Loss of GATA6-mediated up-regulation of UTX promotes pancreatic tumorigenesis and progression

Ubiquitously transcribed tetratricopeptide repeat on chromosome X(UTX),also known as lysine(K)-specific demethylase 6A(KDM6A),functions as a tumor suppressor gene or oncogene depending on the tumor type and context.However,its tumor-suppressive mechanisms remain largely unknown.Here,we investigated the clinical significance and biological effects of UTX expression in pancreatic ductal adenocarcinoma(PDA)and determined the potential mechanisms of its dysregulation.UTX expression and its association with clinicopathologic characteristics of PDA patients were analyzed using immunohistochemistry.UTX mRNA and protein expression and their regulation in PDA cell lines were measured using quantitative polymerase chain reaction and Western blot analyses.The biological functions of UTX in PDA cell growth,migration,and invasion were determined using gain-and loss-of-function assays with both in vitro and in vivo animal models.UTX expression was reduced in human PDA cell lines and specimens.Low UTX expression was associated with poor differentiation and prognosis in PDA.Forced UTX expression inhibited PDA proliferation,migration,and invasion in vitro and PDA growth and metastasis in vivo,whereas knockdown of UTX expression did the opposite.Mechanistically,UTX expression was trans-activated by GATA6 activation.GATA6-mediated PDA progression could be blocked,at least partially,by silencing UTX expression.In conclusion,loss of GATA6-mediated UTX expression was evident in human PDA and restored UTX expression suppressed PDA growth and metastasis.Thus,UTX is a tumor suppressor in PDA and may serve as a prognostic biomarker and therapeutic target.

Integrated separation-electrochemical detection device based on wood column for online identification of enantiomer

Chiral chemicals have attracted significant interest in the pharmaceutical industry,yet the separation methods to get pure enantiomers from racemic mixture are still challenging.To date,the separation of enantiomers still mainly depends on chromatography using high-cost chiral stationary phases.Herein,wood channels were used as the handheld integrated device,and enantiomer separation was simultaneously detected using an electrochemical detector.In this method,a chiral UIO-66(L-UIO-66)modified enantiomer separation zone and carbonized wood based online detection zone are integrated along a single wood column.Based on the in situ separation results from the chronoamperometry data,the wood device shows excellent separation ability for a wide range of electrochemically active enantiomers,including 3,4-dihydroxyphenylalanine,amino acids,ascorbic acid,carnitine,and penicillamine with high chirality purity.The unbiased molecular dynamic simulations indicate that the excellent chiral recognition and separation are attributed to the different barriers from the bound states to the dissociated state of the enantiomers in the homochiral microenvironment of the framework.This integrated enantiomer separation-electrochemical detection device provides a novel,easy,and low-cost platform for the separation of pure enantiomer from racemic mixture.

Objective:Sex-specific differences are observed in various liver diseases,but the influence of sex on the outcomes of hepatocellular carcinoma(HCC)after liver transplantation(LT)remains to be determined.This study is the first Chinese nationwide investigation of the role of sex in post-LT outcomes in patients with HCC.Methods:Data for recipients with HCC registered in the China Liver Transplant Registry between January 2015 and December 2020 were analyzed.The associations between donor,recipient,or donor-recipient transplant patterns by sex and the post-LT outcomes were studied with propensity score matching(PSM).The survival associated with different sex-based donor-recipient transplant patterns was further studied.Results:Among 3,769 patients enrolled in this study,the 1-,3-,and 5-year overall survival(OS)rates of patients with HCC after LT were 96.1%,86.4%,and 78.5%,respectively,in female recipients,and 95.8%,79.0%,and 70.7%,respectively,in male recipients after PSM(P=0.009).However,the OS was comparable between recipients with female donors and male donors.Multivariate analysis indicated that male recipient sex was a risk factor for post-LT survival(HR=1.381,P=0.046).Among the donor-recipient transplant patterns,the male-male donor-recipient transplant pattern was associated with the poorest post-LT survival(P<0.05).Conclusions:Our findings highlighted that the post-LT outcomes of female recipients were significantly superior to those of male recipients,and the male-male donor-recipient transplant pattern was associated with the poorest post-LT survival.Livers from male donors may provide the most benefit to female recipients.Our results indicate that sex should be considered as a critical factor in organ allocation.

