Antibacterial activity and inhibition against Staphylococcus aureus NorA efflux pump by ferulic acid and its esterified derivatives

Objective:To evaluate the inhibitory activity of ferulic acid and four of its esterified derivatives(methyl,ethyl,propyl,and butyl)against resistance mechanisms in Staphylococcus aureus ***:Ferulic acid derivatives were obtained by esterification with methanol,ethanol,propanol,and butanol,and then characterized by hydrogen and carbon-13 nuclear magnetic resonance *** minimum inhibitory concentrations(MIC)of ferulic acid and its esterified derivatives,ethidium bromide,and norfloxacin were obtained using the microdilution test,while the efflux pump inhibition test was conducted by examining reduction in the MICs of norfloxacin and ethidium *** docking was also carried out using the Schrodinger Suite 2015 molecular modeling software.A three-dimensional model of NorA efflux pump was generated using *** best scoring model was used as a receptor for ligand-receptor ***:The methyl and butyl ester derivatives did not demonstrate significant antimicrobial ***,a significant synergic effect was evidenced when norfloxacin was combined with the ethyl and propyl esterified *** docking study demonstrated favorable energy of interaction between ferulate derivatives and NorA,and amino acid residues TYR57,TYR58,and LEU255 were present commonly in stabilizing all *** PCA analysis corroborated the docking hypothesis that the lipophilic character and hydrogen bond interactions were the most relevant characteristics involved with NorA *** pharmacokinetic parameters of ferulic acid derivatives showed good ADMET properties,demonstrating that they can be easily absorbed and have no effect or inhibit the cytochrome P450 enzyme complex,revealing their potential as drug ***:This study provides strong evidence that the molecular basis for this activity is potentially due to the NorA efflux pump.

Integrating Crops-Tree Systems with Outgrower Schemes and Food Security

Outgrower Schemes have been implemented in different parts of the world to provide wood to forest and other companies and as income alternatives for farmers in rural areas. They have been considered by some researchers and practitioners as important advances in the area of planted forest and promoted by governments and forest companies in partnership with rural farmers and communities. Considering the importance of subsistence crops and forest products to smallholders and consumers and the economic viability of agroforestry, attention is given in this paper to Outgrower Schemes that combine tree planting with agroforestry, to contribute to food security and environment preservation in developing countries. The issue of Outgrower Schemes through integrated crops-tree systems was raised in a previous study of Outgrower Schemes in Brazil, commissioned by the FAO1. This paper examines, based on the research done for that study, the views and perspectives of the companies＇ personnel, the tree growers that participate in the programs studied, and other stakeholders＇ related institutions involved in such schemes with regard to the promotion of Outgrower Schemes through integrated crops-tree systems, linking them with agroforestry, which is a way those Outgrower Schemes can contribute to food security and environment preservation goals.

Hesperidin Reduces Cisplatin-lnduced DNA damage in Bone Marrow Cells of Mice

Hesperidin is a bioflavonoid abundantly found in citrus fruits and displays chemoprotective effects against DNA （deoxyribonucleic acid） damage, however there are few reports about hesperidin effects against cisplatin-DNA damage induction. The aim of this work was to evaluate hesperidin antimutagenicity against cisptatin-DNA damage. （1） The antimutagenicity of hesperidin was assayed by bone marrow of mice in vivo using the micronucleus test. Hesperidin pre-treatment protocol reduced the frequency of MNPCE （micronucleated polychromatic erythrocytes） and the dose of 100 mg·kg-1 was highest efficiency, with 65.24% of damage reduction. In the simultaneous treatment protocol, the dose of 200 mg·kg-1 exhibited a more effective reduction of MNPCE, with 94.01% of damage reduction. （2） Hesperidin was also effective in reducing the MNPCE frequency in the post-treatment protocol for all doses, with 77.48%, 82.13% and 90.08% of damage reduction at the doses of 100, 200 and 400 mg·kg-1, respectively. From the study, it can be concluded that hesperidin was able to promote the reduction of micronuclei frequency and DNA damage induced by cisplatin. Hesperidin is a powerful antioxidant compound and its chemoprotective effects on DNA may occur due to its association with the antioxidant cell system which is responsible for eliminate free radicals generated by chemical harmful to DNA.

