Anticorrosion behavior of conversion coatings obtained from unbuffered cerium salts solutions on AA6061-T6

The anticorrosive properties of cerium based conversion coatings deposited on AA6061-T6 alloy by immersion in tmbuffercd cerium chloride and cerium nitrate solutions in the presence of hydrogen peroxide were investigated and characterized by potentiodynamic po- larization （PDP） and electrochemical impedance spectroscopy （EIS） in 0.5 mol/L NaCl aqueous solution. The microstructure and chemical composition of the protective films were examined by scanning electron microscopy （SEM） and energy dispersive X-ray spectroscopy （EDS）. It was found that the best corrosion protection was afforded by the samples treated during 600 s in cerium chloride solution at pH values ~5.5-4, showing higher amounts of cerium and polarization resistance values greater than 10 f~ m2. Moreover, an ennoblement of the corro- sion potential and decreasing of the cathodic and anodic currents were obtained compared with the cerium nitrate solutions application. This behavior was attributed to the influence of the deposition parameters such as type of the salt anion, i.e., chelating effect and chaotropic characteristics, pH fluctuations in the conversion solution and deposition time.

In vitro antileishmanial effects of Physalis angulata root extract on Leishmania infantum

Objective： In the present study, we evaluated the effects of the aqueous extract of Physalis angulata root （AEPa） on Leishmania infanturn proliferation, morphology, and the driving mechanism in leishmanicidal activity and modulatory action on macrophages. Methods： I., infantum promastigotes were treated with 50 and 100 μg/mL AEPa for 72 h and then antipro- mastigote assay was performed by counts in a Newbauer chamber, morphological changes were analyzed by transmission electron microscopy and the mechanism of the leishmanicidal activity was detected. In addition, macrophages were infected with L. infantum and were used to evaluate anti-amastigote activity of AEPa and effects of AEPa on cytokine secretion after 72-hour treatment. Results： Treatment with AEPa reduced the numbers ofL infantum promastigotes （50% inhibitory concen- tration （ICso） = 65.9μg/mL; selectivity index （SI）= 22.1） and amastigotes （ICso = 37.9 μg/mL; SI = 38.5） compared with the untreated control. Amphotericin b reduced 100% of the promastigote numbers after 72 h of treatment （IC50 = 0.2μg/mL）. AEPa induced several morphological changes and increased the production of reactive oxygen species and apoptotic death in promastigotes after treating for 72 h. AEPa （100 μg/mL） promoted tumor necrosis factor-α secretion in macrophages infected with L. infantum after 72 h of treatment, but did not induce an increase in this cytokine in noninfected macrophages. In addition, AEPa showed no cytotoxic effect on J774-A1 cells （50% cytotoxic concentration 〉1000μg/mL）. Conclusion： AEPa presented antileishmanial activity against the promastigotes and amastigotes of I.. infantum without macrophage cytotoxicity; these results show that natural products such as P. angulata have leishmanicidal potential and in the future may be an alternative treatment for leishmaniasis.

Araucaria angustifolia Aboveground Roots Presented High Arbuscular Mycorrhizal Fungal Colonization and Diversity in the brazilian Atlantic Forest

Almost 30 different arbuscular mycorrhizal fungi(AMF) species, distributed in different genera such as Glomus, Acaulospora,Scutellospora, Entrophospora, Ambispora, Kuklospora, Gigaspora, and Archeospora, have been identified in the root zone of Araucaria angustifolia, known as brazil Pine. During our AMF survey in this ecosystem, our attention was called to the presence of many superficially growing Araucaria roots. Our hypothesis was that these roots were colonized with AMF because of the presence of AMF spores in organic material aboveground. Samples of these superficial roots and the organic substrate they were growing on were evaluated for their mycorrhizal status. DNA was extracted from the AMF-colonized superficial roots and submitted to polymerase chain reaction(PCR) amplification using the NS31-AM1 primer pair, followed by cloning and sequencing. We found that the root colonization percentages were between 31% and 52%, and the number of AMF spores in the substrate ranged from 27 to 164 spores per 50 g dry substrate. The phylogenetic analyses and tree construction using maximum parsimony(MP) and neighbor-joining(NJ)methods identified 13 different species of the phylum Glomeromycota belonging to the genera Glomus, Funneliformis, Rhizophagus,Gigaspora, Acaulospora, and Archaeospora, and five isolates were identified only at the genus level. To our knowledge, this is the first report on Araucaria angustifolia with roots growing aboveground, producing runner roots that develop on dead tree trunks and organic material. The higher colonization of the aboveground roots than those commonly found in belowground Araucaria roots suggests that they may present active metabolic uptake of nutrients.

Abscisic Acid, High-Light, and Oxidative Stress Down-Regulate a Photosynthetic Gene via a Promoter Motif Not Involved in Phytochrome- Mediated Transcriptional Regulation

In etiolated seedlings, light perceived by phytochrome promotes the expression of light-harvesting chlorophyll a/b protein of photosystem Ⅱ （Lhcb） genes. However, excess of photosynthetically active radiation can reduce Lhcb expression. Here, we investigate the convergence and divergence of phytochrome, high-light stress and abscisic acid （AbA） signaling, which could connect these processes. Etiolated Arabidopsis thaliana seedlings bearing an Lhcb promoter fused to a reporter were exposed to continuous far-red light to activate phytochrome and not photosynthesis, and treated with AbA. We identified a cis-acting region of the promoter required for down-regulation by AbA. This region contains a CCAC sequence recently found to be necessary for AbI4-binding to an Lhcb promoter. However, we did not find a G-box-binding core motif often associated with the AbI4-binding site in genes promoted by light and repressed by AbI4. Mutations involving this motif also impaired the responses to reduced water potential, the response to high photosynthetic light and the response to methyl viologen but not the response to low temperature or to Norflurazon. We propose a model based on current and previous findings, in which hydrogen peroxide produced in the chloroplasts under high light conditions inter- acts with the AbA signaling network to regulate Lhcb expression. Since the mutation that affects high-light and methyl viologen responses does not affect phytochrome-mediated responses, the regulation by retrograde and phytochrome signaling can finally be separated at the target promoter level.

AIM: To study the effect of the inhibitor of cytochrome P450 known as Lilly 18947 (2,4 dichloro-6 phenyl-phenoxy ethyl diethylamine) on Trypanosoma cruzi. METHODS: Trypanosoma cruzi epimastigotes wen grown in culture, in absence or in presence of drug. The inhibition of its growth was followed by daily counting using a Neubauer chamber. The effect of Lilly 18947 on the parasite ultrastructure was examined by electron microscopy. To test the effect of different concentrations of drug on the parasite cycle, Vero cells were inoculated with trypomastigotes ( RA strain ) and after 72 h the percentage of infected cells and the number of intracellular parasites were estimated and expressed as the endocytic index. RESULTS: Growth of epimastigotes was inhibited by Lilly 18947. Concentrations as low as 50 μmol/L resulted in a complete disappearance of the parasites in culture by the fourth day.
