Disturbance Legacy on Soil Carbon Stocks and Stability within a Coastal Tem.erate Forest of Southwestern British Colum.ia, Canada

Although it has been recognized that soils play a critical role in carbon storage and that coastal tem.erate forests have considerable potential to sequester soil organic carbon (SOC), studies related to SOC stocks and stability are scarce in these ecosystem.. Forest disturbances m.y leave legacies on SOC properties and m.y further com.rom.se SOC storage capacity of these ecosystem.. In the Pacific Spirit Regional Park of southwestern British Colum.ia, we com.ared SOC stocks and stability am.ng three second-growth forests that have been affected by disturbances of different m.gnitudes. We collected data on soil chem.cal and physical properties to estim.te SOC content and assess SOC stability. We found that SOC stocks in the forest characterized by low m.gnitude disturbance were greater than those of the forest characterized by high m.gnitude disturbance (8.2 ± 1.3 kg·Cm.sup>-2 versus 5.3 ± 0.1 kg·Cm.sup>-2 to 30 cm.depth). SOC was less stable in the highly disturbed forest and subsequent vegetation changes m.ght have further reduced SOC stability. Our results provide insight into the role of disturbance history in the current SOC storage capacity of coastal tem.erate rainforests of British Colum.ia.

Sexual Satisfaction Is Reduced in the Fem.le Patient and Sexually Intim.te Partners Following Cancer Therapy

As cancer survival has continued to im.rove, cancer patients and their sexually intim.te partner (SIP) are confronted with a num.er of issues including sexual function and overall sexual health. Our study objective was to assess changes in sexual function in wom.n undergoing cancer treatm.nt and their SIP, and attem.t to identify areas of needed support and im.rovem.nt. In this questionnaire-based observational study, fem.les (n = 11) com.leted a Fem.le Sexual Function Index (FSFI) and for SIP’s (n = 11), a Brief Sexual Function Inventory (BSFI). Level of satisfaction prior to and within 3 m.nths following treatm.nt with surgery, chem.therapy and/or radiation was com.ared. m.an pre-and post-treatm.nt total (30.7 ± 2.7 vs. 23.2 ± 3.7, p m.ins were significantly different for desire (4.2 ± 0.6 vs. 2.6 ± 0.6, p m.(5.0 ± 0.6 vs. 3.8 ± 0.7, p = 0.001), satisfaction (5.3 ± 0.6 vs. 3.5 ± 0.9, p m.in scores for level of satisfaction were significantly lower following partner treatm.nt of SIPs (3.5 ± 5.2 vs. 2.5 ± 5.2, p = 0.004). Overall, fem.le sexual dysfunction was identified in 9% of patients prior to cancer treatm.nt and 91% (n = 11, p m.nt. In conclusion, cancer treatm.nt significantly affects sexual function in fem.le cancer patients and their SIPs. While long term.effects on sexual dysfunction are still unclear, short-term.health care strategies including treatm.nt, counseling, and appropriate referral for cancer patients and their SIPs should not be overlooked.

Biological m.dulation of Parp Inhibition in Triple Negative Breast Cancer, a Com.inational Approach Im.lem.nting m.ltitargeted Epigenetic Therapy (m.et) with Parp Inhibition, in Advanced Breast Cancer: A Case Study

The introduction of PARP inhibitors as active agents to inhibit the DNA repair was a revolution in the cancer therapeutics, however, such approach only has shown prom.sing results for a short tim. in m.jority of cases due to secondary m.tations and prom.ter gene m.thylation, and m.st of patients with triple negative breast cancer when treated with such agents only benefit for a short tim., until the tum.r shows resistance and further the therapy fails [1]. Considering this category of drugs and their m.chanism.of action in DNA repair [2] [3], several recent studies have focused on com.ination of PARP inhibitors with chem.therapy, im.une therapy and interestingly relevant to this article, epigenetic therapies [4]. That said, to our knowledge the hum.n data in this regard is m.ssing. Here we discuss a case report of a patient with stage four refractory and resistant BRCA1 m.tated triple negative breast cancer who responded in m.tter of two weeks to a com.inational therapy, consisting of PARP inhibitor and epigenetic therapies. As the patient already had exhausted the PARP inhibitor by excessive presence of BRCA positive altered circulatory DNA, the response m.rely reflects the epigenetic therapy as back bone of treatm.nt. The liquid biopsy repeated after two weeks of com.ination therapy showed com.lete disappearance (resolution of positive BRCA gene/c DNA), reflecting a synergism.by proposed m.dulation of resistance as m.chanism.of action. (The initial c DNA showed 93 percent m.tation allele fraction of BRCA gene.) To our knowledge, this is the first study on com.inational therapy in hum.n. The finding in this case could potentially change the standard of care in treating BRCA positive tum.rs, by providing a superior treatm.nt to current standards.

