Synergistic Interdecadal Evolution of Precipitation over Eastern China and the Pacific Decadal oscillation during 1951-2015

By using the multi-taper method(MTM)of singular value decomposition(SVD),this study investigates the interdecadal evolution(10-to 30-year cycle)of precipitation over eastern China from 1951 to 2015 and its relationship with the North Pacific sea surface temperature(SST).Two significant interdecadal signals,one with an 11-year cycle and the other with a 23-year cycle,are identified in both the precipitation and SST *** show that the North Pacific SST forcing modulates the precipitation distribution over China through the effects of the Pacific Decadal oscillation(PDo)-related anomalous Aleutian low on the western Pacific subtropical high(WPSH)and Mongolia high(MH).During the development stage of the PDo cold phase associated with the 11-year cycle,a weakened WPSH and MH increased the precipitation over the Yangtze River Basin,whereas an intensified WPSH and MH caused the enhanced rain band to move northward to North China during the decay *** the development stage of the PDo cold phase associated with the 23-year cycle,a weakened WPSH and MH increased the precipitation over North China,whereas an intensified WPSH and the weakened MH increased the precipitation over South China during the decay *** 11-year and 23-year variabilities contribute differently to the precipitation variations in the different regions of China,as seen in the 1998flooding *** 11-year cycle mainly accounts for precipitation increases over the Yangtze River Basin,while the 23-year cycle is responsible for the precipitation increase over Northeast *** results have important implications for understanding how the PDo modulates the precipitation distribution over China,helping to improve interdecadal climate prediction.

Different El Niño Flavors and Associated Atmospheric Teleconnections as Simulated in a Hybrid Coupled Model

A previously developed hybrid coupled model(HCM)is composed of an intermediate tropical Pacific ocean model and a global atmospheric general circulation model(AGCM),denoted as *** this study,different El Niño flavors,namely the Eastern-Pacific(EP)and Central-Pacific(CP)types,and the associated global atmospheric teleconnections are examined in a 1000-yr control simulation of the *** HCMAGCM indicates profoundly different characteristics among EP and CP El Niño events in terms of related oceanic and atmospheric variables in the tropical Pacific,including the amplitude and spatial patterns of sea surface temperature(SST),zonal wind stress,and precipitation *** SST budget analysis indicates that the thermocline feedback and zonal advective feedback dominantly contribute to the growth of EP and CP El Niño events,*** to the shifts in the tropical rainfall and deep convection during EP and CP El Niño events,the model also reproduces the differences in the extratropical atmospheric responses during the boreal *** particular,the EP El Niño tends to be dominant in exciting a poleward wave train pattern to the Northern Hemisphere,while the CP El Niño tends to preferably produce a wave train similar to the Pacific North American(PNA)*** a result,different climatic impacts exist in North American regions,with a warm-north and cold-south pattern during an EP El Niño and a warm-northeast and cold-southwest pattern during a CP El Niño,*** modeling result highlights the importance of internal natural processes within the tropical Pacific as they relate to the genesis of ENSo diversity because the active ocean–atmosphere coupling is allowed only in the tropical Pacific within the framework of the HCMAGCM.

In the face of the elevated incidence and mortality rate of septic shock in the ICU,this retrospective study seeks to investigate the indicative and predictive value of high-mobility group box 1(HMGB1)and miR-146b in patients with septic *** RTPCR was employed in this study to quantify the HMGB1 and miR-146b levels in plasma samples obtained from the patient group and healthy *** investigation involved the comparison between the two groups and tracking changes in the patient group over *** finding revealed that upon admission,the patient group exhibited markedly elevated relative expression levels of HMGB1,which subsequently decreased over ***,the patient group displayed significantly reduced relative expression levels of miR-146b upon admission,which subsequently increased over time compared to the control *** operating characteristic(RoC)curves showed good predictive value for HMGB1 and *** experimental results suggest that HMGB1 and miR-146b serve as valuable and convenient biomarkers for evaluating the severity of septic shock and predicting ***,it is proposed that serum miR-146b may be inducible and potentially exerts a negative regulatory effect on the expression of HMGB1.

