Endoplasmic reticulum stress and autophagy in cerebral ischemia/reperfusion injury:PERK as a potential target for intervention

Several studies have shown that activation of unfolded protein response and endoplasmic reticulum(ER)stress plays a crucial role in severe cerebral ischemia/reperfusion *** occurs within hours after cerebral ischemia,but the relationship between ER stress and autophagy remains *** this study,we established experimental models using oxygen-glucose deprivation/reoxygenation in PC12 cells and primary neurons to simulate cerebral ischemia/reperfusion *** found that prolongation of oxygen-glucose deprivation activated the ER stress pathway protein kinase-like endoplasmic reticulum kinase(PERK)/eukaryotic translation initiation factor 2 subunit alpha(e IF2α)-activating transcription factor 4(ATF4)-C/EBP homologous protein(CHOP),increased neuronal apoptosis,and induced ***,inhibition of ER stress using inhibitors or by si RNA knockdown of the PERK gene significantly attenuated excessive autophagy and neuronal apoptosis,indicating an interaction between autophagy and ER stress and suggesting PERK as an essential target for regulating *** autophagy with chloroquine exacerbated ER stress-induced apoptosis,indicating that normal levels of autophagy play a protective role in neuronal injury following cerebral ischemia/reperfusion *** from this study indicate that cerebral ischemia/reperfusion injury can trigger neuronal ER stress and promote autophagy,and suggest that PERK is a possible target for inhibiting excessive autophagy in cerebral ischemia/reperfusion injury.

DOMAIN DECOMPOSITION WITH NONMATCHING GRIDS FOR EXTERIOR TRANSMISSION PROBLEMS VIA FEM AND DTN MAPPING

In this paper, we are concerned with a non-overlapping domain decomposition method （DDM） for exterior transmission problems in the plane. Based on the natural boundary integral operator, we combine the DDM with a Dirichlet-to-Neumann （DtN） mapping and provide the numerical analysis with nonmatching grids. The weak continuity of the approximation solutions on the interface is imposed by a dual basis multiplier. We show that this multiplier space can generate optimal error estimate and obtain the corresponding rate of convergence. Finally, several numerical examples confirm the theoretical results.

AZD1775 synergizes with SLC7A11 inhibition to promote ferroptosis

Tumor suppressor p53-mediated cell cycle arrest and DNA damage repair may exert cytoprotective effects against cancer therapies, including WEE1 inhibition. Considering that p53 activation can also lead to multiple types of cell death, the role of this tumor suppressor in WEE1 inhibitor-based therapies remains disputed. In this study, we reported that nucleolar stress-mediated p53 activation enhanced the WEE1 inhibitor AZD1775-induced ferroptosis to suppress lung cancer growth. Our findings showed that AZD1775 promoted ferroptosis by blocking cystine uptake, an action similar to that of Erastin. Meanwhile, inhibition of WEE1 by the WEE1 inhibitors or si RNAs induced compensatory upregulation of SLC7A11, which conferred resistance to ferroptosis. Mechanistically, AZD1775 prevented the enrichment of H3K9me3, a histone marker of transcriptional repression, on the SLC7A11 promoter by repressing the expression of the histone methyltransferase SETDB1, thereby enhancing NRF2-mediated SLC7A11 transcription. This finding was also validated using the H3K9me3 inhibitor BRD4770. Remarkably, we found that the nucleolar stress-inducing agent Actinomycin D(Act. D) inhibited SLC7A11 expression by activating p53, thus augmenting AZD1775-induced ferroptosis. Moreover, the combination of AZD1775 and Act. D synergistically suppressed wild-type p53-harboring lung cancer cell growth both in vitro and in vivo. Altogether, our study demonstrates that AZD1775 promotes ferroptosis by targeting cystine uptake but also mediates the adaptive activation of SLC7A11 through the WEE1-SETDB1 cascade and NRF2-induced transcription, and inhibition of SLC7A11 by Act. D boosts the anti-tumor efficacy of AZD1775 by enhancing ferroptosis in cancers with wild-type p53.

Pressureless two-step sintering of ultrafine-grained refractory metals:Tungsten-rhenium and molybdenum

The challenge of sintering ultrafine-grained refractory metals and alloys to full density is hereby addressed by pressureless two-step sintering in tungsten-rhenium alloy and pure molybdenum. Using properly processed nano powders(~50 nm average particle size), we are able to sinter W-10Re alloy to 98.4% density below 1200 ℃ while maintaining a fine grain size of 260 nm, and sinter molybdenum to 98.3% density below 1120 ℃ while maintaining a fine grain size of 290 nm. Compared to normal sintering,two-step sintering offers record-fine grain sizes and better microstructural uniformity, which translates to better mechanical properties with higher hardness(6.3 GPa for tungsten-rhenium and 4.0 GPa for molybdenum, both being the highest in all pressurelessly sintered samples of the respective material system)and larger Weibull modulus. Together with our previous demonstration in tungsten, we believe that twostep sintering is a general effective method to produce high-quality fine-grained refractory metals and alloys, and the lessons learned here are transferable to other materials for powder metallurgy.

