Fibroblast growth factor 21(FGF21)was originally identified as an important meta-bolic regulator which plays a crucial physiological role in regulating a variety of metabolic pa-rameters through the metabolic *** a novel multifunctional endocrine growth factor,the role of FGF21 in the metabolic network warrants extensive *** insight was obtained from the observation that the FGF21-dependent mechanism that regulates lipid metabolism,glycogen transformation,and biological effectiveness occurs through the coordi-nated participation of the liver,adipose tissue,central nervous system,and sympathetic *** review focuses on the role of FGF21-uncoupling protein 1(UCP1)signaling in lipid metabolism and how FGF21 alleviates non-alcoholic fatty liver disease(NAFLD).Additionally,this review reveals the mechanism by which FGF21 governs glucolipid *** research on the role of FGF21 in the metabolic network has mostly focused on the crucial pathway of glucolipid ***21 has been shown to have multiple regulatory roles in the metabolic *** an adequate understanding of the concrete regulatory path-ways of FGF21 in the metabolic network has not been attained,this review sheds new light on the metabolic mechanisms of FGF21,explores how FGF21 engages different tissues and organs,and lays a theoretical foundation for future in-depth research on FGF21-targeted treatment of metabolic diseases.

With the theoretical and technological developments related to cratonic strike-slip faults,the Shuntuoguole Low Uplift in the Tarim Basin has attracted considerable attention *** by multi-stage tectonic movements,the strike-slip faults have controlled the distribution of hydrocarbon resources owing to the special fault characteristics and fault-related *** contrast,the kinematics and formation mechanism of strike-slip faults in buried sedimentary basins are difficult to investigate,limiting the discussion of these faults and hydrocarbon *** this study,we identified the characteristics of massive sigmoidal tension gashes(STGs)that formed in the Shunnan area of the Tarim ***-resolution three-dimensional seismic data and attribute analyses were used to investigate their geometric and kinematic ***,the stress state of each point of the STGs was calculated using seismic curvature ***,the formation mechanism of the STGs and their roles in controlling hydrocarbon migration and accumulation were *** results suggest that:(1)the STGs developed in the Shunnan area have a wide distribution,with a tensile fault arranged in an enéchelon pattern,showing an S-shaped *** STGs formed in multiple stages,and differential rotation occurred along the direction of strike-slip stress during formation.(2)Near the principal displacement zone of the strike-slip faults,the stress value of the STGs was higher,gradually decreasing at both *** shallow layer deformation was greater than the deep layer deformation.(3)STGs are critical for connecting source rocks,migrating oil and gas,sealing horizontally,and developing efficient *** study not only provides seismic evidence for the formation and evolution of super large STGs,but also provides certain guidance for oil and gas exploration in this area.

The evolution of the microstructure and toughness of APL5L X80 pipeline steel after thermal welding simulation was investigated by X-ray diffraction,electron backscatter diffraction,and transmission electron *** results indicated that primary heat-affected zones can be divided into weld,coarse-grained,fine-grained,intercritical,and sub-critical *** microstructure of the weld zone is mainly composed of bainitic ferrite and a small amount of granular bainite;however,the original austenite grains are distributed in the columnar *** structure of the coarse-grained zone is similar to that of the weld zone,but the original austenite grains are *** contrast,the microstructure in the fine-grained zone is dominated by fine granular bainite,and the effective grain size is only 8.15μm,thus providing the highest toughness in the entire heat-affected *** intercritical and subcritical zones were brittle valley regions,and the microstructure was dominated by granular ***,the martensite-austenite(M/A)constituents are present in island chains along the grain boundaries,and the coarse size of the M/A constituents seriously reduces the *** results of the crack propagation analyzes revealed that high-angle grain boundaries can significantly slow down crack growth and change the crack direction,thereby increasing the material *** impact toughness of the low-temperature tempering zone was equivalent to that of the columnar grain zone,and the impact toughness was between those of the critical and fine-grained zones.