Characterization of Flaxseed Oil for Nuclear Magnetic Resonance and Its Encapsulation

Flaxseed (Linum usitatissimum L.) is one of the oldest crops known by humans and it has been used in numerous applications, such as in the textile industry, feed formulation, fertilizers, and paper industry. However, nowadays these seeds have won an important highlight for human consumption due to their active ingredients that make them an excellent functional food. Thus, this study aimed to extract flaxseed oil, an oil rich in omega 3 and 6, characterize it by using nuclear magnetic resonance (NMR) and Fourier-transform infrared spectroscopy (FTIR) and then encapsulate this oil in polycaprolactone (PCL) on the micro scale, using the nanoprecipitation technique and subsequently freeze-drying. To determine the mean diameter, the dynamic light scattering technique (DLS) was used, and to verify whether there was encapsulation, the pulse sequence MSE-FID, an NMR sequence in the time domain, was also used. In addition to the previously mentioned techniques, X-ray diffraction (XRD) was also employed. Flaxseed flour was also analyzed by time-domain NMR and FTIR. The results obtained by NMR show that the oil consists of fatty acid esters in the form of triglycerides in which there is the presence of esters of α-linolenic and linoleic acids, respectively ALA and LA, according to the literature. Regarding the material after encapsulation, it presented a mean diameter of 445.2 ± 41 nm and PDI of 0.674 ± 0.064, therefore classified as microparticles. Finally, using the sequence MSE-FID and the FTIR, it can be concluded that there has been the microencapsulation of flaxseed oil in the particles formed.

本文报告了为创建护理知识网络(Nursing Knowledge Network,NKN)对不同国家护理院校进行环境扫描调查所收集到的信息。采用在线方式(2021年2-10月)进行环境扫描,探索研究领域的确定、现有资源、预期收益、教学研究创新、知识传播障碍及网络的预期贡献。目标信息提供者包括来自13个国家63所大学的200名护理教师。要求每所大学有1名参与者回答问卷。共有42所大学(约70%)回复,包括巴西21所,加拿大4所,葡萄牙和西班牙各3所,哥伦比亚、墨西哥、秘鲁和美国各2所,智利、意大利和巴拉圭各1所。其分享了相关信息和见解:通过护理高等教育机构之间的协作和交流,全球可以加强共同创新和研究合作能力;较少被研究的领域是知识网络生成证据、为当地科学实践提供信息的潜在重点。这些信息将为进一步了解护理发展,以及地方、区域和全球护理证据应用和传播相关的政府政策与计划制订提供参考。

Flash microbiocide:A Rapid and Economic Method for Determination of MBC and MFC

Although nowadays there are methods for determining the Minimal Bactericidal Concentration and Minimal Fungicidal Concentration, it is indispensable that?the establishment of innovative methodologies?could be more practical and cheaper. The new methodology Flash microbiocide is an assay in which one aliquot from 96 well plate of Minimal Inhibitory Concentration test is transferred to another plate containing different culture medium. The correspondence with the reference methods described in the National Committee for Clinical Laboratory Standards (NCCLS-CLSI) document M26-A was achieved, denoting the efficiency of this fast and simple method.

Polymorphisms of folate metabolism genes in patients with cirrhosis and hepatocellular carcinoma

AIM To evaluated the association of the risk factors and polymorphisms in MTHFR C677 T, MTHFR A1298 C, MTR A2756 G and MTRR A66 G *** Patients with cirrhosis(n = 116), hepatocellular carcinoma(HCC)(n = 71) and controls(n = 356) were included. Polymerase chain reaction followed by enzymatic digestion and allelic discrimination technique real-time PCR techniques were used for analysis. MINITAB-14.0and SNPstats were utilized for statistical analysis. RESULTS Showed that age ≥ 46 years(OR = 10.31; 95%CI: 5.66-18.76; P < 0.001) and smoking(OR = 0.47; 95%CI: 0.28-0.78; P = 0.003) were associated with cirrhosis. Age ≥ 46 years(OR = 16.36; 95%CI: 6.68-40.05; P < 0.001) and alcohol habit(OR = 2.01; 95%CI: 1.03-3.89; P = 0.039) were associated with HCC. MTHFR A1298 C in codominant model(OR = 3.37; 95%CI: 1.52-7.50; P = 0.014), recessive model(OR = 3.04; 95%CI: 1.43-6.47; P = 0.0051) and additive model(OR = 1.71; 95%CI: 1.16-2.52; P = 0.0072) was associated with HCC, as well as MTR A2756 G in the additive model(OR = 1.68; 95%CI: 1.01-2.77; P = 0.047), and MTRR A66 G in the codominant model(OR = 3.26; 95%CI: 1.54-6.87; P < 0.001), dominant model(OR = 2.55; 95%CI: 1.24-5.25; P = 0.007) and overdominant model(OR = 3.05; 95%CI: 1.66-5.62; P < 0.001). MTR A2756 G in the additive model(OR = 1.54; 95%CI: 1.02-2.33; P = 0.042) and smokers who presented at least one polymorphic allele for MTRR A66G(OR = 1.71; 95%CI: 0.77-3.82; P = 0.0051) showed increased risk for cirrhosis. There was no association between clinical parameters and polymorphisms. CONCLUSION Age ≥ 46 years, alcohol habit and MTR A2756 G, MTHFR A1298 C and MTRR A66 G polymorphisms are associated with an increased risk of HCC development; age ≥ 46 years, tobacco habit and the MTR A2756 G polymorphism are associated with cirrhosis.

