So far approximately 144,000 species of fungi have been named but sequences of the majority of them do not exist in the public databases.Therefore,the quality and coverage of public barcode databases is a bottleneck that hinders the study of fungi.Cortinarius is the largest genus of Agaricales with thousands of species world-wide.The most diverse subgenus in Cortinarius is Telamonia and its species have been considered one of the most taxonomically challenging in the Agaricales.Its high diversity combined with convergent,similar appearing taxa have earned it a reputation of being an impossible group to study.In this study a total of 746 specimens,including 482 type specimens representing 184 species were sequenced.Also,a significant number of old types were successfully sequenced,105 type specimens were over 50 years old and 18 type specimens over 100 years old.Altogether,20 epi-or neotypes are proposed for recently commonly used older names.Our study doubles the number of reliable DNA-barcodes of species of C.subgenus Telamonia in the public sequence data-bases.This is also the first extensive phylogenetic study of the subgenus.A majority of the sections and species are shown in a phylogenetic context for the first time.Our study shows that nomenclatural problems,even in difficult groups like C.subgenus Telamonia,can be solved and consequently identification of species based on ITS barcodes becomes an easy task even for non-experts of the genus.

fungi play vital roles in ecosystems as endophytes,pathogens and saprobes.The current estimate of fungal diversity is highly uncertain,ranging from 1.5 to 12 million,but only around 150,000 species have been named and classified to date.Since the introduction of DNA based methods for species identification,the number of newly described taxa has increased from approximately 1000 to around 2000 yearly.This demonstrates the importance of DNA based methods to identify and distin-guish species,especially cryptic species.Many novel species from recent studies have been found in historically understudied regions and habitats,but these still represent only a small percentage of the estimated species.In this paper,we examine 16 genera from the top 40 most speciose genera as listed in Species fungorum as case studies to examine the diversity of taxa in each genus.The genera treated herein are Cercospora,Diaporthe,Meliola,Passalora,Phyllachora,Phyllosticta,Pseudocer-cospora,Ramularia(ascomycetes)and Cortinarius,Entoloma,Inocybe,Marasmius,Psathyrella,Puccinia,Russula,Uromyces(basidiomycetes).We critically evaluate the number of species in these genera and correlate these numbers with the number of entries in GenBank.We introduce 18 new species Apiospora multiloculata,Candolleomyces thailandensis,Cortinarius acutoproximus,Cortinarius melleoalbus,Cortinarius pacificus,Cortinarius parvoacetosus,Diaporthe guizhouensis,Entoloma pseudosubcorvinum,Inocybe meirensongia,Marasmius albulus,Marasmius obscuroaurantiacus,Meliola camporesii,Phyl-lachora siamensis,Phyllosticta doitungensis,Picipes yuxiensis,Pseudocercospora vignae,Puccinia maureanui and Russula inornata.We also introduce a new record of Candolleomyces cladii-marisci and Inocybe iringolkavensis.We discuss the genera Colletotrichum and Pleurotus that are speciose,but do not occur in the top 40.We hypothesize whether there might be more species in these genera and discuss why these genera have some of the largest number of species.

We report SiO_(2)-supported monometallic Pt,Pd,Au,Ni,Cu and Co catalysts for proton-driven NAD+regeneration,co-producing H_(2).All metals are fully selective to NAD+where the order of turnover frequencies(Pt>Pd>Cu>Au,Ni and Co)coincides with those otherwise observed in electrochemical hydrogen evolution reactions.This has revealed that NADH is capable of converting the metal sites into a“cathode”without an external potential and the NADH to NAD+reaction involves transferring electron and hydrogen atom separately.Electron-deficient Ptδ+(on CeO_(2))enhances TOf and the heterogeneous Pt/CeO_(2) catalyst is recyclable without losing any activity/selectivity.

