MGMT and MLH1 methylation in Helicobacter pylori-infected children and adults

AIM:To evaluate the association between Helicobacter pylori(***) infection and MLH1 and MGMT methylation and its relationship with microsatellite instability(MSI).METHODS:The methylation status of the MLH1 and MGMT promoter region was analysed by methylation specific methylation-polymerase chain reaction(MSPPcR) in gastric biopsy samples from uninfected or ***-infected children(n = 50),from adults with chronic gastritis(n = 97) and from adults with gastric cancer(n = 92).MLH1 and MGMT mRNA expression were measured by real-time PcR and normalised to a constitutive gene(β actin).MSI analysis was performed by screening MSI markers at 4 loci(Bat-25,Bat-26,D17S250 and D2S123) with PcR;PcR products were analysed by single strand conformation polymorphism followed by silver *** analyses were performed with either the χ 2 test with Yates continuity correction or Fisher's exact test,and statistical significance for expression analysis was assessed using an unpaired Student's ***:Methylation was not detected in the promoter regions of MLH1 and MGMT in gastric biopsy samples from children,regardless of *** infection *** MGMT promoter was methylated in 51% of chronic gastritis adult patients and was associated with *** infection(P c cancer patients,and the difference in the percentage of methylated samples between these patients and those from ***-infected chronic gastritis patients was statistically significant(P cies among ***-infected and non-infected chronic gastritis adult patients were 13% and 7%,*** observed methylation of the MLH1 promoter(39%) and increased MSI levels(68%) in samples from gastric cancer patients in comparison to samples from ***-infected adult chronic gastritis patients(P ctively).The frequency of promoter methylation for both genes was higher in gastric cancer sam

Effects of porosity,dry unit weight,cement content and void/cement ratio on unconfined compressive strength of roof tile waste-silty soil mixtures

One of the conventional ways to improve the mechanical behavior of soils is to mix them with cementing agents such as cement, lime and fly ash. Recently, introduction to alternative materials or sub-products that can be adopted to improve the soil strength is of paramount importance. Therefore, the present study aims to investigate the effects of porosity(h), dry unit weight(gd) of molding, cement content(c)and porosity/volumetric cement content ratio(h/civ) or void/cement ratio on the unconfined compressive strength(quor UcS) of silty soileroof tile waste(RT) mixtures. Soil samples are molded into four different dry unit weights(i.e. 13 kN/m^3, 13.67 kN/m^3, 14.33 kN/m^3 and 15 kN/m^3) using 3%, 6% and 9%cement and 5%, 15% and 30% RT. The results show that with the addition of cement, the strength of the RT esoil mixtures increases in a linear manner. On the other hand, the addition of RT decreases quof the samples at a constant percentage of cement, and the decrease in porosity can increase qu. A dosage equation is derived from the experimental data using the porosity/volumetric cement content ratio(h/c_(iv)) where the control variables are the moisture content, crushed tile content, cement content and porosity.

Optimizing the evolution of strength for lime-stabilized rammed soil

capacitorless Solid-state Power Filter for Single-phase Dc-Ac converters

converters rely on passive filtering as a crucial element due to the high-frequency operational characteristics of power *** filtering methods involve a dual inductor-capacitor(Lc)cell or an inductor-capacitor-inductor(LcL)***,capacitors are susceptible to wear-out mechanisms and failure ***,the necessity for monitoring and regular replacement adds to an elevated cost of ownership for such *** utilization of an active output power filter can be used to diminish the dimensions of the Lc filter and the electrolytic dc-link capacitor,even though the inclusion of capacitors remains an indispensable part of the *** paper introduces capacitorless solid-state power filter(SSPF)for single-phase dc-ac *** proposed configuration is capable of generating a sinusoidal ac voltage without relying on *** proposed filter,composed of a planar transformer and an H-bridge converter operating at high frequency,injects voltage harmonics to attain a sinusoidal output *** design parameters of the planar transformer are incorporated,and the impact of magnetizing and leakage inductances on the operation of the SSPF is *** analysis,supported by simulation and experimental results,are provided for a design example for a single-phase *** total harmonic distortion observed in the output voltage is well below the IEEE 519 *** system operation is experimentally tested under both steady-state and dynamic conditions.A comparison with existing technology is presented,demonstrating that the proposed topology reduces the passive components used for filtering.