Characterization of soil fauna under the influence of mercury atmospheric deposition in Atlantic Forest, Rio de Janeiro, Brazil

The increasing levels of mercury（Hg） found in the atmosphere arising from anthropogenic sources, have been the object of great concern in the past two decades in industrialized countries. Brazil is the seventh country with the highest rate of mercury in the *** major input of Hg to ecosystems is through atmospheric deposition（wet and dry）, being transported in the atmosphere over large distances. The forest biomes are of strong importance in the atmosphere/soil cycling of elemental Hg through foliar uptake and subsequent transference to the soil through litter, playing an important role as sink of this element. Soil microarthropods are keys to understanding the soil ecosystem, and for such purpose were characterized by the soil fauna of two Units of Forest Conservation of the state of the Rio de Janeiro, inwhich one of the areas suffer quite interference from petrochemicals and industrial anthropogenic activities and other area almost exempts of these perturbations. The results showed that soil and litter of the Atlantic Forest in Brazil tend to stock high mercury concentrations, which could affect the abundance and richness of soil fauna, endangering its biodiversity and thereby the functioning of ecosystems.

Development of Safflower Subjected to Nitrogen Rates in Cerrado Soil

In a context in which the nitrogen fertilizer recommendations in the literature for safflower crop show up conflicting, it was aimed by the present study to evaluate the development of safflower when subjected to nitrogen rates in an Oxisol collected under Cerrado vegetation. The experiment was conducted in a greenhouse using pots filled with Oxisol collected in area under Cerrado vegetation in the layer from 0.0 to 0.20 m. Each experimental unit was represented by a plastic pot of 5 dm-3, the experimental design was completely randomized, consisting of six nitrogen rates (0, 60, 120, 180, 240 and 300 mg°dm-3) and six repetitions. It were assessed plant height, number of leaves, chlorophyll content, number of heads, dry mass of shoot and dry mass of root. The results were submitted to variance analysis and, when significant, to regression analysis, both at 5% probability by SISVAR program. Nitrogen rates positively influence the development of safflower, and the one that best promotes this development is between 160 and 190 mg°dm-3.

Knowledge of the mechanisms that trigger infection-related encephalopathies is still very limited and cell therapies are one of the most promising alternatives for neurodegenerative diseases,and its application in infectious diseases can be of great *** stromal cells are cells with great immunomodulatory potential;therefore,their use in clinical and preclinical studies in a variety of diseases,including central nervous system diseases,increased in the last *** stromal cells can exert their beneficial effects through several mechanisms,such as direct cell contact,through surface receptors,and also through paracrine or endocrine *** paracrine mechanism is widely accepted by the scientific community and involves the release of soluble factors,which include cytokines,chemokines and trophic factors,and extracellular *** mini review discusses mesenchymal stromal cells mechanisms of action in neurological disorders,the neuroinflammatory process that takes place in the brain as a result of peripheral inflammation and changes in the brain’s cellular scenario as a common factor in central nervous system diseases,and mesenchymal stromal cells therapy in *** stromal cells have been shown to act in neuroinflammation processes,leading to improved survival and mitigating behavioral *** recently,these cells have been tested in preclinical models of infectious diseases-associated encephalopathies(e.g.,cerebral malaria and sepsis associated encephalopathy)and have shown satisfactory results.