Multi-camera calibration method based on minimizing the difference of reprojection error vectors

In order to achieve a high precision in three-dimensional（3D） multi-camera measurement system, an efficient multi-cameracalibration method is proposed. A stitching method of large scalecalibration targets is deduced, and a fundamental of multi-cameracalibration based on the large scale calibration target is *** avoid the shortcomings of the method, the vector differencesof reprojection error with the presence of the constraint conditionof the constant rigid body transformation is modelled, and mini-mized by the Levenberg-Marquardt （LM） method. Results of thesimulation and observation data calibration experiment show thatthe accuracy of the system calibrated by the proposed methodreaches 2 mm when measuring distance section of 20 000 mmand scale section of 7 000 mm × 7 000 mm. Consequently, theproposed method of multi-camera calibration performs better thanthe fundamental in stability. This technique offers a more uniformerror distribution for measuring large scale space.

The AgCl/Al2O3 catalyst has potential for use in the selective catalytic reduction（SCR） of NOx.A compound hydrocarbon,following oxygenation is used as a type of reducing *** this experiment,the AgCl/Al2O3 catalyst was produced by four different methods,and the differences among their reduction catalysis of NOx were *** was used as a type of reducing agent.X-ray diffraction analysis was performed to study the crystalline structure and scanning electron microscope and transmission electron microscope（TEM） were applied to determine the *** results indicated that,in the range of 350-400℃,there was no significant difference on the NOx reduction rate;however,there was dispersion at high and low temperature *** size of the AgCl particles was about 20-100 nm.

On the heterogeneous microstructure development in the welded joint of 12MnNiVR pressure vessel steel subjected to high heat input electrogas welding

Microstructure features of 12 MnNiVR pressure vessel steel welded joint deposited by the high heat input electrogas welding have been systematically investigated. It is revealed that the welded joint is featured by a heterogeneous juxtaposition. The coarse grained heat-affected zone(CGHAZ) primarily consists of lath bainites and minor granular bainites. The fine grained heat-affected zone(FGHAZ) is dominated by polygonal ferrites, pearlites, and fine cementite particles. Moreover, electron backscatter diffraction results further demonstrate that the CGHAZ is populated by coarse prior austenite grains(PAGs) with high frequency(61.3%) of low angle grain boundaries(LAGBs). On the other hand, the FGHAZ is filled with fine PAGs with a lower frequency(19.6%) of LAGBs. Such microstructural differences may likely contribute to differed mechanical properties for samples tested at designed positions.

State-of-the-art of 3D printing technology of cementitious material—An emerging technique for construction

In recent few years, significant improvement has been made in developing largescale 3 D printer to accommodate the need of industrial-scale 3 D printing. Cementitious materials that are compatible with 3 D printing promote rapid application of this innovative technique in the construction field with advantages of cost effective, high efficiency, design flexibility and environmental friendly. This paper firstly reviews existing 3 D printing techniques that are currently being used in commercial3 D printers. It then summarizes three latest development of largescale 3 D printing systems and identifies their relationships and limiting factors. Thereafter, critical factors that are used to evaluate the workability and printable performance of cementitious materials are specified. Easy-extrusive, easy-flowing, well-buildable, and proper setting time are significant for cementitious material to meet the critical requirements of a freeform construction process. Finally, main advantages, potential applications and the prospects of future research of 3 D printing in construction technology are suggested. The objective of this work is to review current design methodologies and operational constraints of largescale 3 D printing system and provide references for optimizing the performance of cementitious material and promote its responsible use with largescale 3 D printing technology.

历次修法中涉及逮捕的制度机制的演进轨迹和条文变化的内在逻辑表明,为实现“慎捕”目标,《中华人民共和国刑事诉讼法》(以下简称《刑事诉讼法》)已初步构筑起一套结构严谨、衔接有序、层次分明的关于逮捕权的法规范控制体系。该体系在事前的权力配置阶段“两次分离”逮捕权,以具体社会危险性评估为重心改造逮捕要件;在事中的权力行使阶段以“兼听则明”为原则设计了充分听取控辩双方意见的审查逮捕机制;在事后的权力制约阶段要求检察院、法院及时纠正撤销错捕决定,释放犯罪嫌疑人。从司法办案的角度来看,现有的法规范控制体系在规则制定和程序执行时仍有不足和缺漏,如逮捕权分离不彻底,控辩意见“兼听”机制保障不充分,径行逮捕条件设计机械、不周延,当事人对错捕的救济途径缺失等。本轮《刑事诉讼法》修改应坚持“实践—规范”的互动逻辑,关注司法实践,将解决和弥补逮捕中的权力运行失范和权利保障不足等现实问题作为修法方向,破难题、开新局,不断推动中国刑事诉讼法治现代化进程。

Computer simulation for materials processing needs a huge database containing a great deal of various physical properties of materials. In order to employ the accumulated large data on materials heat treatment in the past years, it is significant to develop an intelligent database system. Based on the data mining technology for data analysis, an intelligent database web tool system of computer simulation for heat treatment process named as IndBASEweb-HT was built up. The architecture and the arithmetic of this system as well as its application were introduced.