Recovery rates of persistent post-COVID-19 olfactory dysfunction using psychophysical assessment:A longitudinal cohort study

Objectives:Persistent olfactory dysfunction(OD)following loss of smell associated with SARS-CoV-2 infection is a major feature of long COVID.Perspectives on the prevalence of persistent OD predominantly rely on self-reported olfactory function.few studies have tracked longitudinal rates of recovery using psychophysical assessment among patients presenting for evaluation of persistent OD beyond a window of acute recovery.Data anchored in standardized testing methods are needed to counsel patients who fail to acutely regain their sense of smell.This study aims to quantify the degree of persistent OD in post-COVID-19 patients who experience subjective and psychophysical OD.Methods:We grouped participants presenting for OD evaluation into cohorts based on both subjective and psychophysical olfactory status at a baseline assessment and assessed their olfactory abilities with a visual analogue scale and the Sniffin'Sticks extended test at baseline and 1-year time points.Participants had confirmed a history of COVID-19 by lab evaluation or clinical diagnosis if lab evaluation was not available.Results:Baseline olfactory evaluation was completed by 122 participants,53 of whom completed the 1-year follow-up assessment.Among participants presenting with perceived OD,74.5%had confirmed psychophysical OD at baseline,with 55.1%at 1-year follow-up.Participants had reliable trends in self-rated versus psychophysically tested olfactory function at both time points.The total threshold,discrimination,and identification(TDI)score improved by+3.25 points in the cohort with psychophysical OD(p=0.0005),with this improvement largely attributable to an increase in median threshold scores(+2.75 points;p=0.0004).Conclusions:OD persists in a significant number of patients who fail to acutely recovery their sense of smell after COVID-19,with many demonstrating lingering deficits at 1-year.Improvements in threshold,but not discrimination or identification,most significantly mediate improvement of total TDI score at follow-up.

This is a continuity of a series of taxonomic papers where materials are examined,described and novel combinations are proposed where necessary to improve our traditional species concepts and provide updates on their classification.In addition to extensive morphological descriptions and appropriate asexual and sexual connections,DNA sequence data are also analysed from concatenated datasets(rDNA,TEf-a,RBP2 and b-Tubulin)to infer phylogenetic relationships and substantiate systematic position of taxa within appropriate ranks.Wherever new species or combinations are being proposed,we apply an integrative approach(morphological and molecular data as well as ecological features wherever applicable).Notes on 125 fungal taxa are compiled in this paper,including eight new genera,101 new species,two new combinations,one neotype,four reference specimens,new host or distribution records for eight species and one alternative morphs.The new genera introduced in this paper are Alloarthopyrenia,Arundellina,Camarosporioides,Neomassaria,Neomassarina,Neotruncatella,Paracapsulospora and Pseudophaeosphaeria.The new species are Alfaria spartii,Alloarthopyrenia italica,Anthostomella ravenna,An.thailandica,Arthrinium paraphaeospermum,Arundellina typhae,Aspergillus koreanus,Asterina cynometrae,Bertiella ellipsoidea,Blastophorum aquaticum,Cainia globosa,Camarosporioides phragmitis,Ceramothyrium menglunense,Chaetosphaeronema achilleae,Chlamydotubeufia helicospora,Ciliochorella phanericola,Clavulinopsis aurantiaca,Colletotrichum insertae,Comoclathris italica,Coronophora myricoides,Cortinarius fulvescentoideus,Co.nymphatus,Co.pseudobulliardioides,Co.tenuifulvescens,Cunninghamella gigacellularis,Cyathus pyristriatus,Cytospora cotini,Dematiopleospora alliariae,De.cirsii,Diaporthe aseana,Di.garethjonesii,Distoseptispora multiseptata,Dis.tectonae,Dis.tectonigena,Dothiora buxi,Emericellopsis persica,Gloniopsis calami,Helicoma guttulatum,Helvella floriforma,H.oblongispora,Hermatomyces subiculosa,Juncaceicola italica,Lactarius dirkii,Lentithecium

Cardiac differentiation is modulated by anti-apoptotic signals in murine embryonic stem cells

