Effects of the Syzygium aromaticum L. extract on antioxidation and inhibition of matrix metalloproteinase in human dermal fibroblast

Objective: To investigate cosmetic potential of Syzygium aromaticum L.(S. aromaticum L.) and to determine its antioxidant and anti-wrinkling effects. Methods: Using highperformance liquid chromatography, eugenol component was quantitated. The antioxidant activity of S. aromaticum L. was analyzed by 2,2-diphenyl-1-picrylhydrazyl radical scavenging and superoxide dismutase like activities. To determine cell viability, elastase and matrix metalloproteinase-1(MMP-1) activity, human dermal fibroblasts(HS68) were treated with S. aromaticum L. The inhibitory effect of S. aromaticum L. on tumor necrosis factor alpha induced MMPs expression in HS68 was analyzed by realtime-PcR. Results: The eugenol content was confirmed in S. aromaticum L. S. aromaticum L. was observed to have high 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity and superoxide dismutase like activity. S. aromaticum L. had no cytotoxicity against the HS68 and dose-dependently increased elastase inhibition. Moreover, S. aromaticum L. significantly decreased MMP-1 content and inhibited gene levels of MMP-1, MMP-2, MMP-3 and MMP-9. conclusions: The findings suggest that S. aromaticum L. has great potential as a cosmeceutical ingredient with antioxidant and antiwrinkling effects.

We propose a new framework for entity and event extraction based on generative adversarial imitation learning-an inverse reinforcement learning method using a generative adversarial network(GAN).We assume that instances and labels yield to various extents of difficulty and the gains and penalties(rewards)are expected to be *** utilize discriminators to estimate proper rewards according to the difference between the labels committed by the ground-truth(expert)and the extractor(agent).Our experiments demonstrate that the proposed framework outperforms state-of-the-art methods.

Inorganic silica-titania thin films with thicknesses 150 nm-200 nm are deposited on high purity and polished silicon wafer and silica glass substrates by sol-gel dipping process and are patterned by capillary force lithography technique. Subsequently grating structure is embossed in green stage. The patterned gel films are subjected to stepwise heat treatment to 500 ℃ and above in pure oxygen atmosphere in order to achieve major conversion of mixed-gel to oxide optical films which are characterized by Ellipsometry, Fourier transform infrared spectroscopy （FTIR） and atomic force microscopy （AFM） to optimize the fabrication parameters and to get perfectly matched film. Removal of organics and formation of perfectly inorganic silica-titania network at optimized heat treatment in controlled environment are ensured by FTIR spectral study. The difference in refractive indices between the substrate and coated film as developed waveguides for operating wavelength show the planar waveguide behavior of the films. calculated theoretically matches exactly with the （632.8 nm） and the measured optical properties

AIM： To examine the pathway related to the IL-1β induced activation of mitogen-activated protein （MAP） kinases in cat esophageal smooth muscle cells. METHODS： culture of the esophageal smooth muscle cells from cat was prepared. Specific inhibitors were treated before applying the IL-β3. Western blot analysis was performed to detect the expressions of cOX, iNOS and MAP kinases. RESULTS： In the primary cultured cells, although IL-β3 failed to upregulate the cOX and iNOS levels, the levels of the phosphorylated forms of 1344142 HAP kinase and p38 MAP kinase increased in both concentration- and time-dependent manner, of which the level of activation reached a maximum within 3 and 18 h, respectively. The pertussis toxin reduced the level of p44/42 MAP kinase phosphorylation. Tyrphostin 51 and genistein also inhibited this activation. Neomycin decreased the density of the p44/42 HAP kinase band to the basal level. Phosphokinase c （PKc） was found to play a mediating role in the IL-1β-induced p44/42 MAP kinase activity. In contrast, the activation of p38 MAP kinase was inhibited only by a pretreatment with forskolin, and was unaffected by the other compounds. cONcLUSION： Based on these results, IL-1β-induced p44/42 MAP kinase activation is mediated by the Gi protein, tyrosine kinase, phospholipase c （PLc） and PKc. The pathway for p38 MAP kinase phosphorylation is different from that of p44/42 MAP kinase, suggesting that it plays a different role in the cellular response to IL- 1β.

Hair loss due to medical conditions,such as alopecia,male pattern baldness,and cancer chemotherapy treatment,has been a comm onproblem for many in *** n A(csA),a fungal metabolite,has been reported to be a hair growth modulatory agent and is apotential drug for hair ***,the effect of topical application of csA is limited by its poor water *** deliverysystems developed to enhance its solubility still showed poor skin *** overcome these limitations,in this study,we have developed a novel chitosan nanocapsule platform using Pluro nic F127 and chitosan without any chemical crosslinking or complicated preparati on steps for the enhanced water solubility and high tran sdermal penetration of *** chitosan nanocapsules(chiNcs)optimized in terms of structural stability by using chitosan with various molecular weights ranging from 3 to 100 kda enhanced the skin permeation of csA through humanc.daver skin in *** administration of the csA loaded chiNcs in creased the hair follicles by c.a.7 times higher than that of thecontrol group,and effectively induced hair growth in c57BL/6 mice in *** results suggest that chiNcs could be used as a platform foreffective transdermal delivery of various hydrophobic drugs.

