Genetic Enhancement of Indigenous Cowpea with Gamma-Ray Induced Trait Variation

Production of cowpea (Vigna unguiculata (L.) Walp.), a staple legume crop in Sub-Saharan Africa, faces challenges due to biotic and abiotic constraints. Induced mutagenesis was deployed to create genetic variation in two cowpea varieties (KVX396-4-5-2D and Moussa local). The radio-sensitivity tests led to determe the lethal dose 50 (LD50) corresponding to 230 Gy and 220 Gy for KVX396-4-5-2D and Moussa local varieties, respectively. Dried seeds (M0) of each variety were gamma-ray irradiated with LD50 − 50, LD50 and LD50 + 50. M1 seeds were advanced to generate M2, M3 and M4 mutants using the single-seed-descent method. M4 mutant lines were evaluated in rain-fed conditions using a randomized complete block design to assess phenotypic differences. Data on seven qualitative and eleven quantitative traits were collected. The results indicated that the mutation induced variability in three qualitative traits: in KVX 396-4-5-2D mutant lines, with flower and seed color frequencies at 2.61% and 0.56% respectively, and pod dehiscence at a frequency of 0.24%. While in Moussa local mutants, a pod color changed at a frequency of 17%. ANOVA results revealed significant differences between mutants of both varieties for all quantitative traits, including photosynthetic parameters. Positive correlations were observed between leaf diameter and 100-seed weight, and between branch number and 100-seed weight. Hierarchical clustering revealed three clusters among KVX 396-4-5-2D mutants and six clusters among Moussa local mutants. Early maturity and high foliage were induced traits in Cluster 3 of KVX 396-4-5-2D mutants while high hundred-seed weight was induced in Cluster 6 of Moussa local mutants.

Improvement of Mechanical Qualities of Clay Material through Coconut Fiber Stabilization

The criticisms regularly formulated towards clay or soil, in general, are its weak mechanical qualities and low water quality. Therefore, it is necessary to find techniques to improve the properties of this material, which is widely used worldwide. Here, we propose stabilizing clay with coconut fiber as a solution to enhance its mechanical properties. To do this, we used an experimental method, first determining the geotechnical properties of the clay and then its mechanical properties. The geotechnical study using the Proctor Test revealed that the dry density of the clay is γb = 1.42 g/cm3, and its water content is W = 22.3%. by applying the rolling method, the Atterberg limits were determined: liquid limit Wl = 63.6, plastic limit Wp = 27.9, plasticity index Ip = 35.7, and consistency index Ic = 1.46. With 25 P = 35.7 1.3, according to the water classification, it falls into class A3ts. The mechanical part focused on compression and flexural strengths obtained using a PROETI hydraulic press. We obtained a flexural strength of 0.63 MPa for simple clay (bA);0.89 MPa for clay + 0.25% fiber (bAF1/4);1.68 MPa for clay + 0.5% fiber (bAF1/2);1.87 MPa for clay + 0.75% fiber (bAF3/4);and 3.91 MPa for clay + 1% fiber (bAF1). As for the compression strength, bA = 5.90 MPa, bAF1/4 = 6.395 MPa, bAF1/2 = 6.292 MPa, bAF3/4 = 6.065 MPa, and bAF1 = 5.423 MPa. The addition of fiber has thus improved the mechanical qualities of the simple clay. These stabilized bricks can be used for sustainable and bioclimatic construction, providing higher durability and good comfort.

The addition of surface acoustic wave(SAW)technologies to microfluidics has greatly advanced lab-on-a-chip applications due to their unique and powerful attributes,including high-precision manipulation,versatility,integrability,biocompatibility,contactless nature,and rapid ***,the development of SAW microfluidic devices is limited by complex and time-consuming micro/nanofabrication techniques and access to cleanroom facilities for multistep photolithography and vacuum-based *** simplify the fabrication of SAW microfluidic devices with customizable dimensions and functions,we utilized the additive manufacturing technique of aerosol jet *** successfully fabricated customized SAW microfluidic devices of varying materials,including silver nanowires,graphene,and poly(3,4-ethylenedioxythiophene)polystyrene sulfonate(PEDOT:PSS).To characterize and compare the acoustic actuation performance of these aerosol jet printed SAW microfluidic devices with their cleanroom-fabricated counterparts,the wave displacements and resonant frequencies of the different fabricated devices were directly measured through scanning laser Doppler ***,to exhibit the capability of the aerosol jet printed devices for lab-on-a-chip applications,we successfully conducted acoustic streaming and particle concentration ***,we demonstrated a novel solution-based,direct-write,single-step,cleanroom-free additive manufacturing technique to rapidly develop SAW microfluidic devices that shows viability for applications in the fields of biology,chemistry,engineering,and medicine.