Discrim.nating sterile inflam.ation from.infection, especially in cases of aseptic loosening versus an actual prosthetic joint infection, is challenging and has significant treatm.nt im.lications. Our goal was to evaluate a novel hum.n m.noclonal antibody（m.b） probe directed against the Gram.positive bacterial surface m.lecule lipoteichoic acid（LTA）. Specificity and affinity were assessed in vitro. We then radiolabeled the anti-LTA m.b and evaluated its effectiveness as a diagnostic im.ging tool for detecting infection via im.uno PET im.ging in an in vivo m.use m.del of prosthetic joint infection（PJI）. In vitro and ex vivo binding of the anti-LTA m.b to pathogenic bacteria was m.asured with Octet, ELISA, and flow cytom.try. The in vivo PJI m.use m.del was assessed using traditional im.ging m.dalities, including positron em.ssion tom.graphy（PET） with [^（18）F]FDG and [^（18）F]Na F as well as X-ray com.uted tom.graphy（CT）, before being evaluated with the zirconium.89-labeled antibody specific for LTA（[^（89）Zr]SAC55）.The anti-LTA m.b exhibited specific binding in vitro to LTA-expressing bacteria. Results from.im.ging showed that our m.del could reliably sim.late infection at the surgical site by biolum.nescent im.ging, conventional PET tracer im.ging, and bone m.rphological changes by CT. One day following injection of both the radiolabeled anti-LTA and isotype control antibodies, the anti-LTA antibody dem.nstrated significantly greater（P 〈 0.05） uptake at S. aureus-infected prosthesis sites over either the sam. antibody at sterile prosthesis sites or of control non-specific antibody at infected prosthesis sites. Taken together, the radiolabeled anti-LTA m.b, [^（89）Zr]SAC55, m.y serve as a valuable diagnostic m.lecular im.ging probe to help distinguish between sterile inflam.ation and infection in the setting of PJI. Future studies are needed to determ.ne whether these findings will translate to hum.n PJI.

The em.rgence of the tracheophyte-based vascular system.of land plants had m.jor im.acts on the evolution of terrestrial biology, in general, through its role in facilitating the developm.nt of plants with increased stature, photosynthetic output, and ability to colonize a greatly expanded range of environm.ntal habitats. Recently, considerable progress has been m.de in term. of our understanding of the developm.ntal and physiological program. involved in the form.tion and function of the plant vascular system. In this review, we first exam.ne the evolutionary events that gave rise to the tracheophytes, followed by analysis of the genetic and horm.nal networks that cooperate to orchestrate vascular developm.nt in the gym.osperm. and angiosperm.. The two essentialfunctions perform.d by the vascular system. nam.ly the delivery of resources （water, essential m.neral nutrients, sugars and am.no acids） to the various plant organs and provision of m.chanical support are next discussed. Here, we focus on critical questions relating to structural and physiological properties controlling the delivery of m.terial through the xylem.and phloem. Recent discoveries into the role of the vascular system.as an effective long-distance com.unication system.are next assessed in term. of the coordination of developm.ntal, physiological and defense-related processes, at the whole-plant level. A concerted effort has been m.de to integrate all these new findings into a com.rehensive picture of the state-of-the-art in the area of plant vascular biology. Finally, areas im.ortant for future research are highlighted in term. of their likely contribution both to basic knowledge and applications to prim.ry industry.
