Mirror twin boundary(MTB)brings unique one-dimensional(1D)physics and properties into two-dimensional(2D)transition metal dichalcogenides(TMDCs),but they were rarely observed in non-Mo-based TMDCs.Herein,by post growth Nb doping,high density 4|4E-W and 4|4P-Se mirror twin boundaries(MTBs)were introduced into molecular beam epitaxy(MBE)grown WSe2 monolayers.Of them,4|4E-W MTB with a novel structure was discovered experimentally for the first time,while 4|4P-Se MTBs present a random permutations of W and Nb,forming a 1D alloy system.Comparison between the doped and non-doped WSe2 confirmed that Nb dopants are essential for MTB formation.Furthermore,quantitative statistics reveal the areal density of MTBs is directly proportional to the concentration of Nb dopants.To unravel the injection pathway of Nb dopants,first-principles calculations about a set of formation energies for excess Nb atoms with different configurations were conducted,based on which a model explaining the origin of MTBs introduced by excess metal was built.We conclude that the formation of MTBs is mainly driven by the collective evolution of excess Nb atoms introduced into the lttice of host WSe2 crystal and subsequent displacement of metal atoms(W or Nb).This study provides a novel way to tailor the MTBs in 2D TMDC materials via proper metal doping and presents new opportunities for exploring the intriguing properties.

Pulmonary fibrosis(PF)is a pathological change caused by repeated injuries and repair dysfunction of the alveolar epithelium.Our previous study revealed that the residues Asn3 and Asn4 of peptide DR8(DHNNPQIR-NH_(2))could be modified to improve stability and antifibrotic activity,and the unnatural hydrophobic amino acids a-(4-pentenyl)-Ala and D-Ala were considered in this study.DR3penA(DHa-(4-pentenyl)-ANPQIR-NH_(2))was verified to have a longer half-life in serum and to significantly inhibit oxidative damage,epithelial-mesenchymal transition(EMT)and fibrogenesis in vitro and in vivo.Moreover,DR3penA has a dosage advantage over pirfenidone through the conversion of drug bioavailability under different routes of administration.A mechanistic study revealed that DR3penA increased the expression of aquaporin 5(AQP5)by inhibiting the upregulation of miR-23b-5p and the mitogen-activated protein kinase(MAPK)pathway,indicating that DR3penA may alleviate PF by regulating MAPK/miR-23b-5p/AQP5.Safety evaluation showed that DR3penA is a peptide drug without obvious toxicity or acute side effects and has significantly improved safety compared to DR8.Thus,our findings suggest that DR3penA,as a novel and low-toxic peptide,has the potential to be a leading compound for PF therapy,which provides a foundation for the development of peptide drugs for fibrosis-related diseases.

Organic matter extracted from activated sludge with ammonium hydroxide and its characterization

In order to characterize the organic properties of waste activated sludge in a wastewater treatment plant,organic matter within sludge was extracted with NH 3 ·H 2 O preferentially,and subsequently fractionated into five fractions using XAD-8/XAD-4 resins.Up to a 63.8%-71.1% of organic matter within the sludge could be efficiently extracted by NH 3 ·H 2 O.Fractionation results showed that hydrophobic acid and hydrophilic fraction were two main components among the sludge organic matter （accounting for 32.2% and 48.0% of the bulk organic matter,respectively）,whereas transphilic acid,hydrophobic neutral and transphilic neutral were quite low （accounting for 9.2%,5.8% and 4.8%,respectively）.Despite that the extractant of NH 3 ·H 2 O showed a relatively higher extraction efficiency of the aromatic components,the relatively low aromaticity of the organic fractions implied that those non-aromatic components could also be effectively extracted,especially for neutral and hydrophilic fractions.In addition,acidic fractions contained more aromatic humic-like components,whereas the neutral fractions had a greater content of aromatic proteins and soluble microbial byproduct-like components.Extraction of sludge organics with NH3·H2O and subsequential fractionation using XAD resins could be a novel method for further characterization of sludge organics.

The antimicrobial peptide YD attenuates inflammation via miR-155 targeting CASP12 during liver fibrosis

The antimicrobial peptide APKGVQGPNG(named YD),a natural peptide originating from Bacillus amyloliquefaciens CBSYD1,exhibited excellent antibacterial and antioxidant properties in vitro.These characteristics are closely related to inflammatory responses which is the central trigger for liverfibrosis.However,the therapeutic effects of YD against hepatic fibrosis and the underlying mechanisms are rarely studied.In this study,we show that YD improved liver function and inhibited the progression of liver fibrosis by measuring the serum transaminase activity and the expression of a-smooth muscle actin and collagen I in carbon tetrachloride-induced mice.Then we found that YD inhibited the level of miR-155,which plays an important role in inflammation and liver fibrosis.Bioinformatics analysis and luciferase reporter assay indicate that Casp12 is a new target of miR-155.We demonstrate that YD signifi-cantly decreases the contents of inflammatory cytokines and suppresses the NF-k B signaling pathway.Further studies show that transfection of the miR-155 mimic in RAW264.7 cells partially reversed the YD-mediated CASP12 upregulation,the downregulated levels of inflammatory cytokines,and the inactivation of the NF-k B pathways.Collectively,our study indicates that YD reduces inflammation throughthe mi R-155 e Casp12 e NF-k B axis during liver fibrosis and provides a promising therapeutic candidate for hepatic fibrosis.