BACKGROUND Embryonic stem cells(ESCs)serve as a crucial ex vivo model,representing epiblast cells derived from the inner cell mass of blastocyst-stage embryos.ESCs exhibit a unique combination of self-renewal potency,unlimited proliferation,and pluripotency.The latter is evident by the ability of the isolated cells to differ-entiate spontaneously into multiple cell lineages,representing the three primary embryonic germ layers.Multiple regulatory networks guide ESCs,directing their self-renewal and lineage-specific differentiation.Apoptosis,or programmed cell death,emerges as a key event involved in sculpting and forming various organs and structures ensuring proper embryonic development.How-ever,the molecular mechanisms underlying the dynamic interplay between diffe-rentiation and apoptosis remain poorly understood.AIM To investigate the regulatory impact of apoptosis on the early differentiation of ESCs into cardiac cells,using mouse ESC(mESC)models-mESC-B-cell lym-phoma 2(BCL-2),mESC-PIM-2,and mESC-metallothionein-1(MET-1)-which overexpress the anti-apoptotic genes Bcl-2,Pim-2,and Met-1,respectively.METHODS mESC-T2(wild-type),mESC-BCL-2,mESC-PIM-2,and mESC-MET-1 have been used to assess the effect of potentiated apoptotic signals on cardiac differentiation.The hanging drop method was adopted to generate embryoid bodies(EBs)and induce terminal differentiation of mESCs.The size of the generated EBs was measured in each condition compared to the wild type.At the functional level,the percentage of cardiac differentiation was measured by calculating the number of beating cardiomyocytes in the manipulated mESCs compared to the control.At the molecular level,quantitative reverse transcription-polymerase chain reaction was used to assess the mRNA expression of three cardiac markers:Troponin T,GATA4,and NKX2.5.Additionally,troponin T protein expression was evaluated through immunofluorescence and western blot assays.RESULTS Our findings showed that the upregulation of Bcl-2,Pim-2,and Met-1 genes led to a reduction in th

Radiation Dose from CT-Scan of Childhood’s Head: Results of the first Ivorian Survey in a Single Study Site

Objectives: This study aims to evaluate the level of X-ray doses used in childhood’s head as Local Diagnostic Reference Levels (LDRLs) in computed tomography (CT) at a university hospital in Côte d’Ivoire. The Diagnostic Reference Level (DRL) have been set up and used to prevent unusually high radiation doses used in radiology departments and is therefore an optimization tool for practices and procedures in medical X-ray imaging for the radiation protection of patients. Methods: A prospective study of volume CT dose index (CTDIvol) and dose length product (DLP) was performed on images of childhood’s head obtained from a CT-scanner of 64 bars equipped with the tube current modulation capability and manufactured by Hitachi Medical System. 122 CT-scan data from 55 childhood’s head were analyzed. The scan data were stratified in four age groups: Results: The 75th percentile of CTDIvol and DLP (set as LDRL) obtained with respect to the stratified age groups are: 22.5 mGy and 452.5 mGy∙cm, 27.7 mGy and 690.6 mGy∙cm, 28 mGy and 722.4 mGy∙cm, 33.6 mGy and 736.8 mGy∙cm respectively. These outcome values increase with respect to the age of pediatric patients and are comparable to DRLs values obtained internationally. Conclusions: Obtaining good image quality while using low dose in children’s head computed tomography for radiation protection require to setup more surveys in Côte d’Ivoire for regional and national DRL. We proposed through this survey LDRLs in terms of CTDIvol and DLP, comparable to international DRLs values. This survey will be strengthened by additional surveys in order to obtain national DRLs for the radiation protection of the child patient in Côte d’Ivoire.

Background There is increasing epidemiological evidence supporting the association between onchocerciasis and seizures,reinforcing the concept of onchocerciasis-associated epilepsy(OAE).The aim of this paper is to provide an update on the new knowledge about OAE and to propose recommendations to the World Health Organization how to address this public health problem.Main text During the 2nd International Workshop on OAE held on 19-21 September,2023,in Antwerp,Belgium,participants recognised OAE as a substantial yet neglected public health problem,particularly in areas of sub-Saharan Africa where onchocerciasis remains hyperendemic.Evidence from prospective population-based studies suggest that strengthening onchocerciasis elimination efforts leads to a significant reduction of OAE incidence.There is a need to validate an OAE case definition to estimate the burden of disease and identify onchocerciasis-endemic areas requiring intensification of onchocerciasis elimination programmes and integration of epilepsy care t is expected that raising awareness about OAE will boost the population uptake of ivermectin.The implementation of a commu-nity-based epilepsy treatment programme offering free anti-seizure medications(ASMs)has shown high effectiveness in reducing the frequency of seizures and improving the overall quality of life of people with epilepsy.Conclusions To reduce OAE burden,enhanced collaboration between onchocerciasis and mental health pro-grammes at community,national,and international levels is required.Urgent efforts are needed to ensure the uninter-rupted provision of free ASMs in onchocerciasis-endemic areas.furthermore,OAE should be included in the quantif-cation of the onchocerciasis disease burden.
