To reflect human development,it is critical to create a substrate that can support long-term cell survival,differentiation,and *** are promising materials for 3D ***,a bulk structure consisting of dense polymer networks often leads to suboptimal microenvironments that impedes nutrient exchange and cell-to-cell ***,granular hydrogel-based scaffolds were used to support 3D human induced pluripotent stem cell(hipSC)-derived neural networks.A custom designed 3D printed toolset was developed to extrude hyaluronic acid hydrogel through a porous nylon fabric to generate hydrogel *** and hydrogel granules were combined using a weaker secondary gelation step,forming self-supporting cell laden *** three and seven days,granular scaffolds supported higher cell viability compared to bulk hydrogels,whereas granular scaffolds supported more neurite bearing cells and longer neurite extensions(65.52±11.59μm)after seven days compared to bulk hydrogels(22.90±4.70μm).Long-term(three-month)cultures of clinically relevant hipSC-derived neural cells in granular hydrogels supported well established neuronal and astrocytic colonies and a high level of neurite extension both inside and beyond the *** approach is significant as it provides a simple,rapid and efficient way to achieve a tissue-relevant granular structure within hydrogel cultures.

The interaction of sperm with the egg＇s extracellular matrix, the zona pellucida （Zp） is the first step of the union between male and female gametes. The molecular mechanisms of this process have been studied for the past six decades with the results obtained being both interesting and confusing. In this article, we describe our recent work, which attempts to address two lines of questions from previous studies. First, because there are numerous Zp binding proteins reported byvarious researchers, how do these proteins act together in sperm-Zp interaction？ Second, why do a number of acrosomal proteins have Zp affinity？ Are they involved mainly in the initial sperm-Zp binding or rather in anchoring acrosome reacting/reacted spermatozoa to the Zp？ Our studies reveal that a number of Zp binding proteins and chaperones, extracted from the anterior sperm head plasma membrane, coexist as high molecular weight （HMW） complexes, and that these complexes in capacitated spermatozoa have preferential ability to bind to the Zp. Zonadhesin （ZAN）, known as an acrosomal protein with Zp affinity, is one of these proteins in the HMW complexes. Immunoprecipitation indicates that ZAN interacts with other acrosomal proteins, proacrosin/acrosin and sp32 （ACRBp）, also present in the HMW complexes. Immunodetection of ZAN and proacrosin/acrosin on spermatozoa further indicates that both proteins traffic to the sperm head surface during capacitation where the sperm acrosomal matrix is still intact, and therefore they are likely involved in the initial sperm-Zp binding step.

Meta-analysis comparing differing methods of endoscopic therapy for colorectal lesions

AIM： To compare the outcomes of endoscopic mucosal resection （EMR） and endoscopic submucosal dissection （ESD） of colorectal ***： An electronic systematic literature search of four computerized databases was performed in July 2014 identifying studies reporting the outcomes of colorectal ESD and EMR. The primary outcome measures were en-bloc resection rate, endoscopic clearance rate and lesion recurrence rate of the patients followed up. The secondary outcome was the complication rate （including bleeding, perforation and surgery post EMR or ESD rate）. Statistical pooling and random effects modelling of the studies calculating risk difference, heterogeneity and assessment of bias and quality were ***： Six observational studies reporting the outcomes of 1324 procedures were included. The en-bloc resection rate was 50% higher in the ESD group than in the EMR group （95%CI： 0.17-0.83, p 〈 0.0001, I2 = 99.7%）. Endoscopic clearance rates were also signifcantly higher in the ESD group （95%CI： -0.06-0.02, p 〈 0.0001, I2 = 92.5%）. The perforation rate was 7% higher in the ESD group than the EMR group （95%CI： 0.05-0.09, p 〉 0.05, I2 = 41.1%） and the rate of recurrence was 50% higher in the EMR group than in the ESD group （95%CI： 0.20-0.79, p 〈 0.001, I2 = 99.5%）. Heterogeneity remained consistent when subgroup analysis of high quality studies was performed （with the exception of piecemeal resection rate）, and overall effect sizes remained unchanged for all ***： ESD demonstrates higher en-bloc resection rates and lower recurrence rates compared to colorectal EMR. Differences in outcomes may beneft from increased assessment through well-designed comparative studies.
