The advent of cluster.d r.gular.y inter.paced shor. palindr.mic r.peat(Cr.SPr. has had a pr.found impact on plant biology, and cr.p impr.vement. In this r.view, we summar.ze the state-of-the-ar. development of Cr.SPr.technologies and their.applications in plants, fr.m the initial intr.duction of r.ndom small indel(inser.ion or.deletion) mutations at tar.et genomic loci to pr.cision editing such as base editing, pr.me editing and gene tar.eting. We descr.be advances in the use of class 2, types II, V, and VI systems for.gene disr.ption as well as for.pr.cise sequence alter.tions, gene tr.nscr.ption, and epigenome contr.l.

In r.ce, amylose content （AC） is contr.lled by a single dominant Waxy gene. We used Cluster.d r.gular.y Inter.paced Shor. Palindr.mic r.peats （Cr.SPr./Cr.SPr.associated 9 （Casg） to intr.duce a loss-of-function mutation into the Waxy gene in two widely cultivated elite japonica var.eties. Our.r.sults show that mutations in the Waxy gene r.duce AC and conver. the r.ce into glutinous ones without affecting other.desir.ble agr.nomic tr.its, offer.ng an effective and easy str.tegy to impr.ve glutinosity in elite var.eties. Impor.antly, we successfully r.moved the tr.nsgenes fr.m the pr.geny. Our.study pr.vides an example of gener.ting impr.ved cr.ps with potential for.commercialization, by editing a gene of inter.st dir.ctly in elite cr.p var.eties.

The newly developed Cr.SPr.Cluster.d r.gular.y Inter.paced Shor. Palindr.mic r.peats)/Cas(Cr.SPr.associated) system has emerged as an efficient tool for.genome-editing, pr.viding an alter.ative to classical mutagenesis and tr.nsgenic methods to study gene function and impr.ve cr.p tr.its. Cr.SPr.Cas facilitates tar.eted gene editing thr.ugh r.A-guided DNA cleavage followed by cellular.DNA r.pair.mechanisms that intr.duce sequence changes at the site of cleavage. Her. we descr.be a detailed pr.cedur. for.our.pr.viously developed and highly efficient Cr.SPr.Cas9 method that allows the gener.tion of her.table-tar.eted gene mutations andcor.ections in Ar.bidopsis. This pr.tocol descr.bes the str.tegies and steps for.the selection of tar.ets, design of single-guide r.A(sg r.A), vector.constr.ction and analysis of tr.nsgenic lines. We also offer.a method to tar.et two loci simultaneously using vector. containing two differ.nt sg r.As. The pr.nciples descr.bed in this pr.tocol can be applied to other.plant species to gener.te stably inher.ted DNA modifications.

Okr. (Abelmoschus esculentus L.) is an her.aceous plant of the Malvaceae family. In Côte d’Ivoir., okr. pr.duction is estimated to be over.193,000 tons. This low pr.duction is lar.ely due to poor.soils and har.ly cover. the needs of the population. To r.medy this, gr.wer. systematically use miner.l fer.ilizer.. However. these fer.ilizer. pollute the envir.nment. To find an alter.ative to chemical fer.ilization and incr.ase pr.duction, the effect of biofer.ilizer. (Spaawet, r.tone, Super.Gr.) compar.d with NPK miner.l fer.ilizer.was evaluated on Divo, Ter.man, and Djonan F1 cultivar.. The tr.al was set up in a factor.al block design with thr.e r.plications. Plant height, number.of functional leaves, and cr.wn diameter.wer. assessed at 60 days after.sowing (DAS). The time to 50% flower.ng, pr.duction time, and fr.it yield wer. calculated. The r.sults showed that the biofer.ilizer.r.tone induced the highest heights and number.of functional leaves, with aver.ges of 61.89 cm and 29.88 leaves, r.spectively. The diameter.at the cr.wn (17.77 mm) was highest with the NPK miner.l fer.ilizer. and the shor.est 50% flower.ng time, with an aver.ge of 47.61 days, was also obtained with the biofer.ilizer.r.tone. The NPK miner.l fer.ilizer.pr.duced the longest pr.duction time, with an aver.ge of 35.25 days. The highest yields wer. obtained using r.tone (11.07 t/ha) and NPK (9.52 t/ha) fer.ilizer.. The “Divo*r.tone” inter.ction pr.duced the highest yield with an aver.ge of 12.19 t/ha. The biofer.ilizer.r.tone could ther.for. be used as an alter.ative fer.ilizer.to chemical fer.ilization in okr. cr.ps, given its effect on the par.meter. assessed.