Rapid advancement in aerospace technology has successfully enabled long-term life and economic activities in space,particularly in Low Earth orbit(LEo),extending up to 2000 km from the mean sea ***,the sustainance of the LEo Economy and its Environmental Control and Life Support System(ECLSS)still relies on a regular cargo supply of essential commodities(e.g.,water,food)from Earth,for which there still is a lack of adequate and sustainable *** key challenge in this context is developing water treatment technologies and standards that can perform effectively under microgravity *** this technical challenge will be a milestone in providing a scientific basis and the necessary support mechanisms for establishing permanent bases in outer space and *** identify clues towards solving this challenge,we looked back at relevant scientific research exploring novel technologies and standards for deep space exploration,also considering feedback for enhancing these technologies on *** our findings,we share our outlook for the future of drinking water treatment in *** also bring up a new concept for space aquatic chemistry,considering the closed environment of engineered systems operating in microgravity.

Interleukin-21(IL-21)is produced mainly by activated CD4^(+)T cells and promotes the proliferation,survival and differentiation of immune *** study characterised a high affinity binding receptor of IL-21(IL-21 Rα)from grass carp(Ctenopharyngodon idella,Ci)and analyzed the expression of CiIL-21 and CiIL-21 Rα.It has been shown that they were modulated by PAMPs and during infection with bacterial and viral *** CiIL-21 protein produced in bacteria was active in inducing the expression of genes involved in Th responses.A monoclonal CiIL-21 antibody was generated against the rCiIL-21 and showed good reactive specificity with the rCiIL-21 proteins expressed in *** cells,and HEK293 T cells and the native protein from PHA-stimulated kidney *** addition,the IL-21-producing cells detected by the CiIL-21 monoclonal antibody were found to be increased to in the kidney,spleen and hindgut after infection with Flavobacterium *** data indicate that IL-21 is involved in the regulation of immune response to bacterial and viral infection.

Enhanced editing efficiency in Arabidopsis with a LbCas12a variant harboring D156R and E795L mutations

Cas12a(Cpf1),a Class 2 Type V CRISPR/Cas nuclease,has several unique attributes for genome editing and may provide a valuable alternative to ***,a low editing efficiency due to temperature sensitivity and insufficient cleavage activity of the Cas12a nuclease are major obstacles to its broad *** this report,we generated two variants,ttAsCas12 Ultra and ttLbCas12a Ultra harboring three(E174R,M537R,and F870L)or two(D156R and E795L)mutations,respectively,by combining the mutations from the temperature-tolerant variants ttAsCas12a(E174R)and ttLbCas12a(D156R),and those from the highly active variants AsCas12a Ultra(M537R and F870L)and LbCas12a Ultra(E795L).We compared editing efficiencies of the five resulting Cas12a variants(LbCas12a,ttLbCas12a,ttLbCas12a Ultra,AsCas12a Ultra,and ttAsCas12 Ultra)at six target sites of four genes in Arabidopsis(Arabidopsis thaliana).The variant ttLbCas12a Ultra,harboring the D156R and E795L mutations,exhibited the highest editing efficiency of all variants tested in Arabidopsis and can be used to generate homozygous or biallelic mutants in a single generation in Arabidopsis plants grown at 22 *** addition,optimization of ttLbCas12a Ultra,by varying nuclear localization signal sequences and codon usage,further greatly improved editing ***,our results indicate that ttLbCas12a Ultra is a valuable alternative to Cas9 for editing genes or promoters in Arabidopsis.

