AIM To describe factors associated with treatment failure and frequency of resistance-associated substitutions(RAS).METHOdS Human immunodeficiency virus(HIV)/hepatitis C virus(HCV) coinfected patients starting a first direct-acting antiviral(dAA) regimen before February 2016 and included in the French ANRS CO13 HEPAVIH cohort were eligible. Failure was defined as:(1) non-response [HCV-RNA remained detectable during treatment, at end of treatment(EOT)]; and(2) relapse(HCVRNA suppressed at EOT but detectable thereafter). Sequencing analysis was performed to describe prevalence of drug class-specific RAS. Factors associated with failure were determined using logistic regression *** Among 559 patients, 77% had suppressed plasma HIV-RNA dAA treatment initiation, 41% were cirrhotic, and 68% were HCV treatmentexperienced. Virological treatment failures occurred in 22 patients and were mainly relapses(17, 77%) then undefined failures(3, 14%) and non-responses(2, 9%). Mean treatment duration was 16 wk overall. Posttreatment NS3, NS5 A or NS5 B RAS were detected in 10/14 patients with samples available for sequencing analysis. After adjustment for age, sex, ribavirin use, HCV genotype and treatment duration, low platelet count was the only factor significantly associated with a higher risk of failure(OR: 6.5; 95%CI: 1.8-22.6). CONCLUSION Only 3.9% HIV-HCV coinfected patients failed dAA regimens and RAS were found in 70% of those failing. Low platelet count was independently associated with virological failure.

小胶质细胞NLRP3炎症小体激活正在成为神经退行性变期间神经炎症的关键因素。β-淀粉样蛋白和α-突触核蛋白等致病蛋白的聚集体可触发小胶质细胞NLRP3激活,并导致半胱氨酸天冬氨酸酶激活和白介素-1β分泌。半胱氨酸天冬氨酸酶和白介素-1β均促进肌萎缩侧索硬化(ALS)小鼠SOd1G93A模型的疾病进展,这提示小胶质细胞NLRP3在该进程中起作用。然而,先前的研究表明SOd1G93A模型小鼠的小胶质细胞并不表达NLRP3,并且SOd1G93A蛋白在小胶质细胞中产生白介素-1β不依赖于NLRP3。本研究展示了使用Nlrp3-GFP基因敲入小鼠,在SOd1G93A小鼠中小胶质细胞表达NLRP3。结果表明,聚集和可溶性SOd1G93A均以剂量和时间依赖性方式激活小鼠原代小胶质细胞中的炎性体,导致半胱氨酸天冬氨酸酶和白介素-1β裂解、ASC斑点形成和白介素-1β分泌。重要的是,SOd1G93A不能诱导缺乏Nlrp3的小胶质细胞或用特异性NLRP3抑制剂MCC950预处理的小胶质细胞分泌白介素-1β,这证实NLRP3是介导SOd1诱导的小胶质细胞白介素-1β分泌的关键炎性体复合物。在TdP-43Q331K ALS小鼠模型中也观察到小胶质细胞NLRP3上调,并且TdP-43野生型和突变蛋白也能以NLRP3依赖性方式激活小胶质细胞炎症小体。从机制上,本研究确定活性氧簇和ATP的产生是SOd1G93A介导的NLRP3激活所需的关键事件。总之,本研究的数据表明ALS小胶质细胞表达NLRP3,病理性ALS蛋白激活了小胶质细胞NLRP3炎性体。因此,抑制NLRP3可能是阻止小胶质细胞神经炎症和ALS疾病进展的潜在治疗方法。