Sea surface temperature(SST) varies significantly in the presence of tropical cyclones(Tcs). Using fixed SST throughout the integration of high resolution Tc models is general practice in research and operational endeavor over the North Indian Ocean. The present study is to assess the impact of updating realistic SST in Tc lifetime on track, intensity and rainfall of Tcs. The Hurricane Weather Research Forecast(HWRF) model of single domain with 9 km resolution is used. A total of 31 forecast cases are considered from 6 Tcs during 2007-16 with unique track and intensity characteristics. A set of two numerical experiments are done without(cNTL) and with 6-hourly SST update(SST) in Tc *** track and intensity errors show that there is an improvement of 3–41% in track during 12–120 h forecast length for SST run. The SST runs improve landfall position and time prediction by 20% and 33% respectively. The cross track error of SST run is comparable(44 km) with average errors available for this basin(34 km);and along track errors are improved by 60% as compared to cNTL as well as average errors of the basin. The model is biased to overestimate a weaker Tc and underestimate a stronger Tc, however, the bias is reduced in SST run by 5–51%. The analyses of wind, enthalpy flux and warm core structures provide insight for realistic intensity prediction of SST run unlike cNTL. Rainfall intensity and radial distribution is also improved in SST run. Thus, this study highlights the significance of ocean coupling with Tc models to advance forecast guidance.

Pallid Swifts(Apus pallidus),as other swifts,are birds extremely adapted to an aerial lifestyle,showing unique adaptations that allow them to fly almost *** diet of these non-stopping high-altitudinal aerial birds has been mostly studied through techniques that fail to produce highly resolved prey identifications,and for that have been replaced by molecular techniques,such as DNA *** samples of Pallid Swifts were monthly collected from a colony in the north of Portugal during the breeding *** from the faecal samples was used to sex the birds and to identify the arthropods present in the diet through DNA *** the detected prey items,74 families were identified belonging to 16 orders,with Hymenoptera and Hemiptera being the most frequently *** were seasonal variations in diet richness,composition and prey *** the diet of males and females,although no differences were found between the diet of males and females in terms of composition and richness,there were differences in the size of arthropods preyed by the different sexes,with males feeding on larger *** large seasonal variation in Pallid Swifts’diet during the breeding season is probably a result of spatiotemporal variation in aerial prey,of which swifts likely predate *** no significant differences were detected in diet richness and composition between sexes,the fact that males consumed larger prey may suggest the existence of sexual dietary segregation in this group of *** last,several pest species were found in these swifts’diet,which,if studied through DNA metabarcoding,can be used to monitor small arthropods,including airborne pests.

AIM: To clarify the mechanism underlying the anti-mutagenic and anti-cancer activities of Scorpio water extract (SWE). METHODS: Human hepatoma HepG2 cells were incubated with various concentrations of SWE. After 24-h incubation, cytotoxicity and apoptosis evaluations were determined by MTT and DNA fragmentation assay, respectively. After treatment with SWE, mitochondrial membrane potential (MMP) was determined by measuring the retention of the dye 3,3'-dihexyloxacarbocyanine (DiOc6(3)) and the protein expression including cytochrome c and poly-(ADPribose) polymerase (PARP) were measured by Western blotting. caspase-3 and -9 enzyme activities were measured using specific fluorescence dyes such as Ac-DEVD-AFc and Ac-LEHD-AFc. RESULTS: We found that treatment with SWE induced apoptosis as confirmed by discontinuous DNA fragmentation in cultured human hepatoma HepG2 cells. Our investigation also showed that SWE-induced apoptosis of HepG2 cells were associated with intracellular events including disruption of MMP, increased translocation of cytochrome c from mitochondria to cytosol, activation of caspase-3, and PARP. Pre-treatment of N-acetyl-Asp-Glu-Val-Asp-cHO (Ac-DEVD-cHO), a caspase-3 specific inhibitor, or cyclosporin A (csA), an inhibitor of MMP disruption, completely abolished SWE-induced DNA fragmentation. cONcLUSION: These results suggest that SWE possibly causes mitochondrial damage, leading to cytochrome c release into cytosol and activation of caspases resulting in PARP cleavage and execution of apoptotic cell death in HepG2 cells. These results further suggest that Scorpio may be a valuable agent of therapeutic intervention of human hepatomas.