Influence of organic matter input and temperature change on soil aggregate-associated respiration and microbial carbon use efficiency in alpine agricultural soils

Understanding the dynamics of soil respiration,microbial carbon use efficiency(CUE),and temperature sensitivity(Q_(10))in response to exogenous organic matter(EoM)input,soil aggregate size,and incubation temperature is crucial for predicting soil carbon cycling responses to environmental *** this study,these interactions were investigated by 180-day incubation of soil aggregates supplemented with EoM at various temperatures(5°C,15°C and 25°C).The results reveal an‘L-shaped’trend in soil respiration on the time scale across all treatments,characterized by initial rapid declines followed by *** input and higher temperatures significantly enhance respiration ***,the respiratory rates of soil aggregates of different sizes exhibit distinct patterns based on the presence or absence of *** conditions without the addition of EoM,larger aggregates show relatively lower respiration ***,in the presence of EoM,larger aggregates exhibit higher respiratory ***,Q_(10)decreases with increasing aggregate *** relationship between Q_(10)and the substrate quality index(SQI)supports the carbon quality temperature(CQT)hypothesis,highlighting SQI’s influence on Q_(10)values,particularly during later incubation *** CUE decreases with EoM input and rising ***,aggregate size plays a role in microbial CUE,with smaller aggregates exhibiting higher CUE due to enhanced nutrient *** conclusion,the intricate interplay of EoM input,aggregate size,and temperature significantly shapes soil respiration,microbial CUE,and Q_(10).These findings underscore the complexity of these interactions and their importance in modeling soil carbon dynamics under changing environmental conditions.

CD4 T helper cells are an important group of cells in the immune system of vertebrates and express CD4 receptor on the cell *** mammals,the CD4 receptor is encoded by a single copy gene,whilst in fish,two copies of cd4 genes,namely cd4-1 and cd4-2,are *** this study,the ectodomains of grass carp(Ctenopharyngodon idella,Ci)CD4-1 and CD4-2 were expressed in the *** cells and used to generate monoclonal antibodies in *** blotting,confocal microscopy and flow cytometry were performed to characterize the monoclonal *** has been shown that the CiCD4-1 and CiCD4-2 monoclonal antibodies had good specificity to react with the recombinant ectodomains of CiCD4-1 and CiCD4-2 expressed in the CHo-S cells and the native CD4 molecules of grass *** CiCD4-1 monoclonal antibody did not recognize CiCD4-2 and verse *** addition,the CD4-1 and CD4-2 monoclonal antibodies specifically recognized the CD4-1 and CD4-2 receptors expressed in the HEK293 cells and native molecules of fish ***,the percentages of lymphocytes in immune tissues of healthy fish were analyzed by flow *** was found that 17.6%of lymphocytes were CD4-1^(+)cells and 22.5%were CD4-2^(+)in the head *** the spleen,13.1%of lymphocytes were CD4-1^(+)and 18.6%were CD4-2^(+)while 7.3%of blood lymphocytes were CD4-1^(+)cells and 8.8%were CD4-2^(+)*** availability of the CD4-1 and CD4-2 monoclonal antibodies provides antibody-based tools for further elucidation of the functions of CD4 T cells in grass carp.

PE6c greatly enhances prime editing in transgenic rice plants

Prime editing is a versatile CRISPR/Cas-based precise genome-editing technique for crop *** new types of prime editors(PEs)named PE6a–d were recently generated using evolved and engineered reverse transcriptase(RT)variants from three different *** this study,we tested the editing efficiencies of four PE6 variants and two additional PE6 constructs with double-RT modules in transgenic rice(oryza sativa)***6c,with an evolved and engineered RT variant from the yeast Tf1 retrotransposon,yielded the highest prime-editing *** average fold change in the editing efficiency of PE6c compared with PEmax exceeded 3.5 across 18 agronomically important target sites from 15 *** also demonstrated the feasibility of using two RT modules to improve prime-editing *** results suggest that PE6c or its derivatives would be an excellent choice for prime editing in monocot *** addition,our findings have laid a foundation for prime-editing-based breeding of rice varieties with enhanced agronomically important traits.
