oxygen species (ROS) were assessed. The induction of apoptosis was investigated by assays for caspase activation. Flow cytometry was performed to analyze expression of death receptors 4/5. Results: Treatment of SiHa cells with a combination of sanguinarine and TRAIL led to a significant reduction in cell viability. Significant increase in ROS was observed and caspase activation assays confirmed the induction of apoptosis. Conclusions: The observed synergistic effect of sanguinarine and TRAIL on SiHa cells is promising for the treatment of cervical, and possibly other, HPV-induced cancers. Oxidative stress caused by sanguinarine seems to play a central role in this synergy. The precise link between reactive oxygen species and the possible upregulation of death receptors needs further investigation. This knowledge will enable us to devise more effective treatments for those who suffer from this devastating disease.

A novel tri-layer approach for immobilizing metal nanoparticles in SiO2 supports is *** this work,we show that under rapid heating to temperatures of approximately 1,000 ℃,metal nanoparticles less than 15 nm in size will entrench in the SiO2 layer on a silicon wafer to create pores as deep as 250 *** studied and characterized this entrenching behavior and subsequent nanopore formation for a wide variety of metal nanoparticles,including Au,Ag,Pt,Pd,and *** also demonstrate that an Al2O3 layer acts as a barrier to such pore ***,by creating a tri-layer architecture consisting of SiO2 on Al2O3 on silicon wafers,we can control the depth to which nanoparticles entrench between 3-5 *** small range allows one to entrench particles for the purpose of immobilization but still present them above the *** two advances of moving into the sub-15 nm size regime and of controlled particle immobilization through entrenchment have important implications in studying site-isolated and stabilized metal nanoparticles for applications in sensing,separations,and catalysis.

The polysaccharide chitin is a major scaffolding molecule in the insect *** order to be functional,both chitin amounts and chitin organization have been shown to be important *** great advances in the past decade,the molecular mechanisms of chitin synthesis and organization are not fully ***,we have characterized the function of the Chitinase 6(Cht6)in the formation of the wing,which is a simple flat cuticle organ,in the fruit fly Drosophila *** of Cht6 function by RNA interference during wing development does not affect chitin organization,but entails a thinner cuticle suggesting reduced chitin *** phenotype is opposed to the one reported recently to be caused by reduction of Cht10 *** as a consequence,cuticle permeability to xenobiotics is enhanced in Cht6-less *** also observed massive deformation of these *** addition,the shape of the abdomen is markedly changed upon abdominal suppression of ***,we found that suppression of Cht6 transcript levels influences the expression of genes coding for enzymes of the chitin biosynthesis *** finding indicates that wing epidermal cells respond to activity changes of Cht6 probably trying to adjust chitin ***,in a working model,we propose that Cht6-introduced modifications of chitin are needed for chitin synthesis to proceed *** thickness,according to our hypothesis,is in turn required for correct organ or body part *** molecular mechanisms of this processes shall be characterized in the future.

Risk scoring model for prediction of non-home discharge after transcatheter aortic valve replacement

Background Patients undergoing transcatheter aortic valve replacement(TAVR) are likely to be discharged to a location other than home. We aimed to determine the association between preoperative risk factors and non-home discharge after TAVR. Methods Patients discharged alive after TAVR at three centers were identified from a prospectively maintained database randomly divided into 80% derivation and 20% validation cohorts. Logistic regression models were fit to identify preoperative factors associated with non-home discharge in the derivation cohort. Multivariable models were developed and a nomogram based risk-scoring system was developed for use in preoperative counseling. Results Between June 2012 and December 2018, a total of 1,163 patients had TAVR at three centers. Thirty-seven patients who died before discharge were excluded. Of the remaining 1,126 patients(97%) who were discharged alive, the incidence of non-home discharge was 25.6%(n = 289). The patient population was randomly divided into the 80%(n = 900) derivation cohort and 20%(n = 226) validation cohort. Mean ± SD age of the study population was 83 ± 8 years. In multivariable analysis, factors that were significantly associated with non-home discharge were extreme age, female sex, higher STS scores, use of general anesthesia, elective procedures, chronic liver disease, non-transfemoral approach and postoperative complications. The unbiased estimate of the C-index was 0.81 and the model had excellent calibration. Conclusions One out of every four patients undergoing TAVR is discharged to a location other than home. Identification of preoperative factors associated with non-home discharge can assist patient counseling and postoperative disposition planning.

Power analysis is a key component of planning prospective studies such as clinical ***,some journals in biomedical and psychosocial sciences request power analysis for data already collected and analysed before acceptingmanuscripts for *** have raised concerns about the conceptual basis for such post-hoc power *** recently,Zhang et al showed by using simulation studies that such power analyses do not indicate true power for detecting statistical significance since post-hoc power estimates vary in the range of practical interests and can be very different from the true *** the other hand,journals'request for information about the reliability of statistical findings in a manuscript due to small sample sizes is justified since the sample size plays an important role in the reproducibility of statistical *** problem is the wording of the journals'request,as the current power analysis paradigm is not designed to address journals'concerns about the reliability of the statistical *** this paper,we propose an alternate formulation of power analysis to provide a conceptually valid approach to the journals'wrongly worded but practically significant concern.

North Atlantic subtropical mode water formation controlled by Gulf Stream fronts

The North Atlantic Ocean hosts the largest volume of global subtropical mode waters(STMWs) in the world,which serve as heat,carbon and oxygen silos in the ocean *** are formed in the Gulf Stream region where thermal fronts are pervasive and result in feedback with the ***,their roles in STMW formation have been *** eddy-resolving global climate simulations,we find that suppressing local frontal-scale ocean-to-atmosphere(FOA) feedback leads to STMW formation being reduced almost by *** is because FOA feedback enlarges STMW outcropping,attributable to the mixed layer deepening associated with cumulative excessive latent heat loss due to higher wind speeds and greater air-sea humidity contrast driven by the Gulf Stream *** enhanced heat loss overshadows the stronger restratification induced by vertical eddies and turbulent heat transport,making STMW colder and he *** more realistic representation of FOA feedback,the eddy-present/rich coupled global climate models reproduce the observed STMWs much better than the eddy-free *** improvement in STMW production cannot be achieved,even with the oceanic resolution solely refined but without coupling to the overlying atmosphere in oceanic general circulation *** findings highlight the need to resolve FOA feedback to ameliorate the common severe underestimation of STMW and associated heat and carbon uptakes in earth system mo dels.

Differences in the immunogenicity of engineered circular RNAs

Circular RNAs(circRNAs)are single-stranded RNAs that are covalently closed and lack free *** current understanding of the functions and regulation of circRNAs in immune systems is still in the nascent stages,but several groups have recently investigated the immunogenicity of engineered ***,which factors determine engineered circRNA immunogenicity still remains ***,we review and compare these investigations on engineered circRNA *** discuss the possible factors that may alter the immunogenic potential of a circRNA,such as its generation,purification,primary sequence,secondary structure,and RNA *** also suggest approaches for designing a circRNA with immunostimulatory or immune-evasive properties and recommend future areas for the field to *** the factors underlying engineered circRNA immunogenicity is an important step towards understanding endogenous circRNA immunity and increasing the therapeutic potential of circRNAs.
