Hepato-biliary profile of potential candidate liver progenitor cells from healthy rat liver

AIM： To evaluate the presence of progenitor cells in healthy adult rat liver displaying the equivalent ad- vanced hepatogenic profile as that obtained in humans. METHODS： Rat fibroblastic-like liver derived cells （rFLDC） were obtained from collagenase-isolated liver cell suspensions and characterized and their phenotype profile determined using flow cytometry, immunocyto- chemistry, reverse transcription polymerase chain reac- tion and functional assays. RESULTS： rFLDC exhibit fibroblastoid morphology, ex- press mesenchymal （CD73, CD90, vimentin, m-smooth muscle actin）, hepatocyte （UGTIA1, CK8） and biliary （CK19） markers. Moreover, these cells are able to store glycogen, and have glucose 6 phosphatase activity, but not UGTIA1 activity. Under the hepatogenic differentia- tion protocol, rFLDC display an up-regulation of hepatocyte markers expression （albumin, tryptophan 2,3-di- oxygenase, G6Pase） correlated to a down-regulation of the expression of the biliary marker CK19. CONCLUSION： Advanced hepatic features observed in human liver progenitor cells could not be demonstrated in rFLDC. However, we demonstrated the presence of an original rodent hepato-biliary cell type.

BACKGROUND Gastrointestinal tumors are among the most common cancer types,and early detection is paramount to improve their ***-free DNA(cfDNA)liquid biopsy raises significant hopes for non-invasive early *** To describe current applications of this technology for gastrointestinal cancer detection and *** A systematic review of the literature was performed across the PubMed *** reporting the use of cfDNA liquid biopsy in the screening or diagnosis of gastrointestinal cancers were included in the *** A total of 263 articles were screened for eligibility,of which 13 articles were *** investigated colorectal cancer(5 studies),pancreatic cancer(2 studies),hepatocellular carcinoma(3 studies),and multi-cancer detection(3 studies),including gastric,oesophageal,or bile duct cancer,representing a total of 4824 *** sensitivities ranged from 71% to 100%,and specificities ranged from 67.4% to 100%.Pre-cancerous lesions detection was less performant with a sensitivity of 16.9% and a 100% specificity in one *** study using a large biobank demonstrated a 94.9% sensitivity in detecting cancer up to 4 years before clinical symptoms,with a 61% accuracy in tissue-of-origin *** cfDNA liquid biopsy seems capable of detecting gastrointestinal cancers at an early stage of development in a non-invasive and repeatable manner and screening simultaneously for multiple cancer types in a single blood *** trials in clinically relevant settings are required to determine the exact place of this technology in gastrointestinal cancer screening and diagnosis strategies.

Feed particle selection and nutrient intake altered by pecking stone consumption and beak length in free-range laying hens

The present study investigated the effects of pecking stones on feeding behaviour of hens from 16 to 46 weeks of age. Eighteen flocks of Hy-Line Brown hens were housed in 2 commercial free-range housing systems. Farm A housed 10 flocks of beak trimmed(infrared beak treatment) hens in fixed sheds. Farm B housed 8 flocks of hens with intact beaks in mobile sheds. On each farm, flocks were equally assigned to control groups(no access to pecking stones) and treatment groups(access to pecking stones). Data were evaluated every 10 weeks. At each time point, 10 hens per flock were housed in individual pens, and each hen was provided with 250 g of mash diet and ad libitum water for 24 h. After 24 h, feed samples were collected and used to determine 24-h feed intake. Nutrient and particle selection was measured by subtracting nutrients and particles present in the leftover feed from the vaules obtained in the offered feed and expressed the change(A). In addition, pecking stone consumption was recorded for each *** were analysed separately for each farm using fixed effects of pecking stone availability and hen ***’s rho correlation coefficients and linear regression models were constructed to evaluate the relationship of beak length and pecking stone usage, discrete mean particle size(dMEAN) consumption(△ dMEAN), and △ nutrient intake. Hens with access to pecking stones consumed significantly lower quantities of large feed particles(>2.8 mm) on farm A(P = 0.029) and selected significantly more fine particles, on farm B(P = 0.013). Overall, positive relationships(P = 0.001) between beak length and pecking stone consumption, A dMEAN, and A phosphorus consumption were observed. In conclusion,pecking stone consumption resulted in reduced selection and consumption of feed particles in hens housed on both farms. Further research is warranted to investigate the effect of pecking stones on sensory innervation of the beak.