Morphodynamic response of an embayed beach to different typhoon events with varying intensities

Beach erosion has occurred globally in recent decades due to frequent and severe storms.Dongsha beach,located in Zhujiajian Island,Zhejiang Province,China,is a typical embayed sandy beach.This study focused on the morphodynamic response of Dongsha beach to typhoon events,based on beach topographies and surficial sediment characteristics acquired before and after four typhoon events with varying intensities.The four typhoons had different effects on the topography and sediment characteristics of Dongsha beach.Typhoons Ampil and Danas caused the largest(-51.72 m3/m)and the smallest erosion(-8.01 m3/m),respectively.Remarkable alongshore patterns of beach profile volumetric changes were found after the four typhoon events,with more erosion in the southern and central parts of the beach and few changes in the northern part.Grain size coarsening and poor sorting were the main sediment patterns on the beach influenced by different typhoons.Typhoons that occurred in the same year after another typhoon enhanced the effect of the previous typhoon on sediment coarsening and sorting variability,but this cumulative effect was not found between typhoons that occurred during different years.A comparison of the collected data revealed that the topographic state of the beach before the typhoon,typhoon characteristics,and tidal conditions were possible reasons for the difference in the responses of Dongsha beach to typhoon events.More severe beach erosion was caused by typhoons with higher intensity levels and longer durations,and high tide levels during typhoons can determine the upper limit of the beach profile erosion site.Taken together,these results can be used to improve beach management for storm prevention.

Reducing the negative effects of flywheel disturbance on space camera image quality using the vibration isolation method

Although the performance of space cameras has largely improved, the micro vibration from flywheel disturbances still significantly affects the image quality of these cameras. This study adopted a passive isolation method to reduce the negative effect of flywheel disturbance on image quality. A metal-rubber shock absorber was designed and installed in a real satellite. A finite element model of an entire satellite was constructed, and a transient analysis was conducted afterward. The change in the modulate transfer function was detected using ray tracing and optical transfer function formulas. Experiments based on real products were performed to validate the influence of the metal-rubber shock absorber. The experimental results confirmed the simulation results by showing that the negative effects of flywheel dis- turbance on the image quality of space cameras can be diminished significantly using the vibration isolation method.

ZnNiAl-LDH/Bi_(2)WO_(6)/Cu_(2)O光催化剂的制备及降解性能的研究

采用共沉淀法合成了一系列不同比例ZnNiAl-LDH和Bi_(2)WO_(6)的ZnNiAl-LDH/Bi_(2)WO_(6)/Cu_(2)O复合光催化剂,并通过四环素(TC)降解实验对其光催化性能进行了研究.同时,采用SEM、TEM、HRTEM、XRD、FTIR、UV-Vis、XPS、PL和EPR等方法研究了光催化剂的结构和光学性能.降解实验结果表明:与纯ZnNiAl-LDH、Bi_(2)WO_(6)和Cu_(2)O相比,ZnNiAl-LDH/Bi_(2)WO_(6)/Cu_(2)O复合光催化剂不仅对四环素具有较高的吸附和降解能力,而且能够降低光生电子-空穴对的复合率.其中,5%ZnNiAl-LDH/15%Bi_(2)WO_(6)/80%Cu_(2)O复合光催化剂在可见光条件下对TC的降解效率最高,100 min内TC的降解率可达93%.高光降解性能归因于在Cu_(2)O与Bi_(2)WO_(6)及ZnNiAl-LDH之间存在高效的界面电荷转移和协同作用,从而提高了光生电子空穴对的分离效率.

1300—2019年江苏沿海风暴潮灾害时空分布特征

历史上江苏沿海曾频发2类风暴潮灾害,给当地带来了巨大损失。建立该地区长时间尺度的风暴潮灾害序列对了解过去与预测未来意义重大。以历史文献、地方志、海洋灾害公报等记载的风暴潮资料为基础,建立了风暴潮灾害识别体系,重建了1300—2019年江苏沿海风暴潮灾害序列,分析了风暴潮灾害时空变化规律,探究了其与气候异常和黄河、长江入海口变迁之间的关系。结果表明:江苏沿海地区主要受台风风暴潮灾害的影响,1300—2019年间共发生246次风暴潮灾害,平均周期为2.9年,且近100年来发生频率有增大的趋势;小波分析结果显示,风暴潮灾害年际变化存在17年、61年和115年的时间尺度周期,其中115年周期最为明显。海气相互作用影响下,厄尔尼诺活跃年江苏沿海台风风暴潮灾害偏少;气温与台风风暴潮灾害频次有一定的正相关性,但其影响机制尚不明确。盐城、南通和苏州为风暴潮灾害高发地区,常州和无锡发生频率较小。在气候变暖与江苏海岸冲淤变化的作用下,1855年后盐城和南通地区风暴潮灾害发生频率增加,连云港地区明显增加;随着海岸线的变迁与长江河口的演变,镇江、扬州、泰州与苏锡常地区受风暴潮灾害影响减少。