[背景]在成年人研究中已知杀虫剂ddT（二氯二苯三氯乙烷）水平升高与血压和高血压呈正相关。越来越多的流行病学和毒理学证据表明，成年期高血压也可能受到生命早期和产前环境的影响。[目的]评估产前杀虫剂ddT暴露是否会增加成人高血压患病风险。[方法]检测一个妇女亚组（n=527）的产前血清样本中ddT（p，p＇和o，p＇-）及其代谢产物p，p＇-ddE（二氯二苯二氯乙烯）的浓度，这些妇女在怀孕期间（1959-1967年）参加了旧金山湾地区一项前瞻性儿童健康和发育出生队列研究。2005--2008年，通过电话采访调查年龄为39—47岁的女儿，以获取自述医师诊断为高血压和高血压药物的使用信息。在Cox比例风险模型的基础上，采用多变量回归分析来估计产前ddT暴露与成年后高血压药物治疗之间关联的相对率，以孕产妇、生命早期和成年期暴露调整潜在的混杂因素。[结果]产前p，p＇ddT暴露与高血压相关联[相对于最低三分位数，P，p＇ddT中、高三分位数校正风险比（aHR）分别为：aHR=3．6，95％CI：1．8-7．2；aHR=2．5，95％CI：1．2-5．330调整独立的高血压危险因素以及敏感性分析后，p，p＇ddT与高血压之间的这些关联更强。[结论]这些研究结果提示，ddT暴露与高血压之间的关联可能源自发育的早期。

Background:Cognitive control is defined as the ability to act flexibly in the environment by either behaving automatically or inhibiting said automatic behaviour and it can be measured using an interleaved pro/anti-saccade *** in cognitive control has been attributed to normal aging and neurological illnesses such as Parkinson’s disease(Pd)as well as decline in other cognitive *** parallel might highlight the role played by cognitive control in information processing and working ***,little is known about the relationship between cognitive control and other cognitive processes such as visual memory,decision making,and visual *** thus propose to correlate the incidence of impaired cognitive control with deficits in visual memory,decision making and visual search in three groups:younger adults,older adults and patients with idiopathic ***:Seventy-one participants,namely 34 adults(M=22.75,Sd=3.8),22 older adults(M=67.4,Sd=8.3),and 20 Pd patients(M=65.59,Sd=8.2)performed four tasks:interleaved pro/anti-saccade,visual memory,decision making,and serial and pop-out visual ***:Results show that within each group,anti-saccade error rate(ER)were significantly and negatively correlated with visual memory ER(ryounger=−0.378,P=0.036;rolder=−0.440,Polder=0.046;rPd=−0.609,P=0.016).On the other hand,correct decision-making reaction times(RT)were significantly correlated with anti-saccade ER,and RTs only in older adults(rER=0.529,P=0.014;rRT=0.512,P=0.018)and Pd patients(rER=0.727,P=0.012;rRT=0.769,P=0.001).For visual search,Pd patients showed a significant relationship between RTs for correct pro-saccades and pop-out(r=0.665,P=0.007),and serial(r=0.641,P=0.010)search ***,there was a significant correlation between MoCA scores and anti-saccade RTs(r=−0.559,P=0.030)and ER(r=−0.562,P=0.029)in Pd *** together,these results support the hypothesis of Pd patients’reliance on bottom-up processes as top-down proces

[目的]采用正交设计L25（56）优化农杆菌介导的AtMYB118基因对橡胶树易碎胚性愈伤组织的遗传转化体系,为橡胶树品种遗传改良提供参考.[方法]以75.0 mg/L卡那霉素筛选经过转化的愈伤组织;采用GUS瞬时表达检测方法评价6个因素即预培养时间、农杆菌菌液浓度（Od600）、乙酰丁香酮（AS）浓度、侵染时间、共培养温度和共培养时间对遗传转化的影响.[结果]6个因素显著影响橡胶树长期培养的易碎胚性愈伤组织的转化频率,易碎胚性愈伤组织遗传转化优化条件为：预培养0d,菌液Od600为0.7,侵染时间7 min,共培养时间5d,AS 200 μmol/L,共培养温度25℃.在此优化条件下可获得最高转化频率.经过4～6个月的筛选,获得17个GUS染色阳性的易碎愈伤组织系.经PCR和反转录PCR分析转化愈伤组织,进一步证实uidA、nptⅡ、AtMYB118基因已被整合到愈伤组织基因组中并表达.培养获得1539个胚状体,其中164个为转基因胚状体,转化频率为10.6％;最终获得4株转AtMYB118基因植株.[结论]优化获得可行有效的橡胶树易碎胚性愈伤组织遗传转化体系,可为其品种遗传改良提供技术支持.