Currently, there is still no effective curative treatment for the development of late-stage liver fibrosis. Here, we have illustrated that TB001, a dual glucagon-like peptide-1 receptor/glucagon receptor(GLP-1 R/GCGR) agonist with higher affinity towards GCGR, could retard the progression of liver fibrosis in various rodent models, with remarkable potency, selectivity, extended half-life and low toxicity. Four types of liver fibrosis animal models which were induced by CCl_(4), a-naphthyl-isothiocyanate(ANIT), bile duct ligation(BDL) and Schistosoma japonicum were used in our study. We found that TB001 treatment dose-dependently significantly attenuated liver injury and collagen accumulation in these animal models. In addition to decreased levels of extracellular matrix(ECM) accumulation during hepatic injury, activation of hepatic stellate cells was also inhibited via suppression of TGF-β expression as well as downstream Smad signaling pathways particularly in CCl_(4)-and S. japonicum-induced liver fibrosis. Moreover, TB001 attenuated liver fibrosis through blocking downstream activation of proinflammatory nuclear factor kappa B/NF-kappa-B inhibitor alpha(NFκB/IKBa) pathways as well as cJun N-terminal kinase(JNK)-dependent induction of hepatocyte apoptosis. Furthermore, GLP-1 R and/or GCGR knock-down results represented GCGR played an important role in ameliorating CCl_(4)-induced hepatic fibrosis. Therefore, TB001 can be used as a promising therapeutic candidate for the treatment of multiple causes of hepatic fibrosis demonstrated by our extensive pre-clinical evaluation of TB001.

Background:β-defensins have attracted considerable research interest because of their roles in protecting hosts from various pathogens.This study was conducted to investigate the expression profiles of the porcineβ-defensin114(PBD114)in different breeds and in response to infections.Moreover,the function of PBD114 protein was partially investigated.Methods:Six Tibetan pigs(TP)and six DLY(Duroc×Landrace×Yorkshire)pigs were slaughtered to explore the expression profiles of PBD114 in different breeds and tissues.For infection models,sixteen DLY pigs were divided into two groups and challenged either with sterile saline or E.coli K88.The recombinant protein PBD114(r PBD114)was obtained by using a heterologous expression system in E.coli.Results:PBD114 gene was highly expressed in tissues such as the intestine,liver,spleen,and thymus.Interestingly,the expression level of PBD114 gene was higher in the TP pigs than in the DLY pigs(P<0.05),and was significantly elevated upon E.coli K88 challenge(P<0.05).The nucleotide sequences of PBD114 from Tibetan and DLY pigs was identical,and both showed a 210-bp open reading frame encoding a 69-amino acid mature peptide.To explaore the function of PBD114 protein,PBD114 gene was successfully expressed in E.coli Origami B(DE3)and the molecular weight of the r PBD114 was estimated by SDS-PAGE to be 25 k Da.The r PBD114 was purified and mass spectrometry verified the protein as PBD114.Importantly,r PBD114 showed antimicrobial activities against E.coli DH5αand E.coli K88,and the minimal inhibitory concentrations(MICs)were 64 and 128μg/m L,respectively.Hemolytic and cytotoxicity assays showed that r PBD114 did not affect cell viability under physiological concentrations.Conclusions:PBD114 is an infection response gene that is differentially-expressed between different porcine breeds and tissues.The antimicrobial activity of PBD114 protein,against pathogens such as the E.coli K88,suggested that it may serve as a candidate for the substitution of conventionally used antibiotics.

Personalized nutrition intervention improves nutritional status and quality of life of colorectal cancer survivors in the community: a randomized controlled trial

Objective: To evaluate the effects of personalized nutrition intervention combined with tele-education on the nutritional status and quality of life among colorectal cancer survivors in the community. Methods: In the randomized, parallel-controlled trial, colorectal cancer survivors who met the eligibility criteria were enrolled and randomly 1:1 assigned into nutrition intervention and routine care groups. The routine care group was received routine community follow-up twice a year. In addition to routine care group follow-up, the nutrition intervention group received personalized nutritional interventions, tele-education through WeChat for six months. The nutrition status, dietary intake, and quality of life were measured and compared between the two groups. Results: Of 60 colorectal cancer survivors were randomly assigned, 56 participants were included in the modified intention-to-treat analysis, with nutrition intervention and routine care groups. After the 6-month intervention, the nutrition group had a statistically lower PG-SGA score, higher energy intake, and protein intake compared with the routine care group. Moreover, the nutrition intervention group gained more weight(2.00 kg, 95%CI = 0.25 to 3.00) than routine care group(0.00 kg, 95%CI =-1.75 to 0.00). Meanwhile, compared with the routine care group, the nutrition intervention group had significantly higher global health status, physical functioning, role functioning, emotional functioning, cognitive functioning and social functioning(P<0.05). Conclusions: Personalized nutrition interventions combined with tele-education provided by community health service centers can improve colorectal cancer survivors nutritional status and quality of life of colorectal cancer survivors.