AIM： To characterize changes in ghrelin levels in response to oral glucose tolerance test （OGTT） and to correlate changes in ghrelin levels with changes in insulin and glucose following OGTT in Chinese obese children of Tanner Ⅰ and Ⅱ stage with insulin resistance. METHODS： 22 obese children with insulin resistance state were divided into four groups according to their Tanner stage and gender： boys of Tanner Ⅰ （fir- Ⅰ ）, boys of Tanner Ⅱ（BT-Ⅱ ）, girls of Tanner Ⅰ （GT- Ⅰ ）, girls of Tanner Ⅱ （GT-Ⅱ）. Ghrelin, insulin and glucose were measured at 0, 30, 60 and 120 rain following OGTT. The control children with normal BMI were divided into control boys of Tanner Ⅰ （CBT- Ⅰ, n = 6）, control boys of Tanner Ⅱ （CBT-Ⅱ, n = 5）, control girls of Tanner Ⅰ （CGT- Ⅰ, n = 6）, control girls of Tanner Ⅱ （CGT-Ⅱ, n = 5）. Fasting serum ghrelin levels were analyzed. RESULTS： Ghrelin levels were lower in obese groups. Ghrelin levels of control group decreased in Tanner Ⅱ stage （CGT- Ⅰ vs CGT-Ⅱ t = -4.703, P = 0.001; CBT- Ⅰ vs CBT- Ⅱ t = -4.794, P = 0.001）. Basal ghrelin levels in fir-Ⅱ decreased more significantly than that in BT- Ⅰ group （t = 2.547, P = 0.029）. Ghrelin levels expressed a downward trend after OGTT among obese children. The decrease in ghrelin levels at 60 min with respect to basal values was 56.9% in BT- Ⅰ. Ghrelin concentrations at 0 min correlated directly with glucose level at 0 min in fir- Ⅰ （r = 0.898, P = 0.015）. There wasn＇t a significant correlation of ghrelin changes with glucose changes and insulin changes during OGTT in obese children with insulin resistance. CONCLUSION： In conclusion, in obese children with insulin resistance, ghrelin levels decreased with advancing pubertal stage. Ghrelin secretion suppression following OGTT was influenced by gender and pubertal stage. Baseline ghrelin levels and ghrelin suppression after OGTT did not significantly correlate with the degree of insulin resistance and insulin sensitivity.

Traditional therapies such as surgery and endocrine therapy no longer meet the clinical needs in prostate cancer treatment,and more effective treatments are urgently *** studies have reported that targeted inhibition of the transcription factor cyclin dependent kinase 7(CDK7)could effectively suppress prostate cancer ***,the toxicity of CDK7 inhibitors such as THZ1 is the main limitation of the clinical *** this work,we synthesized Cys8E(C8E)nanoparticles(NPs)loaded with THZ1(C8E@THZ1),a novel GSH-targeting and stimuli-responsive nano-delivery platform,and investigated its anti-tumor potential and biosafety *** vitro,C8E@THZ1 potently inhibited the proliferation and promoted the apoptosis of prostate cancer *** tumor-bearing mice,C8E@THZ1 inhibited tumors by up to 85%,while the damage of THZ1 to liver function was effectively *** results confirmed that inhibition of CDK7 can effectively block the progression of prostate cancer,and that Cys8E NPs is a highly prospective delivery platform to promote the clinical application of CDK7 inhibitors.

Globally,most organic-rich shales are deposited with volcanic ash *** ash,a source for many sedimentary basins,can affect the sedimentary water environment,alter the primary productivity,and preserve the organic matter(OM)through physical,chemical,and biological *** an increasing number of breakthroughs in shale oil exploration in the Bohai Bay Basin in recent years,less attention has been paid to the crucial role of volcanic impact especially its influence on the OM enrichment and hydrocarbon ***,we studied the petrology,mineralogy,and geochemical characteristics of the organic-rich shale in the upper submember of the fourth member(Es_(4)^(1))and the lower submember of the third member(Es_(3)^(3))of the Shahejie Formation,aiming to better understand the volcanic impact on organic-rich shale *** results show that total organic carbon is higher in the upper shale intervals rich in volcanic ash with enriched light rare earth elements and moderate Eu *** indicates that volcanism promoted OM formation before or after the *** positive correlation between Eu/Eu*and Post-Archean Australian Shale indicates hydrothermal activity before the volcanic *** plane graph of the hydrocarbon-generating intensity(S1+S2)suggests that the heat released by volcanism promoted hydrocarbon ***,the nutrients carried by volcanic ash promoted biological blooms during Es_(4)^(1 )and Es_(3)^(3) deposition,yielding a high primary *** blooms consume large amounts of oxygen and form anoxic environments conducive to the burial and preservation of ***,this study helps to further understand the organic-inorganic interactions caused by typical geological events and provides a guide for the next step of shale oil exploration and development in other lacustrine basins in China.

Global warming in tandem with surface albedo reduction caused by black carbon(BC)deposition on glaciers accelerated glacier melting;however,their respective contributions remain *** in the Qilian Mountains are crucial for the development of oases in the Hexi Corridor;however,their area has decreased by more than 20%over the past ***,this study developed a dynamic deposition model for light-absorbing particles(LAPs),coupled with a surface energy and mass balance *** comprehensively assessed the effects of BC and warming on the melting of a typical glacier in the Qilian Mountains based on the coupled *** on the glacier surface caused 13.1%of annual glacier-wide melting,of which directly deposited atmospheric BC reduced the surface albedo by 0.02 and accounted for 9.1%of glacier *** air temperature during 2000-2010 has increased by 1.5°C relative to that during the 1950s,accounting for 51.9%of current glacier ***,BC emission have increased by 4.6 times compared to those of the early Industrial Revolution recorded in an ice core,accounting conservatively for 6.3%of current glacier *** BC emissions has a limited influence on current glacier melting;however,in the long-term,mitigation should exert a noteworthy impact on glacier melting through the self-purification of glaciers.
