Exercise training is critical for the early prevention and treatment of obesity and diabetes mellitus.However,the mechanism with gut microbiota and fecal metabolites underlying the effects of voluntary wheel running on high-fat diet induced abnormal glucose metabolism has not been fully elaborated.C57BL/6 male mice were randomly assigned to 4 groups according to diets(fed with normal chow diet or high-fat diet)and running paradigm(housed in static cage or with voluntary running wheel).An integrative 16S rDNA sequencing and metabolites profiling was synchronously performed to characterize the effects of voluntary wheel running on gut microbiota and metabolites.It showed that voluntary wheel running prevented the detrimental effects of high-fat feeding on glucose metabolism 16S rDNA sequencing showed remarkable changes in Rikenella and Marvinbryantia genera.Metabolic profiling indicated multiple altered metabolites,which were enriched in secondary bile acid biosynthesis signaling.In conclusion,our study indicated that voluntary wheel running significantly improved glucose metabolism and counteracted the deleterious effects of high-fat feeding on body weight and glucose intolerance.We further found that voluntary wheel running could integratively program gut microbiota composition and fecal metabolites changes,and may regulate muricholic acid metabolism and secondary bile acid biosynthesis in high-fat fed mice.

Methane is the second largest anthropogenic greenhouse gas,and changes in atmospheric methane concentrations can reflect the dynamic balance between its emissions and sinks.Therefore,the monitoring of CH_(4) concentration changes and the assessment of underlying driving factors can provide scientific basis for the government’s policy making and evaluation.China is the world’s largest emitter of anthropogenic methane.However,due to the lack of ground-based observation sites,little work has been done on the spatial-temporal variations for the past decades and influencing factors in China,especially for areas with high anthropogenic emissions as Central and Eastern China.Here to quantify atmospheric CH_(4) enhancements trends and its driving factors in Central and Eastern China,we combined the most up-to-date TROPOMI satellite-based column CH_(4)(xCH_(4))concentration from 2018 to 2022,anthropogenic and natural emissions,and a random forest-based machine learning approach,to simulate atmospheric xCH_(4) enhancements from 2001 to 2018.The results showed that(1)the random forest model was able to accurately establish the relationship between emission sources and xCH_(4) enhancement with a correlation coefficient(R^(2))of 0.89 and a root mean-square error(RMSE)of 11.98 ppb;(2)The xCH_(4) enhancement only increased from 48.21±2.02 ppb to 49.79±1.87 ppb from the year of 2001 to 2018,with a relative change of 3.27%±0.13%;(3)The simulation results showed that the energy activities and waste treatment were the main contributors to the increase in xCH_(4) enhancement,contributing 68.00% and 31.21%,respectively,and the decrease of animal ruminants contributed-6.70% of its enhancement trend.

Improving the application of nanomaterials has always been a research hotspot in the field of energetic materials(EMs)due to their obvious catalytic effect on the EMs,especially the uniformly dispersed nanomaterials.However,few studies have reported the dispersion of nanomaterials.In this study,the dispersity and mixing uniformity of nano-CuCr_(2)O_(4)was evaluated based on the difference of solid UV light absorption between the nano-catalytic materials and EMs.The nano-CuCr_(2)O_(4)/ultrafine AP composites with different dispersity of nano-CuCr_(2)O_(4)were prepared by manual grinding and mechanical grinding with different grinding strength and griding time.And then,the absorbance of different samples at 212 nm was obtained by solid UV testing due to the high repeatability of the absorbance at 210-214 nm for three parallel experiments,and the dispersity of different samples was calculated through the established difference equation.Furthermore,the samples were characterized by XRD,IR,SEM,EDS,DSC and TG-MS,which confirmed that different mixing methods did not change the structure of the samples(XRD and IR),and the mixing uniformity improved with the increase of grinding strength and grinding time(SEM and EDS).The scientificity and feasibility of the difference equation were further verified by DSC.The dispersity of nano-CuCr_(2)O_(4)exhibits a positive intrinsic relationship with its catalytic performance,and the uniformly dispersed nano-CuCr_(2)O_(4)significantly reduces the thermal decomposition temperature of ultrafine AP from 367.7 to 338.8℃.The TG-MS patterns show that the dispersed nano-CuCr_(2)O_(4)advanced the thermal decomposition process of ultrafine AP by about 700 s,especially in the high temperature decomposition stage,and the more concentrated energy release characteristic is beneficial to further enhance the energy performance of AP-based propellants.The above conclusions show that the evaluation method of dispersity based on solid UV curves could provide new ideas for the dispersity characterizati