Mycotoxins are fungal secondary metabolites detected in many agricultural commodities, especially cereals. Due to their high consumption of cereals, pigs are exposed to these toxins. In the European Union, regulations and/or recommendations exist in pig feed for aflatoxins, ochratoxin A, fumonisins,zearalenone, and trichothecenes, deoxynivalenol and T-2 toxin. These mycotoxins have different toxic effects, but they all target the immune system. They have immunostimulatory or immunosuppressive effects depending on the toxin, the concentration and the parameter investigated. The immune system is primarily responsible for defense against invading organisms. The consequences of the ingestion of mycotoxin-contaminated feed are an increased susceptibility to infectious diseases, a reactivation of chronic infection and a decreased vaccine efficacy. In this review we summarized the data available on the effect of mycotoxins on the immune system and the consequences for pig health.

Within a given free-range flock,some hens prefer to spend the majority of their time in the shed(stayers),while others frequently access the range(rangers).Laying performance has been associated not only with the development of these sub-populations but also with different body weights(BW).The purpose of this study was to determine if range usage,BW or a combination of both is associated with energy metabolism and as such co ntribute to improved hen ***-eight Lohmann Brown hens at 74 wk of age were selected from a commercial free-range farm based on their BW and range usage over a 56-week *** a 2 × 2 factorial arrangement,hens were either classified as heavy(mean ± SEM;2.01±0.02 kg,n=24) or light(1.68±0,01 kg,n=24),and also classified as rangers(accessed the range for 84.1 % of available days,242±3.75 d;n=24) or stayers(accessed the range for 7.17% of available days;23,4±6.08 d,n=24).Stayers had significantly higher metabolizable energy(ME) intake per metabolic BW per d(0.852 vs.0.798 MJ/kg BW0.75 perd;P=0.025),higher heat production(0.637 vs.0.607 MJ/kg BW0.75 perd;P=0.005),higher heat increment(0.267 vs.0.237 MJ/kg BW0.75 per d;P=0.005) and retained more nitrogen(1.59 vs.1.46 g/hen perd;P=0.023) compared to the *** hens had significantly higher metabolic energy intake per metabolic BW(0.854 vs.0.796 MJ/kg BW0.75 per d;P=0.018),net energy(NE) intake(0.595 vs.0.551 MJ/kg BW0.75 per d;P=0.032),and retained energy(0.225 vs.0.181 MJ/kg BW0.75 per d;P=0.032),as well as lower heat production(0.936 vs.1.003 MJ/hen per d;P=0.002) compared to heavier *** interaction was observed across levels of analysis *** light stayers and light *** light rangers had significantly higher NE intake compared to the light stayers(9.77 vs.9.27 MJ/kg BW0.75 per d;P=0.024).In conclusion,light hens were more energy efficient compared to heavy ***,light rangers had a more efficient feed utilisation compared to the light stayers.

We introduce a nanoscale photonic platform based on gallium phosphide. Owing to the favorable material properties, peak power intensity levels of 50 GW∕cm^2 are safely reached in a suspended membrane. Consequently,the field enhancement is exploited to a far greater extent to achieve efficient and strong light–matter *** an example, parametric interactions are shown to reach a deeply nonlinear regime, revealing cascaded fourwave mixing leading to comb generation and high-order soliton dynamics.

BACKGROUND： Hemodynamic changes accompany the initiation, development and rupture of middle cerebral artery （MCA） aneurysms. The complexity of the intraaneurysmal hemodynamic factors has not been completely clarified by the indirect measures and methods used in previous studies. OBJECTIVE： To evaluate correlations of intraoperative hemodynamic factors to initiation and rupture of MCA aneurysms. DESIGN, TIME AND SETTING： A case-control study was performed at the Department of Neurosurgery, Tiantan Hospital Affiliated to Capital Medical University, China between March and October 2008. PARTICIPANTS： A total of 12 consecutive patients diagnosed with MCA aneurysms （MCA aneurysms group） and five patients without middle cerebral artery aneurysms （with aneurysms located at other arteries, control group） were enrolled at the Department of Neurosurgery, Tiantan Hospital Affiliated to Capital Medical University, China. METHODS： The proximal and distal arteries of MCA aneurysms were exposed visibly in the MCA aneurysm group. The M1 segment of MCA without the aneurysm and the aneurysm on other arteries were also exposed visibly in the control group. Hemodynamic indices were then measured using an intraoperative 16 MHz probe installed in a Multi-Dop TCD8X4 device. MAIN OUTCOME MEASURES： Mean （time-averaged velocity） difference, maximum mean, pulsatility index difference, maximum pulsatility index, resistance index difference, maximum resistance index; correlation of development and rupture of MCA aneurysms to intraoperative hemodynamic factors of the parent artery. RESULTS： A total of 12 patients underwent microsurgery for treatment to occlude 15 MCA aneurysms. Of the 15 MCA aneurysms, 12 were located at the bifurcation, two at the M1 segment and one at the M3 segment; eight of the aneurysms were unruptured and seven were ruptured.