Physiological Mechanism of Salicylic Acid for Alleviation of Salt Stress in Rice

Soil salinity is one of the most important problems of crop production in estuarine and coastal zones. Improvement in salt tolerance of major food crops is an important way for the economic utilization of coastal zones. This study proved that the application of salicylic acid(SA) improved the growth and yield under salt stress conditions and investigated its physiological mechanisms for salt tolerance. The investigation on the effect of SA for salt tolerance during germination showed that the decreased rates of germination and growth(in terms of shoot and root lengths) by the salt stress were significantly increased by the SA application(SA + NaCl). The treatment of SA to the high and low saline soils enhanced the growth, yield and nutrient values of rice. The effects of SA on Na^+, K^+ and Cl~– ionic accumulation were traced under salt stress condition by inductively coupled plasma optical emission spectrometry and ion chromatography. It was revealed that the increased accumulation of Na^+ and Clˉ ions by the salt stress were reduced by SA application. An increased concentration of endogenous SA level was detected from the SA-treated rice varieties(ASD16 and bR26) by liquid chromatography electrospray Ionization-tandem mass spectrometry. The activities of antioxidant enzymes such as superoxide dismutase, catalase and peroxidase were increased by salt stress whereas decreased by the SA application. The study proved that the application of SA could alleviate the adverse effects of salt stress by the regulation of physiological mechanism in rice plants. In spite of salt stress, it can be applied to the coastal and estuarine regions to increase the rice production.

Chronic Hepatitis b in Indian Americans: Lack of Screening and Poor Linkage to Care

background: Chronic hepatitis b (CHb) is a major cause of liver-related morbidity and mortality in the United States (US) and globally. CHb disproportionately affects Asian Americans and many other immigrant minority populations, primarily owing to the high prevalence of CHb in their countries of origin. India is a country with a medium-to-high prevalence of hepatitis b (Hb) (>2%) and has over 40 million people infected with hepatitis b virus (HbV), with more than 115,000 deaths annually from HbV-related complications. Indian Americans are one of the largest immigrant populations in the US but remain underdiagnosed and poorly linked to clinical care. We, therefore, assessed the HbV prevalence and evaluated the linkage-to-care (LTC) among Indian Americans to develop strategic plans to reduce the impact of HbV in the US. Methods: between April 2022 and January 2024, serologic screening and surveys were provided to 328 Indian American adults (age 20 - 80) in New York City. All participants were tested for a triple panel consisting of hepatitis b surface antigen (HbsAg), hepatitis b surface antibody (anti-Hbs), and hepatitis b core IgG antibody (anti-Hbc). A survey was conducted on the subjects chronically infected with HbV regarding their histories of infection. Self-administered questionnaires were employed to evaluate demographic and epidemiologic characteristics. Results: Of 328 screened and evaluated (246 males and 82 females), 10 (3.0%) were HbV-infected, 222 (67.7%) were susceptible to HbV, and 96 (29.3%) were immune. The prevalence of chronic HbV varied between the age groups: 4.6% (age 20 - 40), 3.4% (age 41 - 60), and 1.7% (age 61 - 80). Of 10 chronically infected, only two subjects had been previously diagnosed but were not engaged in care. Conclusion: HbV disproportionately affects Asian Americans, primarily owing to immigration from parts of the world where the disease is endemic. Starting from sissotrin (1), a natural isoflavonoid isolated from Trifolium baccarinii (Fabaceae), one new semisynthetic derivative, 6-nitrobiochanin A (1b) and two known derivatives, 8-nitrobiochanin A (1a) and 2",3",4",6"-tetraacetylsissotrin (1c) have been obtained after performing nitration and acetylation reactions. Their structures were assigned after interpretation of their spectrometric (HR-ESI-MS) and spectroscopic (NMR 1D and 2D) data and by comparison with those reported in the literature. The substrate as well as the semisynthetic derivatives were evaluated for their antibacterial activities against six strains. The results reveal that they are inactive or weakly active on the strains tested with the exception of 8-nitrobiochanin A (1a) which showed moderate activity (MIC = 62.5 μg∙mL−1) on Staphylococcus aureus ATCC 43300.

Genomic insights into hybridization in a localized region of sympatry between pewee sister species(Contopus sordidulus × C. virens) and their chromosomal patterns of differentiation

background: The Great Plains of the United States includes a large number of hybrid and contact zones between bird species. The amount of gene flow between sister species in these zones ranges from very rare hybridization events to widespread and prevalent introgression. Some of these avian systems have been studied extensively, while others have been indeterminate of whether hybridization exists in areas of sympatry. Using genomic-level approaches allows investigation of genomic patterns of hybridization and gene flow between species—or lack ***: We investigated a narrow zone of sympatry in Nebraska, USA between pewee species(Contopus sordidulus and C. virens), for which no hybridization has been confirmed. We used thousands of single nucleotide polymorphisms to identify potential hybridization and investigate genomic patterns of differentiation between these two ***: We found evidence of multiple hybrid individuals in the contact zone. Little genomic variation was fixed between species, but a large proportion had differentiated allele frequencies between species. There was a positive relationship between genetic differentiation and chromosome ***: We provided the first conclusive evidence of hybridization between C. sordidulus and C. virens, in a region where secondary contact likely occurred due to human disturbance and habitat modification. The genomic patterns of differentiation affirm that these species split in the relatively recent past. Finally, the relationship of chromosome size and genetic differentiation may have resulted from differential rates of chromosomal recombination in songbirds and genetic differentiation between species largely due to genetic drift(possibly in concert with selection).