Anaerobic bacteria infections remain a major threat to humans.Oxidative stress mediated by photodynamic therapy(PDT)has shown great potential in antimicrobial applications,attributing to its controllability and non-resistance.However,the hypoxic infection microenvironment severely compromises the therapeutic outcomes.Here,we designed a nanoplatform comprising the initiator of alkyl radicals(R-),2,2’-azobis[2-(2-imidazolin-2-yl)propane]dihydrochloride(AIPH)-loaded oxidized mesoporous carbon nanoparticles(OMCN)and then coated with a phase change material(PCM)nanoparticle for oxygen-independent PDT to combat anaerobic infections.Upon near-infrared(NIR)light irradiation,OMCN mediates the generation of heat to melt PCM,which could trigger the liberation and decomposition of AIPH to produce R.and further induce the oxidative damage to bacteria,and cooperate with optothermal effects to eradicate anaerobic bacteria.As results,the nanoplatform displays strong synergistic antibacterial behavior,effectively eliminating anaerobic Propionibacterium acnes(P.acnes)in vitro under hypoxic conditions,and successfully treating P.acnes-infected cutaneous abscess in mice models.Moreover,no distinct toxicity appears through the entire course of treatment.Collectively,this light-activated oxygen-independent PDT offers a novel approach to combat anaerobic bacteria infections.

The ascendant danger of bacterial infections has resulted in an urgent requirement for developing new antibacterial approaches.Recently,nitric oxide(NO)has shown a broad-spectrum antimicrobial activity while avoiding resistance.However,achieving effective NO-based antibacterial therapies remains challenging,mired by the safety concerns of NO donor,residual toxicity by the ingredients,or complicated procedures.Herein,a self-activated NO-releasing hydrogel depot is fabricated by Ca^(2+)-crosslinked sodium alginate doped with CaO_(2)nanoparticles,L-arginine,and oxidized mesoporous carbon nanoparticles(OMCN)for photothermal enhanced bacteria killing.The locally concentrated H_(2)O_(2),generated from the reaction of CaO_(2)nanoparticles and water,could oxidize L-arginine to release NO.Meanwhile,benefiting from the remarkable photothermal effect of OMCN,the hybrid hydrogel possesses a synergistic sterilization behavior in combating both Gram-negative Escherichia coli and Grampositive Staphylococcus aureus bacteria in vitro.Moreover,the dual therapeutic hydrogel displays high efficiency in treatment of bacteria-infected mice with back wound model while showing no distinct toxicity.In addition,the restricted environment of hydrogel makes it easy to remove all the components from the infected wound,alleviating possible side effects from exogenous H_(2)O_(2).As such,the designed NO-synergistic photothermal hydrogel provides a promising strategy for treating bacterial infections.

Metal-organic frameworks (MOFs) have attracted much attention because of their large surface areas, tunable structures, and potential applications in many areas. In recent years, MOFs have shown much promise in CO2 photoreduction. This review summarized recent research progresses in MOF-based photocatalysts for photocatalytic reduction of CO2. Besides, it discussed strategies in rational design of MOF-based photocatalysts (functionalized pristine MOFs, MOF-photosensitizer, MOF-semiconductor, MOF-metal, and MOF-carbon materials composites) with enhanced performance on CO2 reduction. Moreover, it explored challenges and outlook on using MOF-based photocatalysts for CO2 reduction.

The sluggish reaction kinetics at the oxygen cathode is one of the important issues hindering the commercialization of the metal-air batteries.Although the noble metal can be used as the high-efficiency electrocatalyst to solve the problems to some extent,the high cost and scarcity of these noble-metal catalysts have limited their application in electrocatalysis.In this review,we discussed the mecha-nisms of the ORR and OER,and proposed the principles for the bifunctional electrocatalysts firstly,and then the state-of-the-art bifunctional catalysts,including carbon-based materials and transition-metal-based materials.On the basis of that,the self-supporting 3D noble-metal-free bifunctional ORR/OER catalysts were also discussed.Finally,the perspectives for the bifunctional electrocatalysts were discussed.

Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection disrupts the epithelial barrier and triggers airway inflammation.The envelope(E)protein,a core virulence structural component of coronaviruses,may play a role in this process.Pathogens could interfere with transepithelial Cl^(-)transport via impairment of the cystic fibrosis transmembrane conductance regulator(CFTR),which modulates nuclear factor kB(NF-kB)signaling.However,the pathological effects of SARS-CoV-2 E protein on airway epithelial barrier function,Cl^(-)transport and the robust inflammatory response remain to be elucidated.Here,we have demonstrated that E protein down-regulated the expression of tight junctional proteins,leading to the disruption of the airway epithelial barrier.In addition,E protein triggered the activation of Toll-like receptor(TLR)2/4 and downstream c-Jun N-terminal kinase(JNK)signaling,resulting in an increased intracellular Cl^(-)concentration([Cl^(-)]_(i))via up-regulating phosphodiesterase 4D(PDE4D)expression in airway epithelial cells.This elevated[Cl^(-)]_(i);contributed to the heightened airway inflammation through promoting the phosphorylation of serum/glucocorticoid regulated kinase 1(SGK1).Moreover,blockade of SGK1 or PDE4 alleviated the robust inflammatory response induced by E protein.Overall,these findings provide novel insights into the pathogenic role of SARS-CoV-2 E protein in airway epithelial damage and the ongoing airway inflammation during SARS-CoV-2 infection.

Herb-drug interaction in the protective effect of Alpinia officinarum against gastric injury induced by indomethacin based on pharmacokinetic, tissue distribution and excretion studies in rats

Alpinia officinarum Hance of the Chinese traditional herb for the treatment of emesis, abdominal pain and diarrhea has been used to counteract gastric disease induced by indomethacin in rats without obvious side effects. However, the role of herb-drug interaction between indomethacin and A. officinarum based on pharmacokinetic, tissue distribution and excretion still remains unknown. In this study, an ultra-fast liquid-tandem mass spectrometry(UFLC-MS/MS) method was developed for simultaneous determination of indomethacin and its three metabolites, O-desmethylindomethacin(ODI), deschlorobenzoylindomethacin(NDI) and indomethacin acyl-b-D-glucuronide(IDAbG) by oral administration of indomethacin solution with and without the ethanolic extract of A. officinarum and applied to comparative pharmacokinetic, tissue distribution and excretion studies. Our results clarified that oral administration of A. officinarum produced significant alterations in the pharmacokinetic parameters of indomethacin. And the pharmacokinetic interaction between indomethacin and A. officinarum reduced the systemic exposure of indomethacin and increased its elimination. Tissue distribution results demonstrated that co-administration of A. Officinarum could not reduce the accumulation of indomethacin in the target tissue of the stomach, but could accelerate the excretions of indomethacin and its three metabolites including ODI, NDI and IDAb G in the bile and feces of rats in the excretion study.Therefore, A. Officinarum might have a gastrointestinal protective effect through the interaction role with indomethacin based on the pharmacokinetics and excretion in rats.

RGD-FasL Induces Apoptosis of Pituitary Adenoma Cells

This study was to investigate the cytotoxic effects on pituitary adenoma cell lines GH3/MMQ/AtT20 induced by RGD-FasL and the underlying mechanism. Fas/DcR3 mRNAs were detected by RT-PCR and their surface expressions were measured by flow cytometry. Cytotoxicity exerted by RGD-FasL on tumor cells was measured with MTT assay and the induced apoptosis was determined by agarose gel electrophoresis. The cell cycle and apoptosis was assessed by flow cytometry with PI staining. The expressions of caspase8/9/3, Bcl-2, RANKL and JNK2 were detected by Western blotting. Approximately 13.7% of GH3 cells, 25.5% of MMQ cells, 22.2% of AtT20 cells express Fas, while 23.9% of GH3 cells, 24.1% of MMQ cells, 4.6% of AtT20 cells express DcR3. The cytotoxic effects of FasL/RGD-FasL on tumor cells were all taken in a dose-dependent manner. Cell lines MMQ/AtT20 showed the same sensitivity to RGD-FasL as to FasL, while cell line GH3 was less sensitive to RGD-FasL. The cell cycle analysis indicated that RGD-FasL could inhibit cells in G0/G1 phase and G2/M phase. In MMQ and AtT20 cells treated with RGD-FasL, the AI was not significantly different from that treated with FasL, while in GH3 cells treated with RGD-FasL, the AI was lower than that treated with FasL. The expressions of caspase-8/9/3, RANKL and JNK2 were increased while that of Bcl-2 was decreased after treatment with RGD-FasL, suggesting that RGD-FasL induces apoptosis through caspase activation. We concluded that RGD-FasL could possibly be considered as a novel therapeutical candidate for the treatment of pituitary adenomas. Cellular ＆ Molecular Immunology.