We discuss a recent analysis by Yu et al.[RAA 11,125（2011）] about constraints on the smoothness parameter α and dark energy models using observational H（z） *** is argued here that their procedure is conceptually inconsistent with the basic assumptions underlying the adopted Dyer-Roeder *** order to properly quantify the influence of the H（z） data on the smoothness parameter α,a χ2-test involving a sample of type Ia supernovae and H（z） data in the context of a flat ΛcDM model is *** result is confronted with an earlier approach discussed by santos et al.（2008） without H（z） *** the（Ωm,α） plane,it is found that such parameters are now restricted to be in the intervals 0.66 ≤ α ≤ 1.0 and 0.27 ≤ Ωm ≤ 0.37 within the 95.4 confidence level（2σ）,and,therefore,are fully compatible with the homogeneous *** basic conclusion is that a joint analysis involving H（z） data can indirectly improve our knowledge about the influence of the ***,this happens only because the H（z） data provide tighter constraints on the matter density parameter Ωm.

Evaluation of hypoxia inducible factor targeting pharmacological drugs as antileishmanial agents

Objective:To evaluate whether hypoxia inducible factor(HIF-1α) targeting pharmacological drugs,echinomycin,resveratrol and cdcl_2 which inhibit HIF-1α stimulation,and mimosine,which enhances the stability of HIF-1α present antileishmanial ***:The leishmanicidal effect of drugs was evaluated in mouse macrophages and Balb/c mouse model for cutaneous ***:Resveratrol and cdcl_2 reduced the parasite load [Ic50,(27.3±2.25) μM and(24.8±0.95) μM,respectively].The Ic50 value of echinomycin was(22.7±7.36) nM and mimosine did not alter the parasite load in primary *** macrophage viability Ic50 values for resveratrol,echinomycin and cdcl_2 and mimosine were >40 μM,>100 nM,> 200 μM and>2 000 μM,*** vivo no differences between cutaneous lesions from control,resveratrol-and echinomycin-treated Balb/c mice were ***:Resveratrol,echinomycin and cdcl_2 reduce parasite survival in *** HIF-1α targeting pharmacological drugs require further study to more fully determine their anti-Leishmania potential and their role in therapeutic strategies.

The caO-P_(2)O_(5)-SiO_(2)-MgO system presents several compounds used as biomaterials such as hydroxyapatite(HA),tricalcium phosphate(TcP)and TcP with magnesium substituting partial calcium(TcMP).Theβ-TcMP phase with whitlockite structure has interesting biological features and mechanical properties,meeting the requirements of a bioactive material for bone *** this work,the production of Mg-doped TcP,β-TcMP,has been investigated by crystallization from a glass composed of 52.75 wt%3caO·P_(2)O_(5),30 wt%SiO_(2)and 17.25 wt%MgO(i.e.,31.7 mol%caO,10.6 mol%P_(2)O_(5),26.6 mol%MgO and 31.1 mol%SiO_(2))using heat treatments between 775℃and 1100℃for up to 8 *** devitrification process of the glass has been accompanied by differential scanning calorimetry(DSc),high-resolution X-ray diffraction(HRXRD),relative density and bending strength *** characterization by HRXRD and DSc revealed the occurrence of whitlockite soon after the bulk glass preparation,a transient non-cataloged silicate between 800℃and 1100℃,and the formation of diopside in samples treated at 1100℃as crystalline *** overall crystalline fraction varied from 26%to 70%depending on the heat ***,contraction of the a-axis lattice parameter and expansion of the c-axis lattice parameter of the whitlockite structure have been observed during the heat treatments,which were attributed to theβ-TcMP formation with the partial substitution of ca^(2+)by Mg^(2+).Relative densities near 99%and 97%for the glass and glass-ceramics respectively indicated a discrete reduction as a function of the devitrification *** strengths of 70 MPa and 120 MPa were determined for the glass and glass-ceramic material crystallized at 975℃for 4 h,respectively.

As a European Union(EU)member,Portugal must comply with reductions in plastic *** Portugal,the 330 items/100 m of beach litter,comprising up to 3.9 million pieces and of which 88%is plastic,is higher than the EU median(149 items/100 m)and must be reduced to 20 items/100 m(94%).Integrative measures are needed to reduce littering and improve plastics’use and disposal under the circular *** this 414 kt of plastic packaging waste,163 kt were declared plastic packaging,140 kt subjected to recycling,and 94 kt to energy *** current recycling rate of plastic packaging(34%)should be improved to reach EU recycling averages(42%)and goals and to provide widespread benefits,considering revenues of 167€/*** a net importer of waste,Portugal could benefit from the valorization of imported *** increased recycling,pyrolysis and gasification could provide short-term alternatives for producing value-added substances from plastic waste,such as hydrogen,consistent with the National Plan of Hydrogen and improving ongoing regulations on single-use *** manuscript provides an integrative view of plastics in Portugal,from use to disposal,providing specific recommendations under the circular economy.
